Autism is a developmental disorder as a result of the interaction of many individual genes and is a characterized by the lack of social and communicative behavior, limitations in activities and ...interests. Based on the structure of the genetic basis of autism, it might be considered that families of the autistic individuals may differ from the normal population. In the general population, some milder autistic symptoms were seen at the parents of children with autism like; social and cognitive deficits, communication skills differences, repetitive behaviors, narrow interests, identity needs and tendency to focus on details. These milder behaviours are called Broad Autism Phenotype.Emotions is one of the area where the children with autism have problems. Alexithymic people is another group that are experiencing difficulty in recognizing and expressing their feelings. This study is aimed to determine the relationship between the Broad Autism Phenotype and Alexithymia properties of the parents of autistic individuals. The study is prepared in accordance with the model of relational research with 494 parents.In the study; Parental Information Form, in order to assess broader autism phenotype characteristics, Autism Spectrum Quotient and in order to assess the alexithymic properties, Toronto Alexithymia Scale were used. Finally, there is a significant relationship between Toronto Alexithymia Scale total test score and Autism Spectrum Quotient Social skills, Attention Switching, Communication, Imagination subscale scores of mothers of autistic individuals and Attention Switching, Communication, Imagination subscale scores and Autism Spectrum Quotient total score of fathers
Otizm birçok bağımsız genin etkileşimi sonucu oluşan, sosyal ve iletişimsel davranışlarda yetersizlik, ilgi ve aktivitelerde sınırlılık ile kendini gösteren gelişimsel bir bozukluktur. Otizmin genetik temelli yapısından yola çıkarak, otistik bireylerin ailelerinde normal popülasyona göre farklılıklar olabileceği düşünülebilir. Genel popülasyonda, otistik çocukların ebeveynlerinde görülen sosyal ve bilişsel yetersizlikler, iletişim becerilerinde farklılık, tekrarlayan davranışlar, dar ilgi alanları, aynılık ihtiyacı, bütünden çok ayrıntıya odaklanmaya yatkınlık ile kendini gösteren hafif düzeyli otistik belirtilere geniş otizm fenotipi adı verilmektedir Duygular da otistik çocukların sorun yaşadığı alanlardan biridir. Duygularını fark edip ifade etme güçlüğü yaşayan diğer bir grup aleksitimik bireylerdir. Bu araştırma, otistik bireylerin ebeveynlerinin geniş otizm fenotipi ve aleksitimi özellikleri arasındaki ilişkiyi belirlemek amacı ile ilişkisel tarama modeli kullanılarak 494 ebeveyn ile yapılmıştır. Araştırmada, Ebeveyn Bilgi Formu, geniş otizm fenotipi özelliklerini değerlendirmek amacıyla Otizm Spektrum Anketi ve Toronto Aleksitimi Ölçeği kullanılmıştır. Otistik bireylerin annelerinin Toronto Aleksitimi Ölçeği toplam test puanı ile Otizm Spektrum Anketi Sosyal Beceri, Dikkati Kaydırabilme, İletişim, Hayal Gücü alt ölçek puanları, babalarının ise Dikkati Kaydırabilme, İletişim, Hayal Gücü alt ölçek puanları ve Otizm Spektrum Anketi toplam puanı arasında anlamlı bir ilişki bulunduğu bulgusuna ulaşılmıştır
