This paper presents a stochastic framework for day-ahead scheduling of microgrid energy storage systems in the context of multi-objective (MO) optimization. Operation cost of microgrid in normal ...conditions and load curtailment index in case of unscheduled islanding events (initiated by disturbances in the main grid) are chosen as main criteria of the proposed scheme. In practice, duration of disconnection from the upstream network is unknown in unscheduled islanding incidents and cannot be predicted with certainty. To properly handle the uncertainties associated with time and duration of such events as well as microgrid load and renewable power generation, stochastic models are involved in the MO scheduling framework and they are formulated as mixed integer linear programming problems. The non-dominated sorting genetic algorithm II is employed to effectively cope with the MO optimization problem and a fuzzy decision making approach is employed for appropriate representation of microgrid operator's preferences in compromising between the two objectives. The proposed scheme is implemented on a test microgrid and the obtained results demonstrate the applicability and efficiency of this framework in dealing with conflicting requirements of microgrid security and economic operation.
With the rapid development of electric vehicles, the safe and environmentally friendly disposal of retired lithium batteries (LIBs) is becoming a serious issue. Echelon utilization of the retired ...LIBs is a promising scheme because of its considerable potential for generating economic and environmental value. The most outstanding technical challenge of echelon utilization is how to sort and regroup the large-scale retired LIBs efficiently and accurately. In this paper, the status and challenges of echelon utilization for the retired LIBs are reviewed. First, the criteria, policies, regulations, markets, costs, and values of echelon utilization are summarized comprehensively to illustrate its potential and expose existing problems and pain points. Second, the key technologies related to large-scale echelon utilization of LIBs are detailed; valuable opinions and technical routes are presented for the selection and rapid estimation of sorting indices, the classification and regrouping algorithm, evaluation of the sorting results, and other aspects. In particular, a multilevel and multidimensional fast sorting method is proposed for large-scale echelon utilization of retired LIBs that considers different scenario constraints. Valuable solutions to the key technical problems are given, such as predicting the characteristics of retired LIBs with in-service data and building a fast sorting model from a small number of samples to sort large quantities of LIBs. Finally, the technological prospects of echelon utilization are discussed. Big data and artificial intelligence can be used to promote further development and application of echelon utilization, which may eventually be applied to managing the whole life cycle of LIBs.

Metazoans require the sorting nexin (SNX) protein, SNX27, to recycle hundreds of important transmembrane protein receptors from endosomes to the plasma membrane. Cargo recycling by SNX27 requires its ...interaction with retromer, a heterotrimer known to assemble on membranes with multiple sorting nexins, including SNX-BAR proteins and SNX3. SNX27 has also been functionally linked to SNX-BARs, but the molecular basis of this interaction has been unknown. We identify a direct biochemical interaction between the conserved and flexible SNX1/SNX2 N-terminus and full-length SNX27 using purified proteins in pulldown experiments. Sequence alignments indicate both SNX1 and SNX2 contain two short and conserved stretches of acidic residues bearing a DxF motif in their flexible N-terminal regions. Biochemical pulldown and mapping experiments reveal forty residues in the N-terminus of either SNX1 or SNX2 can mediate binding to SNX27. SNX27 truncation analysis demonstrates the SNX27 FERM domain binds the SNX1 N-terminus. Calorimetry experiments quantified binding between the SNX1 N-terminus and SNX27 in the low micromolar affinity range (KD ∼10 μM) and suggest the second DxF motif may play a more prominent role in binding. Mutation of either DxF sequence in SNX1 abrogates measurable binding to SNX27 in the calorimeter. Modelling from both predicted and experimentally determined structures suggests the SNX27 FERM domain could accommodate both DxF motifs simultaneously. Together, these data suggest SNX27 is directly linked to specific SNX-BAR proteins through binding acidic motifs in the SNX1 or SNX2 N-terminus.
Group scheduling problems have attracted much attention owing to their many practical applications. This work proposes a new bi-objective serial-batch group scheduling problem considering the ...constraints of sequence-dependent setup time, release time, and due time. It is originated from an important industrial process, i.e., wire rod and bar rolling process in steel production systems. Two objective functions, i.e., the number of late jobs and total setup time, are minimized. A mixed integer linear program is established to describe the problem. To obtain its Pareto solutions, we present a memetic algorithm that integrates a population-based nondominated sorting genetic algorithm II and two single-solution-based improvement methods, i.e., an insertion-based local search and an iterated greedy algorithm. The computational results on extensive industrial data with the scale of a one-week schedule show that the proposed algorithm has great performance in solving the concerned problem and outperforms its peers. Its high accuracy and efficiency imply its great potential to be applied to solve industrial-size group scheduling problems.
