Summary
This is the second report of the United Kingdom Primary Immunodeficiency (UKPID) registry. The registry will be a decade old in 2018 and, as of August 2017, had recruited 4758 patients ...encompassing 97% of immunology centres within the United Kingdom. This represents a doubling of recruitment into the registry since we reported on 2229 patients included in our first report of 2013. Minimum PID prevalence in the United Kingdom is currently 5·90/100 000 and an average incidence of PID between 1980 and 2000 of 7·6 cases per 100 000 UK live births. Data are presented on the frequency of diseases recorded, disease prevalence, diagnostic delay and treatment modality, including haematopoietic stem cell transplantation (HSCT) and gene therapy. The registry provides valuable information to clinicians, researchers, service commissioners and industry alike on PID within the United Kingdom, which may not otherwise be available without the existence of a well‐established registry.

Stable flies (Stomoxys calcitrans Diptera: Muscidae L.) are blood-feeding synanthropic pests, which cause significant economic losses in livestock. Stable fly antennae contain olfactory sensilla ...responsive to host and host environment-associated odours. Field observation indicated that the abundance of stable flies increased significantly in grasslands or crop fields when cattle manure slurry was applied. Major volatile compounds emanating from manure slurry were collected and identified. Behavioural responses of stable flies to those compounds were investigated in laboratory bioassays and field-trapping studies. Results from olfactometer assays revealed that phenol, p-cresol and m-cresol were attractive to adult stable flies. When tested individually, attraction was higher with lower dosages. Stable flies were most attracted to blends of phenol and m-cresol or p-cresol. Traps with binary blend lures caught more stable flies in field trials as well.
Since its discovery in North America in 2000, the soybean aphid, Aphis glycines Mat-sumura (Hemiptera: Aphididae), has rapidly become an important pest of soybean Glycine max (L.) Merrill, sometimes ...resulting in significant yield losses. Previous research has documented the toxicity of neonicotinoid seed treatments to soybean aphids, but control under field conditions has been inconsistent. Imidacloprid, a popular neonicotinoid insecticide, has been shown to exhibit antifeedant effects on aphids. Antifeedant activity has not been demonstrated for other neonicotinoids, including thiamethoxam. This research investigated the effects of a thiamethoxam seed treatment on soybean aphid feeding behavior by using electronic penetration graphs (EPG) to visualize stylet penetration behavior. Soybean aphid feeding behavior was assessed for 9 h on thiamethoxam-treated and untreated soybeans (V2 and V4 stages). Because results were inconclusive from initial experiments, a study was conducted to document the effects of thiamethoxam-treated soybeans on soybean aphid survival. The seed treatment was shown to negatively affect aphid survival at 4, 8, and 11 d after aphid introduction. A subsequent EPG study then was designed to document soybean aphid feeding behavior for 15 h, after an initial exposure of 9 h to thiamethoxam-treated soybeans. In this study, the exposed aphids exhibited significant differences in feeding behavior compared with those aphids feeding on untreated soybeans. Soybean aphids on thiamethoxam-treated soybeans spent significantly less time feeding in the sieve element phase, with a greater duration of nonprobing events. These studies suggest soybean aphids are unable to ingest phloem sap, which may be another important element in seed treatment protection.
The western chinch bug, Blissus occiduus Barber (Hemiptera: Blissidae), has emerged as a serious pest of buffalograss, Buchloë dactyloides (Nuttall) Engelmann. In general, neonicotinoid insecticides ...effectively control a variety of turfgrass insects, particularly phloem-feeding pests. However, because of well documented inconsistencies in control, these compounds are generally not recommended for chinch bugs. This study was designed to document the contact and systemic toxicity of three neonicotinoid insecticides (clothianidin, imidacloprid, and thiamethoxam) to B. occiduus. In contact bioassays, thiamethoxam was ≈20-fold less toxic than clothianidin or imidacloprid to B. occiduus nymphs and three-fold more toxic to adults. In adult systemic bioassays, thiamethoxam was up to five-fold more toxic than clothianidin or imidacloprid. Interestingly, thiamethoxam was significantly more toxic to adults than to nymphs in both contact and systemic bioassays. This was not observed with clothianidin or imidacloprid. Bifenthrin, used for comparative purposes, exhibited 1,844-fold and 122-fold increase in toxicity to nymphs and adults, respectively. These results provide the first documentation of the relative toxicity of these neonicotinoid insecticides to B. occiduus.