Altered concentrations of oil, protein or starch in maize (Zea mays L.) kernels can provide value-added products. A progeny developed from the cross between the long-term divergently selected ...strains, Illinois High Oil (IHO) and Illinois Low Oil (ILO), cycle 70, was used to detect quantitative trait loci (QTL) for protein, starch and oil concentration and kernel weight. Recombinant inbred lines (RIL) were derived from an F2 created from one cycle of random mating the F1 (RM1) or after four cycles of random mating the RM1 (RM5). Both per se and corresponding testcrossed lines (TC1 and TC5) were evaluated in four environments. Genetic variance was lower in the testcrosses than in the per se progenies for all traits. Random mating was associated with a reduction in genetic variance for oil concentration between TC1 and TC5. Recombination maps constructed for the RM1 and RM5 revealed an expansion of 2.05 fold after random mating, close to the expected 1.83 fold expansion. Fewer loci were significant in the random mated generations, reflecting recombination between the markers and the QTL or between linked QTL. Using composite interval mapping (CIM), kernel oil concentration QTL were detected in chromosomal bins 2.09, 3.04, 6.04, 8.04, 8.05 and 8.07 of RM1 or TC1. The oil QTL detected in bin 6.04 in RM1 was also detected in TC1 and in previous research, but not in RM5 or TC5. This may be due to break up of marker-QTL associations and/or recombination between QTL
Concentrazioni modificate di olio, proteina e amido nelle cariossidi del mais (Zea mays L.) possono fornire prodotti a valore aggiunto. Una progenie sviluppata a partire dall'incrocio fra le stirpi selezionate a lungo termine per divergenza, Illinois High Oil (IHO) e Illinois Low Oil (ILO), ciclo 70, è stata utilizzata per individuare i loci di caratteri quantitativi (QTL) per la concentrazione di proteina, amido e olio e il peso della cariosside. Sono state derivate linee consanguinee ricombinanti (RIL) da una F2 creata da un ciclo di fecondazione casuale della F1 (RM1) o dopo quattro cicli di fecondazione casuale della RM1 (RM5). Le linee di per sé o sottoposte a testcross (TC1 e TC5) sono state valutate in quattro ambienti. La varianza genetica era inferiore nei testcross rispetto alle linee di per sé riguardo a tutti i caratteri. La fecondazione casuale è stata collegata alla riduzione della varianza genetica per la concentrazione dell'olio fra TC1 e TC5. Le mappe di ricombinazione costruite per la RM1 e la RM5 hanno mostrato un'espansione di 2,05 volte a seguito della fecondazione casuale, vicina all'espansione attesa di 1,83 volte. Meno loci risultavano significativi nelle generazioni fecondate casualmente, riflettendo la ricombinazione fra i marcatori e i QTL o fra i QTL concatenati. Utilizzando il composite internal mapping (CIM), sono stati individuati QTL per la concentrazione di olio nei bin cromosomici 2.09, 3.04, 6.04, 8.04, 8.05 e 8.07 di RM1 o TC1. Il QTL per l'olio individuato nel bin 6.04 in RM1 è stato pure ritrovato in TC1 e in una ricerca precedente, ma non in RM5 o TC5. Questo può essere dovuto alla rottura delle associazioni marcatore-QTL e/o alla ricombinazione fra QTL.

331 goats, from 35 flocks of different areas of Sardinia, were analyzed by 17 microsatellites. The flocks were assigned to three groups: Maltese (M), crossbred population (C) and local Sardinian type ...(S), on the basis of the overall phenotypic profiles and the breeding strategies of flocks. Parameters obtained from statistic analysis (heterozigosity, private alleles, standard genetic distance and Reynolds distance, Gst) showed a reduced genetic variation of group M compared to the other groups as well as the occurrence of a group of flocks whose individuals are assimilable to local Sardinian type. The crossbred population exhibited a high variability and significant distances from the other groups. The results allowed to identify a group of flocks to start a program of safeguard of the autochthonous genetic type. However, it is still to be assessed which role the crossbred populations should assume in a strategy of preservation of the total genetic variability.
