This study evaluates the performance and robustness of 22 established and newly proposed glare prediction metrics. Experimental datasets of daylight-dominated workplaces in office-like test rooms ...were collected from studies by seven research groups in six different locations (Argentina, Denmark, Germany, Israel, Japan and the United States). The variability in experimental setups, locations and research teams allowed reliable evaluation of the performance and robustness of glare metrics for daylight-dominated workplaces. Independent statistical methods were applied to individual datasets and also to one combined dataset to evaluate the performance and robustness of the 22 glare metrics. As performance and robustness are not established in literature, we defined performance as: (1) the ability of the metric value to describe the glare scale (evaluated by Spearman rank correlation), and (2) the ability of the metric to distinguish between disturbing and non-disturbing situations (evaluated by diagnostic receiver operating characteristic curve analysis tests). Furthermore, we defined robustness as the ability of a metric to deliver meaningful results when applied to different datasets and to fail as few as possible statistical tests. Average Spearman rank correlation coefficients in the range of 0.55–0.60 as well as average prediction rates to distinguish between disturbing and non-disturbing glare of 70–75% for several of the metrics indicate their reliability. The results also show that metrics considering the saturation effect as a main input in their equation perform better and are more robust in daylight-dominated workplaces than purely contrast-based metrics or purely empirical metrics. In this study, the daylight glare probability (DGP) delivered the highest performance amongst the tested metrics and was also found to be the most robust. Future research should aim to optimise the terms of glare equations which combine contrast and saturation effects, such as DGP, PGSV or UGRexp, to achieve metrics that also perform reliably in dimmer lighting conditions than the ones explored in this study.
Objective and Background
Human periodontal ligament mesenchymal stem cells (hPDLMSCs) are reported to be responsible for homeostasis and regeneration of periodontal tissue. Although hPDLMSCs are ...commonly cultured in monolayers, monolayer cultures have been reported as inferior to 3‐dimensional cultures such as spheroids, which are spherical clusters of cells formed by self‐assembly. The aim of this study was to examine the osteogenic phenotype of spheroids of hPDLMSCs, compared with monolayer cultures of hPDLMSC, in vitro and in vivo.
Material and Methods
Spheroids were formed using microwell chips that were tagged with polyethylene glycol. Mesenchymal stem cell (MSC) markers in hPDLMSC spheroids were examined by flow cytometer. Real‐time polymerase chain reaction analysis was examined to measure the expressions of stemness markers and osteogenesis‐related genes in monolayer and spheroid‐cultured hPDLMSCs. Immunofluorescence analysis was performed to confirm protein expressions of stemness markers in PDLMSC spheroids. Nodule formation assay, alkaline phosphatase (ALP) activity assay and transplantation assay in a mouse calvarial defect model were performed to confirm the osteogenic potential of hPDLMSC spheroids. To elucidate the mechanism of spheroid culture enhanced osteogenesis in hPDLMSCs with osteoinductive medium (OIM), a small interfering RNA (siRNA) assay targeted with secreted frizzled‐related protein 3 (SFRP3) was examined. The levels of SFRP3 expression in monolayer and spheroid‐cultured hPDLMSCs with OIM were measured by real‐time polymerase chain reaction and western blotting analysis. ALP gene expression and ALP activity were examined in SFRP3‐deficient hPDLMSC spheroids.
Results
The hPDLMSC spheroids expressed MSC markers, which were similar to hPDLMSCs grown in monolayer cultures. Intriguingly, the protein and mRNA expressions of transcription factors that regulate “stemness” were significantly increased in hPDLMSC spheroids, compared with hPDLMSCs in monolayer cultures. Nodule formation by hPDLMSCs was significantly increased in spheroid cultures grown with OIM, compared with monolayer‐cultured hPDLMSCs. ALP activity and expression of osteogenesis‐related genes were also significantly enhanced in hPDLMSC spheroids, compared with monolayer cultures. Treatment with hPDLMSC spheroids significantly enhanced new bone formation in a murine calvarial defect model, compared with hPDLMSCs in monolayer culture. Finally, to elucidate mechanisms by which spheroid culture enhances ALP activation in hPDLMSCs grown with OIM, an siRNA assay was used to manipulate expression of SFRP3, a Wnt signaling antagonist. Knockdown of SFRP3 suppressed ALP gene expression in hPDLMSCs grown in OIM; further, it suppressed ALP activity in spheroid culture. These data suggest that the enhancement of osteogenic potential in hPDLMSC spheroids is regulated through SFRP3‐mediated ALP activation.
