Sub-Saharan West Africa is a vulnerable region where a better quantification and understanding of the impact of climate change on crop yields is urgently needed. Here, we have applied the ...process-based crop model SARRA-H calibrated and validated over multi-year field trials and surveys at eight contrasting sites in terms of climate and agricultural practices in Senegal, Mali, Burkina Faso and Niger. The model gives a reasonable correlation with observed yields of sorghum and millet under a range of cultivars and traditional crop management practices. We applied the model to more than 7000 simulations of yields of sorghum and millet for 35 stations across West Africa and under very different future climate conditions. We took into account 35 possible climate scenarios by combining precipitation anomalies from −20% to 20% and temperature anomalies from +0 to +6 °C. We found that most of the 35 scenarios (31/35) showed a negative impact on yields, up to −41% for +6 °C − 20% rainfall. Moreover, the potential future climate impacts on yields are very different from those recorded in the recent past. This is because of the increasingly adverse role of higher temperatures in reducing crop yields, irrespective of rainfall changes. When warming exceeds +2 °C, negative impacts caused by temperature rise cannot be counteracted by any rainfall change. The probability of a yield reduction appears to be greater in the Sudanian region (southern Senegal, Mali, Burkina Faso, northern Togo and Benin), because of an exacerbated sensitivity to temperature changes compared to the Sahelian region (Niger, Mali, northern parts of Senegal and Burkina Faso), where crop yields are more sensitive to rainfall change. Finally, our simulations show that the photoperiod-sensitive traditional cultivars of millet and sorghum used by local farmers for centuries seem more resilient to future climate conditions than modern cultivars bred for their high yield potential (−28% versus −40% for the +4 °C − 20% scenario). Photoperiod-sensitive cultivars counteract the effect of temperature increase on shortening cultivar duration and thus would likely avoid the need to shift to cultivars with a greater thermal time requirement. However, given the large difference in mean yields of the modern versus traditional varieties, the modern varieties would still yield more under optimal fertility conditions in a warmer world, even if they are more affected by climate change.
Production and wild animals are major sources of human salmonellosis and animals raised for food also play an important role in transmission of antimicrobial resistant Salmonella strains to humans. ...Furthermore, in sub-Saharan Africa non-typhoidal Salmonella serotypes are common bloodstream isolates in febrile patients. Yet, little is known about the environmental reservoirs and predominant modes of transmission of these pathogens. The purpose of this study was to discover potential sources and distribution vehicles of Salmonella by isolating strains from apparently healthy slaughtered food animals and wild hedgehogs and by determining the genetic relatedness between the strains and human isolates. For this purpose, 729 feces samples from apparently healthy slaughtered cattle (n = 304), poultry (n = 350), swine (n = 50) and hedgehogs (n = 25) were examined for the presence of Salmonella enterica in Burkina Faso. The isolates were characterized by serotyping, antimicrobial-susceptibility testing, phage typing, and pulsed-field gel electrophoresis (PFGE) with XbaI and BlnI restriction enzymes.
Of the 729 feces samples, 383 (53%) contained Salmonella, representing a total of 81 different serotypes. Salmonella was present in 52% of the cattle, 55% of the poultry, 16% of the swine and 96% of the hedgehog feces samples. Antimicrobial resistance was detected in 14% of the isolates. S. Typhimurium isolates from poultry and humans (obtained from a previous study) were multiresistant to the same antimicrobials (ampicillin, chloramphenicol, streptomycin, sulfonamides and trimethoprim), had the same phage type DT 56 and were closely related in PFGE. S. Muenster isolates from hedgehogs had similar PFGE patterns as the domestic animals.
Based on our results it seems that production and wild animals can share the same Salmonella serotypes and potentially transmit some of them to humans. As the humans and animals often live in close vicinity in Africa and the hygiene control of the meat retail chain is defective, high Salmonella carriage rates of the animals can pose a major public health risk in Burkina Faso. This underlines the necessity for a joint and coordinated surveillance and monitoring programs for salmonellosis in Africa.

The gaseous atmosphere plays a major role in the quality of the manufactured parts in Laser Powder Bed Fusion (L-PBF) by protecting the metal from high temperature oxidation. If argon and nitrogen ...are the most commonly used gases, helium has almost never been considered as a possible candidate as a chemically inert shielding gas. To provide a better understanding of the influence of the gas atmosphere on the process stability, a comparative study of L-PBF manufacturing under argon and helium atmospheres has been carried out, considering a nickel-based alloy Inconel® 625 and a single bead configuration. To this end, in-situ process measurements were carried out on a dedicated experimental setup. The melt pool behaviour, the expansion of the vapour plume and the amount of spatters were evaluated with high-speed imaging for the two gases considered, together with the final L-PBF bead dimensions. Results were also compared to single fusion beads carried out in an industrial L-PBF machine for a comparable range of volume energy densities. The influence of the shielding atmosphere on L-PBF single beads was as follows: (1) dimensions of beads were shown to be constant whatever the gas; (2) fewer and smaller spatters were produced under helium atmosphere, especially for high volume energy densities. Physical mechanisms were then discussed to understand those specific effects.
