Jedná se o degenerativní, nevyléčitelné, terminální onemocnění. Postihuje až 75 % nemocných na celém světě postižených demencí. Prevalence tohoto onemocnění je vysoká, etiologie stále není známá. Je sledována řada rizikových faktorů. Pilotní studie projektu Epidemiologie a genetika Alzheimerovy choroby s 334 případy a 102 kontrolami z psychiatrických léčeben přináší následující výsledky z dotazníkového šetření a stěru z bukální sliznice. Dotazník se skládá z otázek zaměřených na základní identifikační údaje, vzdělání, životní styl, vaskulární rizikové faktory, zájmy, zaměstnání, výskyt nemoci v rodině, výsledky Minimental State Examination (MMSE). Dále jsou uvedeny výsledky analýzy polymorfizmů genu pro angiotenzinkonvertázu (ACE). Ve výsledcích je naznačeno, že u pacientů s ACH se v anamnéze častěji vyskytuje kardiovaskulární onemocnění a úraz hlavy. Výsledky analýz vztahu polymorfizmu I/D genu pro ACE k Alzheimerově chorobě odpovídají dosud publikovaným výsledkům, tzn., že genotypy II a ID mají vztah k riziku ACH, i když zatím nebyl zjištěn statisticky významný rozdíl ve sledovaných skupinách respondentů.- Alzheimer's disease (AD) is the most common form of dementia. It is a degenerative incurable terminal disease. AD affects a high percentage (75%) of persons with dementia all over the world. Prevalence is very high. Etiology is still unknown. Numerous etiological factors of AD have already been discovered. Pilot evaluation of 334 cases and 102 controls from mental hospitals in project Epidemiology and Genetics of Alzheimer's Disease provided following the results from questionaires and buccal mucosa smears. The questionnaires contain questions about identification, education, lifestyle, vascular risk factors, hobbies, occupation, incidence of the disease in the family, results of Minimental State Examination (MMSE).Results of the analysis of the gene of I/D polymorphism of ACE are presented. Results suggest that persons with AD have often a cardiovascular disease and head injury in their case history. Cilj istraživanja bio je ispitati pojavljuje li se završni stupanj kronične bubrežne bolesti (KBB) u oboljelih od autosomne dominantne policistične bubrežne bolesti (ADPBB) ranije u bolesnika rođenih ...u drugoj polovici 20. stoljeća u odnosu na rođene prije toga vremena. Podaci su prikupljeni za 70 bolesnika liječenih u Kliničkom bolničkom centru Osijek od 1992. do 2012. godine. Preuzeti su iz medicinskih zapisa ili prikupljeni anketom. U mlađih je ispitanika završni stupanj KBB-a nastupio značajno ranije u odnosu na starije (stariji=59±10, mlađi= 47±6, z=- 4,226, P=<0,001). U njih je i ranije postavljena dijagnoza bolesti. Nije bilo razlika u učestalosti komplikacija bolesti između mlađih i starijih bolesnika (hematurija, nefrolitijaza, infekcije mokraćnoga sustava, nefrektomija). U parovima roditelj - dijete, u djece je završni stupanj KBB-a nastupio značajno ranije nego u roditelja. Razlog može biti u genskoj anticipaciji, ali i promjeni prehrane, odnosno većega prehrambenog proteinskog opterećenja u mlađih generacija. Ovo je prvo epidemiološko istraživanje ADPBB-a u Hrvatskoj.

