The incidence of resistant fungal pathogens has been increasing, especially in immuno-compromised people. As such, considerable research has been focused on discovering anti-fungal agents with new ...mechanisms of action and on optimizing the use of existing agents. In this context, interest in the polyene group of anti-fungals has recently been renewed, since they are known to be effective against a broad spectrum of fungal pathogens that only rarely develop a resistance to them. In the past 10 years considerable efforts have been made to improve their efficacy and, simultaneously, to reduce their toxicity. Knowledge about the basic mechanisms of their action will be of crucial importance to further optimizing their use. The mechanisms of polyene action at the membrane level are reviewed here, focusing primarily on their pore-forming activity and on the resulting osmotic responses of artificial lipid vesicles and different eukaryotic cells.

Piezo1 is a membrane nonspecific cation channel involved in red blood cells (RBCs) in the regulation of their volume. Recently, it was shown that it is distributed on the RBC membrane in a nonuniform ...manner. Here it is shown that it is possible to interpret the lateral distribution of Piezo1 molecules on RBC membrane by the curvature dependent Piezo1-bilayer interaction which is the consequence of the mismatch between the intrinsic principal curvatures of the Piezo1 trimer and the principal curvatures of the membrane at Piezo1's location but without its presence. This result supports the previously proposed model for the role of Piezo1 in the regulation of RBC volume.
The cell volume changes induced by hypotonic electrolyte and sucrose solutions were studied in Chinese-hamster-ovary epithelial cells. The effects in the solutions with osmolarities between 32 and ...315 mosM/L and distilled water were analyzed using bright-field and fluorescence confocal microscopy. The changes of the cell volume, accompanied by the detachment of cells, the formation of blebs, and the occurrence of almost spherical vesicle-like cells ("cell-vesicles"), showed significant differences in the long-time responses of the cells in the electrolyte solutions compared with the sucrose-containing solutions. A theoretical model based on different permeabilities of ions and sucrose molecules and on the action of Na
/K
-ATPase pumps is applied. It is consistent with the observed temporal behavior of the cells' volume and the occurrence of tension-induced membrane ruptures and explains lower long-time responses of the cells in the sucrose solutions.
The responses of Chinese hamster ovary epithelial cells, caused by the pore-forming agent nystatin, were investigated using brightfield and fluorescence microscopy. Different phenomena, i.e., the ...detachment of cells, the formation of blebs, the occurrence of "cell-vesicles" and cell ruptures, were observed. These phenomena were compared to those discovered in giant lipid vesicles. A theoretical model, based on the osmotic effects that occur due to the size-discriminating nystatin transmembrane pores in lipid vesicles, was extended with a term that considers the conservation of the electric charge density in order to describe the cell's behavior. The increase of the cellular volume was predicted and correlated with the observed phenomena.
β(2)-glycoprotein I (β(2)GPI) is a plasma protein capable of binding reversibly to membranes, and is classified among the amphitropic proteins. Part of the protein intercalates into the outer ...membrane leaflet, altering the difference between the preferred areas of the membrane leaflets, which results in membrane shape transformations. Budding, as a specific example of such shape transformations, was studied using giant unilamellar vesicles. Our aim was to identify the vesicle parameters that influence the degree of membrane budding by studying this process qualitatively and quantitatively. A simple theoretical model has been developed and assessed against the experimental observations. The results show that β(2)GPI binds in a concentration dependent manner, causing transitions between vesicle shapes with increasing numbers of buds. Higher numbers of buds are characteristic of larger and/or more flaccid vesicles. When the vesicle membrane is strained, a higher β(2)GPI concentration is needed to produce the same effects as on the unstrained vesicle. Vesicles were found to be highly individual in their behaviour, so each was treated individually. Specific vesicle behaviour was found to be the consequence of the neck between the main vesicle body and the buds, which could be either open, closed for the exchange of solution, or closed for the exchange of both solution and membrane.
Activation of carbonyl moiety is one of the most rudimentary approaches in organic synthesis and is crucial for a plethora of industrial-scale condensation reactions. In esterification and aldol ...condensation, which represent two of the most important reactions, the susceptibility of the carbonyl group to nucleophile attack allows the construction of a variety of useful organic compounds. In this context, there is a constant need for development of and improvement in the methods for addition-elimination reactions via activation of carbonyl functionality. In this paper, an advanced methodology for the direct esterification of carboxylic acids and alcohols, and for aldol condensation of aldehydes using widely available, inexpensive, and metal-free 1,3-dibromo-5,5-dimethylhydantoin under neat reaction conditions is reported. The method is air- and moisture-tolerant, allowing simple synthetic and isolation procedures for both reactions presented in this paper. The reaction pathway for esterification is proposed and a scale-up of certain industrially important derivatives is performed.

-bromosuccinimide (NBS) has been promoted ...as the most efficient and selective catalyst among the NXSs in the reaction of direct esterification of aryl and alkyl carboxylic acids. Comprehensive esterification of substituted benzoic acids, mono-, di- and tri-carboxy alkyl derivatives has been performed under neat reaction conditions. The method is metal-free, air- and moisture-tolerant, allowing for a simple synthetic and isolation procedure as well as the large-scale synthesis of aromatic and alkyl esters with yields up to 100%. Protocol for the recycling of the catalyst has been proposed.
