3HBAY K 8644, a new class of pharmacologically potent compounds, the calcium channel activating dihydropyridines (DHP), were demonstrated in cultured myocardial cells.
...3HBAY K 8644 exhibited reversible and saturable binding to myocytes, and specific binding was Ca
2+-dependent. The equilibrium dissociation constant,
K
d, was 35.2 nM, and maximal binding capacity,
B
max, was 1.07 pmol/mg protein. Binding of the
3H-ligand was highly specific for various potently displacing DHP derivatives (either the calcium channel activating BAY K 8644, or the Ca
2+ entry blockers of the nifedipine type) with inhibition constants (
K
i values) in the nanomolar range. BAY K 8644, on the other hand, showed very low affinity to other receptors tested in brain and heart membranes. Displacement potency of BAY K 8644 correlated well with data of the functional pharmacology; e.g., the enhanced myocardial contractility. Results from competition studies using
3HBAY K 8644 and
3Hnimodipine support the conclusion that both the channel activating and inhibiting DHP structures interact with the same specific receptor site that might be associated with the putative Ca
2+-channel.
A specific polyclonal antiserum to human serum carnosinase was raised in rabbits and was used to prepare an agarose-protein A-antibody matrix. An antigen capture procedure showed that sera from ...homocarnosinosis patients, which lack carnosinase activity, contain an immunoreactive protein (M(r) 75,000) indistinguishable from the carnosinase band from normal serum. Other higher primates have active serum carnosinase and a similar immunoreactive M(r) 75,000 protein. The immunoaffinity matrix was used in a facile procedure to isolate pure carnosinase from human plasma with a yield of 69%. The antiserum inhibited human serum carnosinase strongly, but the maximum inhibition attained averaged only 71%. The antiserum inhibited human and chimpanzee serum carnosinases more effectively than gorilla or other higher primate serum carnosinases.
Vitrectomy and the subsequent vitreous fluid exchange alter the chemical and physical properties of the vitreous. This study assessed the time course alterations of total protein, ascorbic acid and ...lactic acid in the rabbit vitreous cavity following vitrectomy. Normal values were 2.2 +/- 1.2 mg/ml, 8.5 +/- 1.0 mg/100 ml and 16.8 +/- 3.5 mumol/ml, respectively. Nine groups of 5-10 adult albino rabbits underwent closed vitrectomy using physiologic saline, Ringer's lactate and BSS plus solutions. Samples were withdrawn at 3 and 24 h and 7 days after vitrectomy. Total protein levels increased in all groups at 3 and 24 h (20-30 mg/ml). Ascorbic acid measurements at 24 h rose only with saline and BSS plus. At 24 h the BSS plus group was the only one demonstrating an increased lactic acid concentration.
An outside rally at the Carnegie Mellon university (CMU) campus took place November 9, 1994. The action was called to protest a recent administration decision to ban students from gaining access to ...certain Internet news groups that contain materials school officials deem pornographic. CMU officials maintain that their decision was necessary to comply with a Pennsylvania law dealing with obscenity. "Our official policy is to obey the law." said Bill Arms, vice president for Computing Services. "There are bboards out there that are used to display sexually explicit material." A letter on behalf of the American Civil Liberties Union (ACLU) addressed to CMU president Robert Mehrabian explains that the "University's plan is inconsistent with the principles of academic freedom and free speech ... and is based on serious misreading of relevant laws." Comparing the Internet to campus libraries, which are exempt from the state's obscenity laws, the ACLU letter notes, "A library free from government control is an essential component of a vibrant university."

This article is a case study on the opening and management of a BBS(Bulletin Board System) computer network for the staff of institution, teacher of special education, and all pernonnel who work for ...the handicapped person in Aichi Prefecture. This BBS network (MARUMONET) got 78membership, 89 research data, and 61 statesment from the network members in fifteen months. Some problems, however, remain unsolved which might hinder the effectiveness of the network. The major problems are as follows; (i)The difficulty of the installation of the hard-and-software of networking for the members who are engaged in the daily work for the handicapped. (ii)Chronic shortage of the research data and the reaction to them which might reinforce the writing response of data. The function of the "local" BBS was discussed.
