Iron(II) phthalocyanine was immobilized onto amidoximated polyacrylonitrile fiber to construct a bioinspired catalytic system for oxidizing organic dyes by H2O2 activation. The amidoxime groups greatly helped to anchor Iron(II) phthalocyanine molecules onto the fiber through coordination interaction, which has been confirmed by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy and UV⿿vis diffuse reflectance spectroscopy analyses. Electron spin resonance studies indicate that the catalytic process of physically anchored Iron(II) phthalocyanine performed via a hydroxyl radical pathway, while the catalyst bonded Iron(II) phthalocyanine through coordination effect could selectively catalyze the H2O2 decomposition to generate high-valent iron-oxo species. This may result from the amidoxime groups functioning as the axial fifth ligands to favor the heterolytic cleavage of the peroxide OO bond. This feature also enables the catalyst to only degrade the dyes adjacent to the catalytic active centers and enhances the efficient utilization of H2O2. In addition, this catalyst could effectively catalyze the mineralization of organic dyes and can be easily recycled without any loss of activity.

In this study, we evaluated the potential roles for two R2R3 MYB transcription factors (TFs), MYB88 and the paralogous FLP (MYB124), in cold stress in apple and Arabidopsis. We found that MYB88 and MYB124 positively regulate freezing tolerance and cold-responsive gene expression in both apple and Arabidopsis.
Chromatin-Immunoprecipitation-qPCR and electrophoretic mobility shift assays showed that MdMYB88/MdMYB124 act as direct regulators of the COLD SHOCK DOMAIN PROTEIN 3 (MdCSP3) and CIRCADIAN CLOCK ASSOCIATED 1 (MdCCA1) genes. Dual luciferase reporter assay indicated that MdCCA1 but not MdCSP3 activated the expression of MdCBF3 under cold stress. Moreover, MdMYB88 and MdMYB124 promoted anthocyanin accumulation and H2O2 detoxification in response to cold.
Taken together, our results suggest that MdMYB88 and MdMYB124 positively regulate cold hardiness and cold-responsive gene expression under cold stress by C-REPEAT BINDING FACTOR (CBF)-dependent and CBF-independent pathways.

We demonstrate the synthesis of antibacterial carbon quantum dots (CQDs) through a one-step pyrolysis of biogenic polyamine (PA) and dopamine (DA) mixture. SPM/DA-CQDs synthesized from DA combined with spermine (SPM) exhibit effective antibacterial activity and high adhesion properties on glass and surfaces of polymeric contact lens material. The antimicrobial activity of SPM/DA-CQDs is primarily due to their ability to disrupt the bacterial membrane, and they possess wide spectrum antimicrobial activity against Gram-negative bacteria (Escherichia coli, Salmonella enterica serovar Enteritidis, and Pseudomonas aeruginosa) and Gram-positive bacteria (Staphylococcus aureus and its multidrug-resistant strain, methicillin-resistant S. aureus). The minimal inhibitory concentration (MIC) of SPM/DA-CQDs against the tested bacteria was >17-fold and >7-fold lower than those of CQDs prepared solely from spermine and dopamine, respectively. The great biocompatibility of our SPM/DA-CQDs is revealed from the in vitro cytotoxicity and hemolysis assays and in vivo evaluation of morphological and physiological changes of rabbit corneas. We further prepared SPM/DA-CQDs-coated coverslips and contact lenses, which display excellent stability and antibiofilm properties. Moreover, in vivo tests show that SPM/DA-CQDs suppress bacterial keratitis (BK) formation in a corneal-injured rabbit model. Our results indicate that SPM/DA-CQDs have great potential as a coating material for inhibition of biofilm formation on contact lenses, and broadly to protect medical devices from contamination.
