Abstract
Background
Basic sanitation could be a potential indicator of the spread of coronavirus disease-2019 (COVID-19) and, in this context, space-time patterns are important tools with which to ...elucidate the spread of disease and identify risk factors. The aim of this study was to assess a possible association between basic sanitation indices and COVID-19 rates in all the 5570 municipalities of Brazil and its spatial distribution.
Methods
Data of COVID-19 cases registered in Brazil from 28 February until 31 May 2020 and independent variables associated with basic sanitation were included.
Results
High incidence rates were significantly associated with precarious water service index (0–25% coverage) and offstandard faecal coliforms index for tap water (5–50% and 75–100% of samples tested). A significant association between high mortality rates and sewage collection (0–25% coverage)/treatment (25–50% coverage) indices was also verified. In addition, clusters with significant spatial autocorrelation were identified mainly in the North and Northeast regions for mortality and incidence rates (high-high risk areas) and for offstandard faecal coliforms index. Those regions are considered the poorest in Brazil, presenting with low incomes, human agglomerations, as well as a poor basic sanitation system, which also hinder the implementation of COVID-19-preventative measures.
Conclusions
A precarious basic sanitation infrastructure could potentially be associated with the high transmission of severe acute respiratory syndrome coronavirus-2 in Brazil.
CD163, receptor for the haptoglobin-hemoglobin complex, is expressed on monocytes/macrophages and neutrophils. A soluble form of CD163 (sCD163) has been associated with the M2 macrophage phenotype, ...and M2 macrophages have been shown to down-modulate inflammatory responses. In particular, previous studies have shown that M2 is closely associated with the most severe clinical presentation of leprosy (i.e. lepromatous leprosy (LL)), as well as tuberculosis. We hypothesized that sCD163 correlates with severity of diseases caused by intracellular pathogens.
To assess this hypothesis, sCD163 levels were measured in the serum of leprosy and visceral leishmaniasis (VL) patients stratified by severity of the clinical presentation. sCD163 levels were significantly higher in patients with these diseases than those observed in healthy control individuals. Further analyses on infection and disease status of leprosy and VL patients revealed a clear association of sCD163 levels with clinical parameters of disease severity. In vitro culture assays revealed that Leishmania infection induced CD163 expression on the surface of both monocyte/macrophages and neutrophils, suggesting these cells as possible sources of sCD163. FACS analyses shows that the cells expressing CD163 produces both TNF-α and IL-4.
Taken together, our results reveal sCD163 as a potential biomarker of severity of diseases caused by intracellular pathogens M. leprae and Leishmania spp. and have a modulatory role, with a mix of an inflammatory property induced by TNF-α release, but that potentially induces an anti-inflammatory T cell response, related to IL-4 release.
Five years after the failure of the Fundão Dam in Brazil, no scientific report has been published regarding the rehabilitation interventions performed by the Renova Foundation, which is solely ...responsible for the rehabilitation of the Doce River Basin. This study aimed to assess the fluvial and riparian rehabilitation efforts performed by Renova, as well as to test and propose alternatives, when appropriate. Before the dam rupture, the fluvial system showed a single‐thread morphology that was sinuous and meandering. After the rupture, changes occurred in the river dynamics with strong sediment deposition in riparian areas, resulting in the loss of several meandering streams, oxbow‐lakes, lagoons, and fluvial islands. There was also a loss of the riparian vegetation, from 61% to 6%, as estimated by the Normalized Difference Vegetation Index (NDVI). Renova subjected nearly 61% of the study area to fluvial rehabilitation techniques, particularly using rip‐rap or larssen sheet piling (37%) and erosion control fibre blankets (17%). However, 50% of these interventions have been unsuccessful because the rip‐rap was poorly positioned and the biodegradable fibre blankets decomposed rapidly. We recommend changing the position of the rip‐rap from convex to concave banks and to avoid using biodegradable fibre blankets. Instead, we encourage the planting of riparian forests with native species to form buffer zone systems. Renova' riparian rehabilitation attempts were found to have low success rates, which was attributed to the unaddressed presence of toxic compounds, such as sodium and ether amine. However, our results proved that riparian rehabilitation using native species is feasible, if physical, chemical, biological, and phyto‐remediation strategies are adopted, to alleviate the ether amine and sodium toxicity.
Understanding the effects of environmental conditions on biodiversity may provide cues regarding the resilience of ecological communities facing human activities in tropical forests. Under this ...scenario, harvestmen are among the most ideal models for understanding the ecological dynamics associated with shifts in environmental conditions. In this study, we evaluated how changes in attributes related to environmental conditions shape harvestmen assemblages in the Atlantic Forest. We assessed the effect of environmental transformation in four forest fragments with different proportions of native forest cover based on their environmental attributes (temperature, humidity, leaf litter depth and cover, and herbaceous vegetation cover) on harvestmen species richness, abundance, and composition. Overall, 865 harvestmen individuals belonging to six families and 41 species were sampled. The effects of environmental conditions on harvestmen assemblages were context‐dependent. In the most conserved fragment, there was an increase in harvestmen richness and abundance with increasing temperature. Furthermore, herbaceous vegetation cover negatively affected harvestmen richness and abundance. Species turnover was the main driver of harvestmen beta diversity in all forest fragments. With the results presented herein, we demonstrate that harvestmen species richness and abundance are modulated by landscape modifications and subsequent alterations in the environment resulting from human‐induced disturbance. Our results highlight the importance of considering environmental transformations at a large spatial scale (i.e., landscape) and local scale (i.e., environmental attributes) to design appropriate conservation strategies for Brazilian Atlantic Forest.

