Low-cost adsorbents, e.g., cow manure-based carbon, provide alternatives to remove veterinary antibiotic sulfamethazine (SMT) from contaminated water bodies. In this study, the chemical structures ...and compositions of cow manure (CM) carbonized at 400, 600, and 800 °C (CM400, CM600, and CM800) were examined using elemental analyzer (EA), Brunauer–Emmett–Teller, and spectroscopic techniques. Adsorptions of SMT on CM samples were conducted as functions of pH, hydrophobicity, and ionic strengths. Results of EA and spectroscopic analyses suggested that the raw CM and CM400 samples contained the highest amounts of O-containing groups and aliphatic domains. Amounts of such two chemical groups decreased as carbonization temperatures increased. The specific surface areas and total pore volumes of CM samples increased significantly when the carbonization temperatures exceeded 600 °C. SMT adsorption on CM samples could be described essentially by the pseudo-second-order kinetic, intra-particle diffusion, and Freundlich isotherm models. EDA) was formed between SMT and CM surfaces enriched with hydrophobic domains. Further, the high adsorption affinity of SMT to the CM600 and CM800 samples was attributed in part to their larger surface areas and total pore volumes. Generally, CM-based materials carbonized >600 °C showed relatively stable structures and exhibited strong aromatic properties. Moreover, maximum adsorption capacities of SMT on the CM800 sample (37–39 mM/kg) were significantly higher than those of other common adsorbents (0.02–35.93 mM/kg).
To maximize spore production by Bacillus amyloliquefaciens B128, and its antagonism to the fungal pathogen Botrytis elliptica B061. In the 5-l stirred-tank bioreactor (STR), with the 0·5 vvm aeration ...rate, an agitation rate of 200 rev min⁻¹ significantly enhanced the spore yield compared to the same in 300 rev min⁻¹ cultivations. In a 20-l airlift bioreactor (ALR) the maximal spore production was further increased with a controlled aeration rate of 2·5 vvm operated in a 24-mesh net-draft tube mode, and no pH control cultivation. This spore yield in the 20-l ALR was five- and eightfold higher; in addition the cultivation period was 19 h shorter, compared to that obtained from shaker flask and in the 5-l STR cultivations respectively. Although culture conditions are still to be optimized, by using an ALR with net-draft tube, a scaling up from shaker flasks and STR to ALR of spore production by the strain B128 is technically feasible. The spore yields obtained using bioreactors were much higher than those previously reported. The freshly produced spore preparations from the B128 strain significantly antagonized the grey mould pathogen B. elliptica.

Diabetes has been a long-standing disorder and its management has been challenging various medical and research experts for several decades because of its complex causative factors and ...pathophysiological processes leading to complications. Medicinal plants have been explored in several countries and traditional Chinese medicine is one of the well-recognized alternative treatment methods. In this study, we have chosen some of the underexplored plant parts of Chinese medicinal herbs and analyzed their antioxidant activity and ability to modulate the expression of fibronectin during high glucose conditions. Extraction of the plant materials with different solvent led to 17 extracts and among which, 3 extracts (2, 12 & 17) were observed to render more than 50 μg/ml vitamin C equivalents of DPPH free radical scavenging ability and 2 of them (2 & 17) showed more than 25 μg/ml of vitamin C equivalents of ferric ion reducing power. Based on the antioxidant activity and comparison of their total phenolic content, we used extracts 2 & 17 to check their effect on fibronectin expression in MES-13 cells under high sugar conditions. We observed that both extracts showed a significant reduction of fibronectin expression compared to untreated cells with high glucose levels. The expression was much lesser than the normal untreated, normal sugar supplemented cells and this was not observed in vitamin C supplemented cells. In conclusion, crude extracts containing a group of phenolic compounds have shown significant effects against fibronectin expression leading to reduced ECM deposition and tissue fibrosis. Further exploration might provide insights into the exact mechanism and checkpoints of the extract that can successfully reduce diabetes-induced renal complications.