Sono stati analizzati 331 caprini, provenienti da 35 allevamenti di diverse aree della Sardegna, con 17 microsatelliti. Gli allevamenti sono stati suddivisi in tre gruppi, Maltese (M), meticcio (C) e tipo Sardo autoctono (S), sulla base dei profili fenotipici degli animali e delle informazioni fornite dall´allevatore riguardo l´origine dei riproduttori utilizzati. Le statistiche calcolate (eterozigosità, alleli privati, distanza genetica standard e di Reynolds, Gst) hanno messo in evidenza una ridotta variabilità genetica del gruppo M rispetto agli altri due, nonché la presenza di un gruppo di allevamenti i cui individui sono assimilabili al tipo Sardo autoctono. Il gruppo meticcio ha mostrato un´elevata variabilità e distanze significative con gli altri 2 gruppi. I risultati hanno consentito di identificare il nucleo di allevamenti da cui partire per la conservazione del tipo genetico autoctono. Resta ancora da definire il ruolo del gruppo meticcio nel preservare la variabilità genetica complessiva
The European corn borer (ECB), Ostrinia nubilalis Huebner, is an important pest of maize in the US. Breeding for resistance to leaf-feeding by ECB can be more efficient if the genetic basis for ...resistance is better understood. The objectives of this study were (i) to locate and characterize the genetic factors controlling resistance to leaf blade feeding by ECB larvae in temperate lines and (ii) to compare the locations of the quantitative trait loci (QTLs) detected in the F2:3 and F6:8 generations of the same maize population derived from the cross of inbred lines Mo17 and H99. One hundred and fifty F2:3 lines and the parents and 185 F6:8 lines and the parents were grown in replicated trials in 1989 and 1995, respectively. Plots were infested with ECB larvae and visual leaf blade feeding ratings were obtained for each plot. Lines were genotyped with RFLP and SSR loci and a linkage map was developed. QTL analysis was performed with composite interval mapping. The heritability of family means was 0.77 in the F2:3 generation and 0.91 in the F6:8 generation. The QTLs explained around 50% of the phenotypic variance in both generations. Five QTLs were detected in each generation. A QTL on linkage group 4 near umc123 and a QTL on linkage group 7 near umc110 were detected in both generations. Significant epistatic interactions among QTLs were detected in both generations. Several loci that showed significant interactions did not have significant main effects, indicating that important epistatic interactions will be missed if only interactions between QTLs with significant main effects are tested. Marker-assisted selection will be most efficient when main effects and interaction effects of QTL are included in the selection criteria
La piralide del mais (ECB), Ostrinia nubilalis Huebner, è un parassita importante del mais negli USA. Il miglioramento genetico per la resistenza ai danni alle foglie determinati da ECB può essere più efficiente se se ne conosce meglio la base genetica. Gli obiettivi di questa ricerca erano: (i) localizzare e caratterizzare i fattori genetici che controllano la resistenza a ECB nelle linee temperate e (ii) confrontare le localizzazioni dei QTL individuati nelle generazioni F2:3 ed F6:8 della stessa popolazione di mais derivata dall'incrocio fra le linee consanguinee Mo17 e H99. In prove replicate nel 1989 e nel 1995, rispettivamente, sono state allevate 150 linee F2:3 e i parentali e 185 linee F6:8 e i parentali. Le parcelle erano infestate da larve di ECB e per ognuna sono stati valutati quantitativamente i danni alle lamine fogliari riscontrabili visivamente. Le linee sono state genotipizzate con loci RFLP e SSR ed è stata sviluppata una mappa di linkage. L'analisi dei QTL è stata effettuata tramite il composite interval mapping. L'ereditabilità delle medie delle famiglie era di 0,77 nella generazione F2:3 e di 0,91 nella F6:8. I QTL spiegavano circa il 50% della varianza fenotipica in ambedue le generazioni. Sono stati individuati 5 QTL in ogni generazione. In ambedue le generazioni è stato individuato un QTL sul gruppo di linkage 4 vicino a umc123 e un QTL sul gruppo di linkage 7 vicino a umc10. In ambedue le generazioni sono state evidenziate interazioni epistatiche significative fra i QTL. Numerosi loci che presentavano interazioni significative non avevano effetti principali significativi, indicando che possono essere ignorate importanti interazioni epistatiche se sono saggiate solamente le interazioni fra QTL con effetti principali significativi. La selezione assistita da marcatori presenta la massima efficienza quanto sono inseriti nei criteri di selezione gli effetti principali e gli effetti di interazione dei QTL.
