Crop ...genetic improvement to address environmental stresses for sustainable food production has been in high demand, especially given the current situation of global climate changes and reduction of the global food production rate/population rate. Recently, the emerging clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein (Cas)-based targeted mutagenesis has provided a revolutionary approach to crop improvement. The major application of CRISPR/Cas in plant genome editing has been the generation of indel mutations via error-prone nonhomologous end joining (NHEJ) repair of DNA DSBs. In this study, we examined the power of the CRISPR/Cas9-based novel approach in the precise manipulation of protein domains of tomato hybrid proline-rich protein 1 (HyPRP1), which is a negative regulator of salt stress responses. We revealed that the precise elimination of SlHyPRP1 negative-response domain(s) led to high salinity tolerance at the germination and vegetative stages in our experimental conditions. CRISPR/Cas9-based domain editing may be an efficient tool to engineer multidomain proteins of important food crops to cope with global climate changes for sustainable agriculture and future food security.
We previously reported that the ethyl acetate (EtOAc) fraction of a 70% ethanol extract of
(ESE) inhibits varicella-zoster virus (VZV) and human cytomegalovirus (HCMV) replication in vitro. PGG ...(1,2,3,4,6-penta-O-galloyl-ß-D-glucose) is a major chemical constituent of the EtOAc fraction of ESE that inhibits VZV but not HCMV replication. In this study, we comprehensively screened the chemical compounds identified in the EtOAc fraction of ESE for potential antiviral properties. Among the examined compounds, quercetin and isoquercitrin displayed potent antiviral activities against both VZV and HCMV with no significant cytotoxic effects. Both compounds strongly suppressed the expression of VZV and HCMV immediate-early (IE) genes. Our collective results indicated that, in addition to PGG, quercetin and isoquercitrin are bioactive compounds in the EtOAc fraction of ESE that effectively inhibit human herpesvirus replication.
The practicability of deep learning techniques has been demonstrated by their successful implementation in varied fields, including diagnostic imaging for clinicians. In accordance with the ...increasing demands in the healthcare industry, techniques for automatic prediction and detection are being widely researched. Particularly in dentistry, for various reasons, automated mandibular canal detection has become highly desirable. The positioning of the inferior alveolar nerve (IAN), which is one of the major structures in the mandible, is crucial to prevent nerve injury during surgical procedures. However, automatic segmentation using Cone beam computed tomography (CBCT) poses certain difficulties, such as the complex appearance of the human skull, limited number of datasets, unclear edges, and noisy images. Using work-in-progress automation software, experiments were conducted with models based on 2D SegNet, 2D and 3D U-Nets as preliminary research for a dental segmentation automation tool. The 2D U-Net with adjacent images demonstrates higher global accuracy of 0.82 than naïve U-Net variants. The 2D SegNet showed the second highest global accuracy of 0.96, and the 3D U-Net showed the best global accuracy of 0.99. The automated canal detection system through deep learning will contribute significantly to efficient treatment planning and to reducing patients' discomfort by a dentist. This study will be a preliminary report and an opportunity to explore the application of deep learning to other dental fields.
The present study investigated whether infants reason about others' social preferences based on the intentions of others' interactive actions. In Experiment 1, 12-month-old infants were familiarized ...with an event in which an agent either successfully helped a circle to climb up a hill (successful-helping condition) or failed to help the circle to achieve its goal (failed-helping condition). During the test, the infants saw the circle approach either the helper (approach-helper event) or the hinderer (approach-hinderer event). In the successful-helping condition, the 12-month-old infants looked for longer at the approach-hinderer event than at the approach-helper event, but in the failed-helping condition, looking times were about equal for the two test events. These results suggest that 12-month-old infants could not infer the circle's preference when the helper's action did not lead to its intended outcome. In Experiment 2, 16-month-olds were tested in the failed-helping condition; they looked longer at the approach-hinderer event than at the approach-helper event, which suggests that they could reason about the third party's social preferences based on the exhibited intentions. In Experiment 3, 12-month-olds were familiarized with events in which the final outcomes of helping and hindering actions were ambiguous. The results revealed that 12-month-old infants are also sensitive to intentions when inferring other's social preferences. The results suggest that by 12-months of age, infants expect an agent to prefer and approach another who intends to help the circle to achieve its goal, regardless of the outcome. The current research has implications for moral reasoning and social evaluation in infancy.
