Hybrids of the species Pseudoplatystoma corruscansvs.Pseudoplatystoma reticulatum are extensively traded because they are more docile and precocious compared to their parents. This study aimed to ...identify the occurrence of hybrid 'Surubim' in rivers of the State of Mato Grosso do Sul, applying molecular techniques. Pseudoplatystoma corruscans, Pseudoplatystoma reticulatum and hybrid 'Surubim' were detected using multiplex PCR and PCR-RFLP in RAG2, GLOBIN, EF1α, 18S rRNA nuclear genes, and mitochondrial 16S rRNA gene. The results revealed the presence of F1 hybrids in rivers and the occurrence of genetic introgression, evidenced by the Post-F1 hybrids. Furthermore, discrepancies between the morphological and genetic identification were observed, demonstrating the effectiveness of using different molecular markers for the correct identification of these hybrids. The necessity of implementation of management and conservation projects to maintain the genetic integrity of native species populations threatened by hybridization in the rivers of Mato Grosso do Sul State is evident.
Híbridos envolvendo as espécies Pseudoplatystoma corruscans vs.Pseudoplatystoma reticulatum são amplamente comercializados por apresentarem maior docilidade e precocidade quando comparados aos parentais. Neste trabalho objetivou-se identificar, por meio de técnicas moleculares, a existência de surubins híbridos em rios do estado do Mato Grosso do Sul. Pseudoplatystoma corruscans, Pseudoplatystoma reticulatum e 'Surubim' híbrido foram identificados utilizando as técnicas de PCR multiplex e PCR-RFLP dos genes nucleares RAG2, GLOBINA, EF1α, 18S rRNA e mitocondrial 16S rRNA. Os resultados demonstraram a presença de híbridos F1 em rios e a ocorrência da introgressão genética, evidenciada pelos híbridos Pós-F1. Além disso, foram observadas discrepâncias entre a identificação morfológica e genética, demonstrando a eficácia da utilização de diversos marcadores moleculares para identificação correta destes híbridos. Torna-se evidente a necessidade da implantação de projetos de manejo e conservação, a fim de manter a integridade genética de populações de espécies nativas em rios do estado do Mato Grosso do Sul, que encontram-se ameaçadas pela hibridação.
This study reports the first cytogenetic characterization of the Amazonian catfish Leiarius marmoratus (“jandiá”) and its F1 (first generation) hybrid “cachandiá” with Pseudoplatystoma reticulatum ...(“cachara”). A diploid number of 56 chromosomes and a single argyrophilic nucleolus organizer region (Ag-NOR) in the short arm of two sub-telocentric chromosomes were observed for both L. marmoratus and P. reticulatum, but with differences in the karyotype formula and the size of the chromosome pair with NORs. The hybrid showed 2n = 56 chromosomes with an intermediate karyotype when compared to the parental species. A single Ag-NOR was maintained in the hybrid but located in two chromosomes with marked differences in size and presenting intraindividual variation in NOR activity (nucleolar dominance). For L. marmoratus and the hybrid, heterochromatic bands were predominately distributed in the terminal, centromeric, and sub-centromeric regions of some chromosomes and 5S rDNA sites located in two distinct sub-telocentric chromosomes, similar to the previously described for P. reticulatum. The data suggested that the hybrid karyotype might be insufficient for a precise discrimination of hybrids, however, Ag-NOR can be used as a chromosome marker to differentiate “cachandiá” from L. marmoratus and P. reticulatum. The current study also provides insights into the chromosomal features of L. marmoratus and contributes with novel cytogenetic information of this native Amazonian catfish included in the Pimelodidae family.
Leporinus friderici is a migratory neotropical fish with elevated ecological and economic importance in Brazil. Microsatellite markers are highly important in population genetic studies, management, ...and conservation programs; however, no markers are available for this species. In this study, seven microsatellite loci, previously developed for Megaleporinus obtusidens, were successfully cross-amplified in L. friderici. Among these loci, five presented moderate to high genetic variability levels, with four to seven alleles per loci and expected heterozygosities varying from ≥ 0.574 to 1.000. These markers represent a valuable tool for the future management and ecological studies involving this species and group of neotropical fishes.
