Trema (Trema orientalis Linn. Blume) merupakan tanaman serba guna karena semua bagian pohon dapat digunakan. Dalam pengembangan tanaman ini, diperlukan benih bermutu, dimana salah satu syarat untuk ...menentukan benih bermutu adalah benih harus berasal dari buah yang sudah masak fisiologis. Tujuan penelitian adalah mengetahui mutu fisik, mutu fisiologis, dan kandungan biokimia (lemak, karbohidrat dan protein) benih trema berdasarkan tingkat kemasakan buah. Benih trema yang digunakan berasal dari Badung, Bali. Buah trema dikelompokkan menjadi 3 warna (hijau, coklat, hitam). Rancangan penelitian menggunakan Rancangan Acak Lengkap dengan parameter yang diamati : ukuran buah dan benih, berat buah dan benih, kadar air buah dan benih, kandungan biokimia (protein, lemak, karbohidrat), daya berkecambah, dan kecepatan berkecambah. Hasil penelitian menunjukkan : (1) Tingkat kemasakan buah berpengaruh nyata terhadap mutu fisik (ukuran buah dan benih, berat buah dan benih, kadar air buah dan benih), fisiologis (daya berkecambah, kecepatan berkecambah), dan kandungan biokimia (lemak, karbohidrat, protein) benih trema, (2) Mutu fisik dan fisiologis benih yang berasal dari buah yang berwarna hitam lebih baik dibandingkan dengan buah berwarna hijau dan coklat, (3) Kandungan lemak, karbohidrat dan protein benih trema berbeda dari masing-masing tingkat kemasakan (hijau, coklat, hitam), (4) Buah trema yang sudah mencapai masak fisiologis adalah buah berwarna hitam, dengan kriteria yaitu : panjang buah 3,87 ± 0,05 mm dan lebar 3,41 ± 0,02 mm; panjang benih 2,10 ± 0,05 mm dan lebar 1,81 ± 0,06 mm; berat 1000 butir buah 25,6883 gr; berat 1000 butir benih 3,8288 gr; kadar air buah 54,74%; kadar air benih 12,03%; daya berkecambah 78%; kecepatan berkecambah 3,05%/Etmal; kandungan karbohidrat 20,10%; kandungan protein 2,84%; kandungan lemak 0,65%.
Establishing the genetic diversity and population structure of a species can guide the selection of appropriate conservation and sustainable utilization strategies. Next-generation sequencing (NGS) ...approaches are increasingly being used to generate multi-locus data for genetic structure determination. This study presents the genetic structure of a fodder species -Trema orientalis based on two genome-wide high-throughput diversity array technology (DArT) markers; silicoDArT and single nucleotide polymorphisms (SNPs). Genotyping of 119 individuals generated 40,650 silicoDArT and 4767 SNP markers. Both marker types had a high average scoring reproducibility (>99%). Genetic relationships explored by principal coordinates analysis (PCoA) showed that the first principal coordinate axis explained most of the variation in both the SilicoDArT (34.2%) and SNP (89.6%) marker data. The average polymorphic information content did not highly differ between silicoDArT (0.22) and SNPs (0.17) suggesting minimal differences in informativeness in the two groups of markers. The, mean observed (H.sub.o) and expected (H.sub.e) heterozygosity were low and differed between the silicoDArT and SNPs respectively, estimated at H.sub.o = 0.08 and H.sub.e = 0.05 for silicoDArT and H.sub.o = 0.23 and H.sub.e = 0.19 for SNPs. The population of T. orientalis was moderately differentiated (F.sub.ST = 0.20-0.53) and formed 2 distinct clusters based on maximum likelihood and principal coordinates analysis. Analysis of molecular variance revealed that clusters contributed more to the variation (46.3-60.8%) than individuals (32.9-31.2%). Overall, the results suggest a high relatedness of the individuals sampled and a threatened genetic potential of T. orientalis in the wild. Therefore, genetic management activities such as ex-situ germplasm management are required for the sustainability of the species. Ex-situ conservation efforts should involve core collection of individuals from different populations to capture efficient diversity. This study demonstrates the importance of silicoDArT and SNP makers in population structure and genetic diversity analysis of Trema orientalis, useful for future genome wide studies in the species.

Central to symbiotic ...signalling are nuclear-associated oscillations in calcium ions (Ca2+), occurring in the root hairs of several legume species in response to the rhizobial Nod factor signal.
In this study we expanded the species analysed for activation of Ca2+ oscillations, including nonleguminous species within the nitrogen-fixing clade.
We showed that Ca2+ oscillations are a common feature of legumes in their association with rhizobia, while Cercis, a non-nodulating legume, does not show Ca2+ oscillations in response to Nod factors from Sinorhizobium fredii NGR234. Parasponia andersonii, a nonlegume that can associate with rhizobia, showed Nod factor-induced calcium oscillations to S. fredii NGR234 Nod factors, but its non-nodulating sister species, Trema tomentosa, did not. Also within the nitrogen-fixing clade are actinorhizal species that associate with Frankia bacteria and we showed that Alnus glutinosa induces Ca2+ oscillations in root hairs in response to exudates from Frankia alni, but not to S. fredii NGR234 Nod factors.
We conclude that the ability to mount Ca2+ oscillations in response to symbiotic bacteria is a common feature of nodulating species within the nitrogen-fixing clade.
Trema orientale (L.) Blume is an important medicinal plant with multiple applications for treating several disease conditions. This study compiled published data on botanical, traditional uses, ...phytochemistry, pharmacology, and antimicrobials, coupled with discussing the conservation and domestication potential of T. orientale. Data were sourced from databases such as Google Scholar, PubMed, Scopus, Elsevier Plants of the World Online (Kew Science), Global Biodiversity Information Facility (GBIF), and World Flora Online (WFO), using key search terms: Trema orientale or orientalis, phytochemistry pharmacology, taxonomy, and domestication with Boolean operators to include and exclude articles for the review. The review indicated that molecular studies have shown that T. orientale is closely related to a sister group of Cannabis through plastome phylogenetic evidence which accounts for its transfer from Ulmaceae to the Cannabaceae family. T. orientale is distributed across several African countries and has recently been assessed as the Least Concern by the IUCN Red List of Threatened Species. Nevertheless, deforestation continues to pose an extinction risk to their population. Currently, 31 compounds have been isolated from different parts of T. orientale justifying many traditional uses accredited to it. T. orientale is considered a dose-dependent safe remedy for the treatment of infectious diseases, cancer, cardiovascular diseases and other disease conditions ascribed to it except for its continuous application. This review underscores the domestication potential of T. orientale including evidence of molecular markers, soil seed banks, and promising outcomes of germination experimentations. This, therefore, presents significant gains toward sustainable utilization of Trema orientale.

