A method for quantifying C10−C13 polychloroalkanes (PCAs) in environmental samples by high-resolution gas chromatography/electron capture negative ion high-resolution mass spectrometry in the ...selected ion monitoring (SIM) mode is presented. The molecular compositions of commercial PCAs and of PCA-containing extracts from environmental samples are first determined by monitoring M − Cl- ions of specific m/z value corresponding to formula groups present and by assuming that the integrated ion signals are proportional to molar concentration weighted by the number of chlorine atoms in the formula group. For quantitative measurement, one specific m/z peak is selected for each analyte and its SIM response is integrated. The integrated SIM responses have a linear dependence upon amount of PCA injected over the range 0.5−500 ng. An analytical protocol is then described. High-resolution (∼12 000 resolving power) mass spectrometry is shown to be desirable to eliminate self-interferences between PCAs at low resolving power (∼1000) and potential interferences from technical chlordane, toxaphene, PCBs, and other organochlorine pesticides. Extraction recoveries of PCAs from fish averaged >80%. The analytical detection limit is ∼60 pg of injected PCA, at a signal-to-noise ratio of 4:1, while the method detection limit was 23 ng/g. As illustrations of the application of the technique to “real world” problems, PCA levels in biota and sediment samples from the mouth of the Detroit River (MI) are reported to be in the 0.3−1.2 μg/g range.
Direct observations of vertical movements of the earth's surface are now possible with space-based GPS networks, and have applications to resources, hazards and tectonics. Here we present data on ...vertical movements of the Earth's surface in New Zealand, computed from the processing of GPS data collected between 2000 and 2015 by 189 permanent GPS stations. We map the geographical variation in vertical rates and show how these variations are explicable within a tectonic framework of subduction, volcanic activity and slow slip earthquakes. Subsidence of >3 mm/yr is observed along southeastern North Island and is interpreted to be due to the locked segment of the Hikurangi subduction zone. Uplift of 1–3 mm/yr further north along the margin of the eastern North Island is interpreted as being due to the plate interface being unlocked and underplating of sediment on the subduction thrust. The Volcanic Plateau of the central North Island is being uplifted at about 1 mm/yr, which can be explained by basaltic melts being injected in the active mantle-wedge at a rate of ∼6 mm/yr. Within the Central Volcanic Region there is a 250 km2 area that subsided between 2005 and 2012 at a rate of up to 14 mm/yr. Time series from the stations located within and near the zone of subsidence show a strong link between subsidence, adjacent uplift and local earthquake swarms.

Tropical cyclones (TCs) are a hazard to life and property and a prominent element of the global climate system; therefore, understanding and predicting TC location, intensity, and frequency is of ...both societal and scientific significance. Methodologies exist to predict basinwide, seasonally aggregated TC activity months, seasons, and even years in advance. It is shown that a newly developed high-resolution global climate model can produce skillful forecasts of seasonal TC activity on spatial scales finer than basinwide, from months and seasons in advance of the TC season. The climate model used here is targeted at predicting regional climate and the statistics of weather extremes on seasonal to decadal time scales, and comprises high-resolution (50 km × 50 km) atmosphere and land components as well as more moderate-resolution (∼100 km) sea ice and ocean components. The simulation of TC climatology and interannual variations in this climate model is substantially improved by correcting systematic ocean biases through “flux adjustment.” A suite of 12-month duration retrospective forecasts is performed over the 1981–2012 period, after initializing the climate model to observationally constrained conditions at the start of each forecast period, using both the standard and flux-adjusted versions of the model. The standard and flux-adjusted forecasts exhibit equivalent skill at predicting Northern Hemisphere TC season sea surface temperature, but the flux-adjusted model exhibits substantially improved basinwide and regional TC activity forecasts, highlighting the role of systematic biases in limiting the quality of TC forecasts. These results suggest that dynamical forecasts of seasonally aggregated regional TC activity months in advance are feasible.
Tectonic tremor is characterized by persistent, low‐frequency seismic energy seen at major plate boundaries. Although predominantly associated with subduction zones, tremor also occurs along the deep ...extension of the strike‐slip San Andreas Fault. Here we present the first observations of tectonic tremor along New Zealand's Alpine Fault, a major transform boundary that is late in its earthquake cycle. We report tectonic tremor that occurred on the central section of the Alpine Fault on 12 days between March 2009 and October 2011. Tremor hypocenters concentrate in the lower crust at the downdip projection of the Alpine Fault; coincide with a zone of high P‐wave attenuation (low Qp) and bright seismic reflections; occur in the 25–45 km depth range, below the seismogenic zone; and may define the deep plate boundary structure extending through the lower crust and into the upper mantle. We infer this tremor to represent slow slip on the deep extent of the Alpine Fault in a fluid‐rich region marked by high attenuation and reflectivity. These observations provide the first indication of present‐day displacement on the lower crustal portion of the Australia–Pacific transform plate boundary.

