Administration of dibutyl phthalate (DBP) to pregnant rats causes reproductive disorders in male offspring, resulting from suppression of intratesticular testosterone, and is used as a model for ...human testicular dysgenesis syndrome (TDS). DBP exposure in pregnancy induces focal dysgenetic areas in fetal testes that appear between e19.5-e21.5, manifesting as focal aggregation of Leydig cells and ectopic Sertoli cells (SC). Our aim was to identify the origins of the ectopic SC. Time-mated female rats were administered 750 mg/kg/day DBP in three different time windows: full window (FW; e13.5-e20.5), masculinisation programming window (MPW; e15.5-e18.5), late window (LW; e19.5-e20.5). We show that DBP-MPW treatment produces more extensive and severe dysgenetic areas, with more ectopic SC and germ cells (GC) than DBP-FW treatment; DBP-LW induces no dysgenesis. Our findings demonstrate that ectopic SC do not differentiate de novo, but result from rupture of normally formed seminiferous cords beyond e20.5. The more severe testis dysgenesis in DBP-MPW animals may result from the presence of basally migrating GC and a weakened basal lamina, whereas GC migration was minimal in DBP-FW animals. Our findings provide the first evidence for how testicular dysgenesis can result after normal testis differentiation/development and may be relevant to understanding TDS in human patients.
The aim was to investigate the incidence of sentinel lymph node (SLN) metastases and the contribution of SLN mapping in presumed low- and intermediate-risk endometrial cancer (EC). A multicenter, ...prospective cohort study in presumed low- and intermediate-risk EC patients was performed. Patients underwent SLN mapping using cervical injections of indocyanine green and a minimally invasive hysterectomy with bilateral salpingo-oophorectomy. The primary outcome was the incidence of SLN metastases, leading to adjusted adjuvant treatment. Secondary outcomes were the SLN detection rate and the occurrence of complications. Descriptive statistics and univariate general linear model analyses were used. A total of 152 patients were enrolled, with overall and bilateral SLN detection rates of 91% and 61%, respectively. At final histology, 78.9% of patients (
= 120) had truly low- and intermediate-risk EC. Macro- and micro-metastases were present in 11.2% (
= 17/152), and three patients had isolated tumor cells (2.0%). Nine patients (5.9%) had addition of adjuvant radiotherapy based on SLN metastases only. In 2.0% of patients with high-risk disease, adjuvant therapy was more limited due to negative SLNs. This study emphasizes the importance of SLN mapping in presumed early-stage, grade 1 and 2 EC, leading to individualized adjuvant management, resulting in less undertreatment and overtreatment.
Chronic pain is a debilitating problem, and insights in the neurobiology of chronic pain are needed for the development of novel pain therapies. A genome-wide association study implicated the 5p15.2 ...region in chronic widespread pain. This region includes the coding region for FAM173B, a functionally uncharacterized protein. We demonstrate here that FAM173B is a mitochondrial lysine methyltransferase that promotes chronic pain. Knockdown and sensory neuron overexpression strategies showed that FAM173B is involved in persistent inflammatory and neuropathic pain via a pathway dependent on its methyltransferase activity. FAM173B methyltransferase activity in sensory neurons hyperpolarized mitochondria and promoted macrophage/microglia activation through a reactive oxygen species-dependent pathway. In summary, we uncover a role for methyltransferase activity of FAM173B in the neurobiology of pain. These results also highlight FAM173B methyltransferase activity as a potential therapeutic target to treat debilitating chronic pain conditions.
Zearalenone, an oestogenic mycotoxin produced by Fusarium sp., occurs naturally in agricultural commodities. Economic losses and health concerns associated to mycotoxins has attracted research ...interest towards exploring novel approaches to detoxify mycotoxin-contaminated food and feed. The aim of the present work was to study the ability of 11 aflatoxin-degrading Bacillus strains to degrade ZEA. In addition, a qualitative assessment of protease, amylase and cellulase activity of the studied Bacillus strains was made. All strains were able to degrade 58–96.9% ZEA after 72 h. Toxicity towards Artemia salina was significantly reduced (P < 0.0001). Degradation extracts fluorescence decreased 50% indicating a probable cleavage of the lactone ring. Strains RC1A, RC3A and RC6A showed a remarkable enzymatic activity, showing potential to be used as feed additives.
•AFB1-degrading Bacillus sp. strains also degrade ZEA in vitro.•ZEA degradation by Bacillus sp. reduced ZEA toxicity towards A. salina.•Bacillus sp. demonstrated protease, amylase and cellulase activity.
