Pattern recognition receptors (PRRs) recognize the conserved molecular structure of pathogens and trigger the signaling pathways that activate immune cells in response to pathogen infection. ...Toll-like receptors (TLRs) are the first and best characterized innate immune receptors. To date, at least 20 TLR types (TLR1, 2, 3, 4, 5M, 5S, 7, 8, 9, 13, 14, 18, 19, 20, 21, 22, 23, 24, 25, and 26) have been found in more than a dozen of fish species. However, of the TLRs identified in fish, direct evidence of ligand specificity has only been shown for TLR2, TLR3, TLR5M, TLR5S, TLR9, TLR21, and TLR22. Some studies have suggested that TLR2, TLR5M, TLR5S, TLR9, and TLR21 could specifically recognize PAMPs from bacteria. In addition, other TLRs including TLR1, TLR4, TLR14, TLR18, and TLR25 may also be sensors of bacteria. TLR signaling pathways in fish exhibit some particular features different from that in mammals. In this review, the ligand specificity and signal pathways of TLRs that recognize bacteria in fish are summarized. References for further studies on the specificity for recognizing bacteria using TLRs and the following reactions triggered are discussed. In-depth studies should be continuously performed to identify the ligand specificity of all TLRs in fish, particularly non-mammalian TLRs, and their signaling pathways. The discovery of TLRs and their functions will contribute to the understanding of disease resistance mechanisms in fish and provide new insights for drug intervention to manipulate immune responses.

Land surface temperature (LST) is one of the most important factors in the land-atmosphere interaction process. Raw measured LSTs may contain biases due to instrument replacement, changes in ...recording procedures, and other non-climatic factors. This study attempts to reduce the above biases in raw daily measurements and achieves a homogenized daily LST dataset over China using 2360 stations from 1960 through 2017. The high-quality land surface air temperature (LSAT) dataset is used to correct the LST warming biases especially evident during cold months in regions north of 40°N due to the replacement of observation instruments around 2004. Subsequently, the Multiple Analysis of Series for Homogenization (MASH) method is adopted to detect and then adjust the daily observed LST records. In total, 3.68 × 10

monthly records (about 20%) are detected. A large number of these effective breakpoints are located over large parts of the Sichuan Basin and southern China. After the MASH procedure, LSTs at more than 80% of the breakpoints are adjusted within +/− 0.5°C, and of the remaining breakpoints, only 10% are adjusted over 1.5°C. Compared to the raw LST dataset over the whole domain, the homogenization significantly reduces the mean LST magnitude and its interannual variability as well as its linear trend at most stations. Finally, we perform preliminary analysis upon the homogenized LST and find that the annual mean LST averaged across China shows a significant warming trend 0.22°C (10 yr)

. The homogenized LST dataset can be further adapted for a variety of applications (e.g., model evaluation and extreme event characterization).
Microbial degradation is a useful tool for inhibiting or preventing polycyclic aromatic hydrocarbons (PAHs) widely distributed in marine environment after oil spill accidents. This study aimed to ...evaluate the potential and diversity of bacteria Bacillus sp. PAH-2 on Benzo (a) anthracene (BaA), Pyrene (Pyr), and Benzo (a) pyrene (BaP), their composite system, aromatic components system, and crude oil. The seven-day degradation rates against BaA, Pyr, and BaP were 20.6%, 12.83%, and 17.49%, respectively. Further degradation study of aromatic components demonstrated PAH-2 had a high degradation rate of substances with poor stability of molecular structure. In addition, the degradation of PAHs in crude oil suggested PAH-2 not only made good use of PAHs in such a more complex structure of pollutants but the saturated hydrocarbons in the crude oil also showed a good application potential.

and purified. Secondly, the immune-stimulatory activities of the first AAP fraction (AAP1) were evaluated in the CTX-treated mice. Results showed that AAP1 significantly enhanced immune organ indexes, remarkably stimulated IFN-γ, IL-2, IL-4, IL-10, and TNF-α levels in the serum, and dramatically up-regulated the mRNA levels of Claudin-1, Occludin and ZO-1. Compared to the CTX group, AAP1 administration restored the gut microbiota composition similar to that of the control group by decreasing the ratio of Firmicutes/Bacteroidetes and increasing the relative abundances of short-chain fatty acid-producing microbiota. This study provides useful information for its further application as an immune-stimulator in foods and drugs.
Rheumatoid arthritis (RA), a kind of autoimmune disorder, is featured by many physical symptoms and proliferation of fibroblast-like synoviocytes (FLSs). The relevance of long non-coding RNAs ...(lncRNAs) in the progression of RA has been probed. Hence, the goal of this report was to investigate the action of plasmacytoma variant translocation 1 (PVT1), a lncRNA, in FLSs and the basic mechanism.
Initially, RA rats were developed to evaluate the expression of PVT1, microRNA-543 (miR-543), and signal peptide-CUB-EGF-like containing protein 2 (SCUBE2) in synovial tissues. Enhancement or loss of PVT1 or miR-543 was achieved to explore their effects on proliferation, cell cycle, and apoptosis of FLSs. The interaction between PVT1 and miR-543 and between miR-543 and its putative target SCUBE2 was examined to elucidate the correlations. Finally, the protein expression of proliferation- and apoptosis-associated genes were assessed by western blot assays.
PVT1 was overexpressed in synovial tissues from RA patients through microarray expression profiles. The PVT1 and SCUBE2 expression was boosted, and miR-543 was reduced in synovial tissues of rats with RA. PVT1 specifically bound to miR-543, and miR-543 negatively regulated SCUBE2 expression. Overexpression of PVT1 or silencing of miR-543 enhanced SCUBE2 expression, thereby promoting proliferation and interleukin-1β (IL-1β) secretion, while inhibiting apoptosis rate of FLSs. Conversely, si-SCUBE2 reversed the role of miR-543 inhibitor.
The key findings support that PVT1 knockdown has the potency to hinder RA progression by inhibiting SCUBE2 expression to sponge miR-543.
Food dyes, or color additives, are often added into foods, cosmetics and beverages during processing to improve the sensory attributes of the final products. However, the toxicity of tartrazine (TZ), ...one of the most common azo-dyes, is still unclear, and needs to be ascertained by further study. Hence, in the present study, we aimed to evaluate the effects of TZ consumption on health by using a teleost, crucian carp (Carassius auratus) as the experimental fish. TZ consumption (1.4, 5.5 and 10 mg/kg bwt/day) could cause severe histopathological and cellular alterations in intestine and liver. The height of intestinal villus, thickness of intestinal muscle, and microvilli density were also affected. With the increasing of TZ concentrations, the activities of antioxidant enzymes (CAT, SOD and GSH-Px), exhibited a decreasing trend, while the contents of MDA elevated. Upregulations of pro-inflammatory cytokines (il6 and tnfα), anti-inflammatory cytokines (il8, and il10) and other immune related genes (complement component 3 (c3), lysozymes (lyz), β-defensin 3 (defb3)), were observed after TZ uptake. In addition, TZ consumption also affected the community structure of the microbiota in the intestine of crucian carp. The amount of some probiotic bacteria (Roseomonas, Rhodococcus and Bacillus) and the bacteria (Bacteroides and Clostridium), producing short chain fatty acids, were significantly reduced, and some pathogenetic microorganisms (e.g. Bdellovibrio and Shewanella) were significantly increased after TZ uptake. In summary, the data in the present study indicate that TZ consumption, even at a low concentration, may lead to adverse effects on fish health. Therefore, in aquaculture, it is necessary to be informed about the hazardous effects of TZ, and more attentions should be focused on using natural substitutes.

