Bu çalışmada Brucella S19 genç aşısı ile aşılanan danalar ile Brucella spp. ile infekte sığırlar ve Brucella Rev1 genç aşısı ile aşılanmış kuzular ile Brucella spp. ile infekte koyunlara ait kan ...serumları; konvansiyonel testler ve Kompetatif-ELISA (C-ELISA) ile incelenerek, oluşan humoral immun yanıt değerlendirilmiştir. Doğal infekte sığırların %94'ü Rose Bengal Plate Test (RBPT), %98'i Serum Aglütinasyon Test (SAT), %92'si Komplement Fikzasyon Test (CFT) ve %95'i C-ELISA ile pozitif bulunmuştur. Aşı yapılan danalara ait antikor titrelerinde test zamanlarına göre testler arasında farklılıklar meydana gelmiştir. Doğal infekte koyunların %91.16'sı RBPT, %91.16'sı SAT, %85.63'ü CFT ve %86.74'ü C-ELISA ile pozitif bulunmuştur. Sonuç olarak brusellozisin teşhisinde, C_ELISA'nın özellikle CFT'ye alternatif olarak kullanılabileceği gösterilmiştir.
In this study, blood sera from calves vaccinated with Brucella S19 vaccine and cattle infected by Brucella spp. and lambs vaccinated with Brucella Rev1 vaccine and sheep infected by Brucella spp. were tested with conventional tests and Competitive-ELISA (C-ELISA) in order to evaluate the humoral immune response. In naturally infected cattle group, 94% with Rose Bengal Plate Test (RBPT), 98% with Serum Agglutination Test (SAT), 92% with Complement Fixation Test (CFT) and 95% with C-ELISA were found positive. Differences between the tests were found in antibody titers on the basis of testing times. In naturally infected sheep group, 91.16% with RBPT, 91.16% with SAT, 85.63% with CFT and 86.74% with C-ELISA were found positive. It was concluded that, in the diagnosis of brucellosis C-ELISA could be used as an alternative test especially to CFT.
In the present study 49 sheep fetuses naturally infected with Brucella melitensis were morphologically and immunohistochemically evaluated in association with bacteriologic culture. B. melitensis was ...isolated from nine fetuses of the 28 cases of which bacteriological analysis was performed and further immunohistochemical investigations were performed to those B. melitensis positive cases. Histopathological changes of nine sheep fetuses were mainly bronchopneumonia in the lungs and lymphoreticular hyperplasia in the liver and spleen. Nine sheep fetal lungs microbiologically infected with B. melitensis were also immunohictochemically positive. However, the antigen was not detected in any of the thymus examined. Intracellular antigenic localization was identified mainly in macrophages, neutrophils and hepatocytes. CD3+ T lymphocytes were determined with varying intensities in the liver, lungs, spleen, kidney, heart, bronchial lymph node, small intestines and abomasum of the aborted sheep fetuses. Local humoral response were limited and reflected as a few lambda light chain positive (λ-Ig) cells in only two lungs and one abomasum addition to spleens and lymph nodes of the aborted fetuses. In the present study determination of the Brucella causative agents with use of immunohistochemistry showed that this method could be useful as complementary method in cases where bacteriologic culture and serology is not available, in cases where sample fixed in formaldehyde or in suspicious cases where culture is negative. Furthermore, abortuses in sheep fetuses could occur not only as a result of plasentitis but also limited humoral and cellular response of the fetuses.
Bu çalışmada Brucella melitensis ile doğal olarak enfekte 49 koyun fötusu üzerinde etkenin varlığı ve enfeksiyona bağlı olarak şekillenen değişiklikler morfolojik, bakteriyolojik ve immunohistokimyasal yöntemlerle incelendi. Bakteriyolojik olarak incelenen 28 fötusun dokuzunda B. melitensis saptandı ve etken izole edilen bu olgular üzerinde immunohistokimyasal incelemeler yapıldı. İncelenen dokuz fötusun akciğerlerinde bronkopnömoni, karaciğer ve dalakta lenfoid ve lenforetiküler hiperplazi dikkati çekti. Bakteriyolojik olarak B. melitensis ile enfekte olduğu belirlenen dokuz koyun fötusunun hepsinin akciğerlerinde immunohistokimyasal olarak pozitif reaksiyon tespit edildi. Fakat incelenen timusların hiçbirinde Brucella antijeni tespit edilmedi. Brucella antijeninin başlıca makrofajlar, nötrofiller ve hepatositlerde intrasellüler olarak lokalize oldukları saptandı. CD3+ T lenfosit infiltrasyonları atık koyun fötuslarının karaciğer, akciğer, dalak, böbrek, kalp, beyin, bronşiyal lenf düğümü, ince bağırsak ve abomazumda değişen yoğunluklarda tespit edildi. Fötuslarda gelişen lokal humoral immun yanıtın dalak ve lenf düğümleri dışında iki fötus akciğer ile bir fötus abomazumunda tespit edilebilen az sayıdaki lambda hafif zincir immunglobulin (λ-Ig) pozitif hücreler ile sınırlı olduğu tespit edildi. Bakteriyolojik ve serolojik incelemelerin mümkün olmadığı, materyalin formaldehidde tespit edildiği ya da bakteriyolojik kültürün negatif olduğu şüpheli durumlarda Brucella etkenlerinin immunohistokimyasal olarak belirlenmesi bu yöntemin tamamlayıcı teşhis metodu olarak kullanılabileceğini göstermesi bakımından önem taşımaktadır. Ayrıca, bu çalışma ile koyun fötuslarında meydana gelen abortusların yalnızca annede gelişen plasentitis nedeni ile değil aynı zamanda brusellozise karşı oluşan sınırlı humoral ve hücresel yanıt nedeni ile gelişebileceği belirlenmiştir.