Recent investigations reveal that lactate is not a waste product but a major energy source for cells, especially in the mitochondria, which can support cellular survival under glucose shortage. ...Accordingly, the new understanding of lactate prompts to target it together with glucose to pursue a more efficient cancer starvation therapy. Herein, zeolitic imidazolate framework‐8 (ZIF‐8) nanoplatforms are used to co‐deliver α‐cyano‐4‐hydroxycinnamate (CHC) and glucose oxidase (GOx) and fulfill the task of simultaneous depriving of lactate and glucose, resulting in a new nanomedicine CHC/GOx@ZIF‐8. The synthesis conditions are carefully optimized in order to yield monodisperse and uniform nanomedicines, which will ensure reliable and steady therapeutic properties. Compared with the strategies aiming at a single carbon source, improved starvation therapy efficacy is observed. Besides, more than boosting the energy shortage, CHC/GOx@ZIF‐8 can block the lactate‐fueled respiration and relieve solid tumor hypoxia, which will enhance GOx catalysis activity, depleting extra glucose, and producing more cytotoxic H2O2. By the synergistically enhanced anti‐tumor effect, both in vitro and in vivo cancer‐killing efficacies of CHC/GOx@ZIF‐8 show twice enhancements than the GOx mediated therapy. The results demonstrate that the dual‐depriving of lactate and glucose is a more advanced strategy for strengthening cancer starvation therapy.
Metal‐organic‐framework nanomedicines encapsulating an inhibitor of monocarboxylate transporters 1 and glucose oxidase are developed for efficient glucose depletion while inhibiting the lactate influx. At meanwhile, inhibiting the lactate influx can improve the intracellular oxygen pressure, which subsequently promotes the catalytic activity of glucose oxidase. Cancer cells are substantially killed from ATP shortage and high ROS levels.
The Universal Soil Loss Equation (USLE) and its revised version (RUSLE) are often used to estimate soil erosion at regional landscape scales. USLE/RUSLE contain parameters for slope length factor (L) ...and slope steepness factor (S), usually combined as LS. However a major limitation is the difficulty in extracting the LS factor. Methods to estimate LS based on geographic information systems have been developed in the last two decades. L can be calculated for large watersheds using the unit contributing area (UCA) or the slope length (λ) as input parameters. Due to the absence of an estimation of slope length, the UCA method is insufficiently accurate. Improvement of the spatial accuracy of slope length and LS factor is still necessary for estimating soil erosion. The purpose of this study was to develop an improved method to estimate the slope length and LS factor. We combined the algorithm for multiple-flow direction (MFD) used in the UCA method with the LS-TOOL (LS-TOOLSFD) algorithms, taking into account the calculation errors and cutoff conditions for distance, to obtain slope length (λ) and the LS factor. The new method, LS-TOOLMFD, was applied and validated in a catchment with complexly variable slopes. The slope length and LS calculated by LS-TOOLMFD both agreed better with field data than with the calculations using the LS-TOOLSFD and UCA methods, respectively. We then integrated the LS-TOOLMFD algorithm into LS-TOOL developed in Microsoft's.NET environment using C# with a user-friendly interface. The method can automatically calculate slope length, slope steepness, L, S, and LS factor, providing the results as ASCII files that can be easily used in GIS software and erosion models. This study is an important step forward in conducting accurate large-scale erosion evaluation.
The design of a nano-system for the detection of intracellular microRNAs is challenging as it must fulfill complex requirements, i.e., it must have a high sensitivity to determine the dynamic ...expression level, a good reliability for multiplex and simultaneous detection, and a satisfactory biostability to work in biological environments. Instead of employing a commonly used physisorption or a full-conjugation strategy, here, a GO-DNA nano-system was developed under graft/base-pairing construction. The common anchor sequence was chemically grafted to GO to base-pair with various microRNA probes; and the hybridization with miRNAs drives the dyes on the probes to leave away from GO, resulting in "turned-on" fluorescence. This strategy not only simplifies the synthesis but also efficiently balances the loading yields of different probes. Moreover, the conjugation yield of GO with a base-paired hybrid has been improved by more than two-fold compared to that of the conjugation with a single strand. We demonstrated that base-paired DNA probes could be efficiently delivered into cells along with GO and are properly stabilized by the conjugated anchor sequence. The resultant GO-DNA nano-system exhibited high stability in a complex biological environment and good resistance to nucleases, and was able to accurately discriminate various miRNAs without cross-reaction. With all of these positive features, the GO-DNA nano-system can simultaneously detect three miRNAs and monitor their dynamic expression levels.
