The Old Rhine downstream of the Kembs diversion dam is one of the largest by-passed river reaches in the world (50km). It offers a unique opportunity to study the morphological effects of by-passing ...and address physical and ecological restoration approaches in regulated rivers. We conduct a space-time analysis of channel adjustment over a period of 80years (1932 to 2011). We examine planform changes (from aerial photographs), erosional and depositional patterns (from vertical profiles), sediment sizes within the active channel and the new established floodplain, and we date riparian vegetation encroachment. Results show that the Old Rhine exhibited rapid response to the completion of the by-passing scheme in the 1950s, with a 26% narrowing in median active channel width between 1956 and 2008, from vegetation encroachment on dewatered channel margins (mostly groyne fields). The narrowing was accompanied by overbank fine sediment deposition (~1.5cmy−1 aggradation since 1950) as well as slight bed degradation (~0.7cmy−1 since 1950). We found no downstream propagation of active channel narrowing over time, nor propagation of bed degradation. The channel was already significantly adjusted prior to the diversion scheme, following the nineteenth century river straightening and groyne construction. By-passing (dewatering) mainly provided new pioneer habitat for synchronous vegetation establishment and promoted channel stability by decreasing sediment transport owing to peak flow reduction. The morphological budget calculated over the past 20years estimated a downstream output for coarse sediments at 16,000m3y−1, with 80% originating from bed degradation and 20% from bank erosion, without significant inputs from upstream. The present-day morphodynamics remain sensitive to changes because of dynamic bed armouring (<2.1). This retrospective analysis permits us to discuss management strategies for altered rivers. Recommended activities are gravel reintroduction with particle-sizes finer than the armoured river bed to enhance sediment transport and promote form dynamism under current flood magnitudes. This should be coupled with river widening to allow for complex depositional patterns that benefit ecological habitats. These actions should not be implemented without prior analysis of local sediment transfer processes based on in situ experiments and modelling.

The Materials Science beamline at the Swiss Light Source has been operational since 2001. In late 2010, the original wiggler source was replaced with a novel insertion device, which allows ...unprecedented access to high photon energies from an undulator installed in a medium‐energy storage ring. In order to best exploit the increased brilliance of this new source, the entire front‐end and optics had to be redesigned. In this work, the upgrade of the beamline is described in detail. The tone is didactic, from which it is hoped the reader can adapt the concepts and ideas to his or her needs.
This study investigated changes in heart rate variability (HRV) in elite Nordic-skiers to characterize different types of "fatigue" in 27 men and 30 women surveyed from 2004 to 2008. R-R intervals ...were recorded at rest during 8 min supine (SU) followed by 7 min standing (ST). HRV parameters analysed were powers of low (LF), high (HF) frequencies, (LF+HF) (ms(2)) and heart rate (HR, bpm). In the 1 063 HRV tests performed, 172 corresponded to a "fatigue" state and the first were considered for analysis. 4 types of "fatigue" (F) were identified: 1. F(HF(-)LF(-))SU_ST for 42 tests: decrease in LFSU (- 46%), HFSU (- 70%), LFST (- 43%), HFST (- 53%) and increase in HRSU (+ 15%), HRST (+ 14%). 2. F(LF(+) SULF(-) ST) for 8 tests: increase in LFSU (+ 190%) decrease in LFST (- 84%) and increase in HRST (+ 21%). 3. F(HF(-) SUHF(+) ST) for 6 tests: decrease in HFSU (- 72%) and increase in HFST (+ 501%). 4. F(HF(+) SU) for only 1 test with an increase in HFSU (+ 2161%) and decrease in HRSU (- 15%). Supine and standing HRV patterns were independently modified by "fatigue". 4 "fatigue"-shifted HRV patterns were statistically sorted according to differently paired changes in the 2 postures. This characterization might be useful for further understanding autonomic rearrangements in different "fatigue" conditions.
Using inelastic neutron scattering, we map a 14 meV coherent resonant mode in the topological Kondo insulator SmB6 and describe its relation to the low energy insulating band structure. The resonant ...intensity is confined to the X and R high symmetry points, repeating outside the first Brillouin zone and dispersing less than 2 meV, with a 5d-like magnetic form factor. We present a slave-boson treatment of the Anderson Hamiltonian with a third neighbor dominated hybridized band structure. This approach produces a spin exciton below the charge gap with features that are consistent with the observed neutron scattering. We find that maxima in the wave vector dependence of the inelastic neutron scattering indicate band inversion.
