Acute myocardial infarction (AMI) is the most common cause of death in the world. Comprehensive risk assessment of patients presenting with chest pain and eliminating undesirable results should ...decrease morbidity and mortality rates, increase the quality of life of patients, and decrease health expenditure in many countries. In this study, the advantages and disadvantages of the enzymatic and nonenzymatic biomarkers used in the diagnosis of patients with AMI are given in historical sequence, and some candidate biomarkers - hFABP, GPBB, S100, PAPP-A, RP, TNF, IL6, IL18, CD40 ligand, MPO, MMP9, cell-adhesion molecules, oxidized LDL, glutathione, homocysteine, fibrinogen, and D-dimer procalcitonin - with a possible role in the diagnosis of AMI are discussed.
The present study was carried out using meta-analyses, reviews of clinical trials, evidence-based medicine, and guidelines indexed in PubMed and Web of Science.
These numerous AMI biomarkers guide clinical applications (diagnostic methods, risk stratification, and treatment). Today, however, TnI remains the gold standard for the diagnosis of AMI. Details in the text will be given of many biomarkers for the diagnosis of AMI.
We evaluated the advantages and disadvantages of routine enzymatic and nonenzymatic biomarkers and the literature evidence of other candidate biomarkers in the diagnosis of AMI, and discuss challenges and constraints that limit translational use from bench to bedside.
The aim of the study was to determine the effects of different and regularly applied exercise programs on irisin, heat shock protein 70 and some biochemical parameters.
120 male university students ...participated in the study. Participants were divided into 4 equal groups as control (C), resistance exercise group (RE), high intensity interval (HIIT) and aerobic exercise group (AE). While the control group did not perform any exercise, the pre-determined exercise programs were applied to the other groups for 8 weeks and 3 days in a week. Blood samples were taken from all participants before and after the exercise program. Cholesterol, High-density Lipoprotein (HDL) and Low-density Lipoprotein (LDL) cholesterol, triglyceride (TG), Creatine kinase (CK), Lactate dehydrogenase (LDH), Irisin and Heat shock protein 70 (HSP70) levels were analyzed in blood samples.
It is determined that there are significant differences in pre-posttest values of the AE group's LDH, cholesterol, HDL-cholesterol, TG and HSP 70 levels, HIIT group's CK, LDH, Cholesterol, HDL-cholesterol, TG, Irisin and HSP70 levels and RE group's CK, LDH, Cholesterol, LDL-cholesterol, TG and Irisin levels (p<0.05).
It can be said that exercise can provide improvements in lipid profile, changes in HSP70 levels may vary depending on muscle damage, the increase of irisin due to exercise.
A fine-tuned pro-inflammatory and anti-inflammatory balance in the follicular unit is essential for cumulus expansion and successful ovulation. While the long pentraxin 3 (PTX3) gene is required for ...the expansion of cumulus cells (CCs), ovulation, resumption of meiosis and fertilization, the vitamin D receptor gene (VDR-X2) is required for intra-follicle redox balance. This study was planned to determine the expression pattern of VDR-X2 and PTX3 mRNA in CCs isolated from germinal vesicle (GV), metaphase I (MI), and metaphase II (MII) oocytes of PCOS patients with ovulatory dysfunction.
The relative expression of CC-PTX3 and CC-VDR-X2 mRNA were evaluated using qRT-PCR in a total of 79 CC samples collected from individual cumulus-oocyte complex of 40 infertile patients (20 PCOS and 20 non-PCOS normal responders) who underwent ovarian stimulation with the GnRH antagonist protocol.
Relative PTX3 mRNA expressions of CCMI-control and CCMII-control showed 3- and 9-fold significant upregulation compared to CCGV-control, respectively. The relative PTX3 mRNA expression of CCMII-control increased approximately three fold compared to CCMI-control. Compared to CCGV-pcos, a 3-fold increase was noted in the relative PTX3 mRNA expression of CCMI-pcos and an approximately 4-fold increase in the PTX3 mRNA expression of CCMII-pcos. Relative PTX3 mRNA expression values of CCMII-pcos and CCMI-pcos were similar. A 6-fold upregulation of relative PTX3 mRNA and a 4-fold upregulation of VDR-X2 mRNA were detected in CCMII-control compared to CCMII-pcos. CC-VDR-X2 expression patterns of the PCOS and control groups overlapped with the CC-PTX3 pattern. Fertilization rates of the PCOS group exhibiting failed transcript expression were similar to normal responders.
