Background and Aims
NAFLD, characterized by aberrant triglyceride accumulation in liver, affects the metabolic remodeling of hepatic and nonhepatic tissues by secreting altered hepatokines. Small ...ubiquitin‐related modifier (SUMO)–specific protease 2 (SENP2) is responsible for de‐SUMOylation of target protein, with broad effects on cell growth, signal transduction, and developmental processes. However, the role of SENP2 in hepatic metabolism remains unclear.
Approach and Results
We found that SENP2 was the most dramatically increased SENP in the fatty liver and that its level was modulated by fed/fasted conditions. To define the role of hepatic SENP2 in metabolic regulation, we generated liver‐specific SENP2 knockout (Senp2‐LKO) mice. Senp2‐LKO mice exhibited resistance to high‐fat diet–induced hepatic steatosis and obesity. RNA‐sequencing analysis showed that Senp2 deficiency up‐regulated genes involved in fatty acid oxidation and down‐regulated genes in lipogenesis in the liver. Additionally, ablation of hepatic SENP2 activated thermogenesis of adipose tissues. Improved energy homeostasis of both the liver and adipose tissues by SENP2 disruption prompted us to detect the hepatokines, with FGF21 identified as a key factor markedly elevated in Senp2‐LKO mice that maintained metabolic homeostasis. Loss of FGF21 obviously reversed the positive effects of SENP2 deficiency on metabolism. Mechanistically, by screening transcriptional factors of FGF21, peroxisome proliferator–activated receptor alpha (PPARα) was defined as the mediator for SENP2 and FGF21. SENP2 interacted with PPARα and deSUMOylated it, thereby promoting ubiquitylation and subsequent degradation of PPARα, which in turn inhibited FGF21 expression and fatty acid oxidation. Consistently, SENP2 overexpression in liver facilitated development of metabolic disorders.
Conclusions
Our finding demonstrated a key role of hepatic SENP2 in governing metabolic balance by regulating liver–adipose tissue crosstalk, linking the SUMOylation process to metabolic regulation.
Solitons are self-sustained wavepackets that occur in many physical systems. Their recent demonstration in optical microresonators has provided a new platform for the study of nonlinear optical ...physics with practical implications for miniaturization of time standards, spectroscopy tools, and frequency metrology systems. However, despite its importance to the understanding of soliton physics, as well as development of new applications, imaging the rich dynamical behavior of solitons in microcavities has not been possible. These phenomena require a difficult combination of high-temporal-resolution and long-record-length in order to capture the evolving trajectories of closely spaced microcavity solitons. Here, an imaging method is demonstrated that visualizes soliton motion with sub-picosecond resolution over arbitrary time spans. A wide range of complex soliton transient behavior are characterized in the temporal or spectral domain, including soliton formation, collisions, spectral breathing, and soliton decay. This method can serve as a visualization tool for developing new soliton applications and understanding complex soliton physics in microcavities.

Measurement of optical and vibrational spectra with high resolution provides a way to identify chemical species in cluttered environments and is of general importance in many fields. Dual-comb ...spectroscopy has emerged as a powerful approach for acquiring nearly instantaneous Raman and optical spectra with unprecedented resolution. Spectra are generated directly in the electrical domain, without the need for bulky mechanical spectrometers. We demonstrate a miniature soliton-based dual-comb system that can potentially transfer the approach to a chip platform. These devices achieve high-coherence pulsed mode locking. They also feature broad, reproducible spectral envelopes, an essential feature for dual-comb spectroscopy. Our work shows the potential for integrated spectroscopy with high signal-to-noise ratios and fast acquisition rates.
Abstract
Background
The emergence of coronavirus disease 2019 (COVID-19) is a major healthcare threat. The current method of detection involves a quantitative polymerase chain reaction (qPCR)–based ...technique, which identifies the viral nucleic acids when present in sufficient quantity. False-negative results can be achieved and failure to quarantine the infected patient would be a major setback in containing the viral transmission. We aim to describe the time kinetics of various antibodies produced against the 2019 novel coronavirus (SARS-CoV-2) and evaluate the potential of antibody testing to diagnose COVID-19.
Methods
The host humoral response against SARS-CoV-2, including IgA, IgM, and IgG response, was examined by using an ELISA-based assay on the recombinant viral nucleocapsid protein. 208 plasma samples were collected from 82 confirmed and 58 probable cases (qPCR negative but with typical manifestation). The diagnostic value of IgM was evaluated in this cohort.
Results
The median duration of IgM and IgA antibody detection was 5 (IQR, 3–6) days, while IgG was detected 14 (IQR, 10–18) days after symptom onset, with a positive rate of 85.4%, 92.7%, and 77.9%, respectively. In confirmed and probable cases, the positive rates of IgM antibodies were 75.6% and 93.1%, respectively. The detection efficiency by IgM ELISA is higher than that of qPCR after 5.5 days of symptom onset. The positive detection rate is significantly increased (98.6%) when combining IgM ELISA assay with PCR for each patient compared with a single qPCR test (51.9%).
Conclusions
The humoral response to SARS-CoV-2 can aid in the diagnosis of COVID-19, including subclinical cases.
The time kinetics of humoral responses against the novel coronavirus (SARS-CoV-2) are characterized in patients with COVID-19 by nucleocapsid-based enzyme-linked immunosorbent assay. The antibody testing can aid in the diagnosis of COVID-19 when combined with quantitative polymerase chain reaction, including in subclinical cases.
Organic electrochemistry has a rich history in organic synthesis and has been considered as a promising alternative to traditional chemical oxidants and reductants because it obviates the use of ...stoichiometric amount of dangerous and toxic reagents. In particular, the electrochemical C—H bonds functionalization is one of the most desirable approaches for the construction of carbon–carbon (C—C) and carbon–heteroatom (C—X) bonds. This review summarizes the substantial progress made in the last few years in C—H functionalization via organic electrochemistry. It is divided into sections on C—C, C—N, C—O, C—S, C–Halogen and C—P bond formation.

