Skin cutaneous melanoma (SKCM) is an aggressive malignant skin tumor. Ferroptosis is an iron-dependent cell death that may mobilize tumor-infiltrating immunity against cancer. The potential mechanism ...of long non-coding RNAs (lncRNAs) in ferroptosis in SKCM is not clear. In this study, the prognostic and treatment value of ferroptosis-related lncRNAs was explored in SKCM, and a prognostic model was established.
We first explored the mutation state of ferroptosis-related genes in SKCM samples from The Cancer Genome Atlas database. Then, we utilized consensus clustering analysis to divide the samples into three clusters based on gene expression and evaluated their immune infiltration using gene-set enrichment analysis (GSEA) ESTIMATE and single-sample gene-set enrichment analysis (ssGSEA) algorithms. In addition, we applied univariate Cox analysis to screen prognostic lncRNAs and then validated their prognostic value by Kaplan-Meier (K-M) and transcripts per kilobase million (TPM) value analyses. Finally, we constructed an 18-ferroptosis-related lncRNA prognostic model by multivariate Cox analysis, and SKCM patients were allocated into different risk groups based on the median risk score. The prognostic value of the model was evaluated by K-M and time-dependent receiver operating characteristic (ROC) analyses. Additionally, the immunophenoscore (IPS) in different risk groups was detected.
The top three mutated ferroptosis genes were TP53, ACSL5, and TF. The SKCM patients in the cluster C had the highest ferroptosis-related gene expression with the richest immune infiltration. Based on the 18 prognosis-related lncRNAs, we constructed a prognostic model of SKCM patients. Patients at low risk had a better prognosis and higher IPS.
Our findings revealed that ferroptosis-related lncRNAs were expected to become potential biomarkers and indicators of prognosis and immunotherapy treatment targets of SKCM.
Background:
Type A acute aortic dissection (TAAAD) is a destructive cardiovascular disease, with high morbidity and mortality rates. Identifying the high-risk TAAAD patients at an early stage is ...urgently necessary.
Methods:
A retrospective study of 160 patients was carried out. The admission data were retrospectively gathered. Logistic regression analysis and receiver operator characteristic curve (AUC) was utilized.
Results:
Compared with the survivor group, the nonsurvivor group was older, had higher D-dimer levels, red blood cell distribution width (RDW) levels and platelet distribution width (PDW) levels, and lower fibrinogen levels, platelet levels and plateletcrit levels. Multivariate analysis displayed that four independent factors, age (hazard ratio (HR): 7.877, 95% confidence interval (CI) 2.740–22.641, p < 0.001), D-dimer (HR: 3.791, 95% CI 1.520–9.452, p = 0.004), RDW (HR: 3.300, 95% CI 1.109–9.825, p = 0.032), PDW (HR: 3.755, 95% CI 1.436–9.815, p = 0.007) were incorporated into the model. The predict accuracy of the model (AUC 0.861, 95% CI 0.798–0.911, p < 0.001) was best.
Conclusions:
Age, D-dimer, RDW and PDW are independent markers of in-hospital death in TAAAD patients and the newly established model has better performance in predicting high-risk patients. This model can be used as a quick screening tool to assess the prognosis of patients in individualizing.

β-catenin is a key signaling molecule in the canonical Wnt pathway, which is involved in animal development. However, little information has been reported for β-catenin in bivalves. In the present ...study, we cloned a homolog of β-catenin from the scallop Chlamys farreri and determined its expression characteristics. The full-length cDNA of β-catenin was 3,353 bp, including a 2,511 bp open reading frame that encoded a predicted 836 amino acid protein. Level of the β-catenin mRNA increased significantly (P<0.05) with C. farreri gonadal development and presented a sexually dimorphic expression pattern in the gonads, which was significantly high in ovaries detected by quantitative real-time polymerase chain reaction (qRT-PCR). Immunohistochemical analysis revealed that the β-catenin was mainly located in germ cells of the gonads, with obvious positive immune signals in the oogonia and oocytes of ovaries as well as in the spermatogonia and spermatocytes of testes, implying β-catenin might be involved in the gametogenesis of C. farreri. Furthermore, when 0.1 µg/mL and 0.2 µg/mL DKK-1 (an inhibitor of the canonical Wnt pathway) were added in vitro to culture medium containing testis cells of C. farreri, the expression of β-catenin decreased significantly detected by qRT-PCR (P<0.05), suggesting the canonical Wnt signal pathway exists in the scallop testis. Similarly, when 50 µM and 100 µM quercetin (an inhibitor of β-catenin) were added in vitro to the culture system, Dax1 expression was significantly down-regulated compared with controls (P<0.05), implying the β-catenin is an upstream gene of Dax1 and is involved in the regulation of C. farreri spermatogenesis.

