Monolayers of transition metal dichalcogenides can exist in several structural polymorphs, including 2H, 1T and 1T'. The low-symmetry 1T' phase has three orientation variants, resulting from the ...three equivalent directions of Peierls distortion in the parental 1T phase. Using first-principles calculations, we predict that mechanical strain can switch the relative thermodynamic stability between the orientation variants of the 1T' phase. We find that such strain-induced variant switching only requires a few percent elastic strain, which is eminently achievable experimentally with transition metal dichalcogenide monolayers. Calculations indicate that the transformation barrier associated with such variant switching is small (<0.2 eV per chemical formula unit), suggesting that strain-induced variant switching can happen under laboratory conditions. Monolayers of transition metal dichalcogenides with 1T' structure therefore have the potential to be ferroelastic and shape memory materials with interesting domain physics.
Next-generation sequencing (NGS) based on genomic DNA has been widely applied for gene rearrangement detection in patients with NSCLC. However, intergenic-breakpoint fusions, in which one or both ...genomic breakpoints localize to intergenic regions, confound kinase fusion detection. We evaluated the function of intergenic-breakpoint fusions with multiplex molecular testing approaches.
NSCLCs with intergenic-breakpoint fusion identified by DNA-based NGS were analyzed by RNA-based NGS, immunohistochemistry (IHC), and fluorescence in situ hybridization.
A total of 26 cases with single intergenic-breakpoint fusion were identified from a large cohort of NSCLCs using DNA-based NGS. Of the 26 cases, RNA-based NGS detected expressed fusion transcripts in 11 cases, but the genomic breakpoint position did not logically predict breakpoint of the fusion transcript in these cases, possibly owing to complex rearrangements (n = 5), alternative splicing (n = 2), and reciprocal rearrangement (n = 4). Nonetheless, no expressed fusion transcript was detected in five cases. Moreover, positive anaplastic lymphoma receptor tyrosine (ALK) finding was observed in three of the remaining 10 cases with IHC but not with RNA-based NGS. Three patients with intergenic-breakpoint ALK fusion with or without RNA-based NGS or IHC confirmation who received crizotinib treatment were found to have partial responses. However, one patient with intergenic-breakpoint ROS1, given the positive fluorescence in situ hybridization result, received crizotinib but developed progressive disease within 1 month, possibly owing to no functional fusion transcript detected by RNA-based NGS.
Intergenic-breakpoint fusions detected by DNA sequencing confound kinase fusion detection in NSCLC, as functional fusion transcripts may be generated or not. Additional validation testing using RNA/protein assay should be performed in intergenic-breakpoint fusion cases to guide optimal treatment.
Landslide susceptibility prediction (LSP) has been widely and effectively implemented by machine learning (ML) models based on remote sensing (RS) images and Geographic Information System (GIS). ...However, comparisons of the applications of ML models for LSP from the perspectives of supervised machine learning (SML) and unsupervised machine learning (USML) have not been explored. Hence, this study aims to compare the LSP performance of these SML and USML models, thus further to explore the advantages and disadvantages of these ML models and to realize a more accurate and reliable LSP result. Two representative SML models (support vector machine (SVM) and CHi-squared Automatic Interaction Detection (CHAID)) and two representative USML models (K-means and Kohonen models) are respectively used to scientifically predict the landslide susceptibility indexes, and then these prediction results are discussed. Ningdu County with 446 recorded landslides obtained through field investigations is introduced as case study. A total of 12 conditioning factors are obtained through procession of Landsat TM 8 images and high-resolution aerial images, topographical and hydrological spatial analysis of Digital Elevation Modeling in GIS software, and government reports. The area value under the curve of receiver operating features (AUC) is applied for evaluating the prediction accuracy of SML models, and the frequency ratio (FR) accuracy is then introduced to compare the remarkable prediction performance differences between SML and USML models. Overall, the receiver operation curve (ROC) results show that the AUC of the SVM is 0.892 and is slightly greater than the AUC of the CHAID model (0.872). The FR accuracy results show that the SVM model has the highest accuracy for LSP (77.80%), followed by the CHAID model (74.50%), the Kohonen model (72.8%) and the K-means model (69.7%), which indicates that the SML models can reach considerably better prediction capability than the USML models. It can be concluded that selecting recorded landslides as prior knowledge to train and test the LSP models is the key reason for the higher prediction accuracy of the SML models, while the lack of a priori knowledge and target guidance is an important reason for the low LSP accuracy of the USML models. Nevertheless, the USML models can also be used to implement LSP due to their advantages of efficient modeling processes, dimensionality reduction and strong scalability.