Antrodia cinnamomea is a valuable fungus. The 4-acetylantroquinonol B (4-AAQB) contained in it has strong anticancer activity. In this study, five kinds of herbs, burdock, wolfberry, coix seed, ...hawthorn, and tangerine peel were selected and processed into powders, aqueous extracts, and ethanolic extracts to investigate the effects on the production of 4-AAQB by A. cinnamomea. A combination strategy was conducted by adding burdock and tangerine peel aqueous extract. By this means, the production of 4-AAQB was improved to 54.5 mg/L, which was approximately 33-fold higher than the control. After analysis of components of the tangerine peel aqueous extract and the addition of the major components, vanillic acid, protocatechuic acid, and their analogs played an important role in the synthesis of 4-AAQB. It was demonstrated that the addition of Chinese herbs facilitated both cell growth of A. cinnamomea and 4-AAQB production, providing a feasible way to increase the yield of 4-AAQB.
The lasing peak of λ=1090nm at T=110K was obtained for a microdisk cavity with three layers of InGaAs quantum dots integrated on AlAs/GaAs distributed Bragg reflector. The refractive indices and ...reflectance spectrum of AlAs/GaAs DBR at T=110K were derived and estimated. The lasing mode was subsequently confirmed as the whispering gallery mode with azimuthal mode number m=13 and could be explained as the valid of bottom DBR mirror.

+)-induced neurotoxicity was studied in SH-SY5Y neuroblastoma cells. In contrast to the cytotoxicity of 0.1% oxygen, 1% oxygen hypoxia for 24
h did not cause significant cell death. A transient increase in LC3-II level, a biomarker of autophagy, was demonstrated during hypoxic treatment. At the same time, 8-h hypoxia increased fluorescence due to monodansylcadaverine, a specific dye for autophagosomes, in the treated cells. Co-incubation with bafilomycin A1 (10 nM) further increased hypoxia-induced LC3-II levels but 3-methyladenine (3-MA; 10
mM) reduced the elevation in LC3-II levels induced by 8-h hypoxia. Moreover, 8-h hypoxia increased free radical formation and nuclear HIF-1α level. Glutathione was found to diminish hypoxia-induced LC3-II elevation. In contrast to the elevated LC3-II level, 8-h hypoxia significantly decreased mitochondrial mass. Furthermore, a rebound elevation in mitochondrial mass was observed under 8-h hypoxia and subsequent 12-h normoxia. Prior hypoxia attenuated the MPP
+-induced elevation in LC3-II levels and cell death. Moreover, hypoxic pretreatment inhibited MPP
+-induced activation of caspase-3 and DNA fragmentation. Co-incubation with 3-MA during hypoxia prevented the protection afforded by hypoxic preconditioning against MPP
+-induced increases in LC3-II levels and neurotoxicity. Taken together, our results suggest that sublethal hypoxia induces autophagy that is mediated by oxidative stress. Furthermore, autophagy may be involved in the protection provided by hypoxic preconditioning against MPP
+-induced neurotoxicity, indicating a neuroprotective role of autophagy in hypoxic preconditioning.

B. subtilis Y-108 thus isolated can be used for deproteinization of crustacean ...wastes in the preparation of chitin. For deproteinization tests, liquid phase fermentation of untreated shrimp shell, crab shell, and lobster shell wastes with this microbe showed protein removal of 88, 67, and 83%, respectively. In contrast, the protein removal of the acid treated wastes was 76, 62, 56%, respectively. B. subtilis Y-108 attained the highest activity. It was as high as 20.2 U/ml. The protease was purified in a three-step procedure involving ammonium sulfate precipitation, DEAE-Sepharose CL-6B ionic exchange chromatography, and Sephacryl S-200 gel permeation chromatography. The enzyme was shown to have a relative molecular weight of 44 kDa by SDS polyacrylamide gel electrophoresis. The protease was most active at pH 8.0 and 50°C with casein as substrate. +2, but inhibited completely by Hg
+2. The protease was also inhibited by metal-chelating agent such as EDTA, sulfhydryl reagents as β-mercaptoethanol, and by cysteine hydrochloride, histidine, glycerol. The EDTA was the most effective inhibitor that caused complete inhibition of protease. It was concluded that this enzyme is a metal-chelator-sensitive neutral protease.

The results indicate that the positive exchange bias is from antiferromagnetic interface moments perpendicular to their ferromagnetic counterparts. With field cooling the alignment is put into a parallel configuration resulting in a negative exchange bias.
