It is not known how ILI S affects atherogenesis. We ... investigated the IL-15 role in the early stages of atherogenesis. As a likely pathway of the IL-15 action, we studied the IL-15 effect on the expression of the proatherogenic chemokine fractalkineand its receptor CX3CR1. Methods: Experiments on isolated mouse-aortic SMC in vitro were followed by experiments on wild type C57žBL6 mice in vivo. In vitro, IL-15 and IL-15Ra expression in SMC was evaluated by PCR. The effect of serum stimulation and IL-15 treatment on SMC proliferation was determined by flow-cytometric cell-cycle analysis. The effect of IL-15 treatment on fractalkine and CX3CR1 expression in SMC was evaluated by real-time PCR. In vivo, mice were subjected to carotid arterial cuff injury. IL-15 and its receptor (IL-15Ra) expression was determined by-PCR. Immunochistochemical detection of IL-15 was also performed. The effect of endogenous IL-15 blockade on intimal thickening was tested separately using treatment with specific anti-IL-15 antibody. Control groups consisted of mice which received treatment with nonspecific IgG and of non-treated mice. Intimalthickness was evaluated 21 days after injury. The effect of endogenous IL-15 blockade on fractalkine and CX3CR1 expression was examined by real-time PCR. ResultsČ In vitro results showed that SMC up-regulate IL-15 and IL-15Ra mRNA expression after serum stimulation. IL-15 treatment attenuated proliferation in serum stimulated SMC by 20% (p < 0.05). In addition, IL-15 down-regulated fractalkine and CX3CR1 expression in SMC. In vivo, IL-15 and IL-15 Ra expression was elevated in injured carotid arteries.
