Background: Ground-level concentrations of ozone (O₃) and fine particulate matter ≤ 2.5 μm in aerodynamic diameter (PM2.5) have increased since preindustrial times in urban and rural regions and are ...associated with cardiovascular and respiratory mortality. Objectives: We estimated the global burden of mortality due to O₃ and PM2.5 from anthropogenic emissions using global atmospheric chemical transport model simulations of preindustrial and present-day (2000) concentrations to derive exposure estimates. Methods: Attributable mortalities were estimated using health impact functions based on long-term relative risk estimates for O₃ and PM2.5 from the epidemiology literature. Using simulated concentrations rather than previous methods based on measurements allows the inclusion of rural areas where measurements are often unavailable and avoids making assumptions for background air pollution. Results: Anthropogenic O₃ was associated with an estimated 0.7 ± 0.3 million respiratory mortalities (6.3 ± 3.0 million years of life lost) annually. Anthropogenic PM2.5 was associated with 3.5 ± 0.9 million cardiopulmonary and 220,000 ± 80,000 lung cancer mortalities (30 ± 7.6 million years of life lost) annually. Mortality estimates were reduced approximately 30% when we assumed low-concentration thresholds of 33.3 ppb for O₃ and 5.8 μg/m³ for PM2.5. These estimates were sensitive to concentration thresholds and concentration—mortality relationships, often by > 50%. Conclusions: Anthropogenic O₃ and PM2.5 contribute substantially to global premature mortality. PM2.5 mortality estimates are about 50% higher than previous measurement-based estimates based on common assumptions, mainly because of methodologic differences. Specifically, we included rural populations, suggesting higher estimates; however, the coarse resolution of the global atmospheric model may underestimate urban PM2.5 exposures.
Dust storms and sandy dust events originating in arid and semi-arid areas can transport particulate material, pollutants, and potential transport long distances from their sources. Exposure to desert ...dust particles is generally acknowledged to endanger human health. However, most studies have examined anthropogenic particulate sources, with few studies considering contributions from natural desert dust. A systematic literature review was undertaken using the ISI Web of Knowledge and PubMed databases with the objective of identifying all studies presenting results on the potential health impact from desert dust particles across the world. This review reveals an imbalance between the areas most exposed to dust and the areas most studied in terms of health effects. Among the human health effects of dust storms are mortality and morbidity, arising from respiratory system, circulatory system, and other diseases. We summarize the quantitative results of current scientific health research and possible pathological mechanisms, and describe some of the many challenges related to understanding health effects from exposures to desert dust particles. Overall, for respiratory and circulatory mortality, both positive and negative associations have been reported for PM10 of desert dust, but only a positive relationship was reported between PM2.5–10 and mortality, and a positive relationship was also reported between PM2.5 and human mortality. Future pathological studies should continue to focus on those mechanisms causing the most harmful effect of desert dust on respiratory and cardiovascular diseases. More attention should also be paid to the association between desert dust and the morbidity of other diseases, such as those affecting the reproductive system and nervous system.
Sirtuin deacetylases and FOXO (Forkhead box, class O) transcription factors have important roles in many biological pathways, including cancer development. SIRT1 and SIRT2 deacetylate FOXO factors to ...regulate FOXO function. Because acetylation and ubiquitination both modify the ɛ-amino group of lysine residues, we investigated whether FOXO3 deacetylation by SIRT1 or SIRT2 facilitates FOXO3 ubiquitination and subsequent proteasomal degradation. We found that SIRT1 and SIRT2 promote FOXO3 poly-ubiquitination and degradation. Proteasome-inhibitor treatment prevented sirtuin-induced FOXO3 degradation, indicating that this process is proteasome dependent. In addition, we demonstrated that E3 ubiquitin ligase subunit Skp2 binds preferentially to deacetylated FOXO3. Overexpression of Skp2 caused poly-ubiquitination of FOXO3 and degradation, whereas knockdown of Skp2 increased the amount of FOXO3 protein. We also present evidence that SCF-Skp2 ubiquitinates FOXO3 directly in vitro. Furthermore, mutating four known acetylated lysine residues (K242, K259, K290 and K569) of FOXO3 into arginines to mimic deacetylated FOXO3 resulted in enhanced Skp2 binding but with inhibition of FOXO3 ubiquitination; this suggests that some or all of these four lysine residues are likely the sites for ubiquitination. In the livers of mice deficient in SIRT1, we detected increased expression of FOXO3, indicating SIRT1 regulates FOXO3 protein levels in vivo. Furthermore, we found that the elevation of SIRT1 and Skp2 expression in malignant PC3 and DU145 prostate cells is responsible for the downregulation of FOXO3 protein levels in these cells. Taken together, our data support the notion that deacetylation of FOXO3 by SIRT1 or SIRT2 facilitates Skp2-mediated FOXO3 poly-ubiquitination and proteasomal degradation.
