In 1926, Professor Paul A. van der Bijl made an address to the South African Association for the Advancement of Science in his capacity as President of Section C of the Association, entitled ...‘Landmarks in the development of the science of plant pathology and of disease control’.1 The talk itself marks a key moment in the development of plant pathology as a discipline in South Africa, as Van der Bijl had been appointed 5 years earlier as the first Professor of Plant Pathology and Mycology in South Africa, at Stellenbosch University. The field was just being established, not only in South Africa, but in many parts of the world, and its distinction from other disciplines was a matter of pride.
A putative male-produced pheromone has recently been described for the global pest of pines, Sirex noctilio, but field-activity has not been demonstrated. This study aimed to investigate the ...pheromone biology of S. noctilio in more detail. Specifically, we i) analysed effluvia and extracts for additional compounds by gas chromatography coupled with electro-antennographic detection (GC-EAD), mass spectrometry (GC-MS) and two dimensional time of flight mass spectrometry (GC X GC TOF MS), ii) conducted dose-response experiments for putative pheromone components, iii) determined the site of synthesis/ storage of the putative pheromone and iv) determined the release rate of the putative pheromone from males and three types of lures. A blend of four compounds was identified, including the previously described (Z)-3-decenol and (Z)-4-decenol, and two new compounds (Z)-3-octenol and (Z)-3-dodecenol. All compounds elicited a response from both male and female antennae, but the strength of the response varied according to sex, compound and dose tested. (Z)-3-Decenol and (Z)-3-octenol at lower and higher doses, respectively, elicited larger responses in males and females than the other two compounds. (Z)-3-Octenol and (Z)-4-decenol generally elicited larger female than male antennal responses. The site of synthesis and/or storage in males was determined to be the hind legs, likely in the leg-tendon gland. The relative release rate of the major compound by male wasps was shown to be 90 ± 12.4 ng/min, which is between 4 and 15 times greater than that observed from typical lures used previously. These observations are consistent with the hypothesis that these compounds may mediate lek formation in S. noctilio males and lek location in females.
Many academies globally interact with young scientists (viewed here as all those who have just completed postgraduate studies to mid-career scientists) only on an ad-hoc basis through prizes or ...once-off engagements. This substantial group of active knowledge producers is often not treated as a core part of the scientific community or as leaders who could shape science and society -- despite the fact that they often drive new developments, and will be the leaders of the scientific community in as little as 5-10 years. This paradigm of interaction between academies of science and young scientists is changing in South Africa, thanks to visionary leadership in the Academy of Science of South Africa (ASSAf) over the last few years. ASSAf today is amongst the most active academies globally in their efforts to promote young scientists. There is no other body in South Africa that is currently doing more to provide platforms for their interaction and engagement, and to give a voice to this critical part of the science community.
The Botryosphaeriales are amongst the most widespread, common and important fungal pathogens of woody plants. Many are also known to exist as endophytes in healthy plant tissues. This special issue ...highlights a number of key themes in the study of this group of fungi. In particular, there have been dramatic taxonomic changes over the past decade; from one family to nine (including two in this special issue) and from 10 to 33 genera known from culture. It is also clear from many studies that neither morphology nor single locus sequence data are sufficient to define taxa. This problem is exacerbated by the increasing recognition of cryptic species and hybrids (as highlighted for the first time in this special issue). It is futile that management strategies, including quarantine, continue to rely on outdated taxonomic definitions and identification tools. This is especially true in light of growing evidence that many species continue to be moved globally as endophytes in plants and plant products. A well defined natural classification and an extensive collection of tools to study the Botryosphaeriaceae, including a growing number of genomes, now provide a springboard for a much deeper exploration of their biology, biogeography and host associations.
The Botryosphaeriaceae are important plant pathogens, but also have the ability to establish asymptomatic infections that persist for extended periods in a latent state. In this study, we used ...comparative genome analyses to shed light on the genetic basis of the interactions of these fungi with their plant hosts. For this purpose, we characterised secreted hydrolytic enzymes, secondary metabolite biosynthetic gene clusters and general trends in genomic architecture using all available Botryosphaeriaceae genomes, and selected Dothideomycetes genomes. The Botryosphaeriaceae genomes were rich in carbohydrate-active enzymes (CAZymes), proteases, lipases and secondary metabolic biosynthetic gene clusters (BGCs) compared to other Dothideomycete genomes. The genomes of Botryosphaeria, Macrophomina, Lasiodiplodia and Neofusicoccum, in particular, had gene expansions of the major constituents of the secretome, notably CAZymes involved in plant cell wall degradation. The Botryosphaeriaceae genomes were shown to have moderate to high GC contents and most had low levels of repetitive DNA. The genomes were not compartmentalized based on gene and repeat densities, but genes of secreted enzymes were slightly more abundant in gene-sparse regions. The abundance of secreted hydrolytic enzymes and secondary metabolite BGCs in the genomes of Botryosphaeria, Macrophomina, Lasiodiplodia, and Neofusicoccum were similar to those in necrotrophic plant pathogens and some endophytes of woody plants. The results provide a foundation for comparative genomic analyses and hypotheses to explore the mechanisms underlying Botryosphaeriaceae host-plant interactions.