Abstract
Disease susceptibility and resistance are important factors for the conservation of endangered species, including elephants. We analyzed pathology data from 26 zoos and report that Asian ...elephants have increased neoplasia and malignancy prevalence compared with African bush elephants. This is consistent with observed higher susceptibility to tuberculosis and elephant endotheliotropic herpesvirus (EEHV) in Asian elephants. To investigate genetic mechanisms underlying disease resistance, including differential responses between species, among other elephant traits, we sequenced multiple elephant genomes. We report a draft assembly for an Asian elephant, and defined 862 and 1,017 conserved potential regulatory elements in Asian and African bush elephants, respectively. In the genomes of both elephant species, conserved elements were significantly enriched with genes differentially expressed between the species. In Asian elephants, these putative regulatory regions were involved in immunity pathways including tumor-necrosis factor, which plays an important role in EEHV response. Genomic sequences of African bush, forest, and Asian elephant genomes revealed extensive sequence conservation at TP53 retrogene loci across three species, which may be related to TP53 functionality in elephant cancer resistance. Positive selection scans revealed outlier genes related to additional elephant traits. Our study suggests that gene regulation plays an important role in the differential inflammatory response of Asian and African elephants, leading to increased infectious disease and cancer susceptibility in Asian elephants. These genomic discoveries can inform future functional and translational studies aimed at identifying effective treatment approaches for ill elephants, which may improve conservation.
Many actors, from the individual to neighborhood to municipal scale, influence the management of trees, grass, and other vegetation on residential properties. Recent work has been directed toward ...understanding the ecological characteristics of residential landscapes and the human drivers of landscape management; however, much of this work remains disciplinarily focused and at a single scale of analysis. This study employs a mixed-method approach to examine household- and neighborhood-scale drivers of urban residential tree species richness and tree canopy structure. A stratified sampling design was used to capture households in homeowners associations (HOAs) and neighborhood associations (NAs) to better understand the informal and formal institutions having an impact on residential tree management practices. We used the social-ecological system (SES) framework to build a classification system for identifying significant variables that influence residential tree composition and cover. Results of this work demonstrated tree species richness and canopy cover to be positively related to tree abundance and housing age, which is suggestive of the legacy effect. Governance variables were more strongly correlated with tree species richness whereas variables reflecting socioeconomic status and education were more strongly correlated with canopy cover. Rule compliance and fitting in with neighborhood landscaping norms was significantly more important to HOA households than NA households. Conversely, opportunities to work together to solve community problems were viewed more positively by NA households than HOA households. For all parcels together, compliance with city and HOA/NA rules was negatively related to tree species richness and canopy cover, a finding that may implicate the nature of rules that focus on tree removal practices and barriers to plant (what and where). This work is timely given the rapid expansion of neighborhood associations in urban areas, which parallels establishment of rules governing residential yard practices. If current arboriculture and urban forestry standards are considered during rule formation and implementation, such rules have the potential to promote species diversity and the sustained provisioning of ecosystem services.
Objective. Cold-inducible RNA-binding protein (CIRBP) has been shown to be involved not only in cooling-induced cellular protection but also as a mediator of sterile inflammation, a critical ...mechanism of the innate immune response in ischemia/reperfusion (I/R) injury. The role of microglia and its activation in cerebral I/R injury warrants further investigation as both detrimental and regenerative properties have been described. Therefore, we investigated the effects of cooling, specifically viability, activation, and release of damage associated molecular patterns (DAMPs) on oxygen glucose deprivation/reperfusion- (OGD/R-) induced injury in murine BV-2 microglial cells. Methods. Murine BV-2 microglial cells were exposed to 2 to 6 h OGD (0.2% O2 in glucose- and serum-free medium) followed by up to 19 h of reperfusion, simulated by restoration of oxygen (21% O2) and nutrients. Cells were maintained at either normothermia (37°C) or cooled to 33.5°C, 1 h after experimental start. Cultured supernatants were harvested after exposure to OGD for analysis of DAMP secretions, including high-mobility group box 1 (HMGB1), heat