•The spectral indices improve the performance of PTFs in estimating of SM.•Soil moisture is more accurately estimated by the combined NDWI and SWCI indices.•The accuracy of the RF for estimating the ...SM is better than the MLR-based PTFs.•Amount of clay in all functions has the most impact on estimating of SM.
To estimate the surface soil moisture (SM) using a combination of new spectral indices and methods of Random Forrest (RF) and Multiple Linear Regression (MLR), 11 pedotransfer functions (PTF1-11) were developed by combining basic soil properties (clay, silt/sand, and bulk density) and spectral indices of Sentinel-2 satellite. In this study, 124 surface soil samples were randomly taken from three regions including Telo in Tehran province, Ivaneki in Semnan province, and Borujerd in Lorestan province, Iran. The results showed that the accuracy of the RF method was considerably higher compared to the MLR method. The SM was better estimated using water spectral indices (such as Normalized Difference Water Index (NDWI) and Surface Water Capacity Index (SWCI)), along with the basic properties of soil as inputs of PTF7. In the training and testing steps, the Root Mean Square Error (RMSE) decreased from 0.041 and 0.05 (cm3 cm−3) in PTF1 to 0.028 and 0.039 in PTF7, respectively. The average values of RMSE, Akaike Information Criterion (AIC), coefficient of determination (R2) and Relative Improvement (RI) of the RF method were 0.028 (cm3 cm−3), −559, 0.79 and 0.001, and 0.038 (cm3 cm−3), −279, 0.73 and −0.006 for the training and testing steps of all PTFs, respectively. While, these values for the MLR method were 0.032 (cm3 cm−3), −542, 0.69 and 0.0003, and 0.043 (cm3 cm−3), −269, 0.63 and 0.002 for the training and testing steps, respectively. Due to the low values of MBE, it was possible to disregard the overestimation of the results. The evaluation of results for predictor importance indicated that among the basic properties, clay percent has a significant effect on estimation of SM. These results show that spectral indices alone are not suitable estimators for SM estimation. It suggests using basic soil properties and spectral indices to estimate the SM.
Summary
Oxidative stress and telomere attrition are considered the driving factors of aging. As oxidative damage to telomeric DNA favors the erosion of chromosome ends and, in turn, telomere ...shortening increases the sensitivity to pro‐oxidants, these two factors may trigger a detrimental vicious cycle. To check whether limiting oxidative stress slows down telomere shortening and related progeria, we have investigated the effect of p66SHC deletion, which has been shown to reduce oxidative stress and mitochondrial apoptosis, on late‐generation TERC (telomerase RNA component)‐deficient mice having short telomeres and reduced lifespan. Double mutant (TERC−/− p66SHC−/−) mice were generated, and their telomere length, fertility, and lifespan investigated in different generations. Results revealed that p66SHC deletion partially rescues sterility and weight loss, as well as organ atrophy, of TERC‐deficient mice, but not their short lifespan and telomere erosion.
Therefore, our data suggest that p66SHC‐mediated oxidative stress and telomere shortening synergize in some tissues (including testes) to accelerate aging; however, early mortality of late‐generation mice seems to be independent of any link between p66SHC‐mediated oxidative stress and telomere attrition.

In the quest for selective G-quadruplex (G4)-targeting chemotypes, natural compounds have been thus far poorly explored, though representing appealing candidates due to the high structural diversity ...of their scaffolds. In this regard, a unique high diversity in-house library composed of ca. one thousand individual natural products was investigated. The combination of molecular docking-based virtual screening and the G4-CPG experimental screening assay proved to be useful to quickly and effectively identify—out of many natural compounds—five hit binders of telomeric and oncogenic G4s, i.e., Bulbocapnine, Chelidonine, Ibogaine, Rotenone and Vomicine. Biophysical studies unambiguously demonstrated the selective interaction of these compounds with G4s compared to duplex DNA. The rationale behind the G4 selective recognition was suggested by molecular dynamics simulations. Indeed, the selected ligands proved to specifically interact with G4 structures due to peculiar interaction patterns, while they were unable to firmly bind to a DNA duplex. From biological assays, Chelidonine and Rotenone emerged as the most active compounds of the series against cancer cells, also showing good selectivity over normal cells. Notably, the anticancer activity correlated well with the ability of the two compounds to target telomeric G4s.