Plate tectonics successfully describes the surface of Earth as a mosaic of moving lithospheric plates. But it is not clear what happens at the base of the plates, the lithosphere-asthenosphere ...boundary (LAB). The LAB has been well imaged with converted teleseismic waves, whose 10-40-kilometre wavelength controls the structural resolution. Here we use explosion-generated seismic waves (of about 0.5-kilometre wavelength) to form a high-resolution image for the base of an oceanic plate that is subducting beneath North Island, New Zealand. Our 80-kilometre-wide image is based on P-wave reflections and shows an approximately 15° dipping, abrupt, seismic wave-speed transition (less than 1 kilometre thick) at a depth of about 100 kilometres. The boundary is parallel to the top of the plate and seismic attributes indicate a P-wave speed decrease of at least 8 ± 3 per cent across it. A parallel reflection event approximately 10 kilometres deeper shows that the decrease in P-wave speed is confined to a channel at the base of the plate, which we interpret as a sheared zone of ponded partial melts or volatiles. This is independent, high-resolution evidence for a low-viscosity channel at the LAB that decouples plates from mantle flow beneath, and allows plate tectonics to work.
This review summarizes and synthesizes the significant amount of data which was generated on mercury (Hg) and persistent organic pollutants (POPs) in Canadian Arctic marine biota since the first ...Canadian Arctic Contaminants Assessment Report (CACAR) was published in 1997. This recent body of work has led to a better understanding of the current levels and spatial and temporal trends of contaminants in biota, including the marine food species that northern peoples traditionally consume. Compared to other circumpolar countries, concentrations of many organochlorines (OCs) in Canadian Arctic marine biota are generally lower than in the European Arctic and eastern Greenland but are higher than in Alaska, whereas Hg concentrations are substantially higher in Canada than elsewhere. Spatial coverage of OCs in ringed seals, beluga and seabirds remains a strength of the Arctic contaminant data set for Canada. Concentrations of OCs in marine mammals and seabirds remain fairly consistent across the Canadian Arctic although subtle differences from west to east and south to north are found in the proportions of various chemicals.
The most significant development since 1997 is improvement in the temporal trend data sets, thanks to the use of archived tissue samples from the 1970s and 1980s, long-term studies using archeological material, as well as the continuation of sampling. These data cover a range of species and chemicals and also include retrospective studies on new chemicals such as polybrominated diphenyl ethers. There is solid evidence in a few species (beluga, polar bear, blue mussels) that Hg at some locations has significantly increased from pre-industrial times to the present; however, the temporal trends of Hg over the past 20–30 years are inconsistent. Some animal populations exhibited significant increases in Hg whereas others did not. Therefore, it is currently not possible to determine if anthropogenic Hg is generally increasing in Canadian Arctic biota. It is also not yet possible to evaluate whether the recent Hg increases observed in some biota may be due solely to increased anthropogenic inputs or are in part the product of environmental change, e.g., climate warming. Concentrations of most “legacy” OCs (PCBs, DDT, etc.) significantly declined in Canadian Arctic biota from the 1970s to the late 1990s, and today are generally less than half the levels of the 1970s, particularly in seabirds and ringed seals. Chlorobenzenes and endosulfan were among the few OCs to show increases during this period while ∑HCH remained relatively constant in most species.
A suite of new-use chemicals previously unreported in Arctic biota (e.g., polybrominated diphenyl ethers (PBDEs), short chain chlorinated paraffins (SCCPs), polychlorinated naphthalenes (PCNs), perfluoro-octane sulfonic acid (PFOS) and perfluorocarboxylic acids (PFCAs)) has recently been found, but there is insufficient information to assess species differences, spatial patterns or food web dynamics for these compounds. Concentrations of these new chemicals are generally lower than legacy OCs, but there is concern because some are rapidly increasing in concentration (e.g., PBDEs), while others such as PFOS have unique toxicological properties, and some were not expected to be found in the Arctic because of their supposedly low potential for long-range transport. Continuing temporal monitoring of POPs and Hg in a variety of marine biota must be a priority.

To identify new susceptibility variants for bladder cancer in a meta-analysis of new and existing genome-wide genotype data.
Data from 32 studies that includes 13,790 bladder cancer cases and 343,502 controls of European ancestry were used for meta-analysis.
Log-additive associations of genetic variants were assessed using logistic regression models. A fixed-effects model was used for meta-analysis of the results. Stratified analyses were conducted to evaluate effect modification by sex and smoking status. A polygenic risk score (PRS) was generated on the basis of known and novel susceptibility variants and tested for interaction with smoking.