Bruselloz dünyada yaygın, önemi bir zoonozdur. Bu hastalık gelişmekte olan ülkelerde özellikle Akdeniz Havzası'nda hiperendemiktir. Birçok organ veya sistem tutulabilir. Santral sinir sistemi ...tutulumu bruselloz vakalarında nadiren saptanmaktadır. Bu yazıda ventrikuloperitoneal şant takılması sonrasında nörobrusellozis gelişen 41 yaşındaki olgu sunulmuştur. Oral doksisiklin ve rifampisin tedavisi sonrasında relaps gelişen hasta antibiyotik tedavisi ve şantın çıkarılması ile başarıyla tedavi edilmiştir. Endemik bölgelerde çalışan hekimlerin nörobrusellozun değişik kliniklerle gelebileceğini göz önünde bulundurularak dikkatli olması gerekmektedir.
Brucellosis is a worldwide important zoonosis. The disease is highly endemic in developing countries, particularly in the Mediterranean basin. Any organ or system of the body can be involved. Nervous system involvement has been rarely detected in the cases with brucellosis. In this report, we present 41-year old patient who developed neurobrucellosis after implantation of a ventriculoperitoneal shunt. The patient relapsed after oral doxycycline - rifampin therapyÊand successfully cured with removal of the shunt and antimicrobial therapy. Physicians should be careful aboutÊ the hetoregenous presentation of neurobrucellosis in the endemic areas.

Brucellosis is a contagious, zoonotic infection caused by Brucella species. Beside the serological and conventional cultural methods, different polymerase chain reaction (PCR) applications can be ...used in diagnosis of brucellosis. The aim of this study was to compare bacteriological culture and PCR methods used for the detection of Brucella spp. in stomach contents of the 31 bovine, 8 ovine and 1 caprine aborted fetuses. At the end of the study, same positive and negative results were obtained from both methods.
Brusellosis, Brucella türlerinin neden olduğu oldukça bulaşıcı, zoonotik bir enfeksiyondur. Brusellozisin teşhisinde, serolojik ve konvansiyonel kültürel yöntemlerin yanı sıra farklı polimeraz zincir reaksiyonu (PZR) uygulamaları da kullanılmaktadır. Bu çalışmada 31 adet sığır, 8 adet koyun ve 1 adet keçi fötal atık mide içeriğinden bakteriyolojik kültür ve PZR ile Brucella spp.'nin belirlenmesi ve her iki teşhis yönteminin karşılaştırılması amaçlanmıştır. Çalışma sonunda her iki yöntemle de benzer pozitif ve negatif sonuçlar elde edilmiştir.
L'accentuation des phénomènes de résistance aux antibiotiques augmente la difficulté d'enrayer les infections bactériennes. Afin de palier ce problème, la mise au point de nouvelles stratégies, telle ...la stratégie antivirulence, est essentielle. Ainsi, ce manuscrit propose une nouvelle approche thérapeutique contre les bactéries à développement intracellulaire. Les analyses génomiques ont permis de mettre en évidence l'histidinol déshydrogénase (HDH, EC 1.1.1.23), enzyme impliquée dans la biosynthèse de l'histidine, comme cible biologique pour la conception de nouveaux agents antibactériens. L'étude de l'inhibition de cette dernière permet une validation de l'approche sur Brucella suis, agent responsable de la brucellose, et un élargissement du spectre d'action des composés mis au point est envisagé par l'inhibition de HDH de Mycobacterium tuberculosis. Les travaux préliminaires nécessaires a cet élargissement sont présentés dans ce manuscrit.
The raise of antibiotic resistances increases the difficulty to eradicate bacterial infections. The development of new therapeutic approaches, such as the antivirulence strategy, is essential to limitate the impact of this phenomenon. This manuscript details a new therapeutic approach against intracellular pathogens. Genomic analyses allowed to discover new targets. The histidinol dehydrogenase (HDH, EC 1.1.1.23), which is an enzyme involved in histidine biosynthesis, has therefore be chosen for the conception of new antibacterial compounds. Inhibition studies of HDH of Brucella suis allows a validation of the strategy. In order to confirm the width of the therapeutic spectrum of synthesised compounds, the inhibition of HDH from Mycobacterium tuberculosis is envisaged and preliminary experiments are presented.

Les bactéries du genre Brucella, responsables de la brucellose, sont pathogènes pour les animaux et occasionnellement pour l'homme. Le coût de cette zoonose mondialement répandue est encore très ...important pour la société aujourd'hui. Ces dernières années, les travaux entrepris sur Brucella ont permis d'avancer dans deux domaines principaux. D'une part, sur le plan de l'organisation du génome, il a été montré que Brucella possède un génome complexe à deux chromosomes. D'autre part, en ce qui concerne l'étude du pouvoir pathogène de cette bactérie, les données les plus récentes suggèrent que Brucella a su développer des mécanismes de virulence communs avec des pathogènes de plantes et des symbiotes du groupe des alpha protéobactéries. L'étude des facteurs de virulence de Brucella est importante dans l'optique du développement d'un vaccin efficace et peut être aussi de nouveaux antibiotiques. L'accès à la séquence du génome de Brucella est un atout important pour le futur
