A new selenium substituted hydroxyapatite nanoparticle, with unique anticancer characteristics toward hepatoma cells, was designed, synthesized, and evaluated as the drug carrier. ...Notably, selenium substitution in the hydroxyapatite crystalline can retard the growth of the hepatoma cells in a nude mice model. Based on the massive tumor necrosis and cell apoptosis, in combination with low blood toxicity and improved organ function, more animals survived with the tumor.
•Selenium-substituted hydroxyapatite can be used as the anticancer carrier.•Selenium addition can retard the growth of the hepatoma cells.•Selenium addition exhibited lower systemic toxicity and improved organ function.•Selenium substitution can cause the massive necrosis and apoptosis in hepatoma nest.•The highest overall survival (100%) was got in selenium treatment group.
Absence of curative treatment creates urgent need for new strategies for unresectable hepatoma. Novel selenium-substituted hydroxyapatite nanoparticles (SeHAN) were designed to serve as anticancer agent. The authors examined the nanoparticles by physicochemical techniques. The in vivo efficacy and toxicity of these nanoparticles were also investigated on a nude mice model of human hepatocellular carcinoma. The results showed that the selenite ions can be incorporated into the hydroxyapatite lattice facilely. They exhibited bundles of needles shape with a size of 160–200nm. In the in vivo study, they showed better survival advantage. The overall survival rate of nude mice in the control, pure hydroxyapatite and SeHAN group were 50.00%, 76.92%, and 100.00% respectively. Blood biochemical studies showed that SeHAN group had significantly lower toxicities on the liver and kidney functions. Histopathological studies confirmed that massive tumor necrosis and calcium deposition were evident after SeHAN treatment. Moreover, immunohistochemistry and Western blot assay showed significantly reduced expression of the Ki-67, VEGF and MMP-9 protein in the SeHAN group. Taken together, these results suggest that the selenium-substituted hydroxyapatite nanoparticles could be a new type of promising anticancer agent to provide both survival advantage and lower toxicity.
In dynamic cardiac cine magnetic resonance imaging, the spatiotemporal resolution is limited by the low imaging speed. Compressed sensing (CS) theory has been applied to improve the imaging speed and ...thus the spatiotemporal resolution. In this paper, we propose a novel technique that employs a patch-based 3-D spatiotemporal dictionary for sparse representations of dynamic image sequence in the CS framework. Specifically, the dynamic image sequence is divided into overlapping patches along both the spatial and temporal directions. The dictionary is used to provide flexible sparse expressions for these patches. The underlying optimization problem is solved by variable splitting and the alternating direction method with multiplier. Experimental results based on in vivo cardiac data demonstrate that the proposed method is able to accelerate cardiac cine imaging by a factor up to 8 and outperforms the existing state-of-the-art CS methods at high accelerations. The method is expected to be useful in dynamic imaging with a higher spatiotemporal resolution.
The combined toxicity of five insecticides (chlorpyrifos, avermectin, imidacloprid, λ-cyhalothrin, and phoxim), two herbicides (atrazine and butachlor) and a heavy metal (cadmium) has been examined ...with the earthworm acute toxicity test. Toxicological interactions of these chemicals in four, five, six, seven, and eight-component mixtures were studied using the combination-index (CI) equation method. In four-component and five-component mixtures, the synergistic effects predominated at lower effect levels, while the patterns of interactions found in six, seven, and eight-component mixtures displayed synergism. The λ-CY+IMI+BUT+ATR+CPF+PHO combination displayed the most strongly synergistic interaction, with CI values ranging from 0.09 to 0.15. The nature of the interaction changes with the effect level and the relevance of synergistic effects increase with the complexity of the mixture. The CI method was compared with the classical models of concentration addition (CA) and independent action (IA) and we found that the CI method could accurately predict the combined toxicity. The predicted synergism resulted from co-existence of the pesticides and the heavy metal especially at low effect levels may have important implications in risk assessment for the real terrestrial environment.
•The combined toxicity of insecticides, herbicides and a heavy metal was examined.•Acute earthworm toxicity assays were conducted in multi-component mixtures.•Enhanced synergistic effects predominated in majority of multi-component mixtures.•Combination index method could more accurately predict the combined toxicity.•Synergism among chemical mixtures has implications for terrestrial risk assessment.