Conclusion
Spheroid cultures of hPDLMSCs may be a novel and useful tool in regenerative medicine.
Context.
The JAXA asteroid sample return mission Hayabusa2 acquired a huge quantity of data from the asteroid (162173) Ryugu during its 1.5 years in asteroid proximity orbit. On December 5, 2020 ...(Japan time), Hayabusa brought back to Earth a 5.4 g sample from Ryugu’s surface.
Aims.
We analyzed the near-infrared spectra of Ryugu, in particular the band at 2.72 μm, with the aim to investigate the hydrogen content of the H
2
O and OH
−
groups in hydrated phyllosilicates on Ryugu’s surface.
Aims.
We applied two different methods, normalized optical path length (NOPL) and effective single-particle absorption thickness (ESPAT), to the 3 μm region absorption band, and we compared the obtained spectral parameters with those obtained from carbonaceous chondrite meteorites whose H content was determined in the laboratory.
Methods.
We derived an exponential correlation between the selected meteorite H content and its respective ESPAT and NOPL parameters. The average value of the H content obtained on Ryugu’s surface with its relative variations, combining the results obtained with the two methods, is 0.52
−0.21
+0.16
wt.%. These methods can be applied to other asteroids that exhibit a 3 μm region absorption band to estimate the mean average of H content.
Results.
The results of the ESPAT and NOPL methods used on the Ryugu spectral data present small variations across Ryugu’s surface and do not show any evident relation with the surface geomorphological structures. Our estimation of the global average H content of Ryugu is in agreement with those of several aqueously altered carbonaceous chondrites measured in the laboratory and is most similar to the H content of heated CM. The study of phyllosilicate H
2
O and OH
−
group hydrogen content on Ryugu and the derived method may be applied to other observed primitive asteroids. The obtained results will allow Solar System evolution models to be constrained and will allow the formation and evolution of the Solar System to be better understood.
In postmortem examinations, the drug analysis of hair is effective for revealing drug-use history. Additionally, a method to estimate the day of death using hair was previously developed by analyzing ...a single hair strand segmented at 0.4-mm intervals (micro-segmental hair analysis). However, for drowned bodies, drugs in the hair may be washed out due to soaking in water for extended periods. To evaluate the possibility of measuring drug distribution in the hair of drowned bodies, drug stability in hair samples soaked in various aqueous solutions was examined. First, reference hair strands of drug users containing specific drugs consistently along the hair shaft were prepared. The participants ingested 4 hay-fever medicines (fexofenadine, epinastine, cetirizine, and loratadine) every day for approximately 4 months before hair collection. Each reference strand was divided into regions, and each region was soaked in different solutions containing various solutes for extended periods up to approximately 2 months. In solutions without divalent ions (Ca
2+
and Mg
2+
), the drug content in the hair decreased up to approximately 5 % with increasing salt concentration and soaking time. However, the decreased drug content was negligible in solutions containing divalent ions, implying that the divalent ions prevented drugs contained in hair from washing out. As natural river and sea waters contain divalent ions, the drugs in hair were hardly washed out even when the hair was soaked for 2 months. Thus, it was concluded that drug-distribution measurements using micro-segmental analysis can also be applied to the hairs of drowned bodies.

Hair and nails are often used to prove drug intake over several months. However, it is impossible to determine the day of drug intake by conventional segmental analysis of bulk samples. To improve ...this segmental analysis, we prepared accurate 0.4-mm hair and 0.2-mm nail segments, which correspond to their respective growth rates of 1–2 days, using a tissue slicer. The aim of this study was to develop an efficient method to extract drugs from a single sub-millimeter segment of hair and nail. Hair and nails were collected from a subject who was administered a single dose of chlorpheniramine. Four drug extraction methods based on different principles such as sonication, microwaves, micropulverization, and alkaline dissolution were compared. Short-duration sonication followed by long-duration soaking served the aim. Drug extraction from a sub-millimeter segment was performed in three steps as follows: a segment was first washed, followed by sonication for 10 min soaking in the extraction solution for 24 h. The drug concentrations in the three extracts from each segment were quantified using high performance liquid chromatography-tandem mass spectrometry. Each concentration was displayed on a single hair strand and a single nail block so that the first, second, and third extracts corresponded to components on the surface, in the outer layer, and within the sample, respectively. The distribution of chlorpheniramine in a hair successfully reflected the intake history. This method can be used in the future to measure the detailed distribution of drugs in a single hair and nail.