Objective: To analyze the spatial pattern of the incidence of COVID-19 in association with social determinants of health (SDH) in the Northeast Region of Brazil during the first year of the pandemic. ...Methods: We conducted an ecological analytical study that included notifications made between 27 March 2020 and 27 March 2021. The data analysis used two global regression models: the ordinary least squares (OLS) and spatial lag model and the geographically weighted multiscale regression model (GWMSR). Results: We observed that the Gini index, illiteracy rate, percentages of people living below the poverty line, people in households who were vulnerable to poverty, and dependent elderly people are predictors of a higher incidence of COVID-19 in Northeast Brazil. Conclusions: Results of this study may contribute to generating new hypotheses for studies focusing on the syndemic process and for the formulation of intersectoral public policies targeting the population at greatest vulnerability to minimize the impact of the disease.
The increase in incidence of infectious diseases worldwide, particularly in developing countries, is worrying. Each year, 14 million people are killed by infectious diseases, mainly HIV/AIDS, ...respiratory infections, malaria and tuberculosis.. Despite the great burden in the poor countries, drug discovery to treat tropical diseases has come to a standstill. There is no interest by the pharmaceutical industry in drug development against the major diseases of the poor countries, since the financial return cannot be guaranteed. This has created an urgent need for new therapeutics to neglected diseases. A possible approach has been the exploitation of the inhibition of unique targets, vital to the pathogen such as the shikimate pathway enzymes, which are present in bacteria, fungi and apicomplexan parasites but are absent in mammals. The chorismate synthase (CS) catalyses the seventh step in this pathway, the conversion of 5-enolpyruvylshikimate-3-phosphate to chorismate. The strict requirement for a reduced flavin mononucleotide and the anti 1,4 elimination are both unusual aspects which make CS reaction unique among flavin-dependent enzymes, representing an important target for the chemotherapeutic agents development. In this review we present the main biochemical features of CS from bacterial and fungal sources and their difference from the apicomplexan CS. The CS mechanisms proposed are discussed and compared with structural data. The CS structures of some organisms are compared and their distinct features analyzed. Some known CS inhibitors are presented and the main characteristics are discussed. The structural and kinetics data reviewed here can be useful for the design of inhibitors.
Gentamicin is an important aminoglycoside antibiotic used for treatment of infections caused by Gram-negative bacteria. Although most of the biosynthetic pathways of gentamicin have been elucidated, ...a remaining intriguing question is how the intermediates JI-20A and JI-20B undergo a dideoxygenation to form gentamicin C complex. Here we show that the dideoxygenation process starts with GenP-catalyzed phosphorylation of JI-20A and JI-20Ba. The phosphorylated products are successively modified by concerted actions of two PLP (pyridoxal 5′-phosphate)-dependent enzymes: elimination of water and then phosphate by GenB3 and double bond migration by GenB4. Each of these reactions liberates an imine which hydrolyses to a ketone or aldehyde and is then reaminated by GenB3 using an amino donor. Importantly, crystal structures of GenB3 and GenB4 have guided site-directed mutagenesis to reveal crucial residues for the enzymes’ functions. We propose catalytic mechanisms for GenB3 and GenB4, which shed light on the already unrivalled catalytic versatility of PLP-dependent enzymes.
The fact that most IoT solutions are provided by third parties, along with the pervasiveness of the collected data, raises privacy and security concerns. There is a need to verify which data is being ...sent to the third party, as well as preventing those channels from becoming an exploitation avenue. We propose to use existing API definition languages to create contracts which define the data that can be transmitted, their format and constraints. To verify the compliance with these contracts, we propose a Network Service architecture which validates REST-like API requests/responses against a Swagger schema. We deal with encrypted traffic using an Service Function Chaining (SFC)-enabled Man-in-the-Middle (MITM), allowing verifications in "real-time." We devised a Proof of Concept and showed that we were able to detect (and stop) contract violations.
The aim of this study was to investigate whether microalgae in Japanese quail feed alters performance, fatty acid profiles in the eggs and egg quality. One hundred quails were distributed in four ...groups and five replicates of five birds per experimental group. The treatments consisted of four levels of Spirulina platensis microalgae (0%, 5%, 10%, and 15%) in the diets. We evaluated the productive performance and chemical‐physical characteristics of eggs, the oxidant/antioxidant status in egg yolks, and the fatty acid profile in the diet and egg yolks. Microalgae in the diet did not influence egg production; however, it increased the yolk index as well as the color intensity of the yolk. Saturated and polyunsaturated fatty acid levels decreased in egg yolks, and monounsaturated fatty acid levels increased in the yolks. Lipid peroxidation levels were lower and total antioxidant capacity was higher in egg yolks of quails receiving microalgae in the diet.
Practical applications
Microalgae in quail diets improves egg quality and provides benefits to consumer health, acting as an antioxidant and immune‐stimulant. Microalgae in quail diets had positive effects on egg quality. This is because it reduced levels of saturated fatty acids that are undesirable, and increased monounsaturated fatty acid levels that are beneficial to the health of consumers. Finally, antioxidants increased in egg yolks, consequently reducing lipid peroxidation.
