We demonstrate the feasibility to control the confined electromagnetic field by surface plasmon poloritons (SPPs) excitation and scattering modulation based on the pre-processed Au nanostructures ...under the irradiation of single-shot femtosecond (fs) laser pulse, and then to control the surface tension-induced molten Si transport due to the directed lateral temperature profile, thus resulting in the formation of Si nanostructures. Annular-shaped fs laser pulse is employed based on nonlinear optical effect of frequency-doubling process to imprint Au planar nanostructures with various morphologies. Because surface plasmon polaritons can be easily excited and controlled on planar nanostructures of noble metals, the localized electromagnetic field can be effectively modulated by controlling the metallic nanostructures. Thus, the quasi-plasmonic Au nanostructures act as precursors for controlling the subsequent electromagnetic field by fs laser irradiation. Upon irradiation of pre-processed Au nanostructures, confined electromagnetic field induces a specific surface tension profile, causing directional transport of molten Si. After solidification, Si nanostructures of controllable morphologies can be fabricated. Simulation results are in agreement with the theoretical mechanism. Moreover, further optimized modulation of nanodome can be achieved by controlling the focus spot control combined with the polarization state control. This paper provides an effective method for enabling a scalable formation of Si nanostructures.
To carry out genetic analysis for 21 patients with methylmalonic acidemia (MMA) and provide genetic counseling for their families.

Among these, c.2000A>G of MMUT, c.298G>T of MMACHC and c.734-7A>G of MMAA gene were unreported previously.
Genetic testing for MMA patients can clarify the cause of the disease and provide a basis for the clinical diagnosis. Discovery of novel variants has enriched the mutational spectrum of MMA.
Carbonic anhydrase IX (CAIX) plays an important role in the growth and survival of tumor cells. MORC2 is a member of the MORC protein family. The MORC proteins contain a CW-type zinc finger domain ...and are predicted to have the function of regulating transcription, but no MORC2 target genes have been identified. Here we performed a DNA microarray hybridization and found CAIX mRNA to be down-regulated 8-fold when MORC2 was overexpressed. This result was further confirmed by northern and western blot analysis. Our results also showed that the protected region 4 (PR4) was important for the repression function of MORC2. Moreover, MORC2 decreased the acetylation level of histone H3 at the CAIX promoter. Meanwhile, trichostatin A (TSA) had an increasing effect on CAIX promoter activity. Among the six HDACs tested, histone deacetylase 4 (HDAC4) had a much more prominent effect on CAIX repression. ChIP and ChIP Re-IP assays showed that MORC2 and HDAC4 were assembled on the same region of the CAIX promoter. Importantly, we further confirmed that both proteins are simultaneously present in the PR4-binding complex. These results may contribute to understanding the molecular mechanisms of CAIX regulation.
Extremely rapid heating and cooling rates during the additive manufacturing (AM) process generate complicated thermal cycles, which affect the microstructure evolution and ultimate mechanical ...properties of the alloy. In this paper, FGH96 blocks with a height of 6 mm were prepared by selective laser melting (SLM) and the microstructure was characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Comparing specimens of varying heights, it was found that subsequent thermal cycles (STC) coarsened some solidified grains and accelerated the grain growth along the build direction, together with an increase in texture intensity and high-angle grain boundaries (HAGBs). After coarsening the grains in the middle portion of the built block, finer grains were observed near the top area due to a faster cooling rate. There were numerous dislocations in the grain because of the occurrence of unequal internal tension. In the middle of the sample with stable thermal cycles, the dislocations were both perpendicular to the grain growth direction and 45° off it. In spite of the texture characteristics, the segregation of elements was also found to be influenced by thermal cycling. Inherent reheating leads to the increase in the Laves phase and the decrease in the γ’ phase as subsequent deposition. This was also one of the reasons why the microhardness of the sample decreased as the building height and the other reason being the decrease in the solution treatment of the later sediments.
We, along with others, have shown previously that P21-activated kinase 1 (Pak1) plays a pivotal role in gastric cancer progression and metastasis. However, whether Pak1 controls gastric cancer ...metastasis by regulating microRNAs (miRNAs) has never been explored. Here, we report a novel mechanism of Pak1 in tumor metastasis. A detailed examination revealed that Pak1 interacts with and phosphorylates the serine 62 residue of ATF2 and then blocks its translocation into the nucleus. We also confirmed that ATF2 binds to the promoter of miR-132 and tightly regulates its transcription, thus explaining the regulatory mechanism of miR-132 by Pak1. miR-132 also significantly reduced cell adhesion, migration, and invasion of gastric cancer cells in vitro and significantly prevented tumor metastasis in vivo. miR-132 specifically inhibited hematogenous metastasis, but not lymph node or implantation metastases. In order to further delineate the effects of the Pak1/ATF2/miR-132 cascade on gastric cancer progression, we identified several targets of miR-132 using a bioinformatics TargetScan algorithm. Notably, miR-132 reduced the expression of CD44 and fibronectin1 (FN1), and such inhibition enabled lymphocytes to home in on gastric cancer cells and induce tumor apoptosis. Taken together, our studies establish a novel cell-signaling pathway and open new possibilities for therapeutic intervention of gastric cancer.
Objective:
There is currently no effective treatment for Japanese encephalitis, which has a high rate of morbidity and mortality. This study assessed the effectiveness of a ganciclovir, ...methylprednisolone, and immunoglobulin combination (TAGMIC) therapy in decreasing cognitive impairment and mortality among patients with Japanese encephalitis.
