Coronavirus disease 2019 (Covid-19) disproportionately results in hospitalization or death in older patients and those with underlying conditions. Sotrovimab is a pan-sarbecovirus monoclonal antibody ...that was designed to prevent progression of Covid-19 in high-risk patients early in the course of disease.
In this ongoing, multicenter, double-blind, phase 3 trial, we randomly assigned, in a 1:1 ratio, nonhospitalized patients with symptomatic Covid-19 (≤5 days after the onset of symptoms) and at least one risk factor for disease progression to receive a single infusion of sotrovimab at a dose of 500 mg or placebo. The primary efficacy outcome was hospitalization (for >24 hours) for any cause or death within 29 days after randomization.
In this prespecified interim analysis, which included an intention-to-treat population of 583 patients (291 in the sotrovimab group and 292 in the placebo group), 3 patients (1%) in the sotrovimab group, as compared with 21 patients (7%) in the placebo group, had disease progression leading to hospitalization or death (relative risk reduction, 85%; 97.24% confidence interval, 44 to 96; P = 0.002). In the placebo group, 5 patients were admitted to the intensive care unit, including 1 who died by day 29. Safety was assessed in 868 patients (430 in the sotrovimab group and 438 in the placebo group). Adverse events were reported by 17% of the patients in the sotrovimab group and 19% of those in the placebo group; serious adverse events were less common with sotrovimab than with placebo (in 2% and 6% of the patients, respectively).
Among high-risk patients with mild-to-moderate Covid-19, sotrovimab reduced the risk of disease progression. No safety signals were identified. (Funded by Vir Biotechnology and GlaxoSmithKline; COMET-ICE ClinicalTrials.gov number, NCT04545060.).

This article investigates whether GVC reconfiguration through the adoption of redistributed manufacturing (RDM) in local production can deliver greater resilience against unexpected, disruptive global events. It proposes actionable steps for managers to ensure more resilient GVCs in the face of global shocks.
Abstract The study aimed to determine the natural history of Staphylococcus aureus nasal colonization in hemodialysis outpatients. Surveillance cultures were taken from patients presenting for ...hemodialysis or routine care to identify S. aureus nasal carriers. A prospective cohort study was performed to identify risks for persistent colonization. Detailed microbiologic and molecular studies of colonizing isolates were performed. Only 23/145 (15.9%) dialysis patients were persistently colonized, and only HIV-positive status was associated with persistence ( P = 0.05). Prior hospitalization was the only risk factor for methicillin-resistant S. aureus carriage (OR 2.5, P = 0.03). In isolates from patients with ≤42 days of vancomycin exposure, vancomycin minimum bactericidal concentrations (MBCs) increased with duration of exposure. Among dialysis patients, S. aureus colonization was limited and transient; only HIV status was associated with persistence. Nevertheless, duration of vancomycin exposure was associated with increasing vancomycin MBCs. Vancomycin exposure in S. aureus carriers may be involved in increasing resistance.
Postpartum depression (PPD) is typically constructed as a medical condition that is defined by prevalence rates, time of onset, duration, symptoms, causes and treatments. Presentations of PPD in ...popular texts have also revealed that it is a site for constructions of mothering/motherhood. While PPD has traditionally been assumed to be the domain of women, researchers have recently focused attention on PPD and men. Using articles on maternal and paternal PPD published in Canadian and American newspapers between 2008 and 2012, we analysed how constructions of mothering/motherhood and fathering/fatherhood were inscribed in this public forum. We show how mothering/motherhood was foregrounded via dominant evaluative discourses and highly imbued with expectations while fathering/fatherhood was kept in the background and characterised by the unavailability of clear expectations. We conclude that maternal PPD remains primary and normative while paternal PPD remains ‘othered’ despite the supposed attention to it in these public texts.

This study investigated the relationship between three dimensions of perfectionism (i.e., self-oriented perfectionism, other-oriented perfectionism, and socially prescribed perfectionism) and ...achievement in a graduate-level research methodology course among 130 psychology students. Achievement, which involved students’ knowledge of research concepts, methodologies, and applications, was measured individually in all sections via comprehensive written midterm and final examinations. A canonical correlation analysis revealed that graduate students with relatively high levels of self-oriented perfectionism and other-oriented perfectionism tended to have the highest levels of achievement in the class, with socially prescribed perfectionism serving as a suppressor variable. Self-oriented perfectionism was the best predictor of performance, highlighting the relative importance of this dimension of perfectionism in the context of learning in research methodology courses. Implications are discussed.