A precondition for colour vision is the presence of at least two spectral types of photoreceptors in the eye. The order Hymenoptera is traditionally divided into the Apocrita (ants, bees, wasps) and ...the Symphyta (sawflies, woodwasps, horntails). Most apocritan species possess three different photoreceptor types. In contrast, physiological studies in the Symphyta have reported one to four photoreceptor types. To better understand the evolution of photoreceptor diversity in the Hymenoptera, we studied the Symphyta Sirex noctilio, which belongs to the superfamily Siricoidea, a closely related group of the Apocrita suborder. Our aim was to (i) identify the photoreceptor types of the compound eye by electroretinography (ERG), (ii) characterise the visual opsin genes of S. noctilio by genomic comparisons and phylogenetic analyses and (iii) analyse opsin mRNA expression. ERG measurements revealed two photoreceptor types in the compound eye, maximally sensitive to 527 and 364 nm. In addition, we identified three opsins in the genome, homologous to the hymenopteran green or long-wavelength sensitive (LW) LW1, LW2 and ultra-violet sensitive (UV) opsin genes. The LW1 and UV opsins were found to be expressed in the compound eyes, and LW2 and UV opsins in the ocelli. The lack of a blue or short-wavelength sensitive (SW) homologous opsin gene and a corresponding receptor suggests that S. noctilio is a UV-green dichromate.

In recent years, forests have been exposed to an unprecedented rise in pests and pathogens. This, coupled with the added challenge of climate change, renders forest plantation stock vulnerable to ...attack and severely limits productivity. Genotypes resistant to such biotic challenges are desired in plantation forestry to reduce losses. Conventional breeding has been a main avenue to obtain resistant genotypes. More recently, genetic engineering has become a viable approach to develop resistance against pests and pathogens in forest trees. Tree genomic resources have contributed to advancements in both these approaches. Genome-wide association studies and genomic selection in tree populations have accelerated breeding tools while integration of various levels of omics information facilitates the selection of candidate genes for genetic engineering. Furthermore, tree associations with non-pathogenic endophytic and subterranean microbes play a critical role in plant health and may be engineered in forest trees to improve resistance in the future. We look at recent studies in forest trees describing defense mechanisms using such approaches and propose the way forward to developing superior genotypes with enhanced resistance against biotic stress.
The advent of simple and affordable tools for molecular identification of novel insect invaders and assessment of population diversity has changed the face of invasion biology in recent years. The ...widespread application of these tools has brought with it an emerging understanding that patterns in biogeography, introduction history and subsequent movement and spread of many invasive alien insects are far more complex than previously thought. We reviewed the literature and found that for a number of invasive insects, there is strong and growing evidence that multiple introductions, complex global movement, and population admixture in the invaded range are commonplace. Additionally, historical paradigms related to species and strain identities and origins of common invaders are in many cases being challenged. This has major consequences for our understanding of basic biology and ecology of invasive insects and impacts quarantine, management and biocontrol programs. In addition, we found that founder effects rarely limit fitness in invasive insects and may benefit populations (by purging harmful alleles or increasing additive genetic variance). Also, while phenotypic plasticity appears important post-establishment, genetic diversity in invasive insects is often higher than expected and increases over time via multiple introductions. Further, connectivity among disjunct regions of global invasive ranges is generally far higher than expected and is often asymmetric, with some populations contributing disproportionately to global spread. We argue that the role of connectivity in driving the ecology and evolution of introduced species with multiple invasive ranges has been historically underestimated and that such species are often best understood in a global context.
Biodiversity and economic losses resulting from invasive plant pests and pathogens are increasing globally. For these impacts and threats to be managed effectively, appropriate methods of ...surveillance, detection and identification are required. Botanical gardens provide a unique opportunity for biosecurity as they accommodate diverse collections of exotic and native plant species. These gardens are also often located close to high-risk sites of accidental invasions such as ports and urban areas. This, coupled with routine activities such as the movement of plants and plant material, and visits by millions of people each year, place botanical gardens at risk to the arrival and establishment of pests and pathogens. Consequently, botanical gardens can pose substantial biosecurity risks to the environment, by acting as bridgeheads for pest and pathogen invasions. Here we review the role of botanical gardens in biosecurity on a global scale. The role of botanical gardens has changed over time. Initially, they were established as physic gardens (gardens with medicinal plants), and their links with academic institutions led to their crucial role in the accumulation and dissemination of botanical knowledge. During the second half of the 20th century, botanical gardens developed a strong focus on plant conservation, and in recent years there has been a growing acknowledgement of their value in biosecurity research as sentinel sites to identify pest and pathogen risks (novel pest-host associations); for early detection and eradication of pests and pathogens; and for host range studies. We identify eight specific biosecurity hazards associated with botanical gardens and note potential management interventions and the opportunities these provide for improving biosecurity. We highlight the value of botanical gardens for biosecurity and plant health research in general, and the need for strategic thinking, resources, and capacity development to make them models for best practices in plant health.