This study assesses the performance of temperature extremes over China in two regional climate models (RCMs), RegCM4 and WRF, driven by the ECMWF’s 20th century reanalysis. Based on the advice of the ...Expert Team on Climate Change Detection and Indices (ETCCDI), 12 extreme temperature indices (i.e., TXx, TXn, TNx, TNn, TX90p, TN90p, TX10p, TN10p WSDI, ID, FD, and CSDI) are derived from the simulations of two RCMs and compared with those from the daily station-based observational data for the period 1981–2010. Overall, the two RCMs demonstrate satisfactory capability in representing the spatiotemporal distribution of the extreme indices over most regions. RegCM performs better than WRF in reproducing the mean temperature extremes, especially over the Tibetan Plateau (TP). Moreover, both models capture well the decreasing trends in ID, FD, CSDI, TX10p, and TN10p, and the increasing trends in TXx, TXn, TNx, TNn, WSDI, TX90p, and TN90p, over China. Compared with observation, RegCM tends to underestimate the trends of temperature extremes, while WRF tends to overestimate them over the TP. For instance, the linear trends of TXx over the TP from observation, RegCM, and WRF are 0.53°C (10 yr)

, respectively. However, WRF performs better than RegCM in reproducing the interannual variability of the extreme-temperature indices. Our findings are helpful towards improving our understanding of the physical realism of RCMs in terms of different time scales, thus enabling us in future work to address the sources of model biases.
The polymeric immunoglobulin receptor (pIgR) plays a pivotal role in mucosal immune response by transporting polymeric immunoglobulins onto the surface of mucosal epithelia to protect animals from ...invading pathogens. In this study, the full-length cDNA of pIgR was firstly cloned in Qihe crucian carp (Carassius auratus), hereafter designated as CapIgR, by using reverse transcription polymerase chain reaction and rapid amplification of cDNA ends. The molecular characterization and expression of CapIgR were investigated. The full-length cDNA sequence of CapIgR was composed of 1409 bp, which included a 112 bp 5ʹ-untranslated region (UTR), a 984 bp ORF, and a 313 bp 3ʹ-UTR, with a putative polyadenylation signal sequence AATAAA located upstream of the poly(A) tail. The deduced amino acid sequence indicated that CapIgR was a single-spanning transmembrane protein with 327 amino acids and possessed a signal peptide, an extracellular region containing two immunoglobulin-like domains, a transmembrane region, and an intracellular region. The mRNA expression levels of CapIgR were detected in different tissues of healthy C. auratus by quantitative real-time PCR, and the highest expression level was found in the liver. After Aeromonas hydrophila challenge, CapIgR expression was upregulated in different tissues at certain time points, and temporal expression changes of CapIgR fluctuated in a time-dependent manner. CapIgR exhibited rapid immune response to A. hydrophila challenge and played an important role in the immune defense of fish. These findings provided insights into the structure, function, and immune defense mechanism of CapIgR in C. auratus. This study can serve as a basis for developing disease control strategies in aquaculture.

Short-chain chlorinated paraffins (SCCPs) were included in the Stockholm Convention in 2017. SCCPs have persistence, bioaccumulation, long-range environmental mobility and biological toxicity, ...significant toxicity to aquatic organisms, and potential carcinogenicity. Little study was on the progress research on the current environmental pollution in China. We reviewed the pollution conditions of SCCPs in air, soil, and water and their accumulation in food and organisms in China, especially for the contaminations of aquatic ecosystem. Meanwhile, we summarize the recent studies on the toxic effects and toxicological mechanisms of SCCPs on aquatic organisms and mammals. Finally, the further direction and trends for SCCP research were proposed. More efforts are necessary to conduct a comprehensive risk assessment and evaluate the relative importance of the various exposure routes.
