To study the micro scale interactions of Mycobacterium with bacteria belonging to other genera by means of Confocal Laser Scanning Microscopy (CLSM), a procedure was developed to non-invasively and ...fluorescently stain Mycobacterium without compromising the signal produced by commonly used fluorescent reporter genes. The procedure makes use of the commercial non-specific nucleic acid stain Syto62 and was optimized to efficiently stain Mycobacterium cells in suspensions and biofilms. The staining procedure was found non-invasive towards overall cell viability, biofilm architecture and fluorescence signals emitted by other organisms expressing the fluorescent reporter genes gfp and dsRed. The procedure was successfully applied to visualize the comportment of the PAH-degrading Mycobacterium sp. VM552 in triple species biofilms containing, in addition to strain VM552, the GFP labeled PAH-degrading Sphingomonas sp. LH128-GFP and DsRed-labeled Pseudomonas putida OUS82(RF), and colonizing a glass substrate coated with phenanthrene crystals in flow chambers. CLSM imaging and subsequent appropriate image processing of the biofilms show that the comportment of strain Mycobacterium sp. VM552 was largely affected by the presence of the other organisms. The data support the value of the staining procedure to study ecological questions about micro scale behavior and niche occupation of Mycobacterium in multi-species systems.

Serum 25(OH)D levels were associated with body mass index (BMI: r = -0.161, P = 0.006), PTH (r = -0.165; P = 0.005), CTX (r = -0.119; P = 0.043) and vBMD at cortical bone (Dcomp: r = 0.132; P = 0.033) and entire bone (D100: r = 0.162 P = 0.009) at the distal radius, but not at the tibia. Age and PTH levels were potential confounding variables, but in the multiple linear regressions only BMI (95% CI: 0.11-4.16; P < 0.01), 25(OH)D (95% CI: -0.007 to 1.70; P = 0.05) and CTX (95% CI: -149.04 to 21.80; P < 0.01) predicted Dcomp, while BMI (95% CI: 1.13-4.18; P < 0.01) and 25(OH)D (95% CI: 0.24-1.52; P < 0.01) predicted D100. NonVFs predominated in patients with 25(OH)D <20 ng/mL (P = 0.013). Logistic regression analysis showed a decrease in the likelihood of presenting grade 2-3 VFs/NonVFs for every increase in 25(OH)D (OR = 0.962, 95% CI: 0.940-0.984; P = 0.001), BMI (OR = 0.932, 95% CI: 0.885-0.981; P = 0.007) and D100 at radius (OR = 0.994, 95% CI: 0.990-0.998; P = 0.005).

