Current strategies to produce homogeneous antibody-drug conjugates (ADCs) rely on mutations or inefficient conjugation chemistries. Here we present a strategy to produce site-specific ADCs using a ...highly reactive natural buried lysine embedded in a dual variable domain (DVD) format. This approach is mutation free and drug conjugation proceeds rapidly at neutral pH in a single step without removing any charges. The conjugation chemistry is highly robust, enabling the use of crude DVD for ADC preparation. In addition, this strategy affords the ability to precisely monitor the efficiency of drug conjugation with a catalytic assay. ADCs targeting HER2 were prepared and demonstrated to be highly potent and specific in vitro and in vivo. Furthermore, the modular DVD platform was used to prepare potent and specific ADCs targeting CD138 and CD79B, two clinically established targets overexpressed in multiple myeloma and non-Hodgkin lymphoma, respectively.
We present a new class of statistical de- anonymization attacks against high-dimensional micro-data, such as individual preferences, recommendations, transaction records and so on. Our techniques are ...robust to perturbation in the data and tolerate some mistakes in the adversary's background knowledge. We apply our de-anonymization methodology to the Netflix Prize dataset, which contains anonymous movie ratings of 500,000 subscribers of Netflix, the world's largest online movie rental service. We demonstrate that an adversary who knows only a little bit about an individual subscriber can easily identify this subscriber's record in the dataset. Using the Internet Movie Database as the source of background knowledge, we successfully identified the Netflix records of known users, uncovering their apparent political preferences and other potentially sensitive information.
Bisphenol A (BPA), a synthetic organic chemical, is creating a new category of ecological and human health challenges due to unintended leakage. Effectively managing the use and leakage of BPA can ...benefit from an understanding of the anthropogenic BPA cycles (i.e., the size of BPA flows and stocks). In this work, we provide a dynamic analysis of the anthropogenic BPA cycles in China for 2000–2014. We find that China’s BPA consumption has increased 10-fold since 2000, to ∼3 million tonnes/year. With the increasing consumption, China’s in-use BPA stock has increased 500-fold to 14.0 million tonnes (i.e., 10.2 kg BPA/capita). It is unclear whether a saturation point has been reached, but in 2004–2014, China’s in-use BPA stock has been increasing by 0.8 kg BPA/capita annually. Electronic products are the biggest contributor, responsible for roughly one-third of China’s in-use BPA stock. Optical media (DVD/VCD/CDs) is the largest contributor to China’s current End-of-Life (EoL) BPA flow, totaling 0.9 million tonnes/year. However, the EoL BPA flow due to e-waste will increase quickly, and will soon become the largest EoL BPA flow. The changing quantities and sources of EoL BPA flows may require a shift in the macroscopic BPA management strategies.

To what extent should multichannel retailers integrate assortments across channels? Previous literature controversially discusses the question of which integration strategy is most successful but arguments are only conceptual, and no empirical assessment exists. This article presents a framework that (a) shows how customers’ perceived shopping benefits of variety, convenience, and reduced risk mediate the impact of multichannel assortment integration (full, asymmetrical, no) on patronage intentions and (b) differentiates the impact for retailer types based on substitutive, complementary, and independent assortment relations. Two large-scale experimental studies empirically investigate whether a dominant integration strategy exists in the context of full and simultaneous information (Study 1) and more uncertain and subsequent information accessibility (Study 2). We consistently find that full integration dominates no integration across assortment relations, but asymmetrical integration—the strategy that is most often realized by multichannel retailers—can have a detrimental impact for substitutive relations compared with no integration. Asymmetrical integration can be more beneficial than full integration for independent relations, while customer outcomes differ less for complementary relations. Researchers and managers can use our findings to understand how shopping benefits of variety, convenience, and reduced risk explain the different customer outcomes of multichannel assortment integration, depending on retailer type.
The thesis will examine how DVD distribution can affect Japanese film dissemination in the UK. The media discourse concerning 4Digital Asia and Third Window proposes that this is the principal factor ...influencing their films' presence in the UK from 2008 to 2010. The distributors' actions establish the UK market that exists for Japanese films, and that these consumer demands are best pursued through the medium of DVD. As a result, the distribution and marketing materials for the film releases often highlight the discs and their DVD labels as much as the production background and content of the films. This fact leads to the following questions: how are DVDs of Japanese films made distinctive? How important is the DVD format itself, as well as the labeling of the films as Japanese? An effective methodological strategy is needed to answer these questions. The thesis' approach allows for an integrated analysis of relevant sources within the fields of Japanese film, DVD media, and film distribution and marketing. DVD packaging, special features, trailers, websites and reviews are all essential parts of 4Digital Asia and Third Window's DVD distribution processes. Multiple analytical approaches can integrate methods of close textual analysis required for the study of various sources that represent film distribution. Therefore, the discourses circulating the DVDs, and the influence of DVD dissemination strategies of Japanese films in the UK, will be investigated. They reveal that the distributors' practices communicate how Japanese cinema is distribution on DVD in the UK, and act as a means of identifying the market that exists for these films.
In 2012, Ciné-Tamaris released Tout(e) Varda, a DVD box set spanning filmmaker Agnès Varda's 60-year career to that date. Although relatively unusual in the degree of control Varda seems to have had ...in curating the collection, this box set is a logical development of Varda's work of the 2000s in which she has increasingly interrogated her oeuvre and career. This article argues for the significance of the collection - and the paratextual material it includes - for an analysis of Varda and her work. But it also seeks to position the analysis within the wider context of DVD scholarship, where - despite a recurring concern with both the commercial and didactic functions of auteurism - the authorial collection has attracted little attention. An analysis of Tout(e) Varda points to the importance of considering the formal qualities of paratextual material as well as their thematic concerns. Tout(e) Varda offers not a definitive commentary on Varda's work, but rather extends its formal and thematic preoccupations, albeit in ways which are at times contradictory, constructing Varda as both an unreliable curator and unreliable narrator.
Barium fluoride (BaF 2 ) is a fast inorganic crystal scintillator. Because of its fast scintillation with sub nanosecond decay time it is considered as a candidate crystal for a very fast crystal ...calorimetry for future HEP experiments at the intensity frontier. Two crucial issues of BaF 2 application are its radiation hardness and its slow scintillation component with 600 ns decay time, which causes pile-up. BaF 2 crystals produced by different vendors were irradiated by γ-rays up to 120 Mrad, and show good radiation hardness. A 20 cm long rare earth doped BaF 2 crystal grown by BGRI shows promising performance in slow component suppression.