► Two novel polysaccharide fractions were isolated from Gomphidius rutilus mycelium. ► GRMP1 possessed better antioxidant activity in vitro than GRMP2. ► GRMP1 and GRMP2 have significant lymphocyte ...proliferation activity.
Two novel polysaccharide fractions, GRMP1 and GRMP2, were isolated from the mycelium of Gomphidius rutilus through submerged fermentation. GRMP1 and GRMP2 had similar average molecular weights (35 and 31kDa, respectively), and were composed of glucose and xylose at molar ratios of 0.46:1 and 0.63:1, respectively. In vitro antioxidant tests showed that GRMP1 and GRMP2 partly scavenged superoxide radical but almost had no scavenging effect on 1,1-diphenyl-2-picrylhydrazyl free radical. The effect of GRMP1 on hydroxyl radicals was stronger than that of GRMP2. Both GRMP1 and GRMP2 had relatively low reducing power and significant lymphocyte proliferation activity. In the presence of concanavalin A or lipopolysaccharide as mitogens for lymphocytes, the lymphocyte proliferation activity increased for GRMP1 but not for GRMP2 within the test dosage range.
Toxoplasma gondii is an obligate intracellular parasitic protozoan that has a wide host range and causes a zoonotic parasitosis called toxoplasmosis. This infection causes significant morbidity, ...costs for care and loss of productivity and suffering. The most effective measures to minimize this parasite’s harm to patients are prompt diagnosis and treatment and preventing infection. A parasite surface antigen, SAG1, is considered an important antigen for the development of effective diagnostic tests or subunit vaccines. This review covers several aspects of this antigen, including its gene structure, contribution to host invasion, mechanisms of the immune responses and its applications for diagnosis and vaccine development. This significant progress on this antigen provides foundations for further development of more effective and precise approaches to diagnose toxoplasmosis in the clinic, and also have important implications for exploring novel measures to control toxoplasmosis in the near future.
China has ambitious renewable energy development plans after undergoing a rapid development phase for wind power. China's prospects for wind development affect the global decarbonization path. The ...question of where to deploy turbines of this magnitude to achieve technical feasibility, high economic competitiveness, and high emission reduction has become an urgent issue for China. Due to regional differences in wind resources and economic development, the deployment of wind power in China requires clarity on the likely siting and investment costs. It requires an accurate assessment of the exploitable possibilities of wind energy resources in China, which provides practical solutions for national and regional wind energy development. In this study, a multi-potential assessment was developed to estimate the feasibility and emission reduction contribution of wind power using a combined GIS-based approach and learning curves. It is found that there are large areas of land and ocean available for wind farms in China, especially the desert in the northwest and the coastal area of Fujian province. The technical potential of onshore and offshore wind power is 8650.33 TWh/yr and 11298.9 TWh/yr, respectively. Moreover, 76.06% and 20.69% of onshore and offshore wind, respectively, could be supplied economically. The optimal areas for building onshore wind by 2030 are located in Inner Mongolia, Gansu, and Xinjiang, while the optimal construction area for offshore wind power is mainly the coastal area of Fujian. Wind energy has a large potential to meet China's electricity demand and is set to be more cost-competitive as time passes. The cost of onshore and offshore wind will decrease by 25–53% and 21–46%, respectively, by 2030. Wind development could contribute 7823.73–10723.50 Mt CO2 reduction during 2021–2030, which could help decarbonize China's energy system.
•A multi-potential assessment is developed to estimate the feasibility and emission reduction of wind power.•China's onshore and offshore wind generation could reach 8650 TWh/yr and 11299 TWh/yr respectively.•76.06% of onshore and 20.69% of offshore wind energy can be supplied economically.•Northwestern China was the most optimal option for wind power operation.•China's wind energy deployment could cut carbon emissions by 7824–10724 Mt CO2 in 2021–2030.
Previous studies had showed that Apelin 13 could protect against apoptosis induced by ischemic/reperfusion (I/R). However, the mechanisms whereby Apelin 13 protected brain I/R remained to be ...elucidated. The present study was designed to determine whether Apelin 13 provided protection through AMPK/GSK-3β/Nrf2 pathway.
In vivo, the I/R model was induced and Apelin 13 was given intracerebroventricularly 15 min before reperfusion. The neurobehavioral scores, infarction volumes, and some cytokines in the brain were measured. For in vitro study, PC12 cells were used. To clarify the mechanisms, proteases inhibitors or siRNA were used. Protein levels were investigated by western blotting.