Tomato spotted wilt virus (TSWV) is a serious threat to both horticultural and ornamental crops. Three fresh-market tomato (Lycopersicon esculentum Mill.) lines were transformed with the ...nucleoprotein gene of TSWV by Agrobacterium tumefaciens. Primary transformants for each line were analysed for transgene integration and expression. Results showed that inserted transgenic DNA was often rearranged. Progenies of selected primary transformants were obtained by self-pollination and tested for resistance to TSWV. A completely resistant line was identified having a single integration locus with multiple rearranged transgene copies and a true-to-type phenotype. It will be further tested for possible inclusion in breeding programs
Il virus dell'avvizzimento a chiazze del pomodoro (TSWV) e' una seria minaccia alle colture, sia ortive, sia ornamentali. Tre linee di pomodoro (Lycopersicon esculentum Mill.) per il consumo fresco sono state trasformate con il gene della nucleoproteina del TSWV mediante Agrobacterium tumefaciens. I trasformanti primari per ogni linea sono stati analizzati riguardo all'integrazione e all'espressione del transgene. I risultati hanno dimostrato che il DNA transgenico inserito era spesso riarrangiato. Mediante autoimpollinazione sono state ottenute progenie di trasformanti primari selezionati, sulle quali e' stata verificata la resistenza a TSWV. E' stata identificata una linea completamente resistente che presentava un singolo locus di integrazione con copie multiple riarrangiate del transgene e un fenotipo uguale al tipo. Esso verra' ulteriormente sperimentato in funzione di un possibile inserimento in programmi di miglioramento genetico

A factorial analysis of 23 linear morphologic traits recorded on 2,800 primiparous cows of Rendena breed was carried out. Factor scores, obtained by explicative latent factors, were utilized in order ...to calculate genetic parameters of factors and, after, to obtain factorial animal breeding values (EBVFs). EBVFs were compared with indexes obtained from single type traits (EBVMs).The correlation analysis showed that EBVFs reflect, in terms both of value and of ranks, the phenotypic link showed by single linear traits with factors (e.g., correlation between EBVM of linear traits related to fleshiness and body condition and EBVF of factor chiefly saturated by these traits, ranging from 0.86 to 0.90). These results suggest the possible utilization of factors in genetic evaluations for linear morphologic records, first of all in situations characterized, as in this case study, by the occurrence of a high number of evaluated traits.
E´ stata condotta l´analisi fattoriale di 23 caratteri morfologici lineari rilevati su 2.800 primipare di razza Rendena. I punteggi fattoriali, ottenuti dai fattori latenti esplicativi, sono stati utilizzati per calcolare i parametri genetici dei fattori stessi e successivamente per ottenere indici genetici fattoriali (EBVF). Gli EBVF sono stati quindi confrontati con indici ottenuti dai singoli punteggi morfologici (EBVM). L´analisi di correlazione ha messo in luce che gli EBVF riflettono, sia in valore, sia in termini di classifiche, il legame fenotipico palesato dai singoli caratteri lineari con i fattori (per esempio, correlazione tra EBVM dei lineari inerenti la muscolosità ed EBVF del fattore saturato principalmente da questi caratteri, compresa tra 0,86 e 0,90). Questi risultati suggeriscono il possibile uso dei fattori nelle valutazioni genetiche per i rilievi morfologici lineari, soprattutto in situazioni contraddistinte, come nel caso oggetto di studio, dalla presenza di un gran numero di caratteri valutati

The approach is based on analysis of phenotypes for a trait of interest (A) obtained as residuals of a linear regression on a positively related trait (B). The study shows, through simulations and an ...application to real data, that such approach increases the detection power of QTLs affecting only A, proportionally to correlation between traits. IT leads, however, the risk of false detection of non-pleiotropic QTLs for A when the locus affects B. It is possible to limit such risk extending the search for QTLs to the related trait. So, the probability of false positive is proportionally reduced to first type error chosen for B.
