Abstract
Radar-based convective modes were assigned to a sample of tornadoes and significant severe thunderstorms reported in the contiguous United States (CONUS) during 2003–11. The significant hail ...(≥2-in. diameter), significant wind (≥65-kt thunderstorm gusts), and tornadoes were filtered by the maximum event magnitude per hour on a 40-km Rapid Update Cycle model horizontal grid. The filtering process produced 22 901 tornado and significant severe thunderstorm events, representing 78.5% of all such reports in the CONUS during the sample period. The convective mode scheme presented herein begins with three radar-based storm categories: 1) discrete cells, 2) clusters of cells, and 3) quasi-linear convective systems (QLCSs). Volumetric radar data were examined for right-moving supercell (RM) and left-moving supercell characteristics within the three radar reflectivity designations. Additional categories included storms with marginal supercell characteristics and linear hybrids with a mix of supercell and QLCS structures. Smoothed kernel density estimates of events per decade revealed clear geographic and seasonal patterns of convective modes with tornadoes. Discrete and cluster RMs are the favored convective mode with southern Great Plains tornadoes during the spring, while the Deep South displayed the greatest variability in tornadic convective modes in the fall, winter, and spring. The Ohio Valley favored a higher frequency of QLCS tornadoes and a lower frequency of RM compared to the Deep South and the Great Plains. Tornadoes with nonsupercellular/non-QLCS storms were more common across Florida and the high plains in the summer. Significant hail events were dominated by Great Plains supercells, while variations in convective modes were largest for significant wind events.
Abstract
This study examines the possibility that supercell tornado forecasts could be improved by utilizing the storm-relative helicity (SRH) in the lowest few hundred meters of the atmosphere ...(instead of much deeper layers). This hypothesis emerges from a growing body of literature linking the near-ground wind profile to the organization of the low-level mesocyclone and thus the probability of tornadogenesis. This study further addresses the ramifications of near-ground SRH to the skill of the significant tornado parameter (STP), which is probably the most commonly used environmental indicator for tornadic thunderstorms. Using a sample of 20 194 severe, right-moving supercells spanning a 13-yr period, sounding-derived parameters were compared using forecast verification metrics, emphasizing a high probability of detection for tornadic supercells while minimizing false alarms. This climatology reveals that the kinematic components of environmental profiles are more skillful at discriminating significantly tornadic supercells from severe, nontornadic supercells than the thermodynamic components. The effective-layer SRH has by far the greatest forecast skill among the components of the STP, as it is currently defined. However, using progressively shallower layers for the SRH calculation leads to increasing forecast skill. Replacing the effective-layer SRH with the 0–500 m AGL SRH in the formulation of STP increases the number of correctly predicted events by 8% and decreases the number of missed events and false alarms by 18%. These results provide promising evidence that forecast parameters can still be improved through increased understanding of the environmental controls on the processes that govern tornado formation.
The life long relationship between herpes simplex virus and its host hinges on the ability of the virus to aggressively replicate in epithelial cells at the site of infection and transport into the ...nervous system through axons innervating the infection site. Interaction between the virus and the sensory neuron represents a pivot point where largely unknown mechanisms lead to a latent or a lytic infection in the neuron. Regulation at this pivot point is critical for balancing two objectives, efficient widespread seeding of the nervous system and host survival. By combining genetic and in vivo in approaches, our studies reveal that the balance between latent and lytic programs is a process occurring early in the trigeminal ganglion. Unexpectedly, activation of the latent program precedes entry into the lytic program by 12 -14hrs. Importantly, at the individual neuronal level, the lytic program begins as a transition out of this acute stage latent program and this escape from the default latent program is regulated by de novo VP16 expression. Our findings support a model in which regulated de novo VP16 expression in the neuron mediates entry into the lytic cycle during the earliest stages of virus infection in vivo. These findings support the hypothesis that the loose association of VP16 with the viral tegument combined with sensory axon length and transport mechanisms serve to limit arrival of virion associated VP16 into neuronal nuclei favoring latency. Further, our findings point to specialized features of the VP16 promoter that control the de novo expression of VP16 in neurons and this regulation is a key component in setting the balance between lytic and latent infections in the nervous system.
