We investigated the responses of
to untreated, ionic liquid and hydrothermally pretreated wheat straw over a 5-day time course using RNA-seq and targeted proteomics. The ionic liquid pretreatment altered the cellulose crystallinity while retaining more of the hemicellulosic sugars than the hydrothermal pretreatment. Ionic liquid pretreatment of straw led to a dynamic induction and repression of genes, which was correlated with the higher levels of pentose sugars saccharified from the ionic liquid-pretreated straw. Hydrothermal pretreatment of straw led to reduced levels of transcripts of genes encoding carbohydrate-active enzymes as well as the derived proteins and enzyme activities. Both pretreatments abolished the expression of a large set of genes encoding pectinolytic enzymes. These reduced levels could be explained by the removal of parts of the lignocellulose by the hydrothermal pretreatment. The time course also facilitated identification of temporally limited gene induction patterns.
The presented transcriptomic and biochemical datasets demonstrate that pretreatments caused modifications of the lignocellulose, to both specific structural features as well as the organisation of the overall lignocellulosic structure, that determined
transcript levels. The experimental setup allowed reliable detection of substrate-specific gene expression patterns as well as hitherto non-expressed genes. Our data suggest beneficial effects of using untreated and IL-pretreated straw, but not HT-pretreated straw, as feedstock for CAZyme production.

We present the data release paper for the Galaxy Zoo: Hubble (GZH) project. This is the third phase in a large effort to measure reliable, detailed morphologies of galaxies by using crowdsourced ...visual classifications of colour composite images. Images in GZH were selected from various publicly-released Hubble Space Telescope Legacy programs conducted with the Advanced Camera for Surveys, with filters that probe the rest-frame optical emission from galaxies out to \(z \sim 1\). The bulk of the sample is selected to have \(m_{I814W} < 23.5\),but goes as faint as \(m_{I814W} < 26.8\) for deep images combined over 5 epochs. The median redshift of the combined samples is \(z = 0.9 \pm 0.6\), with a tail extending out to \(z \sim 4\). The GZH morphological data include measurements of both bulge- and disk-dominated galaxies, details on spiral disk structure that relate to the Hubble type, bar identification, and numerous measurements of clump identification and geometry. This paper also describes a new method for calibrating morphologies for galaxies of different luminosities and at different redshifts by using artificially-redshifted galaxy images as a baseline. The GZH catalogue contains both raw and calibrated morphological vote fractions for 119,849 galaxies, providing the largest dataset to date suitable for large-scale studies of galaxy evolution out to \(z \sim 1\).

Private sector meteorology is a rapidly growing enterprise. It has been estimated that the provision of weather information has, by some estimates, a global market totaling in the billions of ...dollars. Further, the decisions based on such information could easily total trillions of dollars in the U.S. economy alone. The private sector clearly plays an important, and growing, role at the interface of weather research and the weather information needs of society. To date, little information has been paid to the connections of the meteorological research community and the scientific needs of the private sector. Thus, the time is ripe to stimulate a more active dialogue between what is generally considered the 'basic' research community of physical and social scientists and those individuals and businesses that provide weather information to myriad customers across the U.S. economy. In December 2000, the U.S. Weather Research Program (supported by NSF, NOAA, NASA, and the U.S. Navy) sponsored a workshop in Palm Springs, California, to bring together weather researchers and representatives of private sector meteorology to discuss needs, wants, opportunities, and challenges and how to enhance the linkages between the two relatively detached communities. The workshop focused on developing a better understanding of the relations of research and private sector meteorology, which ultimately means a better understanding of one of the important connections of research and societal needs.
We compared the efficacy of continuous intraarterial versus intravenous 6-mercaptopurine (6-MP) infusion in a mongrel canine renal allograft model with regard to overall survival, incidence of ...systemic and renal toxicity, and systemic drug exposure. Arterial anastomoses were done end-to-end, and infusion catheters were placed in the iliac artery or vena cava and connected to a subcutaneously placed programmable pump. A dose of 0.5 mg/kg/day 6-MP did not prolong survival over heparin-treated or untreated controls (MST = 7 days for both groups) when administered either locally or systemically. However, 0.75 mg/kg/day 6-MP i.a. (MST = 20 days) significantly prolonged survival over both untreated (P = 0.007) and heparin-treated controls (P = 0.02), with all dogs eventually dying of rejection. In contrast, 0.75 mg/kg/day i.v. (MST = 7 days) failed to prolong survival over controls (P greater than 0.1) and produced death from systemic toxicity in 3 of 7 animals. Six of 7 dogs receiving 2.0 mg/kg/day 6-MP i.a. (MST = 12 days) developed azotemia secondary to drug-induced nephrotoxicity. Identical renal histologic changes occurred in the same time frame in autotransplants treated similarly. Of 7 animals receiving 2.0 mg/kg/day i.v. (MST = 12 days), 5 died from early, severe systemic drug toxicity and 2 from early rejection. During 6-MP infusion at 0.5 mg/kg/day, systemic exposure was significantly less in the locally treated than in the systemically treated dogs when Cr concentrations were normal or moderately elevated (P less than 0.0005 and P = 0.01, respectively) but not when renal function became severely impaired (P = 0.34). In contrast to i.v. infusion, i.a. 6-MP delivery dissociated immunosuppressive efficacy from systemic toxicity, supporting previous work demonstrating high first-pass renal elimination of 6-MP. We conclude that tightly controlled local delivery of an immunosuppressive agent can effectively prolong graft survival with reduced systemic toxicity in a large animal model employing a pump/catheter system applicable to man.
