Adipose-derived mesenchymal stem cells (ADSCs) are promising candidate for regenerative medicine to repair non-healing bone defects due to their high and easy availability. However, the limited ...osteogenic differentiation potential greatly hinders the clinical application of ADSCs in bone repair. Accumulating evidences demonstrate that circular RNAs (circRNAs) are involved in stem/progenitor cell fate determination, but their specific role in stem/progenitor cell osteogenesis, remains mostly undescribed. Here, we show that circRNA-vgll3 originating from the vgll3 locus markedly enhances osteogenic differentiation of ADSCs; nevertheless, silencing of circRNA-vgll3 dramatically attenuates ADSC osteogenesis. Furthermore, we validate that circRNA-vgll3 functions in ADSC osteogenesis through a circRNA-vgll3/miR-326-5p/integrin α5 (Itga5) pathway. Itga5 promotes ADSC osteogenic differentiation and miR-326-5p suppresses Itga5 translation. CircRNA-vgll3 directly sequesters miR-326-5p in the cytoplasm and inhibits its activity to promote osteogenic differentiation. Moreover, the therapeutic potential of circRNA-vgll3-modified ADSCs with calcium phosphate cement (CPC) scaffolds was systematically evaluated in a critical-sized defect model in rats. Our results demonstrate that circRNA-vgll3 markedly enhances new bone formation with upregulated bone mineral density, bone volume/tissue volume, trabeculae number, and increased new bone generation. This study reveals the important role of circRNA-vgll3 during new bone biogenesis. Thus, circRNA-vgll3 engineered ADSCs may be effective potential therapeutic targets for bone regenerative medicine.
A facile strategy affording high-quality single-crystalline MFI-type nanozeolites (10–55 nm) with hexagonal prism morphology, good monodispersity, high crystallinity, and high product yield (above ...97%) has been developed. This is achieved by synergistically using an l-lysine-assisted approach and a two-step crystallization process in a concentrated gel system (H2O/Si = 9). The morphological evolution of nanosized silicalite-1 is monitored by high-resolution transmission electron microscopy (HRTEM). In this process, metastable irregular nanoparticles are initially obtained at 80 °C as the first step. Consequently, a rearrangement in morphology toward equilibrium crystal shape and without excessive growth for the metastable nanoparticles occurs at 170 °C as the second step. Throughout the whole process, l-lysine acts as an inhibitor to effectively limit the crystal growth of zeolites. Thanks to the high-quality nanosized crystals, the as-prepared ZSM-5 catalysts exhibit superior performance in methanol-to-propylene (MTP) reactions, which deliver a prolonged lifetime of 54 h with a total light olefin selectivity of 74% and a high propylene selectivity of 49% at 470 °C at a high methanol weight hourly space velocity (WHSV) of 7.2 h–1. This synthetic route provides a general strategy for preparing other types of zeolites with good monodispersity, nanosize, high yield, and high crystallinity.
The relationship between dietary glycemic index, glycemic load and risk of coronary heart disease (CHD), stroke, and stroke-related mortality is inconsistent.
We systematically searched the MEDLINE, ...EMBASE, and Science Citation Index Expanded databases using glycemic index, glycemic load, and cardiovascular disease and reference lists of retrieved articles up to April 30, 2012. We included prospective studies with glycemic index and glycemic load as the exposure and incidence of fatal and nonfatal CHD, stroke, and stroke-related mortality as the outcome variable. Pooled relative risks (RR) and 95% confidence intervals (CI) were calculated using random-effects models.
Fifteen prospective studies with a total of 438,073 participants and 9,424 CHD cases, 2,123 stroke cases, and 342 deaths from stroke were included in the meta-analysis. Gender significantly modified the effects of glycemic index and glycemic load on CHD risk, and high glycemic load level was associated with higher risk of CHD in women (RR=1.49, 95%CI 1.27-1.73), but not in men (RR=1.08, 95%CI 0.91-1.27). Stratified meta-analysis by body mass index indicated that among overweight and obese subjects, dietary glycemic load level were associated with increased risk of CHD (RR=1.49, 95%CI 1.27-1.76; P for interaction=0.003). Higher dietary glycemic load, but not glycemic index, was positively associated with stroke (RR=1.19, 95% CI 1.00-1.43). There is a linear dose-response relationship between dietary glycemic load and increased risk of CHD, with pooled RR of 1.05 (95%CI 1.02-1.08) per 50-unit increment in glycemic load level.
High dietary glycemic load is associated with a higher risk of CHD and stroke, and there is a linear dose-response relationship between glycemic load and CHD risk. Dietary glycemic index is slightly associated with risk of CHD, but not with stroke and stroke-related death. Further studies are needed to verify the effects of gender and body weight on cardiovascular diseases.
Identification and monitoring of unstable slopes across wide regions using Synthetic Aperture Radar Interferometry (InSAR) can further help to prevent and mitigate geological hazards. However, the ...low spatial density of measurement points (MPs) extracted using the traditional time-series InSAR method in topographically complex mountains and vegetation-covered slopes makes the final result unreliable. In this study, a method of time-series InSAR analysis using single- and multi-look phases were adopted to solve this problem, which exploited single- and multi-look phases to increase the number of MPs in the natural environment. Archived ascending and descending Sentinel-1 datasets covering Zhouqu County were processed. The results revealed that nine landslides could be quickly identified from the average phase rate maps using the Stacking method. Then, the time-series InSAR analysis with single- and multi-look phases could be used to effectively monitor the deformation of these landslides and to quantitatively analyze the magnitude and dynamic evolution of the deformation in various parts of the landslides. The reliability of the InSAR results was further verified by field investigations and Unmanned Aerial Vehicle (UAV) surveys. In addition, the precursory movements and causative factors of the recent Yahuokou landslide were analyzed in detail, and the application of the time-series InSAR method in landslide investigations was discussed and summarized. Therefore, this study has practical significance for early warning of landslides and risk mitigation.