Background. In 2003, Mali introduced intermittent preventive therapy in pregnancy (ITPp) with sulfadoxine-pyrimethamine (SP) for the control of malaria in pregnancy, consisting of 2 doses of SP given ...in the 2nd and 3rd trimester. This widely used regimen, although very effective, leaves many women unprotected from malaria during the last 4-to-8 weeks of gestation, which is a pivotal period for fetal weight gain. The aim of the study was to compare the efficacy and safety of 3-dose versus 2-dose IPTp-SP for the prevention of placental malaria and associated low birth weight (LBW). Methods. We conducted a parallel-group, open-label, individually randomized controlled superiority trial involving 814 women of all gravidity, enrolled from April 2006 through March 2008. All women were seen at least 3 times and received either 2 (n = 401) or 3 (n = 413) doses of IPTp-SP. The primary endpoint measured was placental malaria, LBW, preterm births, and maternal anemia were secondary endpoints, and severe maternal skin reactions and neonatal jaundice were safety endpoints. Results. Among the 96% of study subjects who were followed up until delivery, the prevalence of placental malaria was 2-fold lower in the 3-dose group (8.0%) than in the 2-dose group (16.7%); the adjusted prevalence ratio (APR) was 0.48 (95% confidence interval CI, 0.32—0.71). LBW and preterm births were also reduced; the prevalence of LBW was 6.6% in the 3-dose group versus 13.3% in the 2-dose group (APR, 0.50; 95% CI, 0.32—0.79), and the prevalence of preterm births was 3.2% versus 8.9% (APR, 0.37; 95% CI, 0.19—0.71). No significant reductions in maternal anemia or differences in safety endpoints were observed. Conclusions. Adding a third dose of ITPp-SP halved the risk of placental malaria, LBW, and preterm births in all gravidae, compared with the standard 2-dose regimen, in this area of highly seasonal transmission with low levels of SP resistance. Methanogenic Archaea are considered as extremely oxygen-sensitive organisms, and their culture is fastidious, requiring specific equipment. We report here conditions allowing the cultivation of ...Methanobrevibacter smithii in an anaerobic chamber without the addition of hydrogen.
We first enriched the stool sample in an anaerobic liquid medium. To cultivate M. smithii with Bacteroides thetaiotaomicron and other hydrogen-producing bacteria on solid medium in an anaerobic chamber, we divided the agar plates into two compartments and seeded each strain on each compartment. Methane production was assessed by gas chromatography, and the growing colonies were authenticated by MALDI-TOF MS.
We successfully cultured M. smithii from a liquid culture medium inoculated with stool collected from a healthy donor in an anaerobic chamber. The isolation in pure culture permitted successful culture on agar medium by our performing a co-culture with B. thetaiotaomicron. We also successfully tested the co-cultivation of M. smithii with other known hydrogen-producing bacteria. Gas chromatographic tests showed that these strains produced hydrogen in different amounts. Agar colonies of methanogens were obtained by co-culture with these bacteria, and methane production was detected.
We propose a new approach to isolate and cultivate new strains of M. smithii by using a co-culture–based technique that can facilitate and make available the isolation of new methanogenic Archaea strains in clinical microbiology laboratories.
Control of gambiense sleeping sickness, a neglected tropical disease targeted for elimination by 2020, relies mainly on mass screening of populations at risk and treatment of cases. This strategy is ...however challenged by the existence of undetected reservoirs of parasites that contribute to the maintenance of transmission. In this study, performed in the Boffa disease focus of Guinea, we evaluated the value of adding vector control to medical surveys and measured its impact on disease burden.
The focus was divided into two parts (screen and treat in the western part; screen and treat plus vector control in the eastern part) separated by the Rio Pongo river. Population census and baseline entomological data were collected from the entire focus at the beginning of the study and insecticide impregnated targets were deployed on the eastern bank only. Medical surveys were performed in both areas in 2012 and 2013.
In the vector control area, there was an 80% decrease in tsetse density, resulting in a significant decrease of human tsetse contacts, and a decrease of disease prevalence (from 0.3% to 0.1%; p=0.01), and an almost nil incidence of new infections (<0.1%). In contrast, incidence was 10 times higher in the area without vector control (>1%, p<0.0001) with a disease prevalence increasing slightly (from 0.5 to 0.7%, p=0.34).
Combining medical and vector control was decisive in reducing T. b. gambiense transmission and in speeding up progress towards elimination. Similar strategies could be applied in other foci.
Mosquitoes in the Anopheles gambiae complex show rapid ecological and behavioral diversification, traits that promote malaria transmission and complicate vector control efforts. A high-density, ...genome-wide mosquito SNP-genotyping array allowed mapping of genomic differentiation between populations and species that exhibit varying levels of reproductive isolation. Regions near centromeres or within polymorphic inversions exhibited the greatest genetic divergence, but divergence was also observed elsewhere in the genomes. Signals of natural selection within populations were overrepresented among genomic regions that are differentiated between populations, implying that differentiation is often driven by population-specific selective events. Complex genomic differentiation among speciating vector mosquito populations implies that tools for genome-wide monitoring of population structure will prove useful for the advancement of malaria eradication.
Hypertension is an important public health challenge in low- and middle-income countries, and in many African countries including Guinea medicinal plants are still widely used for its treatment.
The ...objective of this study was to determine the prevalence of hypertension in two Guinean urban districts (Pounthioun and Dowsare), to describe its management and to collect information on traditional herbal remedies. A total of 316 participants entered the study, 28.2% (89/316) men and 71.8% (227/316) women. Of these, 181 were from Dowsare (50 men and 131 women) and 135 from Pounthioun (39 men and 96 women). The mean age of subjects was 40.8 ± 14.0 years (range18 − 88years), while the majority of subjects (63.3% or 200/316) were 45–74 years old.
The overall prevalence of hypertension was 44.9% (142/316): 46.4% (84/181) from Dowsare and 43.0% (58/135) from Pounthioun. Ethnobotanical investigations among hypertensive patients led to the collection of 15 plant species, among which Hymenocardia acida leaves and Uapaca togoensis stem bark were the most cited. Phytochemical investigation of these two plant species led to the isolation and identification of isovitexin and isoorientin from H. acida, and betulinic acid and lupeol from U. togoensis.
The presence of these constituents in Hymenocardia acida leaves and Uapaca togoensis stem bark may at least in part support their traditional use against hypertension in Guinea.