Abstract Study question Is cervical secretion gene methylation profile associated with embryo implantation outcome, and can be used as a non-invasive biomarker for pregnancy prediction in FET cycles? ...Summary answer Cervical secretion gene methylation profiles show significant associations with implantation outcomes, and can serve as a non-invasive predictor of successful pregnancy in FET cycles. What is known already Cause of implantation failure remains a black box in reproductive medicine, and the exact mechanism of how endometrial receptivity is still behind the curtain. Epigenetic modifications including DNA methylation regulate gene expression and may alter endometrium receptivity. Endometrial DNA methylation changes vigorously only during the reconstruction of a new endometrium throughout each menstrual cycle, suggesting a unique methylation landscape in each regenerated endometrium. Cervical secretions contain implantation-related cytokines and genetic material from endometrium, and the DNA methylation profiles of cervical secretions is highly correlated with endometrium. Therefore, cervical secretions can be used as non-invasive surrogates for investigating endometrial condition. Study design, size, duration This multi-centered, case-control study enrolled 72 infertile females who entered the FET cycle with at least one high-quality blastocyst available between Aug 2017 and Dec 2022. Embryos assessed by Gardner scoring system with grade 3BB or above were defined as high-quality. Cervical secretions were collected on the day right before embryo transfer, and then DNA methylation profile were assessed. Participants/materials, setting, methods These females aged between 20-45 years-old with regular menstruation cycle (24-35 days) and normal BMI (17.5–32.0 kg/m2). Uterine cavity lesions and hydrosalpinx without treatment, adenomyosis, large uterine myoma that distorted endometrium, and thin endometrium (<7mm) were excluded. The association between the cervical secretion gene methylation profile and pregnancy outcome was evaluated via quantitative methylation-specific PCR (qMSP), methylation array, and genome-wide analysis. Different machine learning algorithms were utilized for differential methylation analysis and implantation prediction. Main results and the role of chance Cervical secretion DNA methylome differed between receptive (pregnant) and non-receptive (non-pregnant) FET cycle with genome-wide methylation array. The accuracy of pregnancy outcome prediction using top 2000 differential methylation probes (DMPs) in promoter region can be as high as 86% in all tested sample and 96.4% in pregnant samples. Bioinformatic analysis of six selected candidate genes related to implantation from literature review showed significant differences in DNA methylation between receptive and non-receptive endometrium. The methylation level of the six candidate genes with eight regions were measured by the qMSP platform and showed varying degrees of differential methylation status between the pregnancy and non-pregnancy groups. The AUCs for a single gene/region ranged from 0.53–0.67 for implantation prediction. Ten different machine learning models were applied and the results showed that the combination of candidate genes’ DNA methylation profiles could differentiate pregnant from non-pregnant samples with an accuracy as high as 86.67% and an AUC of 0.81 by logistic regression model. The best model from different machine learning algorithms using selected DMPs of genes related to window of implantation (WOI) can predict ongoing pregnancy with accuracy rates as high as 86.78% and AUC values up to 0.91. Limitations, reasons for caution The results obtained from a relatively small cohort size, and the timing of sample collection is on the day right before embryo transfer. Further investigation of methylation profiles in late follicular phase along with large sample size with euploid embryo transfer are mandatory for validation and clinical applications. Wider implications of the findings We firstly demonstrated the feasibility of cervical secretion methylation profile as a non-invasive testing for pregnancy prediction. The promising result may contribute to predicting endometrial receptivity for cycles with a favorable endometrium and maximize the successful pregnancy rate of embryo transfer. The antioxidant (DPPH radical and superoxide anion scavenging activities), and cytotoxic (in tumor, Jurkat, PC-3, Colon 205, HepG2, and normal PBMCs cells) activities of 16 plant phenolic or related ...compounds were evaluated in vitro. Different categories compounds corresponding to 10 flavonoids, three lignans, two phenolic acids, and a catechin showed significant mean differences in antioxidant and cytotoxic activities. Particularly, the flavonols, quercetin (
3) and tiliroside (
11) possess significant antioxidant activity, as well as cytotoxic activity against Jurkat; and Jurkat and HepG2 cells, respectively. In contrast, the flavanone, 5,7-dimethoxy-3′,4′-methylenedioxyflavanone (
7), and homoisoflavonoid, isobonducellin (
10) shown to have no significant antioxidant activity, but exhibited potent cytotoxic activity in Jurkat and HepG2 cells, while moderate growth inhibition against Colon205 cells. Interestingly, none of these derivatives shown to have toxicity toward normal peripheral blood mononuclear cells, over the concentration range tested (5–200
μM). Cytotoxic activities of some natural flavonoids identified in the medicinal plants were evaluated for the first time.
