Prymnesium parvum is an environmentally harmful algae and well known for its toxic effects to the fish culture. However, there is a dearth of studies on the growth behavior of P. parvum and ...information on how the availability of nutrients and environmental factors affect their growth rate. To address this knowledge gap, we used a uniform design approach to quantify the effects of major nutrients (N, P, Si and Fe) and environmental factors (water temperature, pH and salinity) on the biomass density of P. parvum. We also generated the growth model for P. parvum as affected by each of these nutrients and environmental factors to estimate optimum conditions of growth. Results showed that P. parvum can reach its maximum growth rate of 0.789, when the water temperature, pH and salinity is 18.11 °C, 8.39, and 1.23‰, respectively. Moreover, maximum growth rate (0.895-0.896) of P. parvum reached when the concentration of nitrogen, phosphorous, silicon and iron reach 3.41, 1.05, 0.69 and 0.53 mg/l, respectively. The order of the effects of the environmental factors impacting the biomass density of P. parvum was pH > salinity > water temperature, while the order of the effects of nutrients impacting the biomass density of P. parvum was nitrogen > phosphorous > iron > silicon. These findings may assist to implement control measures of the population of P. parvum where this harmful alga threatens aquaculture industry in the waterbodies such as Ningxia region in China.
Platycodon grandiflorum is an edible and medicinal plant, and polysaccharides are one of its important components. To further improve the utilization rate of P. grandiflorum, we investigated the ...effects of four different extraction methods, including hot water, ultrasonic-assisted, acid-assisted, and alkali-assisted extractions, on the polysaccharides, which were named PG-H, PG-U, PG-C, and PG-A. The findings indicated that the extraction method had a significant impact on the yield, characteristics, and immunoregulatory activity. We observed that the yields decreased in the following order: PG-H, PG-U, PG-C, and PG-A. Galacturonic acid, glucose, galactose, and arabinose were the most prevalent monosaccharides in the four PGs. However, their proportions varied. In addition, the difference between the content of glucose and galacturonic acid was more significant. PG-U had the highest glucose content, whereas PG-C had the lowest. Galacturonic acid content was highest in PG-A, while the lowest in PG-U. The molecular weight decreased in the order of PG-U, PG-H, PG-C, and PG-A; the particle size was in the order of PG-U, PG-A, PG-H, and PG-C. Moreover, the extraction method had a great impact on immunoregulatory activity. The ability to stimulate the immune function of macrophages was as follows: PG-A > PG-C > PG-U > PG-H. The results indicated that PGs, with lower molecular weights and higher GalA content, exhibited better immune-stimulating activity. And more important the AAE method was a good way to extract polysaccharides from Platycodon grandiflorum for use as a functional product and immunological adjuvant.
Hybrid poly(lactic acid)/titania (PLA/TiO2) fibrous membranes exhibiting excellent air filtration performance and good antibacterial activity were prepared via the electrospinning technique. By ...varying the composition of the precursor solutions and the relative humidity, the morphologies of PLA/TiO2 fibers, including the nanopores and nanometer-scale protrusions on the surface of the fibers, could be regulated. The distribution of nanopores and TiO2 nanoparticles on the surface of PLA/TiO2 fibers was investigated. Nitrogen adsorption-desorption analysis revealed that nanopores and nanometer-scale protrusions play an important role in improving the specific surface area and nanopore volume of the relevant PLA/TiO2 fibrous membrane. Filtration performance tests conducted by measuring the penetration of sodium chloride aerosol particles with a 260 nm mass median diameter indicated that fibers with a high surface roughness, large specific surface area, and large nanopore volume greatly improved the particle capture efficiency and facilitated the penetration of airflow. Furthermore, the introduction of TiO2 nanoparticles endows the relevant fibrous membrane with antibacterial properties. The as-prepared PLA/TiO2 fibrous membrane loaded with 1.75 wt% TiO2 nanoparticles formed at a relative humidity of 45% exhibited a high filtration efficiency (99.996%) and a relatively low pressure drop (128.7 Pa), as well as a high antibacterial activity of 99.5%.
