Abstract
Currently, coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has been reported in almost all countries globally. No effective ...therapy has been documented for COVID-19, and the role of convalescent plasma therapy is unknown. In the current study, 6 patients with COVID-19 and respiratory failure received convalescent plasma a median of 21.5 days after viral shedding was first detected, all tested negative for SARS-CoV-2 RNA within 3 days after infusion, and 5 eventually died. In conclusion, convalescent plasma treatment can end SARS-CoV-2 shedding but cannot reduce the mortality rate in critically ill patients with end-stage COVID-19, and treatment should be initiated earlier.

We develop a mathematical theory of symmetry protected trivial (SPT) orders and anomaly-free symmetry enriched topological (SET) orders in all dimensions via two different approaches with ...an emphasis on the second approach. The first approach is to gauge the symmetry in the same dimension by adding topological excitations as it was done in the 2d case, in which the gauging process is mathematically described by the minimal modular extensions of unitary braided fusion 1-categories. This 2d result immediately generalizes to all dimensions except in 1d, which is treated with special care. The second approach is to use the 1-dimensional higher bulk of the SPT/SET order and the boundary-bulk relation. This approach also leads us to a precise mathematical description and a classification of SPT/SET orders in all dimensions. The equivalence of these two approaches, together with known physical results, provides us with many precise mathematical predictions.
A global symmetry (0-symmetry) in an n-dimensional space acts on the whole space. A higher symmetry acts on closed submanifolds (i.e., loops and membranes, etc.), and those transformations form a ...higher group. In this paper, we introduce the notion of algebraic higher symmetry, which generalizes higher symmetry and is beyond higher group. We show that an algebraic higher symmetry in a bosonic system in n-dimensional space is characterized and classified by a local fusion n-category. We find that, when restricted to its symmetric sub-Hilbert space, an algebraic higher symmetry can be fully characterized by a noninvertible gravitational anomaly (i.e., a topological order in one higher dimension). Thus, we also refer to a noninvertible gravitational anomaly as categorical symmetry to stress its connection to symmetry. This provides a holographic and entanglement view of symmetries. For a system with a categorical symmetry, its gapped state must spontaneously break part (not all) of the symmetry, and the state with the full symmetry must be gapless. Using such a holographic point of view, we obtain (1) the gauging of the algebraic higher symmetry; (2) the classification of anomalies for an algebraic higher symmetry; (3) the equivalence between classes of systems, with different (potentially anomalous) algebraic higher symmetries or different sets of low-energy excitations, as long as they have the same categorical symmetry; and (4) the classification of gapped liquid phases for bosonic and/or fermionic systems with a categorical symmetry, as gapped boundaries of a topological order in one higher dimension (that corresponds to the categorical symmetry). This classification includes symmetry protected trivial (SPT) orders and symmetry enriched topological (SET) orders with an algebraic higher symmetry.
Chronic kidney disease (CKD) has emerged as a major public health problem worldwide. It frequently progresses to end-stage renal disease, which is related to very high cost and mortality. Novel ...biomarkers can provide insight into the novel mechanism, facilitate early detection, and monitor progression of CKD and its response to therapeutic interventions. To identify potential biomarkers, we applied an UPLC-HDMS together with univariate and multivariate statistical analyses using plasma samples from patients with CKD of diverse etiologies (100 sera in discovery set and 120 sera in validation set) and two different rat models of CKD. Using comprehensive screening and validation workflow, we identified a panel of seven metabolites that were shared by all patients and animals regardless of the underlying cause of CKD. These included ricinoleic acid, stearic acid, cytosine, LPA(16:0), LPA(18:2), 3-methylhistidine, and argininic acid. The combination of these seven biomarkers enabled the discrimination of patients with CKD from healthy subjects with a sensitivity of 83.3% and a specificity of 96.7%. In addition, these biomarkers accurately reflected improvements in renal function in response to the therapeutic interventions. Our results indicated that the identified biomarkers may improve the diagnosis of CKD and provide a novel tool for monitoring of the progression of disease and response to treatment in CKD patients.
Copper (Cu) is an essential micronutrient for all living organisms but is also highly toxic in excess. Cellular homoeostasis of Cu is maintained by various transporters and metallochaperones. Here, ...we investigated the biological function of OsCOPT7, a member of the copper transporters (COPT) family, in Cu homoeostasis in rice. OsCOPT7 was mainly expressed in the roots and the expression was upregulated by Cu deficiency. OsCOPT7 was localized at the tonoplast and the endoplasmic reticulum. Knockout of OsCOPT7 increased Cu accumulation in the roots but decreased Cu concentrations in the shoots and grain. The knockout mutants contained higher concentrations of Cu in the roots cell sap but markedly lower concentrations of Cu in the xylem sap than wild‐type plants. Seed setting and grain yield were reduced significantly in the knockout mutants grown in a low Cu soil. Knockout mutants were more tolerant to Cu toxicity. Yeast two‐hybrid and bimolecular fluorescence complementation assays showed that OsCOPT7 interacts physically with the rice Cu chaperone antioxidant protein 1 (OsATX1). Taken together, our results indicate that OsCOPT7 is a specific Cu transporter functioning to export Cu from the vacuoles and the ER and plays an important role in controlling the root‐to‐shoot Cu translocation in rice.

