Tribo-dynamics analysis of engine small-end bearings can help to find potential schemes to avoid seizure failure that is common in today's engines under heavy loads and high temperatures. The ...challenge lies in determining real temperature boundary conditions due to the combined effects of frictional heat and environmental sources. This study develops a new in-situ temperature measuring and wireless transmission system, implemented in a full-scale engine under various conditions. Combining simulation and experiment provides a comprehensive understanding of the difficult-to-measure oil temperature and tribo-dynamics behavior. Results show the test bearing upper part reaches 165 °C due to piston heat transfer. Simulated high-temperature oil aggregation on the lower edges corresponds nicely to oil overheating carbonization at the same locations of the test bearing.
•Engine CRSE bearings’ tribo-dynamics model is improved by the real thermal boundary.•A full-scale engine test is conducted using a novel wireless measurement system.•The predicted high-temperature position is consistent with the test one.
Digital innovation ecosystems are currently experiencing a period of growth and are navigating uncertain environments. Improving resilience is an important prerequisite for ensuring sustainable ...developments. This study, based on the technology, organization, and environment (TOE) framework, examines the impact of multilevel antecedent conditions on digital innovation ecosystem resilience using data from 31 Chinese provinces. By applying a necessary condition analysis (NCA) and fuzzy-set qualitative comparative analysis (fsQCA), this study reveals complex causal relationships between five antecedents at the “technology–organization–environment” levels and digital innovation ecosystem resilience, along with the improvement paths of digital innovation ecosystem resilience. The results show the following: Firstly, individual antecedent conditions alone do not constitute necessary conditions for high or non-high digital innovation ecosystem resilience. Secondly, there are five configuration paths leading to high digital innovation ecosystem resilience, namely, a digital technology-enabled organization–environment-driven type (H1a), an organization–environment dual-wheel-driven type (H1b), a digital technology-led environment-driven type (H2), a technology–organization–environment trilateral type (H3), and a pressure–organization-driven type (H4). Thirdly, three configuration paths result in non-high digital innovation ecosystem resilience, exhibiting an asymmetric relationship with paths associated with the configuration paths of high digital innovation ecosystem resilience. Finally, potential substitution relationships exist among antecedent conditions at the technological, organizational, and environmental levels.
Total flavonoids of Rhizoma drynariae (TFRD), a Chinese medicine, is widely used in the treatment of fracture, bone defect, osteoporosis and other orthopedic diseases, and has achieved good effects. ...Purpose of this trial was to explore efficacy of TFRD on bone graft’s mineralization and osteoblasts’ differentiation in Masquelet induced membrane technique in rats. Forty male Sprague-Dawley rats were randomly divided into high dose group (H-TFRD), middle dose group (M-TFRD), low dose group (L-TFRD) and control group (control). The critical size bone defect model of rats was established with 10 rats in each group. Polymethyl methacrylate (PMMA) spacer was implanted into the defect of right femur in rats. After the formation of the induced membrane, autogenous bone was implanted into the induced membrane. After 12 weeks of bone graft, bone tissues in the area of bone graft were examined by X-ray, Micro-CT, hematoxylin-eosin (HE) and Masson trichrome staining to evaluate the growth of the bone graft. -catenin, c-myc, COL1A1, BMP-2 and OPN in bone graft were quantitatively analyzed by Western blot and Immunohistostaining. Osteoblasts were cultured in the medium containing TFRD. Cell Counting Kit-8 (CCK-8) method, Alkaline phosphatase (ALP) and Alizarin Red S (ARS) staining, Western blot, RT-PCR and other methods were used to detect the effects of TFRD on the proliferation of osteoblasts and the regulation of Wnt/β-catenin signaling pathway.
In vivo
experiments showed that the growth and mineralization of bone graft in TFRD group was better. Moreover, the expression of Wnt/β-catenin and osteogenesis-related proteins in bone tissue of TFRD group was more than that in other groups.
In vitro
experiments indicated that osteoblasts proliferated faster, activity of ALP was higher, number of mineralized nodules and proteins related to osteogenesis were more in TFRD group. But blocking Wnt/β-catenin signaling pathway could limit these effects. Therefore, TFRD could promote mineralization of bone graft and differentiation of osteoblasts in a dose-dependent manner during growing period of the bone graft of induced membrane technique, which is partly related to the activation of Wnt/β-catenin signaling pathway.

