Declines in bee populations across the world threaten food security and ecosystem function. It is currently not possible to routinely predict which specific stressors lead to declines in different ...populations or contexts, hindering efforts to improve bee health. Genomics has the potential to dramatically improve our ability to identify, monitor and predict the effects of stressors, as well as to mitigate their impacts through the use of marker-assisted selection, RNA interference and potentially gene editing. Here we discuss the most compelling recent applications of genomics to investigate the mechanisms underpinning bee population declines and to improve the health of both wild and managed bee populations.
Dramatic losses of pollinating insects have become of global concern, as they threaten not only key ecosystem services but also human food production. Recent research provided evidence that ...interactions between ecological stressors are drivers of declining pollinator health and responsible for observed population collapses. We used the honeybee Apis mellifera and conducted a series of experiments to test for long-term effects of a single short exposure to the agricultural pesticide flupyradifurone to a second environmental stressor later in life. To do this, we exposed individuals during their larval development or early adulthood to sublethal dosages of flupyradifurone (0.025 μg for larvae and 0.645 μg for imagos), either pure or as part of an agricultural formulation (Sivanto). We afterwards exposed bees to a second ecological stressor infecting individuals with 10,000 spores of the fungal gut parasite Nosema ceranae. We found that pesticide exposures significantly reduced survival of bees and altered the expression of several immune and detoxification genes. The ability of bees to respond to these latter effects differed significantly between colonies, offering opportunities to breed bees with elevated levels of pesticide tolerance in the future. We conclude that short episodes of sublethal pesticide exposures during development are sufficient to trigger effects later in life and could therefore contribute to the widespread declines in bee health.
Methylation of DNA is an epigenetic mechanism that influences patterns of gene expression. DNA methylation marks contribute to adaptive phenotypic variation but are erased during development. The ...role of DNA methylation in adaptive evolution is therefore unclear. We propose that environmentally-induced DNA methylation causes phenotypic heterogeneity that provides a substrate for selection via forces that act on the epigenetic machinery. For example, selection can alter environmentally-induced methylation of DNA by acting on the molecular mechanisms used for the genomic targeting of DNA methylation. Another possibility is that specific methylation marks that are environmentally-induced, yet non-heritable, could influence preferential survival and lead to consistent methylation of the same genomic regions over time. As methylation of DNA is known to increase the likelihood of cytosine-to-thymine transitions, non-heritable adaptive methylation marks can drive an increased likelihood of mutations targeted to regions that are consistently marked across several generations. Some of these mutations could capture, genetically, the phenotypic advantage of the epigenetic mark. Thereby, selectively favored transitory alterations in the genome invoked by DNA methylation could ultimately become selectable genetic variation through mutation. We provide evidence for these concepts using examples from different taxa, but focus on experimental data on large-scale DNA sequencing that expose between-group genetic variation after bidirectional selection on honeybees, Apis mellifera.
Honeybees communicate the distance and location of resource patches by bee dances, but this spatial information has rarely been used to study their foraging ecology. We analysed, for the first time ...to the best of the authors' knowledge, foraging distances and dance activities of honeybees in relation to landscape structure, season and colony using a replicated experimental approach on a landscape scale. We compared three structurally simple landscapes characterized by a high proportion of arable land and large patches, with three complex landscapes with a high proportion of semi-natural perennial habitats and low mean patch size. Four observation hives were placed in the centre of the landscapes and switched at regular intervals between the six landscapes from the beginning of May to the end of July. A total of 1137 bee dances were observed and decoded. Overall mean foraging distance was 1526.1 ± 37.2 m, the median 1181.5 m and range 62.1-10 037.1 m. Mean foraging distances of all bees and foraging distances of nectar-collecting bees did not significantly differ between simple and complex landscapes, but varied between month and colonies. Foraging distances of pollen-collecting bees were significantly larger in simple (1743 ± 95.6 m) than in complex landscapes (1543.4 ± 71 m) and highest in June when resources were scarce. Dancing activity, i.e. the number of observed bee dances per unit time, was significantly higher in complex than in simple landscapes, presumably because of larger spatial and temporal variability of resource patches in complex landscapes. The results facilitate an understanding of how human landscape modification may change the evolutionary significance of bee dances and ecological interactions, such as pollination and competition between honeybees and other bee species.