Multiple novel bladder cancer susceptibility loci (6p.22.3, 7q36.3, 8q21.13, 9p21.3, 10q22.1, 19q13.33) as well as improved signals in three known regions (4p16.3, 5p15.33, 11p15.5) were identified, bringing the number of independent markers at genome-wide significance (p < 5 × 10−8) to 24. The 4p16.3 (FGFR3/TACC3) locus was associated with a stronger risk for women than for men (p-interaction = 0.002). Bladder cancer risk was increased by interactions between smoking status and genetic variants at 8p22 (NAT2; multiplicative p value for interaction pM-I = 0.004), 8q21.13 (PAG1; pM-I = 0.01), and 9p21.3 (LOC107987026/MTAP/CDKN2A; pM-I = 0.02). The PRS based on the 24 independent GWAS markers (odds ratio per standard deviation increase 1.49, 95% confidence interval 1.44–1.53), which also showed comparable results in two prospective cohorts (UK Biobank, PLCO trial), revealed an approximately fourfold difference in the lifetime risk of bladder cancer according to the PRS (e.g., 1st vs 10th decile) for both smokers and nonsmokers.
We report novel loci associated with risk of bladder cancer that provide clues to its biological underpinnings. Using 24 independent markers, we constructed a PRS to stratify lifetime risk. The PRS combined with smoking history, and other established risk factors, has the potential to inform future screening efforts for bladder cancer.

An eastern Arctic marine food web was analyzed for perfluorooctanesulfonate (PFOS, C8F17SO3 -), perfluorooctanoate (PFOA, C7F15COO-), perfluorooctane sulfonamide (PFOSA, C8F17SO2NH2), and ...N-ethylperfluorooctane sulfonamide (N-EtPFOSA, C8F17SO2NHCH2CH3) to examine the extent of bioaccumulation. PFOS was detected in all species analyzed, and mean concentrations ranged from 0.28 ± 0.09 ng/g (arithmetic mean ± 1 standard error, wet wt, whole body) in clams (Mya truncata) to 20.2 ± 3.9 ng/g (wet wt, liver) in glaucous gulls (Larus hyperboreus). PFOA was detected in approximately 40% of the samples analyzed at concentrations generally smaller than those found for PFOS; the greatest concentrations were observed in zooplankton (2.6 ± 0.3 ng/g, wet wt). N-EtPFOSA was detected in all species except redfish with mean concentrations ranging from 0.39 ± 0.07 ng/g (wet wt) in mixed zooplankton to 92.8 ± 41.9 ng/g (wet wt) in Arctic cod (Boreogadus saida). This is the first report of N-EtPFOSA in Arctic biota. PFOSA was only detected in livers of beluga (Delphinapterus leucas) (20.9 ± 7.9 ng/g, wet wt) and narwhal (Monodon monoceros) (6.2 ± 2.3 ng/g, wet wt), suggesting that N-EtPFOSA and other PFOSA-type precursors are likely present but are being biotransformed to PFOSA. A positive linear relationship was found between PFOS concentrations (wet wt) and trophic level (TL), based on δ15N values, (r 2 = 0.51, p < 0.0001) resulting in a trophic magnification factor of 3.1. TL-corrected biomagnification factor estimates for PFOS ranged from 0.4 to 9. Both results indicate that PFOS biomagnifies in the Arctic marine food web when liver concentrations of PFOS are used for seabirds and marine mammals. However, transformation of N-EtPFOSA and PFOSA and potential other perfluorinated compounds to PFOS may contribute to PFOS levels in marine mammals and may inflate estimated biomagnification values. None of the other fluorinated compounds (N-EtPFOSA, PFOSA, and PFOA) were found to have a significant relationship with TL, but BMFTL values of these compounds were often >1, suggesting potential for these compounds to biomagnify. The presence of perfluorinated compounds in seabirds and mammals provides evidence that trophic transfer is an important exposure route of these chemicals to Arctic biota.
Epigenetic integrity is critical for many eukaryotic cellular processes. An important question is how different epigenetic regulators control development and influence disease. Lysine ...acetyltransferase 8 (KAT8) is critical for acetylation of histone H4 at lysine 16 (H4K16), an evolutionarily conserved epigenetic mark. It is unclear what roles KAT8 plays in cerebral development and human disease. Here, we report that cerebrum-specific knockout mice displayed cerebral hypoplasia in the neocortex and hippocampus, along with improper neural stem and progenitor cell (NSPC) development. Mutant cerebrocortical neuroepithelia exhibited faulty proliferation, aberrant neurogenesis, massive apoptosis, and scant H4K16 propionylation. Mutant NSPCs formed poor neurospheres, and pharmacological KAT8 inhibition abolished neurosphere formation. Moreover, we describe KAT8 variants in 9 patients with intellectual disability, seizures, autism, dysmorphisms, and other anomalies. The variants altered chromobarrel and catalytic domains of KAT8, thereby impairing nucleosomal H4K16 acetylation. Valproate was effective for treating epilepsy in at least 2 of the individuals. This study uncovers a critical role of KAT8 in cerebral and NSPC development, identifies 9 individuals with KAT8 variants, and links deficient H4K16 acylation directly to intellectual disability, epilepsy, and other developmental anomalies.