BACKGROUND AND AIMS: Global warming is gaining significance as a threat to natural and managed ecosystems since temperature is one of the major environmental factors affecting plant productivity. ...Hence, the effects of moderate temperature increase on the growth and development of the tomato plant (Lycopersicon esculentum) were investigated. METHODS: Plants were grown at 32/26 °C as a moderately elevated temperature stress (METS) treatment or at 28/22 °C (day/night temperatures) as a control with natural light conditions. Vegetative growth and reproductive development as well as sugar content and metabolism, proline content and translocation in the androecium were investigated. KEY RESULTS: METS did not cause a significant change in biomass, the number of flowers, or the number of pollen grains produced, but there was a significant decrease in the number of fruit set, pollen viability and the number of pollen grains released. Glucose and fructose contents in the androecium (i.e. all stamens from one flower) were generally higher in the control than METS, but sucrose was higher in METS. Coincidently, the mRNA transcript abundance of acid invertase in the androecium was decreased by METS. Proline contents in the androecium were almost the same in the control and METS, while the mRNA transcript level of proline transporter 1, which expresses specifically at the surface of microspores, was significantly decreased by METS. CONCLUSIONS: The research indicated that failure of tomato fruit set under a moderately increased temperature above optimal is due to the disruption of sugar metabolism and proline translocation during the narrow window of male reproductive development.
Bioactive glasses (BAG) are used as bone-graft substitutes in orthopaedic surgery. A specific BAG scaffold was developed by sintering BAG-S53P4 granules. It is hypothesised that this scaffold can be ...used as a bone substitute to fill bone defects and induce a bioactive membrane (IM) around the defect site. Beyond providing the scaffold increased mechanical strength, that the initial inflammatory reaction and subsequent IM formation can be enhanced by coating the scaffolds with poly(DL-lactide-co-glycolide) (PLGA) is also hypothesised. To study the immunomodulatory effects, BAG-S53P4 (± PLGA) scaffolds were placed on monolayers of primary human macrophage cultures and the production of various pro- and anti-inflammatory cytokines was assessed using reverse transcriptase quantitative polymerase chain reaction (RT-qPCR) and ELISA. To study the osteogenic effects, BAG-S53P4 (± PLGA) scaffolds were cultured with rabbit mesenchymal stem cells and osteogenic differentiation was evaluated by RT-qPCR and matrix mineralisation assays. The scaffold ion release was quantified and the BAG surface reactivity visualised. Furthermore, the pH of culture media was measured. BAG-S53P4 scaffolds had both anti-inflammatory and osteogenic properties that were likely attributable to alkalinisation of the media and ion release from the scaffold. pH change, ion release, and immunomodulatory properties of the scaffold could be modulated by the PLGA coating. Contrary to the hypothesis, the coating functioned by attenuating the BAG surface reactions and subsequent anti-inflammatory properties, rather than inducing an elevated inflammatory response compared to BAG-S53P4 alone. These results further validated the use of BAG-S53P4 (± PLGA) scaffolds as bone substitutes and indicate that scaffold properties can be tailored to a specific clinical need.
Ingested drugs can be taken up into nails and hairs from the bloodstream and these drugs can be stably retained over several months or more. Free edges of nails can often be used as a specimen to ...prove drug intake. However, the mechanism of drug uptake into the nails is unclear. Although it is presumed that there are horizontal uptake routes at the nail root and vertical uptake routes at the nail bed against the direction of growth, three-dimensional distribution analysis is required to verify this phenomenon. Herein, we first developed a method to measure the three-dimensional distribution of drugs in the nails using the combination of micro-segmentation of nails (0.2 × 1.5 × 0.06 mm size) and highly sensitive quantification of drugs by LC-MS/MS. Carbinoxamine was administered as a model compound to a subject in a single dose, and then the free edges of big toenails were collected every several weeks over a year. The three-dimensional distribution of carbinoxamine in each free edge was visualized and arranged along the collection period. Carbinoxamine was localized in the lower layer during the early collection period (up to 78 days after drug ingestion) and in the upper and middle layers later (134 days or later). The changes in the drug distribution in the nails along the collection period implied that two-way drug uptake takes place in the whole nail through both the nail bed and the nail root simultaneously. The developed analytical method could be useful to elucidate the mechanism of drug uptake into the nails.