A red blood cell (RBC) performs its function of adequately carrying respiratory gases in blood by its volume being ∼60% of that of a sphere with the same membrane area. For this purpose, human and ...most other vertebrate RBCs regulate their content of potassium (K+) and sodium (Na+) ions. The focus considered here is on K+ efflux through calcium-ion (Ca2+)-activated Gárdos channels. These channels open under conditions that allow Ca2+ to enter RBCs through Piezo1 mechanosensitive cation-permeable channels. It is postulated that the fraction of open Piezo1 channels depends on the RBC shape as a result of the curvature-dependent Piezo1-bilayer membrane interaction. The consequences of this postulate are studied by introducing a simple model of RBC osmotic behavior supplemented by the dependence of RBC membrane K+ permeability on the reduced volume (i.e., the ratio of cell volume to its maximal possible volume) of RBC discoid shapes. It is assumed that because of its intrinsic curvature and strong interaction with the surrounding membrane, Piezo1 tends to concentrate in the dimple regions of these shapes, and the fraction of open Piezo1 channels depends on the membrane curvature in that region. It is shown that the properties of the described model can provide the basis for the formation of the negative feedback loop that interrelates cell volume and its content of potassium ions. The model predicts the relation, valid for each cell in an RBC population, between RBC volume and membrane area, thus explaining the large value of the measured membrane area versus the volume correlation coefficient. The mechanism proposed here for RBC volume regulation is in accord with the loss of this correlation in RBCs of Piezo1 knockout mice.
A sufficiently large force acting on a single point of the fluid membrane of a flaccid phospholipid vesicle is known to cause the formation of a narrow bilayer tube (tether). We analyze this ...phenomenon by means of general mathematical methods allowing us to determine the shapes of strongly deformed vesicles including their stability. Starting from a free vesicle with an axisymmetric, prolate equilibrium shape, we consider an axial load that pulls (or pushes) the poles of the vesicle apart. Arranging the resulting shapes of strained vesicles in dependence of the axial deformation and of the area difference of monolayers, phase diagrams of stable shapes are presented comprising prolate shapes with or without equatorial mirror symmetry. For realistic values of membrane parameters, we study the force–extension relation of strained vesicles, and we demonstrate in detail how the initially elongated shape of an axially stretched vesicle transforms into a shape involving a membrane tether. This tethering transition may be continuous or discontinuous. If the free vesicle is mirror symmetric, the mirror symmetry is broken as the tether forms. The stability analysis of tethered shapes reveals that, for the considered vesicles, the stable shape is always asymmetric (polar), i.e., it involves only a single tether on one side of the main vesicle body. Although a bilayer tube formed from a closed vesicle is not an ideal cylinder, we show that, for most practical purposes, it is safe to assume a cylindrical geometry of tethers. This analysis is supplemented by the documentation of a prototype experiment supporting our theoretical predictions. It shows that the currently accepted model for the description of lipid-bilayer elasticity (generalized bilayer couple model) properly accounts for the tethering phenomenon.
Bioactive compounds from natural sources are of great importance because of their potential pharmacological activity and tremendous structural diversity. In this study, the chemical composition of ...different moss extracts of Hedwigia ciliata P. Beauv. have been examined, as well as their antioxidant, antineurodegenerative/anti-neuroinflammatory, antidiabetic, and antiproliferative potential. The extracts were prepared by Soxhlet extractor using solvents of different polarity. Chemical characterization of the extracts revealed the presence of phenolics and flavonoid compounds, together with triterpenoids as secondary metabolites of high biological activity. Significant antioxidant properties of all the extracts were exhibited using the β-carotene assay. The highest activities were found for water:ethanol extract (with the highest inhibition rate of 96%), but also significant inhibition was measured for ethanol and ethyl acetate extracts (80% and 70%, respectively). Confirmation of biocompatibility of investigated moss extracts has been performed using normal human fibroblast cell line, MRC-5. The H. ciliata extracts exhibited significant antiproliferative activity (~ 50%) against the MDA-MB-231 (human breast adenocarcinoma cell line), which has not previously been reported elsewhere. The Griess assay confirmed the potential anti-neuroinflammatory activity of the extracts, as significant effects in reducing NO production by LPS-stimulated BV2 (normal murine microglia cell line) was observed. This data is in line with noted antineurodegenerative potential measured by the inhibition of acetylcholinesterase (with the highest inhibition rate of 60% for ethyl acetate extract) and tyrosinase (with the highest inhibition rate of 70% for ethanol extract). Additionally, the H. ciliata extracts exhibited significant antidiabetic effect mediated by α-glucosidase inhibition (with the highest inhibition rate of 80% for ethyl acetate extract). The obtained data suggest the presence of immunomodulatory effects of the moss extracts in vitro, which allows the design of new experiments aimed at detecting and characterizing bioactive compounds of the extracts and additionally elucidate detailed mechanisms of their effects.