Honey bees (Apis mellifera) and other pollinator populations are declining worldwide, and the reasons remain controversial. Based on laboratory testing, fungicides have traditionally been considered ...bee-safe. However, there have been no experimental tests of the effects of fungicides on colony health under field conditions, and limited correlational data suggests there may be negative impacts on bees at levels experienced in the field. We tested the effects of one of the most commonly used fungicides on colony health by feeding honey bee colonies pollen containing Pristine® (active ingredients: 25.2% boscalid, 12.8% pyraclostrobin) at four levels that bracketed concentrations we measured for pollen collected by bees in almond orchards. We also developed a method for calculating per-bee and per-larva dose. Pristine® consumption significantly and dose-dependently reduced worker lifespan and colony population size, with negative health effects observed even at the lowest doses. The lowest concentration we tested caused a 15% reduction in the worker population at an estimated dosage that was three orders of magnitude below the estimated LD15 values for previous acute laboratory studies. The enhanced toxicity under field conditions is at least partially due to activation of colonial nutritional responses missed by lab tests. Pristine® causes colonies to respond to perceived protein malnutrition by increasing colony pollen collection. Additionally, Pristine induces much earlier transitioning to foraging in individual workers, which could be the cause of shortened lifespans. These findings demonstrate that Pristine® can negatively impact honey bee individual and colony health at concentrations relevant to what they experience from pollination behavior under current agricultural conditions.

Capsule: Honey bee colonies exposed to concentrations of a mito-toxic fungicide in pollen at concentrations similar to what they receive while foraging in a sprayed orchard experienced a dose-dependent reduction in worker lifespan, reducing colony populations. Lethal effects were observed at concentrations three orders of magnitude lower than in lab studies demonstrating the need for re-evaluation of fungicide safety.
Plants may benefit from limiting the community of generalist floral visitors if the species that remain are more effective pollinators and less effective pollenivores. Plants can reduce access to ...pollen through altered floral cues or morphological structures, but can also reduce consumption through direct pollen defenses. We observed that Eucera (Peponapis) pruinosa, a specialist bee on Cucurbita plants, collected pure loads of pollen while generalist honey bees and bumble bees collected negligible amounts of cucurbit pollen, even though all groups of bees visited these flowers. Cucurbit flowers have no morphological adaptations to limit pollen collection by bees, thus we assessed their potential for physical, nutritional, and chemical pollen traits that might act as defenses to limit pollen loss to generalist pollinators. Bumble bee (Bombus impatiens) microcolonies experienced reduced pollen consumption, mortality, and reproduction as well as increased stress responses when exposed to nutritional and mechanical pollen defenses. These bees also experienced physiological effects of these defenses in the form of hindgut expansion and gut melanization. Chemical defenses alone increased the area of gut melanization in larger bees and induced possible compensatory feeding. Together, these results suggest that generalist bumble bees avoid collecting cucurbit pollen due to the physiological costs of physical and chemical pollen defenses.
Insects rely more on chemical signals (semiochemicals) than on any other sensory modality to find, identify, and choose mates. In most insects, pheromone production is typically regulated through ...biosynthetic pathways, whereas pheromone sensory detection is controlled by the olfactory system. Orchid bees are exceptional in that their semiochemicals are not produced metabolically, but instead male bees collect odoriferous compounds (perfumes) from the environment and store them in specialized hind-leg pockets to subsequently expose during courtship display. Thus, the olfactory sensory system of orchid bees simultaneously controls male perfume traits (sender components) and female preferences (receiver components). This functional linkage increases the opportunities for parallel evolution of male traits and female preferences, particularly in response to genetic changes of chemosensory detection (e.g. Odorant Receptor genes). To identify whether shifts in pheromone composition among related lineages of orchid bees are associated with divergence in chemosensory genes of the olfactory periphery, we searched for patterns of divergent selection across the antennal transcriptomes of two recently diverged sibling species Euglossa dilemma and E. viridissima.
We identified 3185 orthologous genes including 94 chemosensory loci from five different gene families (Odorant Receptors, Ionotropic Receptors, Gustatory Receptors, Odorant Binding Proteins, and Chemosensory Proteins). Our results revealed that orthologs with signatures of divergent selection between E. dilemma and E. viridissima were significantly enriched for chemosensory genes. Notably, elevated signals of divergent selection were almost exclusively observed among chemosensory receptors (i.e. Odorant Receptors).