The fact that relative CC-PTX3 and CC-VDR mRNA expression does not increase during the transition from MI to MII stage in PCOS as in normal responders suggests that PTX3 and VDR expression may be defective in cumulus cells of PCOS patients with ovulatory dysfunction.
Abstract
Objectives
This study was aimed to investigate the diagnostic values of serum levels of asprosin and neutrophil gelatinase-associated lipocalin (NGAL) in Retinopathy of prematurity (ROP) and ...to assess the role of these biomarkers on the development and progression of the condition.
Methods
This study was carried out from April 2020 to February 2021 in the department of ophthalmology of a tertiary hospital in Turkey. Thirty patients diagnosed with ROP and 30 healthy newborns were included the study. Serum NGAL and asprosin levels were determined via ELISA.
Results
The median serum NGAL levels were found to be similar between the ROP group and the control group (p=0.595). Median asprosin levels were significantly higher in patients diagnosed with ROP 46.58 (12.70–142.28) ng/mL compared to healthy subjects 13.05 (10.92–17.73) ng/mL (p=0.001). The optimal cut-off value of asprosin by ROC analysis was 30 ng/mL (AUC: 0.754, p=0.001) for diagnosing ROP. Serum asprosin levels were positively correlated with serum ALP levels and inversely correlated with gestational week, uric acid and AST values (all, p<0.005).
Conclusions
Our results demonstrated that asprosin, but not NGAL, could be a biomarker for the diagnosis of ROP.
INTRODUCTION: In 2016, the maximum allowable CV% and allowable total error limits (TEa-TR) for 15 test parameters were specified by the Ministry of Health of Turkey and put into practice. Our study ...aimed to assess the analytical performance of 15 test parameters studied in our laboratory in terms of the TEa-TR criteria and demonstrate the effect of the method of CV% use in the TAE formula on the performance of tests when two-level internal quality control (IQC) was employed. METHODS: Total analytical error (TAE) was calculated using the formula TAE%=(CV%×1.65)+bias%. Two different procedures were followed for TAE calculation. TAE1 and TAE2 were separately calculated for each control level using the CV% values obtained from the data of IQC at two different levels used for each test. In addition, TAE3 values were found for each test parameter in line with the total CV% value (Total CV%=√(CV12+CV22) obtained using the IQC data from two different levels. RESULTS: When CV% values were separately taken for each control level, the maximum allowable CV% value was exceeded in two tests at IQC Level 2, whereas the TEa-TR limit was exceeded in two tests at IQC Level 1 and in 2 tests at IQC Level 2 during the 6-month period. According to the TEa-TR formula, the allowable CV% was exceeded 19 times in 13 out of 15 tests, and TEa limits were exceeded 15 times in 10 tests during the 6-month period DISCUSSION AND CONCLUSION: When the total CV% was calculated from Level 1 and Level 2 according to the TEa-TR formula, both the maximum allowable CV% and allowable TEa% limits were exceeded many times. However, when the CV% and TAE values were calculated separately for different levels of IQC samples, the analytical performance of the tests were found better. We think that calculating the TAE and CV% values separately is more useful in showing the analytical performance of the test at different levels.
Schizophrenia, particularly the form related to excessive dopamine (DA), is a chronic psychotic disorder affecting millions of people worldwide. Renalase metabolizes its catecholamine (CA) ...substrates, including DA, suggesting that there might be an association between renalase levels and schizophrenia occurrence. Therefore, the current study aimed to evaluate the renalase and CA levels in the serum of patients with schizophrenia.
The study was conducted with thirty-three schizophrenia patients and an age- and gender-matched group of thirty-one controls. Renalase and CA levels were measured by using an enzyme-linked immunosorbent assay (ELISA).
Renalase levels were significantly lower in the schizophrenia patients than in the control group (
<0.05), whereas DA levels were significantly higher (
<0.05). The epinephrine (Epi) levels of both groups were similar (
=0.186), while the norepinephrine levels in patients with schizophrenia were significantly lower than those in the control group (
<0.05). The areas under the curves for the renalase-dopamine, renalase-norepinephrine and renalase-epinephrine ratios were 0.805, 95% confidence interval (CI): 0.699-0.912 (p<0.001); 0.726, 95% CI: 0.594-0.859 (p=0.032); and 0.656, 95% CI: 0.520-0.791 (p=0.02).
