Fertile queens and sterile workers are alternative forms of the adult female honeybee that develop from genetically identical larvae following differential feeding with royal jelly. We show that ...silencing the expression of DNA methyltransferase Dnmt3, a key driver of epigenetic global reprogramming, in newly hatched larvae led to a royal jelly-like effect on the larval developmental trajectory; the majority of Dnmt3 small interfering RNA-treated individuals emerged as queens with fully developed ovaries. Our results suggest that DNA methylation in Apis is used for storing epigenetic information, that the use of that information can be differentially altered by nutritional input, and that the flexibility of epigenetic modifications underpins, profound shifts in developmental fates, with massive implications for reproductive and behavioral status.
Honey bees are crucial for pollination services globally and produce important hive products including honey, royal jelly, pollen, and propolis that are being used commercially in food, cosmetics, ...and alternative medicinal purposes. Among the bee products, royal jelly (RJ) has long attracted scientists' interest because of its importance in honey caste differentiation. The present research was carried out to determine the acceptance rate of queen cells, and RJ production between the hygienic and non-hygienic lines. Further, this study unveils the effect of pollen substitute diets on the queen cell acceptance rate and RJ yields between both bee stocks. Results showed that the uncapped brood cells and dead brood's removal percentage was significantly more in hygienic bee colonies in comparison to non-hygienic bee colonies (p < 0.05). The average percentage of larval acceptance was statistically higher in hygienic lines (64.33 ± 2.91%) compared to non-hygienic lines (29.67 ± 1.20%). Similarly, the RJ mean weight per colony differed statistically between both bee stocks (p<0.001), which were 12.23 ± 0.52 g and 6.72 ± 0.33 g, respectively. Moreover, our results demonstrated that a significant difference was observed in larval acceptance rate, RJ yields (per colony and per cup) between both bee stocks those fed on various diets. However, no significant difference was recorded in RJ yields (per colony and per cup) between both bee stock that feeds on either commercially available pollen or pollen substitute. This study may provide future applications in helping bee breeders to choose the bees that carry a higher level of hygienic behavior with high RJ production traits.
Acute bee paralysis virus (ABPV), Kashmir bee virus (KBV) and Israeli acute paralysis virus (IAPV) are part of a complex of closely related viruses from the Family Dicistroviridae. These viruses have ...a widespread prevalence in honey bee (
Apis mellifera) colonies and a predominantly sub-clinical etiology that contrasts sharply with the extremely virulent pathology encountered at elevated titres, either artificially induced or encountered naturally. These viruses are frequently implicated in honey bee colony losses, especially when the colonies are infested with the parasitic mite
Varroa destructor. Here we review the historical and recent literature of this virus complex, covering history and origins; the geographic, host and tissue distribution; pathology and transmission; genetics and variation; diagnostics, and discuss these within the context of the molecular and biological similarities and differences between the viruses. We also briefly discuss three recent developments relating specifically to IAPV, concerning its association with Colony Collapse Disorder, treatment of IAPV infection with siRNA and possible honey bee resistance to IAPV.
Here we report the genome sequence of the honeybee Apis mellifera, a key model for social behaviour and essential to global ecology through pollination. Compared with other sequenced insect genomes, ...the A. mellifera genome has high A+T and CpG contents, lacks major transposon families, evolves more slowly, and is more similar to vertebrates for circadian rhythm, RNA interference and DNA methylation genes, among others. Furthermore, A. mellifera has fewer genes for innate immunity, detoxification enzymes, cuticle-forming proteins and gustatory receptors, more genes for odorant receptors, and novel genes for nectar and pollen utilization, consistent with its ecology and social organization. Compared to Drosophila, genes in early developmental pathways differ in Apis, whereas similarities exist for functions that differ markedly, such as sex determination, brain function and behaviour. Population genetics suggests a novel African origin for the species A. mellifera and insights into whether Africanized bees spread throughout the New World via hybridization or displacement.

•Through pollination, wild and managed bees are drivers of human and environmental health.•As OneHealth model systems, bees allow fundamental insights in topics including.•Ecological ...immunology, Social immunology, Disease transmission, Vector-borne transmission.
The OneHealth approach aims to further our understanding of the drivers of human, animal and environmental health, and, ultimately, to improve them by combining approaches and knowledge from medicine, biology and fields beyond. Wild and managed bees are essential pollinators of crops and wild flowers. Their health thus directly impacts on human and environmental health. At the same time, these bee species represent highly amenable and relevant model organisms for a OneHealth approach that aims to study fundamental epidemiological questions. In this review, we focus on how infectious diseases of wild and managed bees can be used as a OneHealth model system, informing fundamental questions on ecological immunology and disease transmission, while addressing how this knowledge can be used to tackle the issues facing pollinator health.
Recently, environmental risk and toxicity of neonicotinoid insecticides to honey bees have attracted extensive attention. However, toxicological understanding of neonicotinoid insecticides on gut ...microbiota is limited. In the present study, honey bees (Apis mellifera L.) were exposed to a series of nitenpyram for 14 days. Results indicated that nitenpyram exposure decreased the survival and food consumption of honey bees. Furthermore, 16S rRNA gene sequencing revealed that nitenpyram caused significant alterations in the relative abundance of several key gut microbiotas, which contribute to metabolic homeostasis and immunity. Using high-throughput RNA-Seq transcriptomic analysis, we identified a total of 526 differentially expressed genes (DEGs) that were significantly altered between nitenpyram-treated and control honey bee gut, including several genes related to metabolic, detoxification and immunity. In addition, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed nitenpyram affected several biological processes, of which most were related to metabolism. Collectively, our study demonstrates that the dysbiosis of gut microbiota in honey bee caused by nitenpyram may influence metabolic homeostasis and immunity of bees, and further decrease food consumption and survival of bees.

Nitenpyram affected metabolism and immunity putatively caused by disordered gut microbiota, further decreased food consumption and survival of bees.
Emerging diseases are among the greatest threats to honey bees. Unfortunately, where and when an emerging disease will appear are almost impossible to predict. The arrival of the parasitic Varroa ...mite into the Hawaiian honey bee population allowed us to investigate changes in the prevalence, load, and strain diversity of honey bee viruses. The mite increased the prevalence of a single viral species, deformed wing virus (DWV), from ~10 to 100% within honey bee populations, which was accompanied by a millionfold increase in viral titer and a massive reduction in DWV diversity, leading to the predominance of a single DWV strain. Therefore, the global spread of Varroa has selected DWV variants that have emerged to allow it to become one of the most widely distributed and contagious insect viruses on the planet.

Foto: Pixabay CC0.