The number of studies using third-generation sequencing utilising Pacific Biosciences (PacBio) and Oxford Nanopore Technologies (ONT) is rapidly increasing in many different research areas. Among ...them, plant full-length single-molecule transcriptome studies have mostly used PacBio sequencing, whereas ONT is rarely used. Therefore, in this study, we examined ONT RNA sequencing methods in plants. We performed a detailed evaluation of reads from PacBio, Nanopore direct cDNA (ONT Dc), and Nanopore PCR cDNA (ONT Pc) sequencing including characteristics of raw data and identification of transcripts. In addition, matched Illumina data were generated for comparison.
ONT Pc showed overall better raw data quality, whereas PacBio generated longer read lengths. In the transcriptome analysis, PacBio and ONT Pc performed similarly in transcript identification, simple sequence repeat analysis, and long non-coding RNA prediction. PacBio was superior in identifying alternative splicing events, whereas ONT Pc could estimate transcript expression levels.
This paper made a comprehensive comparison of PacBio and nanopore-based RNA sequencing of the
transcriptome, the results indicate that ONT Pc is more cost-effective for generating extremely long reads and can characterise the transcriptome as well as quantify transcript expression. Therefore, ONT Pc is a new cost-effective and worthwhile method for full-length single-molecule transcriptome analysis in plants.
The severity of global climate change is exemplified by the significant increase in extreme precipitation events, leading to an urgent need for accurate rainfall prediction models to mitigate flood ...disasters that adversely affect economic and social development. With the rapid progress of machine learning in the big data era, novel solutions to regression problems are being proposed. In this paper, we try to construct and evaluate different rainfall prediction models based on specific humidity, relative humidity, horizontal and vertical water vapor flux, and lifting index as variables, using four classic machine learning algorithms: linear regression, random forest regression, support vector regression, and Bayesian ridge regression. The grid search method is employed for hyperparameter tuning, significantly improving the models' prediction accuracy and generalization ability. Evaluation of the predictive performance of the models on nine typical regions in China, including Zhengzhou, Beijing, and Chengdu, demonstrates that the random forest regression model has the highest predictive accuracy, with an average fitting degree of 0.8 or above, followed by support vector regression and Bayesian ridge regression models. Conversely, the linear regression model may have the poorest predictive performance. Therefore, the random forest regression model is recommended for future precipitation prediction, providing a valuable solution to various regression problems. The appropriate selection of variables for prediction and grid search for hyperparameter tuning are possibly the highlights of this paper.
Homeostasis of bone metabolism is regulated by the central nervous system, and mood disorders such as anxiety are associated with bone metabolism abnormalities, yet our understanding of the central ...neural circuits regulating bone metabolism is limited. Here, we demonstrate that chronic stress in crewmembers resulted in decreased bone density and elevated anxiety in an isolated habitat mimicking a space station. We then used a mouse model to demonstrate that GABAergic neural circuitry in the ventromedial hypothalamus (VMH) mediates chronic stress-induced bone loss. We show that GABAergic inputs in the dorsomedial VMH arise from a specific group of somatostatin neurons in the posterior region of the bed nucleus of the stria terminalis, which is indispensable for stress-induced bone loss and is able to trigger bone loss in the absence of stressors. In addition, the sympathetic system and glutamatergic neurons in the nucleus tractus solitarius were employed to regulate stress-induced bone loss. Our study has therefore identified the central neural mechanism by which chronic stress-induced mood disorders, such as anxiety, influence bone metabolism.
The research on video analytics especially in the area of human behavior recognition has become increasingly popular recently. It is widely applied in virtual reality, video surveillance, and video ...retrieval. With the advancement of deep learning algorithms and computer hardware, the conventional two-dimensional convolution technique for training video models has been replaced by three-dimensional convolution, which enables the extraction of spatio-temporal features. Specifically, the use of 3D convolution in human behavior recognition has been the subject of growing interest. However, the increased dimensionality has led to challenges such as the dramatic increase in the number of parameters, increased time complexity, and a strong dependence on GPUs for effective spatio-temporal feature extraction. The training speed can be considerably slow without the support of powerful GPU hardware. To address these issues, this study proposes an Adaptive Time Compression (ATC) module. Functioning as an independent component, ATC can be seamlessly integrated into existing architectures and achieves data compression by eliminating redundant frames within video data. The ATC module effectively reduces GPU computing load and time complexity with negligible loss of accuracy, thereby facilitating real-time human behavior recognition.
Abstract
Ferroptosis is a form of programmed cell death characterized by intracellular iron accumulation and lipid peroxidation, and earlier studies identified glutathione peroxidase 4 (GPX4) as an ...essential regulator of this process. Ferroptosis plays an essential role in tumors, degenerative diseases, and ischemia-reperfusion injury. However, researchers have found that inhibition of GPX4 does not entirely suppress ferroptosis in certain diseases, or cells express resistance to ferroptosis agonists that inhibit GPX4. As research progresses, it has been discovered that there are multiple regulatory pathways for ferroptosis that are independent of GPX4. The study of GPX4-independent ferroptosis pathways can better target ferroptosis to prevent and treat various diseases. Here, the currently inhibited pulmonary GPX4-dependent ferroptosis pathways will be reviewed.