The absence of tumor antigens leads to a low response rate, which represents a major challenge in immune checkpoint blockade (ICB) therapy. Pyroptosis, which releases tumor antigens and ...damage‐associated molecular patterns (DAMPs) that induce antitumor immunity and boost ICB efficiency, potentially leads to injury when occurring in normal tissues. Therefore, a strategy and highly efficient agent to induce tumor‐specific pyroptosis but reduce pyroptosis in normal tissues is urgently required. Here, a smart tumor microenvironmental reactive oxygen species (ROS)/glutathione (GSH) dual‐responsive nano‐prodrug (denoted as MCPP) with high paclitaxel (PTX) and photosensitizer purpurin 18 (P18) loading is rationally designed. The ROS/GSH dual‐responsive system facilitates the nano‐prodrug response to high ROS/GSH in the tumor microenvironment and achieves optimal drug release in tumors. ROS generated by P18 after laser irradiation achieves controlled release and induces tumor cell pyroptosis with PTX by chemo‐photodynamic therapy. Pyroptotic tumor cells release DAMPs, thus initiating adaptive immunity, boosting ICB efficiency, achieving tumor regression, generating immunological memory, and preventing tumor recurrence. Mechanistically, chemo‐photodynamic therapy and control‐release PTX synergistically induce gasdermin E (GSDME)‐related pyroptosis. It is speculated that inspired chemo‐photodynamic therapy using the presented nano‐prodrug strategy can be a smart strategy to trigger pyroptosis and augment ICB efficiency.

Drought refers to a meteorological disaster that causes insufficient soil moisture and damage to crop water balance due to long-term lack of precipitation. With the increasing shortage of water ...resources, drought has become one of the hot issues of global concern. The standardized precipitation index (SPI) and standardized precipitation evapotranspiration index (SPEI) can effectively reflect the changes in drought characteristics of different geomorphologies in Sichuan on time and space scales, to explore the difference in drought characteristics between different physiognomy types in Sichuan Province, We calculated the SPI and SPEI values based on the data of 44 meteorological stations in Sichuan Province from 1961 to 2019 and used Mann-Kendall trend test and multivariable linear regression method (MLR) to quantify the significance of the drought characteristic trends at different time and space scales. The results as follow: (1) The SPEI drought trend in plain and hilly regions was greater than that in plateau and mountain regions on all time scales (- 0.039 year
for 1-month in hilly, - 0.035 year
for 1-month in plain, - 0.14 year
for 1-month in plateau, - 0.026 year
for 1-month in mountain) and the magnitude of trend of eastern (- 4.4 to 0.1 year
) was lager than western (- 2.1 to 2.7 year
), means that the drought trends transfer from northwest to east. (2) The drought intensity in the western region gradually increased (0.54-1.05) and drought events mainly occurred in the southwest plateau and central mountainous regions (24-47 times), means that drought meteorological hotspots were mainly concentrated in the Sichuan basin. (3) The MLR indicated altitude (H) is not the main influencing factor that causes the spatial unevenness of precipitation in Sichuan Province, but altitude (H), temperature (T), longitude (L
) and latitude (L
) can co-determined the precipitation. The results of this study are instructive and practical for drought assessment, risk management and application decision-making in Sichuan Province, and have guiding significance for agricultural disaster prevention, mitigation and agricultural irrigation in Sichuan Province.
Black phosphorus (BP) has attracted great attentions in the last few years, but their applications in optoelectronics have been strongly hindered by the poor environmental stability, especially under ...light irradiation. Herein, the fabrication of surface passivated few‐layer BP assisted by polymer ionic liquids (PILs) is reported. The PIL‐modified 2D BP exhibits excellent stability under ambient conditions, with negligible deterioration in 100 d. Furthermore, the PILs attached on the BP surface provide reliable and flexible electrical contact between the few‐layer BP and other device components. As a proof of concept, PIL‐modified BP nanosheets are successfully applied in flexible photodetectors, which show high flexibility, good detectivity with no obvious performance deterioration in 120 h. This work demonstrates that the PIL modification can endow BP nanosheets with excellent environmental stability and good conductivity, so that may significantly expand the applications of BP in flexible optoelectronics.
An efficient strategy to fabricate polymer ionic liquid (PIL)‐passivated 2D BP is developed. The PIL‐modified 2D BP shows negligible structural deterioration in 100 d under ambient condition. Photodetectors based on PIL‐modified 2D BP exhibit good performance and excellent stability.
