Pearl powder is a famous traditional Chinese medicine that has a long history in treating palpitations, insomnia, convulsions, epilepsy, ulcers, and skin lightining. Recently, several studies have ...demonstrated the effects of pearl extracts on protection of ultraviolet A (UVA) induced irritation on human skin fibroblasts and inhibition of melanin genesis on B16F10
melanoma cells. To further explore the effect we focused on the whitening efficacy of pearl hydrolyzed conchiolin protein (HCP) on
melanoma MNT-1 cells under the irritation of alpha-melanocyte-stimulating hormone (
-MSH) or endothelin 1 (ET-1) to evaluate the intracellular tyrosinase and melanin contents, as well as the expression levels of tyrosinase (
), tyrosinase related protein 1 (
), and dopachrome tautomerase (
) genes and related proteins. We found that HCP could decrease the intracellular melanin content by reducing the activity of intracellular tyrosinase and inhibiting the expression of TYR, TRP-1, DCT genes and proteins. At the same time, the effect of HCP on melanosome transfer effect was also investigated in the co-culture system of immortalized
keratinocyte HaCaT cells with MNT-1. The result indicated that HCP could promote the transfer of melanosomes in MNT-1 melanocytes to HaCaT cells, which might accelerate the skin whitening process by quickly transferring and metabolizing melanosomes during keratinocyte differentiation. Further study is needed to explore the mechanism of melanosome transfer with depigmentation.
The identification of germplasm resources is an important aspect of sugarcane breeding. The aim of this study was to introduce a new method for identifying Saccharum spontaneum and its progeny. ...First, we cloned and sequenced nuclear ribosomal DNA internal transcribed spacer (nrDNA-ITS) sequences from 20 Saccharum germplasms. Analysis of these nrDNA-ITS sequences showed a stable mutation at base 89. Primers (FO13, RO13, FI16, and RI16) were then designed for tetra-primer amplification refractory mutation system (ARMS) PCR based on mutations at base 89 of the nrDNA-ITS sequence. An additional 71 Saccharum germplasms were identified using this tetra-primer ARMS PCR method, which confirmed that the method using the described primers successfully identified Saccharum spontaneum and progeny. These results may help improve the efficiency of modern molecular breeding of sugarcane and lay a foundation for identification of sugarcane germplasms and the relationships among them.
Rockbursts are hazardous phenomena of sudden and violent rock failure in deep underground excavations under high geostress conditions, which poses a serious threat to geotechnical engineering. The ...occurrence of rockbursts is influenced by a combination of factors. Therefore, it is necessary to find an efficient method to assess rockburst grades. In this paper, we propose a novel method that enhances the VIKOR method using a novel combination of weight and generalized weighted Mahalanobis distance. The combination weights of the evaluation indicators were calculated using game theory by combining subjective experience and objective data statistical characteristics. By introducing the generalized weighted Mahalanobis distance, the VIKOR method is improved to address the issues of inconsistent dimensions, different importance, and inconsistent correlation among indicators. The proposed method can deal with the complexity of the impact factors of rockburst evaluation and classify the rockburst intensity level. The results show that the accuracy of the improved VIKOR method with the distance formula is higher than that of the unimproved VIKOR method; the evaluation accuracy of the improved VIKOR method with the generalized weighted Mahalanobis distance is 91.67%, which outperforms the improved VIKOR methods with the Euclidean and Canberra distances. This assessment method can be easily implemented and does not depend on the discussion of the rockburst occurrence mechanism, making it widely applicable for engineering rockburst evaluation.
Anaplastic thyroid carcinoma (ATC) is a refractory and poor prognosis tumor Present study aimed to investigate the underlying biological functions and pathways involved in the development of ATC and ...to identify potential hub genes and candidate biomarkers of ATC.
Bioinformatics analyses were performed to identify the differentially expressed genes (DEGs) between ATC tissue samples and adjacent normal tissue samples. Protein-protein interaction (PPI) networks of the DEGs were constructed using Search Tool for the Retrieval of Interacting Genes online tool and Cytoscape software and divided into sub-networks using the Molecular Complex Detection (MCODE) plug-in. DEGs in each module was analyzed by enrichment analysis of the KEGG Orthology Based Annotation System (KOBAS) web software version 3.0. Eventually, the hub genes from bioinformatics analysis were verified by qRT-PCR assay in different ATC cell lines.