The lateral flow immunosensor (LFI) is a widely used diagnostic tool for biomarker detection; however, its sensitivity is often insufficient for analyzing targets at low concentrations. Here, an ...electrochemiluminescent LFI (ECL‐LFI) is developed for highly sensitive detection of troponin I (TnI) using Ru(bpy)32+‐loaded mesoporous silica nanoparticles (RMSNs). A large amount of Ru(bpy)32+ is successfully loaded into the mesoporous silica nanoparticles with excellent loading capacity and shows strong ECL signals in reaction to tripropylamine. Antibody‐immobilized RMSNs are applied to detect TnI by fluorescence and ECL analysis after a sandwich immunoassay on the ECL‐LFI strip. The ECL‐LFI enables the highly sensitive detection of TnI‐spiked human serum within 20 min at femtomolar levels (≈0.81 pg mL−1) and with a wide dynamic range (0.001–100 ng mL−1), significantly outperforming conventional fluorescence detection (>3 orders of magnitude). Furthermore, TnI concentrations in 35 clinical serum samples across a low range (0.01–48.31 ng mL−1) are successfully quantified with an excellent linear correlation (R2 = 0.9915) using a clinical immunoassay analyzer. These results demonstrate the efficacy of this system as a high‐performance sensing strategy capable of capitalizing on future point‐of‐care testing markets for biomolecule detection.
An electrochemiluminescent (ECL) lateral flow immunosensor (ECL‐LFI) is developed by incorporating an electrode in the lateral flow testing format for highly sensitive biomolecule detection. The synthesized luminescent nanoparticles are used as an ECL signal generator after the sandwich immunoassay. The ECL‐LFI enables highly sensitive and quantitative detection of troponin I, indicating its great potential as a powerful point‐of‐care testing device.
The Korea Institute of Atmospheric Prediction Systems (KIAPS) began a national project to develop a new global atmospheric model system in 2011. The ultimate goal of this 9-year project is to replace ...the current operational model at the Korea Meteorological Administration (KMA), which was adopted from the United Kingdom’s Meteorological Office’s unified model (UM) in 2010. The 12-km Korean Integrated Model (KIM) system, consisting of a spectral-element non-hydrostatic dynamical core on a cubed sphere grid and a state-of-the-art physics parameterization package, has been launched in a real-time forecast framework, with initial conditions obtained via the advanced hybrid four-dimensional ensemble variational data assimilation (4DEnVar) over its native grid. A development strategy for KIM and the evolution of its performance in medium-range forecasts toward a world-class global forecast system are described. Outstanding issues in KIM 3.1 as of February 2018 are discussed, along with a future plan for operational deployment in 2020.
Osteoarthritis (OA) is the most common degenerative joint disease; however, its etiopathogenesis is not completely understood. Here we show a role for NUDT7 in OA pathogenesis. Knockdown of NUDT7 in ...normal human chondrocytes results in the disruption of lipid homeostasis. Moreover, Nudt7
mice display significant accumulation of lipids via peroxisomal dysfunction, upregulation of IL-1β expression, and stimulation of apoptotic death of chondrocytes. Our genome-wide analysis reveals that NUDT7 knockout affects the glycolytic pathway, and we identify Pgam1 as a significantly altered gene. Consistent with the results obtained on the suppression of NUDT7, overexpression of PGAM1 in chondrocytes induces the accumulation of lipids, upregulation of IL-1β expression, and apoptotic cell death. Furthermore, these negative actions of PGAM1 in maintaining cartilage homeostasis are reversed by the co-introduction of NUDT7. Our results suggest that NUDT7 could be a potential therapeutic target for controlling cartilage-degrading disorders.
