This book is the result of an ethnographic study on the impact of Black cultural diversity on social action. The ethnography has three important characteristics. First, it incorporates the multiple ...perspectives of the ethnographer with the diverse voices of the people through an unusual form of reflexivity that provides additional insight for the descriptions, analyses, and conclusions of the book. This epistemological method is used to challenge traditional structures of ethnographies. Secondly, it argues for the consideration of non-traditional approaches to studying the Black experience - a focus away from race relations and issues of class and an emphasis on intragroup interaction and diversity. Thirdly, it investigates the processes, social institutions, and structures within the Black community of a small college town that influence social change and social action since the Civil Rights Movement of the 1960s.
Books about Oxford have generally focused on the University rather than the city. This original book on the local politics of Oxford City from 1830 to 1980 is based on a comprehensive analysis of ...primary sources and tells the story of the city’s progressive politics. The book traces this history from Chartism and electoral reform in the mid-nineteenth century, through the early years of socialism to the impact of communism in the interwar period, the struggle between nuclear disarmers and Gaitskellites in the 1960s and the impact of the new revolutionary left in the late 1970s. Patient-reported outcome scores are the mainstay method for quantifying success following arthroplasty. However, it is unclear when a “successful outcome” is achieved. We calculated threshold values ...for the Oxford Hip and Knee Score (OHS and OKS) representing achievement of a successful treatment at 12-month follow-up.
Questionnaires were administered to patients undergoing total hip (THA) or knee (TKA) arthroplasty before and 12 months after surgery alongside questions assessing key aspects of treatment success. A composite success criterion was used to perform receiver operator characteristic analysis. Thresholds providing maximum sensitivity and specificity were determined for the total sample and subgroups defined by presurgery scores.
Data were available for 3203 THA and 2742 TKA patients. Applying the composite treatment success criterion, 67.3% of the TKA and 77.6% of the THA sample reported treatment success. Accuracy for predicting treatment success was high for the OHS and OKS (both areas under the curve, 0.87). For the OHS, a threshold value of 37.5 points showed highest sensitivity and specificity in the total sample, while for the OKS the optimal threshold was 32.5 points. Depending on presurgery scores, optimal thresholds varied between 32.5 and 38.5 for the OHS and 28.5 and 36.5 for the OKS.
This is the first study to apply a composite “success” anchor to the OHS and OKS to evaluate outcome following total joint arthroplasty. Notably fewer patients report a “successful outcome” using a composite outcome threshold than report being “satisfied.”
•Direct sequencing of native RNA molecules using Oxford Nanopore Technologies enabled straight inspection of RNA modifications, and offered a promising alternative solution.•N6-methyladenosine is ...known to cause characteristic changes and increased base call errors of ONT signals compared with non-modified adenosines, based on which, the m6A sites can be identified directly from ONT signals.•Using the m6A sites revealed by Illumina technology as benchmark, we showed that, the accuracy of ONT-based m6A site prediction can be further increased by integrating additional information from the primary sequences of RNA (AUROC of 0.918), compared with using ONT signals only (AUROC 0.878 using Base call error features, and 0.804 using Tombo features).•Our method provides a new perspective for more reliable mining of the relatively noisy ONT signals.
Traditional epitranscriptome profiling approach relies on specific antibodies or chemical treatments to capture modified RNA molecules and then applies high throughput sequencing to identify their transcriptomic locations. However, due to the lack of suitable or high-quality antibodies, only a small proportion of the 170 documented RNA modifications were profiled with those approaches. Direct sequencing of native RNA molecules using Oxford Nanopore Technologies (ONT) enabled straight inspection of RNA modifications and offered a promising alternative solution. N6-methyladenosine (m6A) is known to cause characteristic changes and increased base call errors of ONT signals compared with non-modified adenosines, based on which, the m6A sites can be identified directly from ONT signals. Meanwhile, a number of studies have shown that it is possible to predict m6A sites from RNA primary sequences. Using the m6A sites revealed by Illumina technology as benchmark, we showed that, the accuracy of ONT-based m6A site prediction can be further increased by integrating additional information from the primary sequences of RNA (AUROC of 0.918), compared with using ONT signals only (AUROC 0.878 using Base call error features, and 0.804 using Tombo features), providing a new perspective for more reliable mining of the relatively noisy ONT signals.
We propose LongQC as an easy and automated quality control tool for genomic datasets generated by third generation sequencing (TGS) technologies such as Oxford Nanopore technologies (ONT) and SMRT ...sequencing from Pacific Bioscience (PacBio). Key statistics were optimized for long read data, and LongQC covers all major TGS platforms. LongQC processes and visualizes those statistics automatically and quickly.
Background: Given the demonstrated utility of Third Generation Sequencing Pacific Biosciences (PacBio) and Oxford Nanopore Technologies (ONT) long reads in many studies, a comprehensive analysis and ...comparison of their data quality and applications is in high demand.
Methods: Based on the transcriptome sequencing data from human embryonic stem cells, we analyzed multiple data features of PacBio and ONT, including error pattern, length, mappability and technical improvements over previous platforms. We also evaluated their application to transcriptome analyses, such as isoform identification and quantification and characterization of transcriptome complexity, by comparing the performance of size-selected PacBio, non-size-selected ONT and their corresponding Hybrid-Seq strategies (PacBio+Illumina and ONT+Illumina).
Results: PacBio shows overall better data quality, while ONT provides a higher yield. As with data quality, PacBio performs marginally better than ONT in most aspects for both long reads only and Hybrid-Seq strategies in transcriptome analysis. In addition, Hybrid-Seq shows superior performance over long reads only in most transcriptome analyses.
Conclusions: Both PacBio and ONT sequencing are suitable for full-length single-molecule transcriptome analysis. As this first use of ONT reads in a Hybrid-Seq analysis has shown, both PacBio and ONT can benefit from a combined Illumina strategy. The tools and analytical methods developed here provide a resource for future applications and evaluations of these rapidly-changing technologies.

Deixis is essential to creating effective communication. This research aimed (i) to determine the number of each type of deixis contained within E-mails from Oxford University Publications Office and ...(ii) the meanings contained there in. This research used qualitative descriptive method. The basic theory used by researchers is the theory of Yule. Yule described deixis as divided into 3, namely person, spatial, and temporal.The data used in this study came from one of the sources who received an E-mail from Oxford University and interview data. The result of the research was there are 2 types of deixis in E-mail, namely person deixis and spatial deixis. In person deixis was found 29 data with 3 forms of pronouns, namely You, Your, We, Our, Us, Their, and It. In spatial deixis was found 3 data with 2 forms, namely below and this. From the interview data, the researchers found that the language used in international campuses generally easy to comprehend, particularly noting the helpfulness of deixis in grasping message meanings, albeit sometimes requiring repeated reading for full understanding, with messages from Oxford University being notably straightforward.