L´approccio si basa sull´analisi dei fenotipi per un carattere d´interesse (A) ottenuti come residui della regressione lineare su un carattere positivamente correlato (B). Il lavoro dimostra, attraverso simulazioni e con un´applicazione a dati reali, che tale approccio aumenta la potenza di individuazione di QTL con solo effetto su A, in misura proporzionale alla correlazione tra caratteri. Esso comporta tuttavia il rischio di concludere erroneamente sull´esistenza di QTL non pleiotropici per A quando il locus influenza B. E´ possibile limitare tale rischio estendendo la ricerca di QTL al carattere correlato. In tal modo, l´effettiva probabilità di false identificazioni si riduce proporzionalmente al rischio di errore di prima specie scelto per B

The aim of this research was to study mRNA transcriptome of pig muscle by SAGE (Serial Analysis of Gene Expression) method, originally developed for human. Longissimus dorsi samples were taken from ...9-month old pigs by biopsy and, after extraction of mRNA and retrotranscription to cDNA, were analyzed by SAGE. By the utilized method, libraries of 17 bp tags (LongSAGE) were developed, with a variation of the original protocol, by which 10 bp tags were obtained, too shorts for a correct localization on a not much characterized genome. Furthermore, some adjustments were made to original method, to increase the control among different steps. At the present time, 750 tags corresponding to genes expressed in muscle have been identified; their abundance varied from 1 to 43 pairs of transcripted mRNA. The preliminary results reported in the present study indicate that SAGE method can be efficiently used to associate transcriptome variability with phenotypic traits and to compare different physiological conditions in swine.
Lo scopo della ricerca è stato di studiare il profilo trascrizionale di mRNA di muscolo suino adattando il metodo SAGE, sviluppato per l´uomo. Allo scopo, sono stati prelevati dei campioni di longissimus dorsi da suini di 9 mesi mediante biopsia e, dopo estrazione dell´mRNA e retrotrascrizione in cDNA, sono stati analizzati mediante SAGE. Il metodo utilizzato ha previsto la costruzione di librerie di tag a 17 bp (LongSAGE), una variante dell´originale protocollo che produceva tag da 10 bp, troppo corti per la corretta localizzazione su un genoma ancora poco caratterizzato. Inoltre, sono state apportate alcune modifiche alla metodica originale per aumentare il controllo tra le numerose fasi procedurali. Allo stato, sono stati identificati oltre 750 tag corrispondenti a geni espressi nel muscolo, variabili per quantità da 1 a 43 coppie di mRNA trascritto. I risultati preliminari indicano la possibilità di utilizzare il metodo SAGE per gli studi di associazione con i caratteri fenotipici e per confrontare condizioni fisiologiche diverse nel suino
Lactobacilli are of great commercial value because they are widely used in fermented foods as probiotics or starter cultures. Traditional methods of bacterial enumeration, identification and ...characterisation are often insufficient for monitoring specific strains in complex, mixed-strain microbial consortia of lactobacilli. In the past few years, due to the use of molecular methods, knowledge about the microbial diversity of the genus Lactobacillus has dramatically increased. The molecular techniques described in this review can enhance the development of taxonomy and molecular microbial ecology of lactobacilli by providing ways of characterising mixed microbial communities of this industrially important group of micro-organisms
I lattobacilli hanno un notevole impatto commerciale in virtu' del loro vasto impiego come starter o colture probiotiche in svariati alimenti fermentati. I metodi tradizionali di conta, identificazione e caratterizzazione risultano spesso insufficienti per valutare la dinamica di particolari ceppi o specie in popolazioni microbiche complesse o in colture miste di lattobacilli. Ultimamente lo sviluppo esponenziale di tecniche di identificazione e biotipizzazione molecolari ha permesso di incrementare le conoscenze sulla diversita' microbica nel genere Lactobacillus. Le tecniche descritte nel presente articolo potranno contribuire a un ulteriore sviluppo della tassonomia e dell'ecologia microbica molecolare dei lattobacilli, indicando nel contempo efficaci strumenti per una loro caratterizzazione all'interno di comunita' microbiche