Ampelopsis grossedentata, an indigenous plant in southern China, has been used for treating pharyngitis in traditional Chinese medicine for hundreds of years. In this study, we explored the ...anti-inflammatory activity of dihydromyricetin (1), its major bioactive component, and the underlying mechanism of this action. We demonstrated that 1 suppressed the levels of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) as well as increased the level of the anti-inflammatory cytokine interleukin-10 (IL-10) in lipopolysaccharide (LPS)-treated mice. Moreover, 1 was found to markedly inhibit the production of nitric oxide (NO) and the levels of TNF-α, IL-1β, and IL-6, whereas it increased the level of IL-10 in LPS-induced RAW 264.7 macrophage cells. Compound 1 also reduced the protein expression of inducible nitric oxide synthase (iNOS), TNF-α, and cyclooxygenase-2 (COX-2) in macrophage cells. Furthermore, 1 suppressed the phosphorylation of NF-kappa B (NF-κB) and IκBα as well as the phosphorylation of p38 and JNK but not ERK1/2 in LPS-stimulated macrophages. Taken together, the present results suggest that 1 exerts its topical anti-inflammatory action through suppressing the activation of NF-κB and the phosphorylation of p38 and JNK. Thus, 1 may be a potentially useful therapeutic agent for inflammatory-related diseases.
The welfare and health of laying hens in the multi-tier system raise concern in public. The flock distributions during feeding time at 51 and 89 weeks were studied in a multi-tier system. ...Furthermore, the ultra-high frequency radio frequency identification (UHF RFID) equipment was used to identify the transition between tiers and time spent in each tier of 48 focal hens (12 hens from each tier-group of the multi-tier system) at 92 weeks of age. The body weight, tibia size (length and width), body damage (comb and rear part) and feather condition (neck, breast, back, tail, cloaca and wings) of focal hens from different tier-groups were further compared. The results showed that the spatial distribution in flocks changed from top to bottom with increasing age. The hens at 51 weeks of age were mainly distributed in the 4th tier (19.6%±5.0% in 1st tier, 9.6%±1.1% in 2nd tier, 23.6%±2.9% in 3rd tier and 47.3%±2.6% in 4th tier), and hens at 89 weeks of age were mainly distributed in the lower tiers (33.5%±1.5% in 1st tier, 31.9%±5.1% in 2nd tier, 15.7%±3.4% in 3rd tier and 16.6%±3.1% in 4th tier). The spatial distribution of hens at 89 weeks of age was more even than that at 51weeks of age. At 92 weeks of age, the proportion of time spent in original tier of four tier-groups were 91.0%±5.7%, 51.9%±5.7%, 59.0%±7.0% and 63.0%±6.7%, respectively. Focal hens preferred to stay in the original tier and spent significantly less time in other tiers (P < 0.05). There was no significant difference in body weight, body damage score, tibia width and the partial feather scores (neck, breast, tail and cloaca) of focal hens among four tier-groups (P > 0.05). However, focal hens from 1st tier had worse feather scores on wings and back, and shorter tibia length compared to other tiers suggesting that there were more lower ranking birds that located in lower tier to avoid competition, but had equal access to resource, which is good for their welfare and health. In summary, the overcrowding situation was improved near the end of the laying cycle in the multi-tier system, thereby mitigating the potential negative effects to the lower ranking hens and maintain a satisfactory level of welfare and health for laying hens near the end of the laying cycle.
Mineral particles or particulate matters(PMs) emitted during agricultural activities are major recurring sources of atmospheric aerosol loading.However,precise PM inventory from agricultural tillage ...and harvest in agricultural regions is challenged by infrequent local emission factor(EF) measurements.To understand PM emissions from these practices in northeastern China,we measured EFs of PM_(10) and PM_(2.5) from three field operations(i.e.,tilling,planting and harvesting) in major crop production(i.e.,corn and soybean),using portable real-time PM analyzers and weather station data.County-level PM_(10) and PM_(2.5) emissions from agricultural tillage and harvest were estimated,based on local EFs,crop areas and crop calendars.The EFs averaged(107 ± 27),(17 ± 5) and 26 mg/m~2 for field tilling,planting and harvesting under relatively dry conditions(i.e.,soil moisture 〈15%),respectively.The EFs of PM from field tillage and planting operations were negatively affected by topsoil moisture.The magnitude of PM_(10) and PM_(2.5) emissions from these three activities were estimated to be 35.1 and 9.8 kilotons/yr in northeastern China,respectively,of which Heilongjiang Province accounted for approximately45%.Spatiotemporal distribution showed that most PM_(10) emission occurred in April,May and October and were concentrated in the central regions of the northeastern plain,which is dominated by dryland crops.Further work is needed to estimate the contribution of agricultural dust emissions to regional air quality in northeastern China.
Abstract
We present an unprecedented effort to map anthropogenic emissions of air pollutants at 1 km spatial resolution in the contiguous United States (CONUS). This new dataset, Neighborhood ...Emission Mapping Operation (NEMO), is produced at hourly intervals based on the United States Environmental Protection Agency (US EPA) National Emission Inventories 2017. Fine-scale spatial allocation was achieved through distributing the emission sources using 108 spatial surrogates, factors representing the portion of a source in each 1 km grid. Gaseous and particulate pollutants are speciated into model species for the Carbon Bond 6 chemical mechanism. All sources are grouped in 9 sectors and stored in NetCDF format for air quality models, and in shapefile format for GIS users and air quality managers. This dataset shows good consistency with the USEPA benchmark dataset, with a monthly difference in emissions less than 0.03% for any sector. NEMO provides the first 1 km mapping of air pollution over the CONUS, enabling new applications such as fine-scale air quality modeling, air pollution exposure assessment, and environmental justice studies.