The results showed that Apelin 13 treatment significantly reduced infarct size, improved neurological outcomes, decreased brain edema, and inhibited cell apoptosis, oxidative stress, and neuroinflammation after I/R. Apelin 13 significantly increased the expression of Nrf2 and the phosphorylation levels of AMPK and GSK-3β. Furthermore, in cultured PC12 cells, the same protective effects were also observed. Silencing Nrf2 gene with its siRNA abolished the Apelin 13's prevention of I/R-induced PC12 cell injury, oxidative stress, and inflammation. Inhibition of AMPK by its siRNA decreased the level of Apelin 13-induced Nrf2 expression and diminished the protective effects of Apelin 13. The interplay relationship between GSK-3β and Nrf2 was also verified with relative overexpression. Using selective inhibitors, we further identified the upstream of AMPK/GSK-3β/Nrf2 is AR/Gα/PLC/IP3/CaMKK.
In conclusion, the previous results showed that Apelin 13 protected against I/R-induced ROS-mediated inflammation and oxidative stress through activating the AMPK/GSK-3β pathway by AR/Gα/PLC/IP3/CaMKK signaling, and further upregulated the expression of Nrf2-regulated antioxidant enzymes.
Background:Ischemic postconditioning (IPostC) is an endogenous protective mechanism to reduce ischemia-reperfusion (I/R) injury. However, whether IPostC protects aged cardiomyocytes against I/R ...injury is not fully understood. Considering the protective function of microRNA 30a (miR-30a) against ischemia-induced injury in H9C2 cells, its role in the protective effects of IPostC on I/R injury of aged cardiomyocytes was investigated further. Methods and Results:To mimic I/R and IPostC in vitro, the aged cardiomyocyte model for hypoxia postconditioning (HPostC) treatment was established by 9 days of incubation with 8 mg/mL D-galactose and then followed by exposure to hypoxic environment. HPostC significantly alleviated hypoxia/reoxygenation (H/R) injury and reduced autophagy of aged cardiomyocytes, as evidenced by decreased LC3B-II expression and increased p62 by Western blot. Quantified by quantitative real-time polymerase chain reaction (qRT-PCR), miR-30a was increased in aged cardiomyocytes treated with HPostC compared with I/R injury group. Overexpression of miR-30a by LV3-rno-miR-30a mimic promoted cardioprotective effect of HPostC in aged cardiomyocytes by suppressing BECN1-mediated autophagy, all of which was abrogated by knockdown of miR-30a expression. Epigenetic analyses demonstrated that HPostC reduced DNA methyltransferase 3b-mediated DNA hypomethylation levels at miR-30a promoter, leading to upregulation of miR-30a.Conclusions:HPostC protected aged cardiomyocytes survival against H/R injury via DNMT3b-dependent activation of miR-30a. miR-30a could be a potential therapeutic target for ischemic myocardial infarction.
The natural co-occurrence of multiple mycotoxins has been reported in cereals and cereal products worldwide. Even though the dietary exposure to mycotoxins constitutes a serious human health, most ...reports are limited to the toxic effect of individual mycotoxins. The purpose of the present study was to assess the combined toxic effects of zearalenone (ZEN) and fumonisin B1 (FB1) and the potential interaction of their mixture on zebrafish (Danio rerio) embryos. Our results showed that ZEN possessed the higher toxicity to embryonic zebrafish (7-day LC50 value of 0.78 mg a.i. L−1) compared with FB1 (7-day LC50 value of 227.7 mg a.i. L−1). The combination of ZEN and FB1 exerted an additive effect on zebrafish embryos. Meanwhile, the activities of antioxidant CAT, caspase-3, and detoxification enzyme CYP450, as well as the expressions of six genes (Mn-sod, cas9, bax, cc-chem, ERα, and crh) associated with oxidative stress, cellular apoptosis, immune system, and endocrine system were prominently altered in the mixture exposure compared with the corresponding single treatment group of ZEN or FB1. Taken together, the regulatory standards of mycotoxins in food and feed should be updated based on the mixture effects of mycotoxins, and there is an increased need on effective detoxification methods for controlling and reducing the toxicity of multiple mycotoxins in animal feed and throughout the food supply chain.