The Bay of Biscay (North-East Atlantic) has long been subjected to intense direct and indirect human activities that lead to the excessive degradation and sometimes overexploitation of natural ...resources. Fisheries management is gradually moving away from single-species assessments to more holistic, multi-species approaches that better respond to the reality of ecosystem processes. Quantitative modelling methods such as Ecopath with Ecosim can be useful tools for planning, implementing and evaluating ecosystem-based fisheries management strategies. The aim of this study was therefore to model the energy fluxes within the food web of this highly pressured ecosystem and to extract practical information required in the diagnosis of ecosystem state/health. A well-described model comprising 30 living and two non-living compartments was successfully constructed with data of local origin, for the Bay of Biscay continental shelf. The same level of aggregation was applied to primary producers, mid-trophic-levels and top-predators boxes. The model was even more general as it encompassed the entire continuum of marine habitats, from benthic to pelagic domains. Output values for most ecosystem attributes indicated a relatively mature and stable ecosystem, with a large proportion of its energy flow originating from detritus. Ecological network analysis also provided evidence that bottom-up processes play a significant role in the population dynamics of upper-trophic-levels and in the global structuring of this marine ecosystem. Finally, a novel metric based on ecosystem production depicted an ecosystem not far from being overexploited. This finding being not entirely consistent over indicators, further analyses based on dynamic simulations are required.
Monitoring the health status of marine mammals is a priority theme that France aims to develop with the other European Union Member States in the next two years, in the context of the Marine Strategy ...Framework Directive. With approximately 5,000 km of coastline and for nearly ten years, France has been recording an average of 2,000 strandings per year, which are monitored by the National Stranding Network, managed by Pelagis, the observatory for the conservation of marine mammals from La Rochelle University and the French National Center for Scientific Research. Since 1972, this network has successively evolved from spatial and temporal faunistic description to, nowadays, the detection of major causes of mortality. It now aims to carry out epidemiological studies on a population scale. Thus, a strategy to strengthen the monitoring of marine mammals’ health status based on stranding data has been developed. This strategy will allow for a more accurate detection of anthropogenic cause of death as well as those of natural origin. It will allow the monitoring of time trends and geographical differences of diseases associated with conservation and public health issues while ensuring the early detection of emerging and/or zoonotic diseases of importance. It will also allow a better assessment of the consequences of human activities on these animal populations and on the environment. Thus, this strategy is fully in line with the “One Health” approach which implies an integrated vision of public, animal and environmental health. It is broken down into four surveillance modalities: (1) general event-based surveillance (GES); (2) programmed surveillance (PS); (3) specific event-based surveillance (SES); (4) and in the longer term, syndromic surveillance (SyS). This article describes the French strategy as well as these different surveillance modalities, the levels of examinations and the associated sampling protocols and finally, the method of standardisation of the data collected. The objective is to present the strategy developed at the French level in order to integrate it into a future strategy shared at the European level to standardise practices and especially complementary analysis, necessary for a better evaluation of the health status of these mobile marine species.
Over the past decade, the distribution of harbour porpoises Phocoena phocoena has undergone a southward shift in the North Sea, which has led to an increase in the number of stranded porpoises in its ...southern part. Since the changes in distribution and relative abundance of porpoises may be linked to the changes in prey availability, the aim of the present work was to investigate whether any changes in the feeding habits of harbour porpoises along the North Sea occurred in the past decade. The diet of harbour porpoises stranded along the southern North Sea (northern France and Belgian coast) was assessed through 3 complementary methods: stomach content analysis, stable isotopes (carbon and nitrogen) analysis determined from muscle samples, and fatty acids analysis determined from blubber samples. Fatty acid patterns and stable isotope values from 52 porpoises were compared to 14 potential prey species collected from the southern North Sea. Our results showed that the diet of porpoises along the southern North Sea comprises fish species that are among the most abundant and widely distributed in the area, except for the sardine Sardina pilchardus that appeared to be a new potential prey. Moreover, our results suggested that the decline in sandeel (Ammodytidae) in the northern parts of the North Sea along with the re-invasion of the southern North Sea by sardine species might affect the distribution of harbour porpoises.

However, pelagic trawl fisheries that target sea bass have negative operational interactions with common dolphins (Delphinus delphis). Our goal was to determine the diet of adult sea bass in the Bay of Biscay from stomach-content and stable-isotope analyses, and explore the dietary overlap between sea bass and common dolphins. We found that sea bass primarily target small pelagic fish, most notably mackerel (Scomber scombrus), scads (Trachurus spp.), anchovy (Engraulis encrasicolus), and sardine (Sardina pilchardus). These four species also dominated the diets of common dolphins. This overlap in feeding preferences could increase the risk of dolphins being caught by trawl fisheries while feeding among sea bass, and may be an underlying mechanism to explain the high rate of common dolphin bycatch observed in the pelagic trawl fishery for sea bass in the Bay of Biscay. Understanding the foraging ecology and trophic interactions of predator species is an essential step for identifying and resolving management issues in the northeastern Atlantic and other marine ecosystems.

A scanning-less single-photon counting system for FLIM and fluorescence anisotropy wide-field imaging is described and characterized in this paper. The two polarizations of the fluorescence are ...divided by a Glan prism and acquired at the same time by the QA detector.