Our results suggest that rapid changes in the chemosensory gene family occurred among closely related species of orchid bees. These findings are consistent with the hypothesis that strong divergent selection acting on chemosensory receptor genes plays an important role in the evolution and diversification of insect pheromone systems.
Imidacloprid is the most widely used insecticide in the world. In this study, we used spraying methods to simulate field exposures of bees to formulated imidacloprid (Advise® 2FL) alone and binary ...mixtures with seven pesticides from different classes. Synergistic toxicity was detected from mixtures of Advise (58.6 mg a.i./L imidacloprid)+Domark (512.5 mg a.i. /L tetraconazole), Advise+Transform (58.5 mg a.i./L sulfoxaflor), and Advise+Vydate (68 mg a.i./L oxamyl), and mortality was significantly increased by 20%, 15%, and 26% respectively. The mixtures of Advise+Bracket (88.3 mg a.i./L acephate) and Advise+Karate (62.2 mg a.i./L L-cyhalothrin) showed additive interaction, while Advise+Belay (9.4 mg a.i./L clothianidin) and Advise+Roundup (1217.5 mg a.i./L glyphosate) had no additive/synergistic interaction. Spraying bees with the mixture of all eight pesticides increased mortality to 100%, significantly higher than all other treatments. Except Bracket which significantly suppressed esterase and acetylcholinesterase (AChE) activities, other treatments of Advise-only and mixtures with other pesticides did not suppress enzyme activities significantly, including invertase, glutathione S-transferase (GST), and esterase and AChE. Immunity-related phenoloxidase (PO) activities in survivors tended to be more variable among treatments, but mostly still statistically similar to the control. By using specific enzyme inhibitors, we demonstrated that honey bees mainly rely on cytochrome P450 monooxygenases (P450s) for detoxifying Advise, while esterases and GSTs play substantially less roles in the detoxification. This study provided valuable information for guiding pesticide selection in premixing and tank mixing in order to alleviate toxicity risk to honey bees. Our findings indicated mixtures of Advise with detoxification-enzyme-inducing pesticides may help bees to detoxify Advise, while toxicity synergists may pose further risk to bees, such as the Bracket which not only suppressed esterase and AChE activities, but also increased toxicity to bees.
Brazil is the second largest producer of genetically modified plants in the world. This agricultural practice exposes native pollinators to contact and ingestion of Bacillus thuringiensis proteins ...(e.g. Cry toxins) from transgenic plants. Furthermore, native bees are also exposed to various herbicides applied to crops, including glyphosate. Little is known about the possible effects of glyphosate and Cry proteins on stingless bees, especially regarding exposure at an immature stage. Here, we show for the first time that glyphosate is lethal, and that Cry toxins (Cry1F, Cry2Aa) alter the development of the stingless bee Melipona quadrifasciata upon contamination of larval food. Glyphosate was very toxic to the bee larvae, killing all of them within only a few days of exposure. Bees treated with Cry2Aa proteins had a higher survival rate and were delayed in their development, compared to the negative controls. Those treated with the Cry1F protein also suffered delays in their development, compared to the negative controls. In conclusion, the proteins Cry1F, Cry2Aa, and the herbicide glyphosate were highly toxic to the stingless bee M. quadrifasciata, causing lethal or sublethal effects which can severely impair colony growth and viability, and reduce pollination ability.

Land-use change and intensification threaten bee populations worldwide, imperilling pollination services. Global models are needed to better characterise, project, and mitigate bees' responses to ...these human impacts. The available data are, however, geographically and taxonomically unrepresentative; most data are from North America and Western Europe, overrepresenting bumblebees and raising concerns that model results may not be generalizable to other regions and taxa. To assess whether the geographic and taxonomic biases of data could undermine effectiveness of models for conservation policy, we have collated from the published literature a global dataset of bee diversity at sites facing land-use change and intensification, and assess whether bee responses to these pressures vary across 11 regions (Western, Northern, Eastern and Southern Europe; North, Central and South America; Australia and New Zealand; South East Asia; Middle and Southern Africa) and between bumblebees and other bees. Our analyses highlight strong regionally-based responses of total abundance, species richness and Simpson's diversity to land use, caused by variation in the sensitivity of species and potentially in the nature of threats. These results suggest that global extrapolation of models based on geographically and taxonomically restricted data may underestimate the true uncertainty, increasing the risk of ecological surprises.