Endosomal recycling of transmembrane proteins requires sequence-dependent recognition of motifs present within their intracellular cytosolic domains. In this study, we have reexamined the role of ...retromer in the sequence-dependent endosome-to-trans-Golgi network (TGN) transport of the cation-independent mannose 6-phosphate receptor (CI-MPR). Although the knockdown or knockout of retromer does not perturb CI-MPR transport, the targeting of the retromer-linked sorting nexin (SNX)-Bin, Amphiphysin, and Rvs (BAR) proteins leads to a pronounced defect in CI-MPR endosome-to-TGN transport. The retromer-linked SNX-BAR proteins comprise heterodimeric combinations of SNX1 or SNX2 with SNX5 or SNX6 and serve to regulate the biogenesis of tubular endosomal sorting profiles. We establish that SNX5 and SNX6 associate with the CI-MPR through recognition of a specific WLM endosome-to-TGN sorting motif. From validating the CI-MPR dependency of SNX1/2-SNX5/6 tubular profile formation, we provide a mechanism for coupling sequence-dependent cargo recognition with the biogenesis of tubular profiles required for endosome-to-TGN transport. Therefore, the data presented in this study reappraise retromer's role in CI-MPR transport.
The endosomal sorting complexes required for transport (ESCRTs) catalyse one of the most unusual membrane remodelling events in cell biology. ESCRT-I and ESCRT-II direct membrane budding away from ...the cytosol by stabilizing bud necks without coating the buds and without being consumed in the buds. ESCRT-III cleaves the bud necks from their cytosolic faces. ESCRT-III-mediated membrane neck cleavage is crucial for many processes, including the biogenesis of multivesicular bodies, viral budding, cytokinesis and, probably, autophagy. Recent studies of ultrastructures induced by ESCRT-III overexpression in cells and the in vitro reconstitution of the budding and scission reactions have led to breakthroughs in understanding these remarkable membrane reactions.
The sorting nexins (SNXs) are a family of peripheral membrane proteins that direct protein trafficking decisions within the endocytic network. Emerging evidence in yeast and mammalian cells ...implicates a subgroup of SNXs in selective and non-selective forms of autophagy. Using siRNA and CRISPR-Cas9, we demonstrate that the SNX-BAR protein SNX4 is needed for efficient LC3 (also known as MAP1LC3) lipidation and autophagosome assembly in mammalian cells. SNX-BARs exist as homo- and hetero-dimers, and we show that SNX4 forms functional heterodimers with either SNX7 or SNX30 that associate with tubulovesicular endocytic membranes. Detailed image-based analysis during the early stages of autophagosome assembly reveals that SNX4-SNX7 is an autophagy-specific SNX-BAR heterodimer, required for efficient recruitment and/or retention of core autophagy regulators at the nascent isolation membrane. SNX4 partially colocalises with juxtanuclear ATG9A-positive membranes, with our data linking the autophagy defect upon SNX4 disruption to the mis-trafficking and/or retention of ATG9A in the Golgi region. Taken together, our findings show that the SNX4-SNX7 heterodimer coordinates ATG9A trafficking within the endocytic network to establish productive autophagosome assembly sites, thus extending knowledge of SNXs as positive regulators of autophagy.

The retromer complex, which recycles the cation-independent mannose 6-phosphate receptor (CI-MPR) from endosomes to the trans-Golgi network (TGN), is thought to consist of a cargo-selective ...VPS26-VPS29-VPS35 trimer and a membrane-deforming subunit of sorting nexin (SNX)-Bin, Amphyphysin, and Rvs (BAR; SNX-BAR) proteins. In this study, we demonstrate that heterodimers of the SNX-BAR proteins, SNX1, SNX2, SNX5, and SNX6, are the cargo-selective elements that mediate the retrograde transport of CI-MPR from endosomes to the TGN independently of the core retromer trimer. Using quantitative proteomics, we also identify the IGF1R, among more potential cargo, as another SNX5 and SNX6 binding receptor that recycles through SNX-BAR heterodimers, but not via the retromer trimer, in a ligand- and activation-dependent manner. Overall, our data redefine the mechanics of retromer-based sorting and call into question whether retromer indeed functions as a complex of SNX-BAR proteins and the VPS26-VPS29-VPS35 trimer.
Macroautophagy is a physiological cellular response to nutrient stress, which leads to the engulfment of cytosolic contents by a double-walled membrane structure, the phagophore. Phagophores seal to ...become autophagosomes, which then fuse with lysosomes to deliver their contents for degradation. Macroautophagy is regulated by numerous cellular factors, including the Class III PI3K (phosphoinositide 3-kinase) Vps34 (vacuolar protein sorting 34). The autophagic functions of Vps34 require its recruitment to a complex that includes Vps15, Beclin-1 and Atg14L (autophagy-related 14-like protein) and is known as Vps34 Complex I. We have now identified NRBF2 (nuclear receptor-binding factor 2) as a new member of Vps34 Complex I. NRBF2 binds to complexes that include Vps34, Vps15, Beclin-1 and ATG-14L, but not the Vps34 Complex II component UVRAG (UV radiation resistance-associated gene). NRBF2 directly interacts with Vps15 via the Vps15 WD40 domain as well as other regions of Vps15. The formation of GFP-LC3 (light chain 3) punctae and PE (phosphatidylethanolamine)-conjugated LC3 (LC3-II) in serum-starved cells was inhibited by NRBF2 knockdown in the absence and presence of lysosomal inhibitors, and p62 levels were increased. Thus NRBF2 plays a critical role in the induction of starvation-induced autophagy as a specific member of Vps34 Complex I.