Russian wheat aphid, Diuraphis noxia (Mordvilko), feeding injury on ‘Betta’ wheat isolines with the Dn1 and Dn2 genes was compared by assessing chlorophyll and carotenoid concentrations, and aphid ...fecundity. The resistant Betta isolines (i.e., Betta-Dn1 and Betta-Dn2) supported similar numbers of aphids, but had significantly fewer than the susceptible Betta wheat, indicating these lines are resistant to aphid feeding. Diuraphis noxia feeding resulted in different responses in total chlorophyll and carotenoid concentrations among the Betta wheat isolines. The infested Betta-Dn2 plants had higher levels of chlorophylls and carotenoids in comparison with uninfested plants. In contrast, infested Betta-Dn1 plants had the same level of chlorophyll and carotenoid in comparison with uninfested plants. Our data provide essential information on the effect of D. noxia feeding on chlorophyll and carotenoid concentrations for Betta wheat and its isolines with D. noxia-resistant Dn1 and Dn2 genes.
Laboratory bioassays were conducted to characterize the acute toxicity of 7 monoterpenoids to tracheal mites, Acarapis woodi (Rennie), and their honey bee, Apis mellifera L., hosts. Citral, thymol, ...carvacrol, alpha -terpineol, pulegone, d-limonene, and menthol were applied as fumigants to mite-infested honey bees. Thymol and menthol were the most toxic compounds to honey bees, and alpha -terpineol was the least toxic. Menthol, citral, thymol, and carvacrol were more toxic to tracheal mites than to honey bees. Pulegone, d-limonene, and alpha -terpineol were more toxic to honey bees than to tracheal mites. Menthol was 18.9 times more toxic to tracheal mites than to honey bees at the LC sub(50) concentrations; however, as the concentration increased, bee mortality increased more rapidly than mite mortality, and menthol was only 5.7 times more toxic at the LC sub(90) concentrations. Probit regressions for bee and mite mortality were parallel for citral and thymol. Citral and thymol were 2.9 (2.5-3.3) and 2.0 (1.0-3.6) times more toxic to tracheal mites, respectively, at all concentrations estimated.
Background. Group A streptococcus (GAS) causes illness ranging from uncomplicated pharyngitis to life-threatening necrotizing fasciitis, toxic shock, and rheumatic fever. Attempts to develop an M ...protein–based vaccine have been hindered by the fact that some M proteins elicit both protective antibodies and antibodies that cross-react with human tissues. New molecular techniques have allowed the previous obstacles to be largely overcome. Methods. The vaccine is comprised of 4 recombinant proteins adsorbed to aluminum hydroxide that contain N-terminal peptides from streptococcal protective antigen and M proteins of 26 common pharyngitis, invasive, and/or rheumatogenic serotypes. Thirty healthy adult subjects received intramuscular 26-valent GAS vaccine (400 µg) at 0, 1, and 4 months, with clinical and laboratory follow-up for safety and immunogenicity using assays for tissue cross-reactive antibodies, type-specific M antibodies to 27 vaccine antigens, and functional (opsonization) activity of M protein antibodies. Results. The incidence of local reactogenicity was similar to that for other aluminum hydroxide–adsorbed vaccines in adults. No subject developed evidence of rheumatogenicity or nephritogenicity, and no induction of human tissue–reactive antibodies was detected. Overall, 26 of 27 antigenic peptides evoked a >4-fold increase in the geometric mean antibody titer over baseline. The mean log2 fold-increase in serum antibody titer (± standard error of the mean) for all 27 antigens was 3.67 ± 0.21. A significant mean log2 reduction in streptococcal bacterial counts in serum samples obtained after immunization was seen in opsonization assays for all M serotypes. Conclusions. On the basis of epidemiological data demonstrating that the majority of cases of pharyngitis, necrotizing fasciitis, and other invasive streptococcal infections are caused by a limited number of serotypes, this 26-valent vaccine could have significant impact on the overall burden of streptococcal disease.
Big-eyed bugs have been well documented as predators on a diverse group of arthropod prey in turfgrasses; however, little is known about the big-eyed bug species associated with buffalograss, or ...their feeding habits relative to the western chinch bug, Blissus occiduus Barber. This research documented that Geocoris uliginosus (Say) was the predominant big-eyed bug species associated with buffalograss, obtained information on its feeding behavior, and characterized predation rates. Laboratory studies documented G. uliginosus, as a predator of B. occiduus. While all life stages of B. occiduus were attacked by G. uliginosus, predation was greater on 1st through 4th instars than on 5th instars or adults. Low 5th instar and adult chinch bug mortality was likely the result of their larger biomass, as well as their superior size and strength compared to younger (1st through 4th instar) chinch bugs. The mean number of 1st through 3rd instar chinch bugs consumed by G. uliginosus at each evaluation period was higher than for 5th instar or adult chinch bugs. Based on this research, chinch bug management decisions should take into consideration big-eyed bug densities, especially when the majority of B. occiduus are early (1st–2nd) instars.