Abstract
A sample of 22 901 tornado and significant severe thunderstorm events, filtered on an hourly 40-km grid, was collected for the period 2003–11 across the contiguous United States (CONUS). ...Convective mode was assigned to each case via manual examination of full volumetric radar data (Part I of this study), and environmental information accompanied each grid-hour event from the hourly objective analyses calculated and archived at the Storm Prediction Center (SPC). Sounding-derived parameters related to supercells and tornadoes formed the basis of this investigation owing to the dominance of right-moving supercells in tornado production and the availability of supercell-related convective parameters in the SPC environmental archive. The tornado and significant severe thunderstorm events were stratified by convective mode and season. Measures of buoyancy discriminated most strongly between supercell and quasi-linear convective system (QLCS) tornado events during the winter, while bulk wind differences and storm-relative helicity were similar for both supercell and QLCS tornado environments within in each season. The larger values of the effective-layer supercell composite parameter (SCP) and the effective-layer significant tornado parameter (STP) favored right-moving supercells that produced significant tornadoes, as opposed to weak tornadoes or supercells that produced only significant hail or damaging winds. Additionally, mesocyclone strength tended to increase with increasing SCP for supercells, and STP tended to increase as tornado damage class ratings increased. The findings underscore the importance of convective mode (discrete or cluster supercells), mesocyclone strength, and near-storm environment (as represented by large values of STP) in consistent, real-time identification of intense tornadoes.
Novel fluorescent Langmuir-Blodgett (LB) films have been constructed from three different amphiphilic dicynaoquinodimethanes (DADQs). The DADQs varied in functional group structure, which had an ...impact on the LB film structure and the fluorescence properties. As the fluorescence of DADQs competes with non-radiative decay (conformational change), the packing and/or free volume in the LB film will influence the average fluorescence lifetime and integrated intensity. The pristine (blank) LB films were then exposed to a selection of non-fluorescent target analytes (some with environmental relevance) and the fluorescence was measured and analyzed relative to the pristine LB film. Exposure of the LB films to selected target analytes results in a modulation of the fluorescence, both with respect to average fluorescence lifetime and integrated intensity. The modulation of the fluorescence is different for different DADQ LB films and can be attributed to restricted non-radiative decays or charge transfer reactions between target analyte and DADQ LB film. The response from the DADQ LB films shows that these systems can be developed into sensing surfaces based on fluorescence measurements.
Infection and life-long residence in the human nervous system is central to herpes simplex virus (HSV) pathogenesis. Access is gained through innervating axonal projections of sensory neurons. This ...distinct mode of entry separates the viral genome from tegument proteins, including the potent transactivator of viral IE genes, VP16. This, in turn, promotes a balance between lytic and latent infection which underlies the ability of the virus to invade, disseminate, and set up a large reservoir of latent infections. In the mouse ocular model, TG neurons marked as either "latent" or "lytic" at 48 h postinfection indicated that these programs were selected early and were considered distinct and mutually exclusive. More recently, a temporal analysis of viral program selection revealed a default latent-like state that begins at ~18 h postinfection and in individual neurons, precedes entry into the viral lytic cycle. Studies using refined viral mutants demonstrated that transition out of this latent program depended upon the transactivation function of VP16. Pursuit of the apparent incongruity between the established leaky-late kinetics of VP16 expression with a "preimmediate-early" function led to the discovery of an unrecognized regulatory feature of the HSV-1 VP16 promoter near/downstream of its TATA box. Among three potential sites identified was a putative Egr-1/Sp1 site. Here, we report that a refined mutation of this site, while having no impact on replication in cultured cells or cornea, resulted in ~100-fold reduction in lytic infection in TG
. Notably, the HSV-2 VP16 promoter has 13 direct tandem-repeats
of its TATA box forming multiple potential overlapping Egr-1/Sp1 sites. Thus, despite different structures, these promoters might share function in directing the preimmediate-early VP16 protein expression. To test this, the HSV-1 VP16 promoter/5'UTR was replaced by the HSV-2 VP16 promoter/5'UTR in the HSV-1 backbone. Compared to the genomically repaired isolate, the HSV-2 VP16 promoter/5'UTR (1) accelerated the transition into the lytic cycle, and enhanced (2) virulence, and (3) entry into the lytic cycle following a reactivation stressor. These gain-of-function phenotypes support the hypothesis that the VP16 promoter regulates the latent/lytic boundary in neurons and that the HSV-1 and HSV-2 promoter/5'UTRs encode distinct thresholds for this transition.