The high DA levels in patients with schizophrenia might be due to low renalase levels. Renalase enzyme levels may play a substantial role in the pathophysiology of schizophrenia. Thus, this enzyme might be a new future target for the treatment and diagnosis of schizophrenia after intrabrain renalase and DA dynamics have been further evaluated.
Purpose. Pseudoexfoliation syndrome (PEX) is an eye disease that develops under the influence of regional population differences, genetic factors, age, and environmental factors and is characterized ...by visualization of a gray-white fibrogranular substance in the lens anterior capsule and/or pupil margin during anterior segment examination. The underlying biochemical mechanisms of the disease have not yet been fully elucidated. Therefore, this study was designed to show the changes in aqueous humor and blood serum levels of matrix metalloproteinases (decorin and tenascin C), total antioxidants (TAS), and total oxidants (TOS) in both cataract patients who have unilateral PEX material and cataract patients who do not have unilateral PEX material. Methods. Biological samples were simultaneously collected from 22 cataract patients who had unilateral pseudoexfoliation (PEX patients) and 22 cataract patients who did not have unilateral pseudoexfoliation (control patients). From the collected biological samples, decorin (DEC) and tenascin C (TN-C) were measured with the enzyme-linked immunosorbent assay (ELISA) method, and TAS and TOS were measured with an autoanalyzer. Results. When decorin, tenascin C, and TOS values of PEX patients were compared with those of control patients, there was a statistically significant increase in all three parameters. Conversely, TAS values showed a statistically significant decrease in PEX patients compared to controls. DEC, TN-C, TAS values, and TOS values were significantly higher in aqueous fluid than in blood in both the PEX patient and control groups. Conclusions. We suggest that parameters such as DEC, TN-C, TAS, and TOS play a role in the etiopathology of pseudoexfoliation syndrome. Thus, bringing these increased levels of extracellular proteins and TOS and decreased levels of TAS back to within physiological limits can mediate the reorganization of the blood-aqueous fluid barrier and slow the progression of pseudoexfoliation syndrome.
Aim: Insufficient oxygen supply to organs and tissues due to reduced arterial or venous blood flow results in ischaemia, during which, although ATP production stops, AMP and adenosine continue to be ...produced from ATP. The fate of irisin, which causes the production of heat instead of ATP during ischaemia, is unknown. Iloprost and sildenafil are two pharmaceutical agents that mediate the resumption of reperfusion (blood supply) via vasodilatation during ischaemic conditions. Our study aimed to explore the effects of iloprost and sildenafil on irisin levels in the heart, liver and kidney tissues and whether these pharmaceutical agents had any impact on serum irisin and nitric oxide levels in rats with induced experimental myocardial ischaemia. Methods: The study included adult male Sprague-Dawley rats aged 10 months and weighing between 250 and 280 g. The animals were randomly allocated to eight groups, with five rats in each group. The groups were: sham (control), iloprost (ILO), sildenafil (SIL), ILO + SIL, myocardial ischaemia (MI), MI + ILO, MI + SIL and MI + ILO + SIL. The treatment protocols were implemented before inducing ischaemia, which was done by occluding the left coronary artery with a plastic ligature for 30 minutes. Following the reperfusion procedure, all rats were sacrificed after 24 hours, and their heart, liver and kidney tissues and blood samples were collected for analyses. An immunohistochemical method was used to measure the change in irisin levels, the ELISA method to quantify blood irisin levels, and Griess’ assay to determine nitric oxide (NO) levels in the serum and tissue. Myocardial ischaemia was confirmed based on the results of Masson’s trichrome staining, as well as levels of troponin and creatine kinase MB. Results: Irisin levels in biological tissue and serum dropped statistically significantly in the ischaemic group (MI), but were restored with ILO and SIL administration. Individual SIL administration was more potently restorative than individual ILO administration or the combined administration of the two agents. NO level, on the other hand, showed the opposite tendency, reaching the highest level in the MI group, and falling with the use of pharmaceutical agents. Conclusions: Individual or combined administration of ILO and SIL reduced myocardial ischaemia and NO levels, and increased irisin levels. Elevated levels of irisin obtained by drug administration could possibly contribute to accelerated wound recovery by local heat production. Sildenafil was more effective than iloprost in eliminating ischaemia and may be the first choice in offsetting the effects of ischaemia in the future.