Background: G-quadruplex (G4) forming sequences are recurrent in telomeres and promoter regions of several protooncogenes. In normal cells, the transient arrangements of DNA in G-tetrads may regulate ...replication, transcription, and translation processes. Tumors are characterized by uncontrolled cell growth and tissue invasiveness and some of them are possibly mediated by gene expression involving G-quadruplexes. The stabilization of G-quadruplex sequences with small molecules is considered a promising strategy in anticancer targeted therapy. Methods: Molecular virtual screening allowed us identifying novel symmetric bifunctionalized naphtho1,2-b:8,7-b’dithiophene ligands as interesting candidates targeting h-Telo and c-MYC G-quadruplexes. A set of unexplored naphtho-dithiophene derivatives has been synthesized and biologically tested through in vitro antiproliferative assays and spectroscopic experiments in solution. Results: The analysis of biological and spectroscopic data highlighted noteworthy cytotoxic effects on HeLa cancer cell line (GI50 in the low μM range), but weak interactions with G-quadruplex c-MYC promoter. Conclusions: The new series of naphtho1,2-b:8,7-b’dithiophene derivatives, bearing the pharmacophoric assumptions necessary to stabilize G-quadruplexes, have been designed and successfully synthesized. The interesting antiproliferative results supported by computer aided rational approaches suggest that these studies are a significant starting point for a lead optimization process and the isolation of a more efficacious set of G-quadruplexes stabilizers.
Background
Dorsal pons cavernoma can be approached through telo-velar approach instead of transvermian approach, with lower risk of neurological deficits since it uses natural clefts to reach the ...floor of the fourth ventricle.
Materials and methods
We present our surgical technique for telo-velar approach to address pathologies of the dorsal pons, assisted by neuronavigation and neuromonitoring. This surgical technique is illustrated by a surgical video of a dorsal pons cavernoma.
Conclusion
Dorsal pons cavernomas can be reached through telo-velar approach after suboccipital midline craniotomy. The accurate patient positioning, cisternal dissection, and neuromonitoring use are mandatory to avoid neural injuries and identify the safe entry points into the brainstem.
Two new Ru(II) complexes, Ru(bpy)2L3+ (1 and 2), containing guanidinium ligands have been synthesized and characterized. Both complexes show good selectivity for h-telo G-quadruplex DNA compared to ...promoters and duplex DNA. Display omitted
Two new Ru(II) complexes containing guanidinium groups have been synthesized, characterized and analyzed according to their interactions with different G-quadruplexes and duplex DNA. A FRET assay and a competitive FRET assay showed that both complexes promote the formation and stabilization of the human telomeric (h-telo) G-quadruplex and exhibit higher selectivity compared to promoters (such as c-myc, c-kit and bcl2) or duplex DNA. After binding to G-quadruplex, the two complexes have shown different DNA affinity and fluorescence enhancement. CD analyses further indicate that the two complexes display the ability to induce and stabilize the formation of antiparallel G-quadruplex structures in K+, Na+ or ion-free buffers. The binding stoichiometry with h-telo was of the order of three ruthenium complexes per quadruplex.
Obesity is a significant factor related to metabolic disturbances that can lead to metabolic syndrome (MetS). Metabolic dysregulation causes oxidative stress, which affects telomere structure. The ...current study aimed to evaluate the relationships between telomere length, oxidative stress and the metabolically healthy and unhealthy phenotypes in healthy young men. Ninety-eight participants were included in the study (49 healthy slim and 49 obese patients). Study participants were divided into three subgroups according to body mass index and metabolic health. Selected oxidative stress markers were measured in serum. Relative telomere length (rTL) was measured using quantitative polymerase chain reaction. The analysis showed associations between laboratory markers, oxidative stress markers and rTL in metabolically healthy and unhealthy participants. Total oxidation status (TOS), total antioxidant capacity (TAC) and rTL were significantly connected with metabolically unhealthy obesity. TAC was associated with metabolically healthy obesity. Telomeres shorten in patients with metabolic dysregulation related to oxidative stress and obesity linked to MetS. Further studies among young metabolically healthy and unhealthy individuals are needed to determine the pathways related to metabolic disturbances that cause oxidative stress that leads to MetS.