H2 photoproduction, hydrogenase activities and PSII photochemical activities in Chlorella protothecoides under sulfur (S–) or nitrogen (N–) deprivation or simultaneous N-limitation and S-deprivation ...were studied. C. protothecoides pre-cultured in full nutrient TAP medium containing 7 mM NH4Cl was found to produce a detectable but low level of H2, once the cells were inoculated either in S-free or N-free medium. However, cells pre-grown in a low concentration of NH4Cl (0.35 and 0.7 mM) generated a large amount of H2 after transfer to N-limited and S-free medium. The maximal H2 outputs of ∼233.7 and ∼129.1 ml/l were obtained within 100 h in the cultures exposed to S-deprived medium containing 0.35 mM and 0.7 mM NH4Cl, with the average H2 production rates being ∼2.19 and ∼1.37 ml/l/h, respectively. Our studies further indicated that N-limitation resulted in considerable starch accumulation, chlorophyll synthesis reduction, photosynthetic electron transfer block and oxygen evolving complex (OEC) injury, as well as attenuation in PSII oxygenic activity. Significant starch degradation was not observed during the H2 photoevolution process. Attenuation of PSII O2 evolution favored a rapid establishment of anaerobiosis for hydrogenase induction. Meanwhile, a constant high level of hydrogenase activities in C. protothecoides exposed to simultaneous N-limitation and S-deprivation were measured. Based on the above results, a possible mechanism of high H2 photoproduction in C. protothecoides exposed to N-limitation and S-deprivation was discussed. Low net photosynthetic oxygenic rates, together with high hydrogenase activities were thought to contribute to the enhancement of H2 photoproduction by C. protothecoides.
▶ H2 production in Chlorella protothecoides was enhanced under N-limitation and S-deprivation. ▶ The optimal nitrogen concentration to attain maximal H2 yield was 0.35 mM NH4Cl. ▶ N-limitation leads to reduced PSII oxygenic activity favored anoxia establishment.
Mechanical heart valve replacement (MHVR) entails lifetime oral anticoagulation to eliminate thrombosis. However, adverse events may still occur despite proper anticoagulation therapy. In this study, ...we investigated whether D-dimer can predict the clinical events in post-MHVR patients during oral anticoagulation therapy.This was a single-center, prospective study. In all, 772 patients who underwent MHVR in the Wuhan Asia Heart Hospital from January 2013 to May 2014 were screened. Patients were assigned to the abnormal D-dimer group and the normal D-dimer group according to the D-dimer levels measured 3 months after the beginning of the oral anticoagulation therapy regime. All patients were followed up for 24 months or until the observation of the endpoints, which included thrombotic events, bleeding events, and all-cause deaths.A total of 718 patients were included in the analysis: 91 had abnormal D-dimer levels, and 627 had normal D-dimer levels. In all, 53 events were observed during 24 months. Compared with the normal D-dimer group, patients with abnormal D-dimer levels had a higher incidence of thrombotic events (10 versus 14; hazard ratio (HR): 5.36; 95% confidence interval (CI): 2.38-12.1; P < 0.001), all-cause mortality (8 versus 13; HR: 4.65; 95% CI: 1.93-11.2; P < 0.001), and a higher incidence of total events (16 versus 37; HR: 3.26; 95% CI: 1.81-5.86; P < 0.001). No significant difference was observed in bleeding events (2 versus 21; HR: 0.72; 95% CI: 0.17-3.07; P = 0.66).D-dimer may be a useful marker to predict thrombotic events and all-cause deaths in post-MHVR patients during oral anticoagulation therapy (ClinicalTrials.gov; NCT01996657).
Background
The role of the extracellular matrix (ECM) in oncogenic contexts has been studied previously, but the expression patterns and functional role of keratocan, a classical small leucine-rich ...proteoglycan found in the ECM, in tumors remain poorly understood. As pancreatic cancer (PC) is characterized by desmoplasia in the ECM, this study sought to assess the significance of keratocan in PC.
Objective
In this study, Gene Expression Profiling Interactive Analysis (GEPIA) was first used to analyze the expression pattern of keratocan in PC. Keratocan, P53, and P21 levels were evaluated in PDAC patient tissues and the role of keratocan was additionally directly assessed via transfecting PDAC cell lines with a pENTER-human keratocan construct.
Results
Patients with PC showing high levels of keratocan had low survival rates. A significantly upregulated expression of keratocan was observed in the PC tumor tissues in comparison to healthy paracancerous tissues. Keratocan upregulation in BxPC-3 and PANC-1 cells markedly enhanced their proliferation and migration, but reduced cellular apoptosis. Furthermore, the P53 and P21 expression levels were significantly reduced in pancreatic ductal adenocarcinoma cells overexpressing keratocan. P53 and P21 were downregulated in PC tumor tissues.
Conclusions
All results showed that keratocan played important roles in the promoting of PC.

These quantitative research results can provide references for relevant scholars' follow-up research and government departments to formulate and modify health policies or measures.
The annular flume is an ideal hydrodynamic test device for studying river sediment, and it has been widely used in recent years to study the movement patterns of sediment and other particulate ...matter. Annular flumes have made outstanding contributions to research in fields related to sediment transport and the diffusion and migration of pollutants. The existence of circumfluence structures in annular flumes leads to complex and variable flow structures. To obtain a more stable and controllable water flow structure, a sophisticated three-dimensional mathematical model based on the Fluent software was established to study the development law of water flow structure in the flume by changing the size of the annular flume speed ratio. The results show the following: (1) The overall trend of the simulation results basically matched with the measured results; the average relative error was 3.54% and the Nash efficiency coefficient was 0.9934, close to 1. The model calculation data were highly credible. (2) The axial flow velocity of the water tank gradually showed a “U”-shape distribution with the increase in the speed ratio. (3) When the speed ratio was R ≤ 0.17 (where the speed ratio R refers to the ratio of annular groove to shear ring speed), there was only one vortex in the tank; when the speed ratio was R > 0.17, there were multiple vortices in the tank, and the flow pattern was more complicated. (4) When the rotational speed ratio R = 0.28, the secondary flow intensity of the annular flume reached the lowest point, which was only 39.28% of the secondary flow intensity of the conventional annular flume. (5) It was determined that the annular flume water flow structure was most stable and controllable when the rotational speed ratio R = 0.24. The results of the study can provide a further theoretical basis for research on sediment dynamics and its related fields conducted by applying an annular flume.