Aims. We aim at detecting water vapor in the atmosphere of the hot Jupiter HD 209458 b and perform a multi-band study in the near infrared with CARMENES. Methods. The water vapor absorption lines ...from the atmosphere of the planet are Doppler-shifted due to the large change in its radial velocity during transit. This shift is of the order of tens of km s−1, whilst the Earth’s telluric and the stellar lines can be considered quasi-static. We took advantage of this shift to remove the telluric and stellar lines using SYSREM, which performs a principal component analysis including proper error propagation. The residual spectra contain the signal from thousands of planetary molecular lines well below the noise level. We retrieve the information from those lines by cross-correlating the residual spectra with models of the atmospheric absorption of the planet. Results. We find a cross-correlation signal with a signal-to-noise ratio (S/N) of 6.4, revealing H2O in HD 209458 b. We obtain a net blueshift of the signal of –5.2 −1.3+2.6 $^{+2.6}_{-1.3}$ −1.3+2.6 km s−1 that, despite the large error bars, is a firm indication of day- to night-side winds at the terminator of this hot Jupiter. Additionally, we performed a multi-band study for the detection of H2O individually from the three near infrared bands covered by CARMENES. We detect H2O from its 0.96–1.06 μm band with a S/N of 5.8, and also find hints of a detection from the 1.06–1.26 μm band, with a low S/N of 2.8. No clear planetary signal is found from the 1.26–1.62 μm band. Conclusions. Our significant H2O signal at 0.96–1.06 μm in HD 209458 b represents the first detection of H2O from this band individually, the bluest one to date. The unfavorable observational conditions might be the reason for the inconclusive detection from the stronger 1.15 and 1.4 μm bands. H2O is detected from the 0.96–1.06 μm band in HD 209458 b, but hardly in HD 189733 b, which supports a stronger aerosol extinction in the latter, in line with previous studies. Future data gathered at more stable conditions and with larger S/N at both optical and near-infrared wavelengths could help to characterize the presence of aerosols in HD 209458 b and other planets.

The crustacean amphipod
Ampelisca brevicornis
was chosen as the bioindicator species. Organisms were exposed for 10 days to sediment spiked with pharmaceutical compounds frequently used and previously detected in the environment: carbamazepine (CBZ), ibuprofen (IBP), fluoxetine (FX), 17α-ethynylestradiol (EE2), propranolol (PRO), and caffeine (CAF). Short-term bioassay to evaluate amphipod mortality was recommended to assess pollution by CBZ, FX, and PRO. IBP and PRO were metabolized by phases I and II detoxification enzymatic activities. Oxidative stress was caused by PRO and CAF. Contrary to expected results, DNA damage (strand breaks) decreased after the exposure of amphipods to sediment spiked with IBP, FX, EE2, PRO, and CAF (including environmental concentrations). FX was neurotoxic to amphipods. The battery of biomarkers tested allowed the assessment of bioavailability, oxidative stress, genotoxicity, and neurotoxicity of the pharmaceuticals analysed. The results of this study suggested that pharmaceutical products at concentrations currently found in the environment might cause a wide variety of adverse effects (based on laboratory studies). The results obtained here are useful for environmental risk assessment of marine sediments contaminated by pharmaceuticals. Nevertheless, more research is needed using field-based marine sediments.
The European Space Agency's Rosetta mission encountered the main-belt asteroid (2867) Steins while on its way to rendezvous with comet 67P/Churyumov-Gerasimenko. Images taken with the OSIRIS ...(optical, spectroscopic, and infrared remote imaging system) cameras on board Rosetta show that Steins is an oblate body with an effective spherical diameter of 5.3 kilometers. Its surface does not show color variations. The morphology of Steins is dominated by linear faults and a large 2.1-kilometer-diameter crater near its south pole. Crater counts reveal a distinct lack of small craters. Steins is not solid rock but a rubble pile and has a conical appearance that is probably the result of reshaping due to Yarkovsky-O'Keefe-Radzievskii-Paddack (YORP) spin-up. The OSIRIS images constitute direct evidence for the YORP effect on a main-belt asteroid.
The Galaxy And Mass Assembly (GAMA) survey furnishes a deep redshift catalog that, when combined with the Wide-field Infrared Survey Explorer (WISE), allows us to explore for the first time the ...mid-infrared properties of > 110,000 galaxies over 120 deg super(2) to z Asymptotically = to 0.5. In this paper we detail the procedure for producing the matched GAMA-WISE catalog for the G12 and G15 fields, in particular characterizing and measuring resolved sources; the complete catalogs for all three GAMA equatorial fields will be made available through the GAMA public releases. The wealth of multiwavelength photometry and optical spectroscopy allows us to explore empirical relations between optically determined stellar mass (derived from synthetic stellar population models) and 3.4 mu m and 4.6 mu m WISE measurements. Similarly dust-corrected H alpha -derived star formation rates can be compared to 12 mu m and 22 mu m luminosities to quantify correlations that can be applied to large samples to z < 0.5. To illustrate the applications of these relations, we use the 12 mu m star formation prescription to investigate the behavior of specific star formation within the GAMA-WISE sample and underscore the ability of WISE to detect star-forming systems at z ~ 0.5. Within galaxy groups (determined by a sophisticated friends-of-friends scheme), results suggest that galaxies with a neighbor within 100 h super(-1) kpc have, on average, lower specific star formation rates than typical GAMA galaxies with the same stellar mass.