Ginkgo biloba, which contains flavonoids as bioactive components, is widely used in traditional Chinese medicine. Increasing the flavonoid production of medicinal plants through genetic engineering generally focuses on the key genes involved in flavonoid biosynthesis. However, the molecular mechanisms underlying such biosynthesis are not yet well understood. To understand these mechanisms, a combination of second-generation sequencing (SGS) and single-molecule real-time (SMRT) sequencing was applied to G. biloba. Eight tissues were sampled for SMRT sequencing to generate a high-quality, full-length transcriptome database. From 23.36 Gb clean reads, 12,954 alternative polyadenylation events, 12,290 alternative splicing events, 929 fusion transcripts, 2,286 novel transcripts, and 1,270 lncRNAs were predicted by removing redundant reads. Further studies reveal that 7 AS, 5 lncRNA, and 6 fusion gene events were identified in flavonoid biosynthesis. A total of 12 gene modules were revealed to be involved in flavonoid metabolism structural genes and transcription factors by constructing co-expression networks. Weighted gene coexpression network analysis (WGCNA) analysis reveals that some hub genes operate during the biosynthesis by identifying transcription factors (TFs) and structure genes. Seven key hub genes were also identified by analyzing the correlation between gene expression level and flavonoids content. The results highlight the importance of SMRT sequencing of the full-length transcriptome in improving genome annotation and elucidating the gene regulation of flavonoid biosynthesis in G. biloba by providing a comprehensive set of reference transcripts.
The internal–external composite meshing mechanism should be the best way to improve the transmission performance of a silent chain drive, but there is no internal–external composite meshing Hy-Vo ...chain under the classical design approach. In this research, by building the Hy-Vo chain meshing system, the difference between the meshing of the chain–sprocket and the meshing of the rack–gear is deeply analyzed. By rigidifying the external meshing Hy-Vo chain as a rack, the fluctuation range of the pitch line of the external meshing Hy-Vo chain is analyzed. Based on the relations between the actual chain pitch line and the rigid chain pitch line, the design method for the internal meshing profile of the internal–external composite meshing Hy-Vo chain is deduced, the necessary conditions are obtained, and the meshing characteristics are analyzed. By conducting comparative multi-body dynamics experiments, the results prove that the design method of the internal–external composite meshing Hy-Vo chain proposed in this study is effective and feasible. The results also show that the description of the internal–external composite meshing in the classical meshing theory is not completely correct. In fact, the internal–external composite meshing mechanism cannot reduce the meshing impact of the Hy-Vo chain system and can only improve the system polygon action, the meshing performance, and the transmission stability.
RNA sequencing can simultaneously identify exonic polymorphisms and quantitate gene expression. Here we report RNA sequencing of developing maize kernels from 368 inbred lines producing 25.8 billion ...reads and 3.6 million single-nucleotide polymorphisms. Both the MaizeSNP50 BeadChip and the Sequenom MassArray iPLEX platforms confirm a subset of high-quality SNPs. Of these SNPs, we have mapped 931,484 to gene regions with a mean density of 40.3 SNPs per gene. The genome-wide association study identifies 16,408 expression quantitative trait loci. A two-step approach defines 95.1% of the eQTLs to a 10-kb region, and 67.7% of them include a single gene. The establishment of relationships between eQTLs and their targets reveals a large-scale gene regulatory network, which include the regulation of 31 zein and 16 key kernel genes. These results contribute to our understanding of kernel development and to the improvement of maize yield and nutritional quality.
Summary
Spinal cord injury (SCI) is a disease involving gastrointestinal disorders. The underlying mechanisms of the potential protective effects of electroacupuncture (EA) and 5‐hydroxytryptamine ...(5‐HT) system on SCI remain unknown. We investigated whether EA improves gut microbial dysbiosis in SCI and regulates the 5‐HT system. 16S rDNA gene sequencing was applied to investigate alterations in the gut microbiome of the rats. Faecal metabolites and the expression of the 5‐HT system were detected. EA and faecal microbiota transplantation (FMT) treatment facilitated intestinal transmission functional recovery and restored the colon morphology of SCI rats. The composition of the intestinal microbiota, including numbers of phylum Proteobacteria, class Clostridia, order Bacteroidales, and genus Dorea, were amplified in SCI rats, and EA and FMT significantly reshaped the intestinal microbiota. SCI resulted in disturbed metabolic conditions in rats, and the EA and FMT group showed increased amounts of catechin compared with SCI rats. SCI inhibited 5‐HT system expression in the colon, which was significantly reversed by EA and FMT treatment. Therefore, EA may ameliorate SCI by modulating microbiota and metabolites and regulate the 5‐HT system. Our study provides new insights into the pathogenesis and therapy of SCI from the perspective of microbiota and 5‐HT regulation.