Non-small cell lung cancer (NSCLC) is a highly lethal disease and the majority of NSCLC patients are desperate for therapies that can effectively target their cancer and ultimately improve their ...overall survival. Docetaxel (DTX) represents the first-line of the antitumor agent that is used to treat NSCLC; however, it has poor solubility in water and unsatisfactory encapsulation efficiency. In our study, exosomes were isolated from A549 cancer cells by ultracentrifugation and then characterized using transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), and Western blot (WB). The particle size changes of EXO and EXO-DTX were measured daily for seven days to test the stability. DTX was selected payload by electroporation (EXO-DTX). For the in vitro evaluation, cell proliferation, cell cycle, cell apoptosis, reactive oxygen species (ROS) assay and cellular uptake were evaluated in the A549 cells. Also, this study evaluated the target and therapeutic effect of DTX as an antitumor agent in vivo. As a result, EXO-DTX with a particle size of 149.5 nm were successfully prepared and the cytotoxicity of the EXO-DTX was much greater than that of DTX monomers. Exosomes significantly increased the cellular uptake in vitro evaluation and showed better targeting to tumor tissue compared to the free DTX in the mice. We also explored the potential of tumor cell-derived exosomes as a drug delivery agent to target the parent cancer. Hence, we conclude that exosomes might be used as a potential antitumor drug delivery system (DDS).
For hundreds of years, Atractylodes chinensis (DC.) Koidz. (AK) has been widely used as a treatment for spleen and stomach diseases in China. The AK polysaccharides (AKPs) have been thought to be the ...important bioactive components. In this stud, the impacts of different extraction methods were analyzed. The differences between AKPs extracted by hot water extraction (HWE), AKPs extracted by ultrasonic extraction (UAE), and AKPs extracted by enzyme extraction (EAE) were compared in terms of yield, total carbohydrate content, molecular weight distribution, monosaccharide composition, and synergistic activity of the AKPs with apatinib were determined. The results indicated that the yield of the polysaccharide obtained from HWE was higher than that of UAE and EAE. However, activity assays indicated that UAE-AKPs and HWE-AKPs enhanced apoptosis of human gastric cancer cells (SGC-7901) treated with apatinib and UAE-AKPs showed the strongest synergistic activities. This is also in agreement with the fact that UAE-AKPs have a smaller molecular weight, β-configuration, and higher galactose content. These findings suggested that UAE is an efficient and environmentally friendly method for producing new polysaccharides from Atractylodes chinensis (DC.) Koidz. for the development of natural synergist and for the treatment of gastric cancer.
The Diannong River, a valuable river and lake resource of the northern Ningxia Yellow River Irrigation Area, plays an instrumental role in regional flood control, drought resistance, climate ...regulation, and biodiversity conservation. Phytoplankton and zooplankton, as crucial elements of the aquatic ecosystem, have their distribution patterns evaluated and potential influencing factors identified, thereby enhancing the understanding of community distribution patterns. Nested structures and interspecies interaction relationships bear significant implications for community distribution patterns, functions, and stability. The upstream section of the Diannong River in Yinchuan City was chosen as the study object. Water samples were collected in January, April, July, and October 2021, and the community composition of phytoplankton and zooplankton was analyzed using relative abundance, density, and biomass. The distribution matrix temperature and bipartite network methodologies were deployed to investigate their nested pattern and interaction network seasonal characteristics. The findings indicate that the water environment of the Diannong River’s upstream section displays pronounced spatiotemporal heterogeneity, characterized by weak alkalinity and high fluoride content. The plankton community composition and relative abundance showed marked differences among the distinct sampling periods. The temperature of the random distribution matrix shows a significant difference compared to the zero-sum model, revealing a notable nested pattern in plankton in the Diannong River’s upstream section. The bipartite network suggests that the plankton composition was the simplest in January and the most complex in July, with the fiercest species competition observed in January and the lowest levels of species specificity, vulnerability, and generality. Water temperature (WT), dissolved oxygen (DO), total phosphorus (TP), available phosphorus (AP), CODCr, F−, and Cl− constitute the environmental parameters influencing the overall structure of the phytoplankton community in the Diannong River’s upstream section, whereas zooplankton did not present a significant correlation with water environmental factors.