Methods:
We retrospectively assessed the clinical data of 31 patients diagnosed with Japanese encephalitis, who were admitted to an intensive care unit. Patients were divided into the TAGMIC and non-TAGMIC group according to their treatment regime. We compared the 60-day, 6-month, and overall mortality and survival curves between groups. We also compared Barthel Index scores, Montreal Cognitive Assessment (MoCA) scores, and diffusion tensor imaging (DTI) results.
Results:
There was no significant difference in the 30-day mortality rate or Kaplan–Meier survival curve between groups. The 60-day, 6-month, and overall mortality rates in the TAGMIC group were significantly reduced (
P
= 0.043,
P
= 0.018, and
P
= 0.018, respectively) compared with the non-TAGMIC group (0, 0, 0 vs. 31.25, 37.5, 37.5%, respectively). The 60-day, 6-month, and overall Kaplan–Meier survival curves were significantly different between groups (
P
= 0.020,
P
= 0.009,
P
= 0.009, respectively). There was no significant difference in the Barthel Index scores of surviving patients. Among the five patients who underwent MoCA and DTI, four had a score of 0/5 for delayed recall (no cue), while the remaining patient had a score of 2/5. All five patients were able to achieve a score of 5/5 with classification and multiple-choice prompts, and had sparse or broken corpus callosum (or other) fibre bundles.
Conclusion:
TAGMIC treatment can reduce mortality due to severe Japanese encephalitis. The memory loss of surviving patients is mainly due to a disorder of the memory retrieval process, which may be related to the breakage of related fibre bundles.
To analyze the clinical phenotype and genotypes of two children with Carnitine-acylcarnitine translocase deficiency (CACTD).
Two children diagnosed with CACTD at the Gansu Provincial Maternal and ...Child Health Care Hospital respectively on January 3 and November 19, 2018 were selected as the study subjects. Trio-whole exome sequencing (trio-WES) was carried out, and candidate variants were validated through Sanger sequencing and pathogenicity analysis.
Both children were males and had manifested mainly with hypoglycemia. Trio-WES and Sanger sequencing showed that child 1 had harbored compound heterozygous variants of the SLC25A20 gene, namely c.49G>C (p.Gly17Arg) and c.106-2A>G, which were inherited from his father and mother, respectively. Child 2 had harbored homozygous c.199-10T>G variants of the SLC25A20 gene, which were inherited from both of his parents. Among these, the c.106-2A>G and c.49G>C variants were unreported previously. Based on the guidelines from the American College of Medical Genetics and G
Drought remains one of the main factors that negatively affect plant growth and development. Caragana korshinskii is widely distributed on the Loess Plateau, China, where it mediates soil and water ...loss and helps prevent desertification. However, little is known about the stress response mechanisms of C. korshinskii in water-starved environments. MicroRNAs (miRNAs) have been implicated in the regulation of plant responses to several types of biotic and abiotic stress. Here, we describe the miRNAs of wild C. korshinskii from Huangling, Yulin, and Dalad Banner, which occur along a precipitation gradient. Using next-generation sequencing technology, we obtained a total of 13 710 681, 15 048 945, and 15 198 442 reads for each location, respectively; after filtering and BLAST analysis, 490 conserved miRNAs and 96 novel miRNAs were characterized from the sRNAome data, with key functions determined using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses. We also designed stem-loop qRT-PCR to validate the expression patterns of 5 conserved miRNAs (miR390, miR398, miR530, miR2119, and miR5559) that obviously responded to water stress in plants grown both under natural and experimental water stress conditions and found that the expression levels of miR2119 and miR5559 were negatively correlated with their predicted target genes. This study is the first to identify miRNAs from wild C. korshinskii and provides a basis for future studies of miRNA-mediated gene regulation of stress responses in C. korshinskii.
To explore the genetic basis for a Chinese pedigree affected with co-morbid Ornithine carbamoyl transferase deficiency (OTCD) and MECP2 duplication syndrome.
A proband who was admitted to the ...Neonatal Intensive Care Unit of Gansu Provincial Maternal and Child Health Care Hospital on December 19, 2017 was selected as the study subject. High-throughput sequencing and multiplex ligation-dependent probe amplification (MLPA) were carried out for her pedigree, and short tandem repeat-based linkage analysis and chromosome copy number variation sequencing (CNV-seq) were used for the prenatal diagnosis.
The proband, a 3-day-old female, was found to harbor heterozygous deletion of exons 7-9 of the OTC gene. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), the variant was classified as likely pathogenic (PVS1+PM2_Supporting+PP4). The proband was diagnosed with OTCD , which was in keeping with her acute encephalopathy and metabolic abnormalities (manifesting as hyperammonemia, de
The structural color based on the Fabry–Perot (F–P) resonator has been extensively applied lithography-free and tunable color displays. Conventional F–P cavity-based structural color technology ...exhibits a wide half maximum full width (fwhm), thus causing low color saturation. In this study, a Sc0.2Sb2Te3(SST) based structure of multi-order F–P cavity resonance was proposed to obtain high-saturation colors. The surface absorber of the multi-order F–P resonator structure was coated with an SST film, such that the reflection effect at nonresonant wavelengths was reduced. Moreover, ITO layer stacking served as F–P cavity resonance for multi-level modulation, and only a resonant wavelength was allowed to reflect. On that basis, the fwhm of nearly 25 nm and a peak reflectance of 90 was achieved. With the above structure, the color saturation can be dynamically regulated by the phase state of the SST. It is noteworthy that 60% sRGB color gamut space and 50% aRGB color gamut space can be currently achieved. The proposed modulation subsurface is expected to expand the color range of high-level and micro-nano display technology.