RESUMO: Leporinus friderici é um peixe neotropical migratório com elevada importância ecológica e econômica no Brasil. Os marcadores microssatélites são conhecidos por sua importância em estudos genéticos populacionais, programas de manejo e conservação, no entanto, não existem marcadores disponíveis para esta espécie. Neste estudo, sete locos microssatélites, previamente desenvolvidos para Megaleporinus obtusidens foram amplificados com sucesso em L. friderici. Dentre esses loci, cinco apresentaram variabilidade genética moderada a alta, com quatro a sete alelos por loci e heterozigosidades esperadas variando de ≥ 0,574 a 1.000. Esses marcadores representam uma ferramenta valiosa para futuros manejos e estudos ecológicos envolvendo esta espécie e este grupo de peixes neotropicais.
Unraveling adaptive genetic variation represents, in addition to the estimate of population demographic parameters, a cornerstone for the management of aquatic natural living resources, which, in ...turn, represent the raw material for breeding programs. The turbot (Scophthalmus maximus) is a marine flatfish of high commercial value living on the European continental shelf. While wild populations are declining, aquaculture is flourishing in southern Europe. We evaluated the genetic structure of turbot throughout its natural distribution range (672 individuals; 20 populations) by analyzing allele frequency data from 755 single nucleotide polymorphism discovered and genotyped by double‐digest RAD sequencing. The species was structured into four main regions: Baltic Sea, Atlantic Ocean, Adriatic Sea, and Black Sea, with subtle differentiation apparent at the distribution margins of the Atlantic region. Genetic diversity and effective population size estimates were highest in the Atlantic populations, the area of greatest occurrence, while turbot from other regions showed lower levels, reflecting geographical isolation and reduced abundance. Divergent selection was detected within and between the Atlantic Ocean and Baltic Sea regions, and also when comparing these two regions with the Black Sea. Evidence of parallel evolution was detected between the two low salinity regions, the Baltic and Black seas. Correlation between genetic and environmental variation indicated that temperature and salinity were probably the main environmental drivers of selection. Mining around the four genomic regions consistently inferred to be under selection identified candidate genes related to osmoregulation, growth, and resistance to diseases. The new insights are useful for the management of turbot fisheries and aquaculture by providing the baseline for evaluating the consequences of turbot releases from restocking and farming.
ABSTRACT This study aimed to impute the genetic makeup of individual fishes of Serrasalmidae family on the basis of body weight and morphometric measurements. Eighty-three juveniles, belonging to the ...genetic groups Pacu, Pirapitinga, Tambaqui, Tambacu, Tambatinga, Patinga, Paqui and Piraqui, were separated into 16 water tanks in a recirculation system, with two tanks per genetic group, where they remained until they reached 495 days of age. They were then weighed and analyzed according to the following morphometric parameters: Standard Length (SL), Head Length (HL), Body Height (BH), and Body Width (BW). The identity of each fish was confirmed with two SNPs and two mitochondrial markers. Two analyses were performed: one for the validating the imputation and another for imputing a genetic composition of animals considered to be advanced hybrids (post F1). In both analyses, we used linear mixed models with a mixture of normal distributions to impute the genetic makeup of the fish based on phenotype. We applied the mixed models method, whereby the environmental effects were estimated by the Empirical Best Linear Unbiased Estimator (EBLUE) and genetic effects are considered random, obtaining the Empirical Best Linear Unbiased Predictor (EBLUP) from the general (GCA) and the specific (SCA) combining ability effects. The results showed that validation of the genetic makeup imputation based on body weight can be used because of the strong correlation between the observed and imputed genotype. The fish classified as advanced hybrids had a genetic composition with a high probability of belonging to known genotypes and there was consistency in genotype imputation according to the different characteristics used.
This work was carried out to predict the combining abilities, both general and specific, for performance traits and bodily yields of Serrasalmidae. Ninety‐six 30‐day‐old juveniles were purchased from ...two commercial fish farms, 12 of each of the following eight genetic groups: pacu, pirapitinga, tambaqui, tambacu, tambatinga, patinga, paqui and piraqui. Six fish from each genetic group were grown in 500‐L fibreglass tanks (two tanks per genetic group) until they were 495 days old. At the end of the growth period fish were weighed, subjected to morphometric analysis and processed to obtain their bodily yields. Two nuclear markers and one mitochondrial marker were used to confirm the identity of the animals. Combining abilities were obtained using the method proposed by Griffing in 1956 (Australian Journal of Biological Science, 4, 463–493) adapted to a mixed models analysis, environmental effects were estimated by the empirical best linear unbiased estimator method and genetic effects (general and specific combining abilities) were estimated with the empirical best linear unbiased predictor. Predictions of the combining abilities of advanced hybrids were obtained by the mixed models mixture method with normal distributions. Tambaqui showed higher general and specific combining abilities than the other groups for most of the variables, making it the most important genetic group. General combining ability makes a greater contribution to phenotypic variance than specific combining ability for most variables, indicating a predominance of genes with an additive effect in the control of evaluated traits.