Pentatricopeptide repeat (PPR) is a triangular pentapeptide repeat domain that plays a vital role in plant growth. In this study, we seek to identify PPR coding genes and proteins using a mixture of ...feature extraction methods. We use four single feature extraction methods focusing on the sequence, physical, and chemical properties as well as the amino acid composition, and mix the features. The Max-Relevant-Max-Distance (MRMD) technique is applied to reduce the feature dimension. Classification uses the random forest, J48, and naïve Bayes with 10-fold cross-validation.
Combining two of the feature extraction methods with the random forest classifier produces the highest area under the curve of 0.9848. Using MRMD to reduce the dimension improves this metric for J48 and naïve Bayes, but has little effect on the random forest results.

The H3-subtype of avian influenza virus (AIV) is one of the most frequently detected low pathogenic avian influenza virus (LPAIV) subtypes in birds and fowls, causing substantial economic loss to the ...poultry industry. Most importantly, besides poultry, mammals could also be infected with it, such as swines, canines, equines, felines, and humans, posing a serious public health threat. This allows the virus to persist widely in poultry and wild birds for a long time, where it may mix with other subtypes, providing conditions for viral recombination or reassortment. Currently, the monitoring of H3-subtype AIV is inadequate, and there is a lack of effective prevention and control measures for H3-subtype AIV. Here, the epidemiology, phylogeny, and genetic variation of H3-subtype AIV were analyzed, and nonsynonymous and synonymous substitution rates (dN/dS) were calculated. Through these steps, we aimed to clarify the current epidemiological feature and evolutionary characteristics of H3-subtype AIV, and provide an operative reference for future scientific control of H3-subtype AIV.
Plant phenotypes respond to environmental change, an adaptive capacity that is at least partly transgenerational. However, epigenetic components of this interplay are difficult to measure. Depletion ...of the nuclear-encoded protein MSH1 causes dramatic and heritable changes in plant development, and here we show that crossing these altered plants with isogenic wild type produces epi-lines with heritable, enhanced growth vigour. Pericentromeric DNA hypermethylation occurs in a subset of msh1 mutants, indicative of heightened transposon repression, while enhanced growth epi-lines show large chromosomal segments of differential CG methylation, reflecting genome-wide reprogramming. When seedlings are treated with 5-azacytidine, root growth of epi-lines is restored to wild-type levels, implicating hypermethylation in enhanced growth. Grafts of wild-type floral stems to mutant rosettes produce progeny with enhanced growth and altered CG methylation strikingly similar to epi-lines, indicating a mobile signal when MSH1 is downregulated, and confirming the programmed nature of methylome and phenotype changes.
Copy number variation (CNV) is the most prevalent type of genetic structural variation that has been recognized as an important source of phenotypic variation in humans, animals and plants. However, ...the mechanisms underlying the evolution of CNVs and their function in natural or artificial selection remain unknown. Here, we generated CNV region (CNVR) datasets which were diverged or shared among cattle, goat, and sheep, including 886 individuals from 171 diverse populations. Using 9 environmental factors for genome-wide association study (GWAS), we identified a series of candidate CNVRs, including genes relating to immunity, tick resistance, multi-drug resistance, and muscle development. The number of CNVRs shared between species is significantly higher than expected (P<0.00001), and these CNVRs may be more persist than the single nucleotide polymorphisms (SNPs) shared between species. We also identified genomic regions under long-term balancing selection and uncovered the potential diversity of the selected CNVRs close to the important functional genes. This study provides the evidence that balancing selection might be more common in mammals than previously considered, and might play an important role in the daily activities of these ruminant species.
