Subclinical hypothyroidism (SCH) has been shown to be associated with microbiota. However, the association between SCH and oral microbiota has not yet been elucidated. The results of our previous ...clinical studies showed that Prevotella intermedia was abundant in the oral microbiota of SCH patients. This study aimed to investigate the relationship between SCH and oral microbiota, verify the pathogenicity of P. intermedia in SCH, and preliminarily explore the possible mechanism. The SCH mouse model with oral application of P. intermedia was established, and the variance in the mouse oral microbiota and changes in thyroid function and metabolism were detected in mice. Student’s t test and analysis of variance were used for statistical analysis. Oral application of P. intermedia changed the composition of the oral microbiota of SCH mice, which enhanced the damage to the thyroid and decreased the expression of functional genes of the thyroid. Moreover, P. intermedia decreased oxygen consumption and aggravated glucose and lipid metabolism disorders in SCH mice. Glucose tolerance and insulin tolerance decreased, and the triglyceride content of the liver and inflammatory infiltration in adipose tissue increased in SCH mice after P. intermedia stimulation. Mechanistically, P. intermedia increased the proportion of CD4+ T cells in cervical lymph nodes and thyroids in SCH mice. Th1 cells were suggested to play an important role in the pathogenesis of SCH involving P. intermedia. In conclusion, P. intermedia aggravated SCH manifestations, including thyroid dysfunction and glucose and lipid metabolism disorders, by causing immune imbalance in mice. This study sheds new light on the pathogenesis of SCH from the perspective of oral microbiota.
In the era of the Social Web, crowdfunding has become an increasingly more important channel for entrepreneurs to raise funds from the crowd to support their startup projects. Previous studies ...examined various factors such as project goals, project durations, and categories of projects that might influence the outcomes of the fund raising campaigns. However, textual information of projects has rarely been studied for analyzing crowdfunding successes. The main contribution of our research work is the design of a novel text analytics-based framework that can extract latent semantics from the textual descriptions of projects to predict the fund raising outcomes of these projects. More specifically, we develop the Domain-Constraint Latent Dirichlet Allocation (DC-LDA) topic model for effective extraction of topical features from texts. Based on two real-world crowdfunding datasets, our experimental results reveal that the proposed framework outperforms a classical LDA-based method in predicting fund raising success by an average of 11% in terms of F1 score. The managerial implication of our research is that entrepreneurs can apply the proposed methodology to identify the most influential topical features embedded in project descriptions, and hence to better promote their projects and improving the chance of raising sufficient funds for their projects.

We present a measurement of R_{K^{*}}, the branching fraction ratio B(B→K^{*}μ^{+}μ^{-})/B(B→K^{*}e^{+}e^{-}), for both charged and neutral B mesons. The ratio for the charged case R_{K^{*+}} is the ...first measurement ever performed. In addition, we report absolute branching fractions for the individual modes in bins of the squared dilepton invariant mass q^{2}. The analysis is based on a data sample of 711 fb^{-1}, containing 772×10^{6} BBover ¯ events, recorded at the ϒ(4S) resonance with the Belle detector at the KEKB asymmetric-energy e^{+}e^{-} collider. The obtained results are consistent with standard model expectations.
KDM2B (also known as FBXL10) controls stem cell self-renewal, somatic cell reprogramming and senescence, and tumorigenesis. KDM2B contains multiple functional domains, including a JmjC domain that ...catalyzes H3K36 demethylation and a CxxC zinc-finger that recognizes CpG islands and recruits the polycomb repressive complex 1. Here, we report that KDM2B, via its F-box domain, functions as a subunit of the CUL1-RING ubiquitin ligase (CRL1/SCF(KDM2B)) complex. KDM2B targets c-Fos for polyubiquitylation and regulates c-Fos protein levels. Unlike the phosphorylation of other SCF (SKP1-CUL1-F-box)/CRL1 substrates that promotes substrates binding to F-box, epidermal growth factor (EGF)-induced c-Fos S374 phosphorylation dissociates c-Fos from KDM2B and stabilizes c-Fos protein. Non-phosphorylatable and phosphomimetic mutations at S374 result in c-Fos protein which cannot be induced by EGF or accumulates constitutively and lead to decreased or increased cell proliferation, respectively. Multiple tumor-derived KDM2B mutations impaired the function of KDM2B to target c-Fos degradation and to suppress cell proliferation. These results reveal a novel function of KDM2B in the negative regulation of cell proliferation by assembling an E3 ligase to targeting c-Fos protein degradation that is antagonized by mitogenic stimulations.
MicroRNA (miRNA)107 expression is downregulated but high mobility group box 1 (HMGB-1), Toll-like receptors (TLRs), and receptor for advanced glycation end products (RAGE) are upregulated in ...osteoarthritic (OA) cartilage. We investigated mir-107/HMGB-1 signaling in OA after hyperbaric oxygen (HBO) treatment.