The identification of fish species using traditional methods is generally based only on morphological characteristics and these methods are currently under review. This kind of identification of ...hybrid fishes solely based on their morphologies may be unreliable, especially when the samples include juveniles and post-F1 lineage fishes. Therefore, in the present study, we used molecular markers to accurately identify the fish species of economic interest that are used as materials in the projects developed in research institutions. We evaluated six lots of fishes sampled from different research centers, containing a total of 84 specimens acquired from private fish farms that were considered to be the representatives of pure species. Genetic analyses of all the specimens revealed that, globally, 22 samples (26.2%) were interspecific hybrids, while 20 (90.9%) samples were surprisingly characterized as post-F1 hybrids. This result confirms that hybrids are sold in markets without adequate labeling and also indicates the lack of proper control of the commercialization and management of stocks and products in fish farms. In addition, we determined that molecular diagnosis was an extremely effective and necessary method to test the reliability of biological materials currently used in scientific research.
The cytogenetic characteristics of Pseudoplatystoma corruscans and Pseudoplatystoma reticulatum and their F1, F2 and backcross hybrids were assessed by using chromosome banding techniques. The ...diploid number of 56 chromosomes was constant in all species and lineages, with a karyotypic formula containing 20 metacentric, 12 submetacentric, 12 subtelocentric and 12 acrocentric chromosomes. Nucleolar organizer regions (NORs) were identified in two subtelocentric chromosomes in the parents and hybrids, with partial nucleolar dominance in F1 and F2 specimens. Heterochromatic blocks were detected in the terminal and centromeric regions of some chromosomes in all individuals. For parental and hybrid lineages, 18S ribosomal clusters corresponding to NORs and 5S ribosomal genes were identified in distinct pairs of chromosomes. The striking conservation in the chromosomal macrostructure of the parental species may account for the fertility of their F1 hybrids. Similarly, the lack of marked alterations in the chromosomal structure of the F1 hybrids could account for the maintenance of these features in post-F1 lineages.
Herein, we have developed molecular markers for nuclear genes to use in multiplex‐PCR and PCR‐RFLP, with the goal of characterising hybrid lines derived from crosses between pintado Pseudoplatystoma ...corruscans and cachara P. reticulatum. These markers, together with others described previously, were used to perform molecular identification analyses as genetic subsidies for Brazilian aquaculture. These analyses were performed due to the problems of high mortality in the offspring reported by the aquaculturist. From a total of 16 broodstock samples, 13 were genetically identified as hybrids; surprisingly, nine of these hybrids were found to be post‐F1 lineages. These data show that the fertility of these animals can seriously affect the cultivated stocks, thus causing financial damage in this aquaculture system. The establishment of PCR‐RFLP and multiplex‐PCR as molecular techniques allows for both the correct management of these animals and the routine monitoring of production and trade of fish hybrids in aquaculture. Consequently, such tools will enable a sustainable development in the aquaculture industry.

Molecular markers (Multiplex-PCR and PCR-RFLP of nuclear and mitochondrial genes) were used to assess the frequency of hybrids in wild stocks of two catfish species: Pseudoplatystoma corruscans and Pseudoplatystoma reticulatum. Analysis of fish from the Paraguay–Parana hydrographic basin (Brazil) revealed a low frequency of hybrids in the Parana River (3.60%), which might be the result of natural hybridisation. However, the frequency of hybrids was higher in the Mogi Guaçu (50.00%) and Aquidauana (30.75%) Rivers, whose aquaculture systems support the majority of Brazil's fish farms, suggesting that the existence of hybrids in wild populations may be due to introduction or escapes from farm stocks. Furthermore, the identification of a post-F1 individual in the Mogi-Guaçu River provides evidence of genetic introgression. These results show that safeguards are needed to ensure the correct handling of hybrids and the protection of native species.
