The neotropical Oscar Astronotus ocellatus has been translocated worldwide as an ornamental fish species with significant economic importance. Twelve adult individuals of A. ocellatus collected from ...aquaria shops in northern and eastern India were examined for monogenean parasites. Based on morphological methods, all fish samples were found to be infested with Gussevia asota, with a 100% prevalence and high mean intensity 30.16 ± 9.70 (18-48, n=12). Molecular analysis of the partial 28S rRNA gene sequence (675 bp), followed by phylogenetic analysis, confirmed the parasite's identity further. This is the first report of G. asota in India and highlights the need to update and strengthen the quarantine procedures concerning the import of ornamental fish.

This study aims to evaluate the applicability of the three mitochondrial genes 16S rRNA (16S), cytochrome b (cyt b), and cytochrome oxidase subunit I (COI) for the identification of 50 European marine fish species by combining techniques of "DNA barcoding" and microarrays. In a DNA barcoding approach, neighbour Joining (NJ) phylogenetic trees of 369 16S, 212 cyt b, and 447 COI sequences indicated that cyt b and COI are suitable for unambiguous identification, whereas 16S failed to discriminate closely related flatfish and gurnard species. In course of probe design for DNA microarray development, each of the markers yielded a high number of potentially species-specific probes in silico, although many of them were rejected based on microarray hybridisation experiments. None of the markers provided probes to discriminate the sibling flatfish and gurnard species. However, since 16S-probes were less negatively influenced by the "position of label" effect and showed the lowest rejection rate and the highest mean signal intensity, 16S is more suitable for DNA microarray probe design than cty b and COI. The large portion of rejected COI-probes after hybridisation experiments (>90%) renders the DNA barcoding marker as rather unsuitable for this high-throughput technology.
Based on these data, a DNA microarray containing 64 functional oligonucleotide probes for the identification of 30 out of the 50 fish species investigated was developed. It represents the next step towards an automated and easy-to-handle method to identify fish, ichthyoplankton, and fish products.
Exposure to high concentrations of crude oil produces a lethal syndrome of heart failure in fish embryos. Mortality is caused by cardiotoxic polycyclic aromatic hydrocarbons (PAHs), ubiquitous ...components of petroleum. Here, we show that transient embryonic exposure to very low concentrations of oil causes toxicity that is sublethal, delayed, and not counteracted by the protective effects of cytochrome P450 induction. Nearly a year after embryonic oil exposure, adult zebrafish showed subtle changes in heart shape and a significant reduction in swimming performance, indicative of reduced cardiac output. These delayed physiological impacts on cardiovascular performance at later life stages provide a potential mechanism linking reduced individual survival to population-level ecosystem responses of fish species to chronic, low-level oil pollution.
The extensive use of pharmaceuticals has resulted in the intensive contamination of water bodies. Tetracycline is a type of antibiotic and its potential toxicity is causing environmental concern. The ...effects of developmental toxicity and the mechanisms of tetracycline on fish embryos are not well understood. Zebrafish embryos are used in this study to investigate the developmental toxicity of this compound. Four hour post-fertilization (hpf) zebrafish embryos are exposed to different concentrations of tetracycline until 96 hpf. The larvae display developmental delay phenotypes, including hatching delay, shorter body length, increased yolk sac area and uninflated swim bladder upon exposure to tetracycline. Delayed yolk sac absorption and swim bladder deficiency at 96 hpf are observed in the zebrafish larvae upon exposure to 20 μg/L of tetracycline. To test whether tetracycline causes oxidative damage and the resulting oxidative stress-induced apoptosis, the generation of reactive oxygen species (ROS), Acridine Orange staining and real time polymerase chain reaction have been performed in this study. The results indicate that tetracycline exposure results in significant increases in ROS production and cell apoptosis, mainly in the tail areas at 96 hpf. The gene expression pattern demonstrates that tetracycline induces ROS which causes apoptosis in the zebrafish larvae, and the results also indicate that caspase-dependent apoptotic pathways may greatly contribute to tetracycline-induced apoptosis in the early-life stages of the zebrafish. In addition, we have investigated the effects of tetracycline on marker genes related to resistance mechanisms and gene regulating drug biotransformation. The results of these gene expression studies indicate that tetracycline could induce zebrafish to resist pharmaceuticals and Cytochrome P450s that are involved in the biotransformation of tetracycline in zebrafish larvae. The overall results indicate that tetracycline can produce oxidative stress and induce apoptosis, which brings about significant developmental delay in zebrafish embryos.
The increase in reports of novel diseases in a wide range of ecosystems, both terrestrial and marine, has been linked to many factors including exposure to novel pathogens and changes in the global ...climate. Prevalence of skin cancer in particular has been found to be increasing in humans, but has not been reported in wild fish before. Here we report extensive melanosis and melanoma (skin cancer) in wild populations of an iconic, commercially-important marine fish, the coral trout Plectropomus leopardus. The syndrome reported here has strong similarities to previous studies associated with UV induced melanomas in the well-established laboratory fish model Xiphophorus. Relatively high prevalence rates of this syndrome (15%) were recorded at two offshore sites in the Great Barrier Reef Marine Park (GBRMP). In the absence of microbial pathogens and given the strong similarities to the UV-induced melanomas, we conclude that the likely cause was environmental exposure to UV radiation. Further studies are needed to establish the large scale distribution of the syndrome and confirm that the lesions reported here are the same as the melanoma in Xiphophorus, by assessing mutation of the EGFR gene, Xmrk. Furthermore, research on the potential links of this syndrome to increases in UV radiation from stratospheric ozone depletion needs to be completed.