MiR-107 mimic was transfected and the HMGB-1 was analyzed in OA chondrocytes. MiRNA targets were identified using bioinformatics and a luciferase reporter assay. After HBO treatment, the mRNA or protein levels of HMGB-1, RAGE, TLR2, TLR4, and inducible nitric oxide (NO) synthase (iNOS) and phosphorylation of mitogen-activated protein kinase (MAPK) were evaluated. The secreted HMGB-1 and matrix metalloproteases (MMPs) levels were quantified. Finally, we detected the HMGB-1 and iNOS expression in rabbit cartilage defects.
Overexpression of miR-107 suppressed HMGB-1 expression in OA chondrocytes. The 3′UTR of HMGB-1 mRNA contained a ‘seed-matched-sequence’ for miR-107. MiR-107 was induced by HBO and a marked suppression of HMGB-1 was observed simultaneously in OA chondrocytes. Knockdown of miR-107 upregulated HMGB-1 expression in hyperoxic cells. HBO downregulated the mRNA and protein expression of HMGB-1, RAGE, TLR2, TLR4, and iNOS, and the secretion of HMGB-1. HBO decreased the nuclear translocation of nuclear factor (NF)-κB, downregulated the phosphorylation of MAPK, and significantly decreased the secretion of MMPs. Morphological and immunohistochemical observation demonstrated that HBO markedly enhanced cartilage repair and the area stained positive for HMGB-1 and iNOS tended to be lower in the HBO group.

Gas sensors fabricated with nanowires as the detecting elements are powerful due to their many improved characteristics such as high surface-to-volume ratios, ultrasensitivity, higher selectivity, ...low power consumption, and fast response. This paper gives an overview on the recent process of the development of nanotechnology and nanowire-based gas sensors. The two basic approaches, top-down and bottom-up, for synthesizing nanowires are compared. The conduction mechanisms, sensing performances, configurations, and sensing principles of different nanowire gas sensors and arrays are summarized and discussed. Meanwhile, an emerging nanowires fabrication method and a self-powered nanowire pH sensor are highlighted. The scientific and technological challenges in the field are discussed at the end of the review.
The previous observational studies could not overcome the effects of confounding variables and reverse causality. We aimed to determine whether there is a causal relationship between systemic lupus ...erythematosus and osteoporosis in East Asian and European populations, respectively, by two-sample Mendelian Randomization analysis. We obtained and downloaded data from publicly available genome-wide association study databases and analyses for East Asian and European populations, including systemic lupus erythematosus (SLE), osteoporosis (OP), multisite bone mineral density (BMD), and OP with fracture. After screening for instrumental single-nucleotide polymorphisms (SNPs) significantly correlated to SLE, the inverse-variance weighted (IVW) method was used for calculating the ratio and 95% confidence interval, besides utilizing MR-Egger, weighted median, and weighted mode to assess the robustness of the primary outcome. Moreover, multiple analyses, including MR-PRESSO, MR-Egger intercept, Cochran's Q test, as well as "leave-one-out" sensitivity, were used for evaluating horizontal pleiotropy, heterogeneity, and stability. Finally, we exchanged exposure and outcome and performed a reverse MR analysis. IVW (OR = 1.05, 95% CI = 1.01–1.09, P = 0.009) indicated a significant positive correlation between genetically predicted SLE and OP in East Asians. Furthermore, neither heterogeneity nor horizontal pleiotropy was observed. In Europe, there was no significant genetically predicted causal relation between SLE and OP. Bi-directional MR analysis showed no reverse causality between SLE and OP. In the East Asian population, genetically predicted SLE may have had a positive causal relationship with OP. In Europe, there is insufficient evidence for a potential causal relation between SLE and OP or BMD and fracture, and the correlations currently observed may be attributed to a variety of confounder variables.
2D Ruddlesden–Popper perovskites (RPPs) have recently drawn significant attention because of their structural variability that can be used to tailor optoelectronic properties and improve the ...stability of derived photovoltaic devices. However, charge separation and transport in 2D perovskite solar cells (PSCs) suffer from quantum well barriers formed during the processing of perovskites. It is extremely difficult to manage phase distributions in 2D perovskites made from the stoichiometric mixtures of precursor solutions. Herein, a generally applicable guideline is demonstrated for precisely controlling phase purity and arrangement in RPP films. By visually presenting the critical colloidal formation of the single‐crystal precursor solution, coordination engineering is conducted with a rationally selected cosolvent to tune the colloidal properties. In nonpolar cosolvent media, the derived colloidal template enables RPP crystals to preferentially grow along the vertically ordered alignment with a narrow phase variation around a target value, resulting in efficient charge transport and extraction. As a result, a record‐high power conversion efficiency (PCE) of 14.68% is demonstrated for a (TEA)2(MA)2Pb3I10 (n = 3) photovoltaic device with negligible hysteresis. Remarkably, superior stability is achieved with 93% retainment of the initial efficiency after 500 h of unencapsulated operation in ambient air conditions.
A generic guideline for accurately controlling phase purity and arrangement in 2D perovskite films is provided by utilizing coordination engineering of a single‐crystal precursor solution. The resulting films with narrow distribution and preferentially perpendicular crystal orientation result in a significant improvement in device performance and stability, which is not typically found in conventional stoichiometric precursors.