With the rapid development of the Internet of Vehicles, at the same time, due to the limited storage space of vehicles and more and more data requirements, vehicles need more and more ...vehicle-to-vehicle collaboration. At this time, a vehicular ad-hoc network (VANET) is required for inter-vehicle communication cooperation, and link breaks often occur due to the rapid movement of vehicles and the uncertainty of vehicle communication quality, which affects communication quality. Because of these situations, this paper proposes a vehicle cooperative communication method based on fuzzy logic and signal game strategy, obtains more real vehicle position and speed information on the Kalman filter, and then selects relay nodes to assist in requesting information message distribution of fuzzy logic. Each time the vehicle receives a request, the first data information is returned to the signal game. If all the information requests are not received the first time, the information requesting vehicle will send the second information request according to the signal strength and the beliefs observed on the previous vehicle. The experimental verification shows that this method can realize the function of vehicle cooperative communication, solve the problems of low VANET link stability and broadcast storm, and has a good supporting role in establishing the practicality of vehicle cooperation in VANET.

Chronic renal failure (CRF) is a major public health problem worldwide. Earlier studies have revealed salutary effects of rhubarb extracts in CRF. In this study, we employed lipidomic and metabolomic ...approaches to identify the plasma biomarkers and to determine the effect of treatment with petroleum ether, ethyl acetate and n-butanol extracts of rhubarb in a rat model of CRF with adenine-induced chronic tubulointerstitial nephropathy. In addition, clinical biochemistry, histological evaluation and pro-fibrotic protein expression were analyzed. Significant changes were found between the CRF and control groups representing characteristic phenotypes of rats with CRF. Treatment with the three rhubarb extracts improved renal injury and dysfunction, either fully or partially reversed the plasma metabolites abnormalities and attenuated upregulation of pro-fibrotic proteins including TGF-β1, α-SMA, PAI-1, CTGF, FN and collagen-1. The nephroprotective effect of ethyl acetate extract was better than other extracts. The differential metabolites were closely associated with glycerophospholipid, fatty acid and amino acid metabolisms. The results revealed a strong link between renal tubulointerstitial fibrosis and glycerophospholipid metabolism and L-carnitine metabolism in the development of CRF. Amelioration of CRF with the three rhubarb extracts was associated with the delayed development and/or reversal the disorders in key metabolites associated with adenine-induced CRF.
Early detection is critical in prevention and treatment of kidney disease. However currently clinical laboratory and histopathological tests do not provide region-specific and accurate biomarkers for ...early detection of kidney disease. The present study was conducted to identify sensitive biomarkers for early detection and progression of tubulo-interstitial nephropathy in aristolochic acid I-induced rats at weeks 4, 8 and 12. Biomarkers were validated using aristolochic acid nephropathy (AAN) rats at week 24, adenine-induced chronic kidney disease (CKD) rats and CKD patients. Compared with control rats, AAN rats showed anemia, increased serum urea and creatinine, progressive renal interstitial fibrosis, activation of nuclear factor-kappa B, and up-regulation of pro-inflammatory, pro-oxidant, and pro-fibrotic proteins at weeks 8 and 12. However, no significant difference was found at week 4. Metabolomics identified 12-ketodeoxycholic acid, taurochenodesoxycholic acid, LPC(15:0) and docosahexaenoic acid as biomarkers for early detection of tubulo-interstitial nephropathy. With prolonging aristolochic acid I exposure, LPE(20:2), cholic acid, chenodeoxycholic acid and LPC(17:0) were identified as biomarkers for progression from early to advanced AAN and lysoPE(22:5), indoxyl sulfate, uric acid and creatinine as biomarkers of advanced AAN. These biomarkers were reversed by treatment of irbesartan and ergone in AAN rats at week 24 and adenine-induced CKD rats. In addition, these biomarkers were also reversed by irbesartan treatment in CKD patients.
Renal fibrosis is an inevitable outcome of various manifestations of progressive chronic kidney diseases (CKD). The need for efficacious treatment regimen against renal fibrosis can therefore not be ...overemphasized. Here we show a novel protective role of Bacteroides fragilis (B. fragilis) in renal fibrosis in mice. We demonstrate decreased abundance of B. fragilis in the feces of CKD patients and unilateral ureteral obstruction (UUO) mice. Oral administration of live B. fragilis attenuates renal fibrosis in UUO and adenine mice models. Increased lipopolysaccharide (LPS) levels are decreased after B. fragilis administration. Results of metabolomics and proteomics studies show decreased level of 1,5-anhydroglucitol (1,5-AG), a substrate of SGLT2, which increases after B. fragilis administration via enhancement of renal SGLT2 expression. 1,5-AG is an agonist of TGR5 that attenuates renal fibrosis by inhibiting oxidative stress and inflammation. Madecassoside, a natural product found via in vitro screening promotes B. fragilis growth and remarkably ameliorates renal fibrosis. Our findings reveal the ameliorative role of B. fragilis in renal fibrosis via decreasing LPS and increasing 1,5-AG levels.