Thirty hub genes were selected and three modules were built by the Cytoscape software from the PPI network. Seven genes (CDK1, CCNB2, BUB1B, CDC20, RRM2, CHEK1 and CDC45) were screened from thirty hub genes. Enrichment analysis showed that these hub genes were primarily accumulated in ‘cell cycle’, ‘p53 signaling pathway’, ‘viral carcinogenesis’, ‘pyrimidine metabolism’ and ‘ubiquitin mediated proteolysis’. The results of qRT-PCR indicated that seven hub genes were unregulated in three ATC cell lines compared with normal thyroid gland cell.
These findings suggest that CDK1, CCNB2, BUB1B, CDC20, RRM2, CHEK1 and CDC45 may serve as novel diagnosis biomarkers and potential therapeutic target for ATC.
► A new method was established for the simultaneous analysis of three groups of analytes. ► More qualitative information can be attained by the proposed HPLC-DAD–ESI/MS method. ► The method was first ...used to determine the amount of 10 bioactive components in citrus. ► The results provide the proof for evaluating the quality of citrus newly cultivated. ► Products containing the above bioactive components can be evaluated by the method.
A simple and accurate method has been developed to simultaneously separate and determine 10 bioactive compounds in citrus fruits by high-performance liquid chromatography coupled with diode array detection and electrospray ionisation mass spectrometry (HPLC-DAD–ESI/MS). This HPLC assay was performed on a reversed-phase C18 column with acetonitrile and 0.1% (v/v) aqueous formic acid as mobile phase. DAD has been performed at 273, 283 and 324nm for quantification of the alkaloids, flavanones and hydroxycinnamic acids. MS was also employed to identify the each analyte. Ten analytes (naringin, hesperidin, neohesperidin, ferulic acid, p-coumaric acid, chlorogenic acid, caffeic acid, octopamine, synephrine and tyramine) demonstrated good linearity (r⩾0.9990) in a relatively wide concentration range. The method revealed high average recovery (range, 92.1–97.9%) and good precision with interday and intraday variations with less than 4.71%. The limits of detection (LODs) ranged from 0.02 to 0.11μg/ml, while the limits of quantification (LOQs) were defined in the range of 0.08–0.39μg/ml. The proposed method has been successfully applied to analyse three types of bioactive constituents in eight citrus hybrids pulps and eight citrus hybrids peels, which has been successfully cultivated in China.
The shallow-water hydrothermal vent system of Kueishan Island has been described as one of the world's most acidic and sulfide-rich marine habitats. The only recorded metazoan species living in the ...direct vicinity of the vents is
a brachyuran crab endemic to marine sulfide-rich vent systems. Despite the toxicity of hydrogen sulfide,
occupies an ecological niche in a sulfide-rich habitat, with the underlying detoxification mechanism remaining unknown. Using laboratory and field-based experiments, we characterized the gills of
that are the major site of sulfide detoxification. Here sulfide is oxidized to thiosulfate or bound to hypotaurine to generate the less toxic thiotaurine. Biochemical and molecular analyses demonstrated that the accumulation of thiosulfate and hypotaurine is mediated by the sodium-independent sulfate anion transporter (SLC26A11) and taurine transporter (Taut), which are expressed in gill epithelia. Histological and metagenomic analyses of gill tissues demonstrated a distinct bacterial signature dominated by
. Our results suggest that thiotaurine synthesized in gills is used by sulfide-oxidizing endo-symbiotic bacteria, creating an effective sulfide-buffering system. This work identified physiological mechanisms involving host-microbe interactions that support life of a metazoan in one of the most extreme environments on our planet.