A fundamental question in herpes simplex virus (HSV) pathogenesis is the consequence of viral reactivation to the neuron. Evidence supporting both post-reactivation survival and demise is published. ...The exceedingly rare nature of this event at the neuronal level in the sensory ganglion has limited direct examination of this important question. In this study, an in-depth in vivo analysis of the resolution of reactivation was undertaken. Latently infected C57BL/6 mice were induced to reactivate in vivo by hyperthermic stress. Infectious virus was detected in a high percentage (60-80%) of the trigeminal ganglia from these mice at 20 hours post-reactivation stimulus, but declined by 48 hours post-stimulus (0-13%). With increasing time post-reactivation stimulus, the percentage of reactivating neurons surrounded by a cellular cuff increased, which correlated with a decrease in detectable infectious virus and number of viral protein positive neurons. Importantly, in addition to intact viral protein positive neurons, fragmented viral protein positive neurons morphologically consistent with apoptotic bodies and containing cleaved caspase-3 were detected. The frequency of this phenotype increased through time post-reactivation. These fragmented neurons were surrounded by Iba1+ cells, consistent with phagocytic removal of dead neurons. Evidence of neuronal destruction post-reactivation prompted re-examination of the previously reported non-cytolytic role of T cells in controlling reactivation. Latently infected mice were treated with anti-CD4/CD8 antibodies prior to induced reactivation. Neither infectious virus titers nor neuronal fragmentation were altered. In contrast, when viral DNA replication was blocked during reactivation, fragmentation was not observed even though viral proteins were expressed. Our data demonstrate that at least a portion of reactivating neurons are destroyed. Although no evidence for direct T cell mediated antigen recognition in this process was apparent, inhibition of viral DNA replication blocked neuronal fragmentation. These unexpected findings raise new questions about the resolution of HSV reactivation in the host nervous system.
Abstract
In this study, a 13-yr climatology of tornado event and warning environments, including metrics of tornado intensity and storm morphology, is investigated with particular focus on the ...environments of tornadoes associated with quasi-linear convective systems and right-moving supercells. The regions of the environmental parameter space having poor warning performance in various geographical locations, as well as during different times of the day and year, are highlighted. Kernel density estimations of the tornado report and warning environments are produced for two parameter spaces: mixed-layer convective available potential energy (MLCAPE) versus 0–6-km vector shear magnitude (SHR6), and mixed-layer lifting condensation level (MLLCL) versus 0–1-km storm-relative helicity (SRH1). The warning performance is best in environments characteristic of severe convection (i.e., environments featuring large values of MLCAPE and SHR6). For tornadoes occurring during the early evening transition period, MLCAPE is maximized, MLLCL heights decrease, SHR6 and SRH1 increase, tornadoes rated as 2 or greater on the enhanced Fujita scale (EF2+) are most common, the probability of detection is relatively high, and false alarm ratios are relatively low. Overall, the parameter-space distributions of warnings and events are similar; at least in a broad sense, there is no systematic problem with forecasting that explains the high overall false alarm ratio, which instead seems to stem from the inability to know which storms in a given environment will be tornadic.
Abstract
The southeastern United States has become a prime area of focus in tornado-related literature due, in part, to the abundance of tornadoes occurring in high-shear low-CAPE (HSLC) ...environments. Through this analysis of 4133 tornado events and 16 429 tornado warnings in the southeastern United States, we find that tornadoes in the Southeast do indeed have, on average, higher shear and lower CAPE than tornadoes elsewhere in the contiguous United States (CONUS). We also examine tornado warning skill in the form of probability of detection (POD; percent of tornadoes receiving warning prior to tornado occurrence) and false alarm ratio (FAR; percent of tornado warnings for which no corresponding tornado is detected), and find that, on average, POD is better and FAR is worse for tornadoes in the Southeast than for the CONUS as a whole. These measures of warning skill remain consistent even when we consider only HSLC tornadoes. The Southeast also has nearly double the CONUS percentage of deadly tornadoes, with the highest percentage of these deadly tornadoes occurring during the spring, the winter, and around local sunset. On average, however, the tornadoes with the lowest POD also tend to be those that are weakest and least likely to be deadly; for the most part, the most dangerous storms are indeed being successfully warned.
We describe here a facile route for the in situ modification of the surface properties of fibers produced by electrospinning polystyrene containing small quantities of compatible polymer additives, ...end-functionalized with 1–3 fluoroalkyl groups. Such additives undergo spontaneous surface segregation during the electrospinning process, resulting in fibers with low surface energy, fluorine-rich, superhydrophobic surfaces. Surface properties were analyzed using static contact angle measurements (with water as the contact fluid) and X-ray photoelectron spectroscopy. We report the effect of a number of parameters on the surface properties of the resulting polystyrene fibers including the molecular weight and concentration of functionalized additive, the number of fluoroalkyl groups, the effect of annealing, and spinning solvent. The majority of the fibers were successfully produced using THF as the spinning solvent and fibers with a contact angle of ∼150° were attainable. However, preliminary investigations using a blend of polystyrene and 4 wt % of such an additive, end-functionalized with 3 C8F17 groups in a mixed solvent of DMF/THF (3:1 v/v), resulted in a mat of fibers with a superhydrophobic surface and a contact angle of 158°.