All forms of tro - phoblastic disease are characterized by rare incidence. This explains a potential risk of diagnostic and therapeutic errors which can lead to very serious consequences. The contemporary trend across different countries is to establish definite criteria for adequate management of patients with this specific disease which also allow the comparison of treatment results. Jejich působení na mnoha úrovních organismu koresponduje s bio-psycho-sociálním konceptem zdraví a nemoci. Mozek, chování a imunita jsou úzce propojeny a reagují na environmentální kontext. Účinky chronického psychosociálního stresu popisuje model alostatické zátěže, jenž ozřejmuje dynamičnost nastavení biologických procesů a význam mozku pro zpětnovazebnou regulaci v adaptaci na prostředí. Osobnost jedince, jeho kognitivní a emoční charakteristiky, kvalita sociálních vztahů a socioekonomické faktory modifikují stresovou zátěž a mají přímý i nepřímý vliv na zdraví. Řada účinků psychosociálních faktorů je zprostředkována epigeneticky; v období raného vývoje tento mechanismus formuje vulnerabilitu vůči rozvoji patologií souvisejících se stresem. Psychické, genetické, behaviorální a environmentální vlivy spolu průběžně interagují. Nepřímé účinky psychosociálních faktorů spočívají v modifikaci chování a životního stylu. Toto působení je obousměrné, neboť životní styl zpětně ovlivňuje psychické a fyzické zdraví. Přístupy, zaměřené na podporu odolnosti vůči stresu, mají důležité místo v prevenci.- Psychosocial factors significantly participate in the pathogenesis of complex diseases. Their effects correspond to the bio-psycho-social concept of health and disease on many levels. Brain functions, behaviour and immunity are closely linked and responsive to the environmental context. The effects of chronic psychosocial stress are described by the allostatic load model which highlights dynamic adjustment of biological processes and the importance of the brain for feedback regulation in adapting to the environment. The personality of an individual, his cognitive and emotional characteristics, quality of social relationships and socioeconomic factors modify stress load and have a direct and indirect impact on health. Many of the effects of psychosocial factors are mediated epigenetically. In the early development epigenetic mechanisms form the vulnerability to stress-related pathologies. Psychological, genetic, behavioural and environmental factors continuously interact. The indirect effects of psychosocial factors are contained in the modification of behaviour and lifestyle. These effects are bidirectional, because lifestyle in turn has an impact on mental and physical health. Receptori KIR imaju utjecaj na tijek i ishod transplantacije te su odgovorni za stvaranje aloreaktivnih stanica NK prilikom transplantacije krvotvornih matičnih stanica (TKMS). Kako bi se istražila uloga receptora KIR u imunologiji transplantacijske reakcije, provedeno je temeljno istraživanje polimorfizama i učestalosti gena, genotipova i haplotipova KIR na reprezentativnom uzorku hrvatske populacije (N=125). Retrogradnom analizom 111 bolesnika liječenih TKMS od srodnog i nesrodnog davatelja, istražen je utjecaj gena KIR na čimbenike ishoda TKMS: preživljavanje, GvHD i postizanje punog kimerizma. Dobiveni rezultati ukazuju da ligandi HLA-C skupine C1 i C2 primatelja kao i genotip KIR primatelja i davatelja imaju pozitivan učinak na preživljavanje i pojavu GvHD-a, dok na postizanje punog kimerizma nemaju nikakav učinak.- Killer cell immunoglobulin-like receptors (KIR) are family of inhibitory and activatory receptors expressed on natural killer cells (NK) with the basic role of regulation the NK cell activity. KIRs have influence on the hematopoietic stem cell transplantation (HSCT) outcome and are responsible for generating alloreactive NK cells. To explore the KIR gene role in HSCT, we performed the basic investigation of gene polymorphisms and KIR gene, haplotype and genotype frequencies in the group of healthy unrelated individuals (N=125) that represents the Croatian population and serve as a control group in the further studies. The role of KIR genes in HSCT in terms of disease free survival, the intensity of GvHD and chimerism was evaluated by retrograde analyses of 111 patients and their related and unrelated donors. Rozsáhlé genomové asociační studie se ukazují jako velice přínosné v identifikaci lokusů spojených s kardiovaskulárními onemocněními a souvisejícími rizikovými faktory. Ještě zbývá mnoho práce v identifikaci určujících genů a jejich kauzálních variant, stejně jako v definici mechanizmů, které ovlivňují vývoj a progresi onemocnění. Z výsledků studií lokusů spojených s dříve identifikovanými onemocněními se očekává zlepšení našich znalostí o patofyziologii kardiovaskulárních onemocnění a možnost identifikovat nové cíle pro léčbu. V oblasti aterotrombózy a trombofilie jsou významné výsledky z asociačních studií zaměřených na oblast ischemické choroby srdeční, ischemické cévní mozkové příhody a žilního tromboembolizmu. The genomewide association approach has proved successful in identifying loci associated with cardiovascular disease and related risk factors. However, much work remains to identifyning the culprit genes and causal variants as well as the mechanisms whereby they influence disease development and progression. In-depth studies of previously identified disease-associated loci are expected to improve our understanding of the pathophysiology of cardiovascular disease and identify novel targets for treatment. In the field of atherothrombosis and thrombophilia are significant results from association studies focused on the area of coronary artery disease, ischemic stroke, venous thromboembolism.