In this paper, we study a promising plate-type heat exchanger, the printed circuit heat exchanger (PCHE), which has high compactness and is suitable for high-pressure conditions as a vaporizer during ...vaporization. The thermal hydraulic performance of supercritical produce liquefied natural gas (LNG) in the zigzag channel of PCHE is numerically investigated using the SST κ-ω turbulence model. The thermo-physical properties of supercritical LNG from 6.5 MPa to 10MPa were calculated using piecewise-polynomial approximations of the temperature. The effect of the channel bend angle, mass flux and inlet pressure on local convection heat transfer coefficient, and pressure drop are discussed. The heat transfer and pressure loss performance are evaluated using the Nusselt and Euler numbers. Nu/Eu is proposed to evaluate the comprehensive heat transfer performance of PCHE by considering the heat transfer and pressure drop characteristics to find better bend angle and operating conditions. The supercritical LNG has a better heat transfer performance when bend angle is less than 15° with the mass flux ranging from 207.2 kg/(m2·s) to 621.6 kg/(m2·s), which improves at bend angle of 10° and lower compared to 15° at mass flux above 414.4 kg/(m2·s). The heat transfer performance is better at larger mass flux and lower operating pressures.
Wasteless mining has become the mainstream mode of China’s mining industry, which is an environmentally friendly and sustainable mining method. To effectively evaluate the construction of a green ...mine, the wasteless mining evaluation method was proposed, based on the intuitionistic fuzzy set theory and the Vise Kriterijumska Optimizacija I Kompromisno Resenje (VIKOR) decision-making method in this study. Firstly, the intuitionistic fuzzy sets for six indicators (such as waste rock utilization, tailing sand utilization and the current mining situation) of the Dongguashan Copper Mine were established, based on the intuitionistic fuzzy set theory and the national standard system. Secondly, referring to the green mine evaluation system, the index weights were calculated with the Analytic Hierarchy Process (AHP) method. Finally, the above intuitionistic fuzzy set matrix was ranked by the VIKOR method. The calculation results show that the group utility value of this mine is 0.1943 ∈ (0,0.2), which is at level I; the individual regret value is 0.0916 ∈ (0.075,0.15), which is at level II; the compromise value is 0.2193 ∈ (0.2,0.4), which is at level II. Through comprehensive assessment, the final evaluation of the wasteless mining in the Dongguashan Copper Mine is Grade II. The Dongguashan Copper Mine is one of the first mining enterprises subject to the transformation into wasteless mining in China. The results of the final evaluation are consistent with the current status of wasteless mining in the Dongguashan Copper Mine.
This study proposed a multirange attribute object classification method based on a vectorization attribute circle to solve the classification problem of multiple slope stability. There are many ...factors affecting slope stability, and these factors are difficult to assign a value representation; however, they can usually be easily expressed as a range of attribute fields to adapt to this situation and propose a method based on an attribute circle. First, this paper discusses the basic properties of an attribute circle, and subsequently discusses the method, derivation process, and properties of an attribute circle. Finally, the section of the Hai Bamboo Highway on the 318 national road line in Ganzi Prefecture, Sichuan province, China, is selected as an engineering case and this study uses this method to classify 16 slopes according to four attributes. By continuously raising the threshold until all objects are classified, the classification results are the same as the actual stability in the slope sample instance. This method provides a new idea for the study of slope stability state classification.
The accessory nidamental gland (ANG) is part of the reproduction organ in the majority of female cephalopods, including the bigfin reef squid Sepioteuthis lessoniana, an economically important ...fishery product. Microbes in Alphaproteobacteria, Gammaproteobacteria, and Verrucomicrobia have been suggested to play a role in the maturation of the S. lessoniana ANG and are responsible for its color. However, the bacterial composition and dynamics of the different maturation stages of the ANG remain unclear. In the present study, we surveyed ANG-associated bacterial dynamics in wild-caught S. lessoniana at various developmental stages in different populations over 3 years. The results obtained showed that the ANG bacterial community shifted gradually and decreased in diversity throughout maturation. Verrucomicrobia occupied the ANG during the early stages in large numbers, and was replaced by Bacteroidia, Alphaproteobacteria, and Gammaproteobacteria in the later stages. Flavobacteriales and Alphaproteobacteria both appeared to contribute to pigmentation, while Bacteroidia, Alphaproteobacteria, and Gammaproteobacteria may be involved in enriching the heme biosynthesis pathway in the ANG with the maturation of S. lessoniana. The present results provide an open question of whether S. lessoniana actively selects the bacterial community in the ANG to adjust to its surrounding environment.