shock protein 70 (HSP70), and CIRBP, and cytotoxicity was assessed by lactate dehydrogenase releases after exposure to OGD and reperfusion. Intracellular cold-shock proteins CIRBP and RNA-binding motif 3 (RBM3) as well as caspases 9, 8, and 3 were also analyzed via Western blot analysis. Furthermore, inducible nitric oxide synthase (iNOS), ionized calcium-binding adaptor molecule 1 (Iba1), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1β (IL-1β), interleukin-1α (IL-1α), monocyte chemotactic protein 1 (MCP-1), transforming growth factor β (TGFβ), CIRBP, and RBM3 gene expressions were assessed via reverse transcription polymerase chain reaction, and TNF-α, IL-6, and IL-1β releases into the cultured supernatants were assessed via enzyme-linked immunosorbent assays (ELISA). Results. Prolonged exposure to OGD resulted in increased BV-2 necrotic cell death, which was attenuated by cooling. Cooling also significantly induced cold-shock proteins CIRBP and RBM3 gene expressions, with CIRBP expression more rapidly regulated than RBM3 and translatable to significantly increased protein expression. DAMPs including HMGB-1, HSP70, and CIRBP could be detected in cultured supernatants after 6 h of OGD with CIRBP release being significantly attenuated by cooling. Exposure to OGD suppressed cytokine gene expressions of IL-1β, TNF-α, MCP-1, and TGFβ independently of temperature management, whereas cooling led to a significant increase in IL-1α gene expression after 6 h of OGD. In the reperfusion phase, TNF-α and MCP-1 gene expressions were increased, and cooling was associated with significantly lower TGFβ gene expression. Interestingly, cooled Normoxia groups had significant upregulations of microglial activation marker, Iba1, IL-1β, and TNF-α gene expressions. Conclusion. BV-2 microglial cells undergo necrotic cell death resulting in DAMP release due to OGD/R-induced injury. Cooling conveyed neuroprotection in OGD/R-injury as observable in increased cell viability as well as induced gene expressions of cold shock proteins. As cooling alone resulted in both upregulation of microglial activation, expression of proinflammatory cytokines, and cold shock protein transcript and protein expression, temperature management might have ambiguous effects in sterile inflammation. However, cooling resulted in a significant decrease of extracellular CIRBP, which has recently been characterized as a novel DAMP and a potent initiator and mediator of inflammation.
An unexplained >4σ discrepancy persists between "beam" and "bottle" measurements of the neutron lifetime. A new model proposed that conversions of neutrons n into mirror neutrons n^{'}, part of a ...dark mirror sector, can increase the apparent neutron lifetime by 1% via a small mass splitting Δm between n and n^{'} inside the 4.6 T magnetic field of the National Institute of Standards and Technology Beam Lifetime experiment. A search for neutron conversions in a 6.6 T magnetic field was performed at the Spallation Neutron Source which excludes this explanation for the neutron lifetime discrepancy.
Genome architecture and organization play critical roles in cell life. However, it remains largely unknown how genomic loci are dynamically coordinated to regulate gene expression and determine cell ...fate at the single cell level. We have developed an inducible system which allows Simultaneous Imaging and Manipulation of genomic loci by Biomolecular Assemblies (SIMBA) in living cells. In SIMBA, the human heterochromatin protein 1α (HP1α) is fused to mCherry and FRB, which can be induced to form biomolecular assemblies (BAs) with FKBP-scFv, guided to specific genomic loci by a nuclease-defective Cas9 (dCas9) or a transcriptional factor (TF) carrying tandem repeats of SunTag. The induced BAs can not only enhance the imaging signals at target genomic loci using a single sgRNA, either at repetitive or non-repetitive sequences, but also recruit epigenetic modulators such as histone methyltransferase SUV39H1 to locally repress transcription. As such, SIMBA can be applied to simultaneously visualize and manipulate, in principle, any genomic locus with controllable timing in living cells.
The study of railway tracks under high speed trains is one of the most important researches in the domain of transport. A reduced scale experiment with three sleepers is presented to study the ...dynamic behavior and the settlement of ballasted tracks. A large number of trains passing at high speeds are simulated by signals, applied with the help of hydraulic jacks, having the shape of the letter M and representing the passages of bogies on sleepers. This experiment offers results such as displacements, accelerations, pressures and settlements that allow to better understand the dynamic behavior of a portion of a ballasted railway track at reduced scale and to estimate the settlement versus the number of load cycles. It was found that mechanical properties such as the global stiffness of the track can have important variations during the experiment. The settlement was also found to be a function of the acceleration of sleepers and above all it was observed, for accelerations above a critical value, that the increase of settlement per cycle was very high.