The aim of this study was to investigate the clinical characteristics of neonates born to SARS-CoV-2 infected mothers and increase the current knowledge on the perinatal consequences of COVID-19. ...Nineteen neonates were admitted to Tongji Hospital from January 31 to February 29, 2020. Their mothers were clinically diagnosed or laboratory-confirmed with COVID-19. We prospectively collected and analyzed data of mothers and infants. There are 19 neonates included in the research. Among them, 10 mothers were confirmed COVID-19 by positive SARS-CoV-2 RT-PCR in throat swab, and 9 mothers were clinically diagnosed with COVID-19. Delivery occurred in an isolation room and neonates were immediately separated from the mothers and isolated for at least 14 days. No fetal distress was found. Gestational age of the neonates was 38.6±1.5 weeks, and average birth weight was 3293±425 g. SARS-CoV-2 RT-PCR in throat swab, urine, and feces of all neonates were negative. SARS-CoV-2 RT-PCR in breast milk and amniotic fluid was negative too. None of the neonates developed clinical, radiologic, hematologic, or biochemical evidence of COVID-19. No vertical transmission of SARS-CoV-2 and no perinatal complications in the third trimester were found in our study. The delivery should occur in isolation and neonates should be separated from the infected mothers and care givers.
Various bentonite-sludge biochar composites were fabricated by a sequence of loading and pyrolysis for the simultaneous removal of norfloxacin (NOR) and copper (Cu2+) from an aqueous solution. The ...morphology and characteristics of obtained composites were reflected through cation exchange capacity (CEC), BET specific surface area (SBET), SEM, XRD, FTIR and XPS. The isothermal adsorption results showed that Sips adsorption model fitted better for the adsorption of NOR and Cu2+ during co-adsorption. The theoretical maximum adsorption capacity of BT:2 SB (the mass ratio of bentonite to sludge is 1:2) for NOR and Cu2+ was 89.36 mg g−1 and 104.10 mg g−1 at 25 °C in the co-adsorption system. The thermodynamic results showed the capture of NOR and Cu2+ was spontaneous, accompanied by an endothermic reaction with different randomness. In the co-adsorption system, the two were antagonistic to each other due to competition for the adsorption sites of hydroxyl, carboxylic acid and negatively charged provided by bentonite-sludge biochar. This study suggests that using natural mineral as a pyrolysis improver for sludge biochar can product the value-enhanced biochar for simultaneous removal of antibiotic and metal contaminants.

The identification of essential proteins can help in understanding the minimum requirements for cell survival and development to discover drug targets and prevent disease. Nowadays, node ranking ...methods are a common way to identify essential proteins, but the poor data quality of the underlying PIN has somewhat hindered the identification accuracy of essential proteins for these methods in the PIN. Therefore, researchers constructed refinement networks by considering certain biological properties of interacting protein pairs to improve the performance of node ranking methods in the PIN. Studies show that proteins in a complex are more likely to be essential than proteins not present in the complex. However, the modularity is usually ignored for the refinement methods of the PINs.
Based on this, we proposed a network refinement method based on module discovery and biological information. The idea is, first, to extract the maximal connected subgraph in the PIN, and to divide it into different modules by using Fast-unfolding algorithm; then, to detect critical modules according to the orthologous information, subcellular localization information and topology information within each module; finally, to construct a more refined network (CM-PIN) by using the identified critical modules.
To evaluate the effectiveness of the proposed method, we used 12 typical node ranking methods (LAC, DC, DMNC, NC, TP, LID, CC, BC, PR, LR, PeC, WDC) to compare the overall performance of the CM-PIN with those on the S-PIN, D-PIN and RD-PIN. The experimental results showed that the CM-PIN was optimal in terms of the identification number of essential proteins, precision-recall curve, Jackknifing method and other criteria, and can help to identify essential proteins more accurately.
To explore the inhibition mechanism of Saccharomyces boulardii (S. boulardii) on ulcerative colitis (UC) carcinogenesis.
C57BL/6 mice were treated with azoxymethane and dextran sulfate sodium ...(AOM/DSS) to develop a UC carcinogenesis model. The treatment group was lavaged with S. boulardii (5 × 10
CFU/d) for 12 weeks. The mice were sacrificed and the tumor load in the treatment group was compared with that of a control group. The levels of TNF-α and IL-6 in colon tissue were measured by enzyme-linked immunosorbent assays. The influence of S. boulardii on TNF-α and IL-6 regulation was also investigated using different colon cell lines. Differences in intestinal microbiota in both stool and intestinal mucosa samples were assessed using 16S rDNA sequencing.
S. boulardii treatment reduced AOM/DSS-induced UC carcinogenesis in mice, as indicated by the reduced tumor load and reduced TNF-α and IL-6 levels in vivo, as well its effects on TNF-α and IL-6 activities in vitro. Significant changes in both fecal and mucosal microbiota were observed among the control, the AOM/DSS treated, and AOM/DSS plus S. boulardii treated groups. For fecal microbiota, the AOM/DSS treated group was lower in Lactobacillus, but higher in Oscillibacter and Lachnoclostridium than the control group. After intervention with S. boulardii, the percentage of Bacillus and Lactococcus increased, but Lachnoclostridium, Oscillibacter, Bacteroides, and Pseudomonas decreased. For the intestinal mucosal microbiota, the AOM/DSS treated group was lower in Bifidobacterium and Ruminococcaceae_UCG-014 and higher in Alloprevotella than the control group. After S. boulardii exposure, the percentage contributions of Lachnoclostridium and Lachnospiraceae_NK4A136 increased.
S. boulardii effectively reduced UC carcinogenesis in an AOM/DSS induced mice model. This positive result can likely be attributed to the reduction of TNF-α and IL-6 levels or the blockade of their function combined with alterations to the intestinal microbiota.