Trema orientalis (
MLETO) Linn. From the high-resolution mass spectrometry (HRMS) analysis, 21 nucleated flavonoids were screened out, of which only apigeniflavan was selected for further studies because it followed Lipinski’s rule and showed no toxicity. The pharmacokinetics and physiochemical characteristics of apigeniflavan were performed using the online web servers pkCSM, Swiss ADME, and ProTox-II. This is the first in silico study to report the efficiency of apigeniflavan in pancreatic cancer treatment. The targets of apigeniflavan were fetched from SwissTargetPrediction database. The targets of pancreatic cancer were retrieved from DisGeNET and GeneCards. The protein–protein interaction of the common genes using Cytoscape yielded the top five hub genes: KDR, VEGFA, AKT1, SRC, and ESR1. Upon molecular docking, the lowest binding energies corresponded to best docking score which indicated the highest protein–ligand affinity. Kyoto Encyclopaedia of Genes and Genomes (KEGG) database was employed to see the involvement of hub genes in pathways related to pancreatic cancer. The following, pancreatic cancer pathway, MAPK, VEGF, PI3K–Akt, and ErbB signaling pathways, were found to be significant. Our results indicate the involvement of the hub genes in tumor growth, invasion and proliferation in the above-mentioned pathways, and therefore necessitating their downregulation. Moreover, apigeniflavan can flourish as a promising drug for the treatment of pancreatic cancer in future.
Invasive species are frequently found in recently disturbed sites. To examine how these disturbance-dependent invasive species exploit resource pulses resulting from disturbance, twelve physiological ...and morphological traits, including age-dependent responsiveness in leaf traits to nitrogen pulse, were compared between Bischofia javanica, an invasive tree species in Ogasawara islands, and three native Ogasawara species, each having a different successional status. When exposed to a nitrogen pulse, invasive B. javanica showed higher increases in photosynthetic capacity, leaf area, epidermal cell number and cell size in leaves of broad age classes, and root nitrogen absorption ability than two native mid-/late or late-successional species, but showed no particular superiority to a native pioneer species in these responses. Under low nitrogen, however, it showed the largest relative growth rate among the four species, while the native pioneer showed the lowest growth. From these results, we concluded that the combination of moderately high responsiveness to resource pulses and the ability to maintain steady growth under resource limitations may give B. javanica a competitive advantage over a series of native species with different successional status from early to late-successional stages.

Understanding how mutualistic interactions are stabilized in the presence of cheaters is a major question in evolutionary biology. The legume–rhizobia mutualism has become a model system for studying how plants control cheating partners. However, the generality and evolutionary origins of these control mechanisms are intensely debated. In this Opinion article, we argue that a novel system – the Parasponia–rhizobia mutualism – will significantly advance research in mutualism stability. Parasponia is the only non-legume lineage to have evolved a rhizobial symbiosis, which provides an evolutionary replicate to test how rhizobial exploitation is controlled. Evidence also suggests that this symbiosis is young. This allows studies at an earlier evolutionary stage in mutualisms, so the origin of control mechanisms can be better understood.
Poisoning by Trema micrantha commonly causes hepatocellular necrosis in cattle, sheep, and goats and edema and cerebral hemorrhage in horses. This plant can cause toxic pneumopathy in sheep, and ...there is only one report of the natural form and one of the experimental form in the State of Rio Grande do Sul. This study aimed to report an outbreak of the respiratory form of natural poisoning by T. micrantha in sheep. Six sheep developed clinical respiratory signs after consumption of the plant and four of them died and two recovered after treatment with dexamethasone. The sheep presented tachypnea, noisy breathing, edema of the face, eyelids, and vulva, and subcutaneous emphysema on the face and neck. Necropsy (Sheep 2, 3, and 4) showed uncollapsed, heavy, diffuse red lungs with evident costal impressions and a moderate amount of serosanguineous fluid flowed at section. The liver had a moderate diffuse evident lobular pattern. The histopathology of the lungs of the three necropsied sheep showed congestion and edema with the formation of hyaline membranes within accentuated diffuse alveoli, in addition to thickening of the alveolar septa due to mild to moderate diffuse type II pneumocyte hyperplasia and also mild to moderate diffuse infiltrate of macrophages, lymphocytes, plasma cells, and neutrophils in the lumen of alveoli, bronchi, and bronchioles. Sheep 3 also showed type II pneumocytes with enlarged and hyperchromatic nuclei, sometimes binucleated with evident nucleoli, and, in some regions, the pneumocytes were desquamated to the alveolar lumen forming small syncytia and mild multifocal hyperplasia in the bronchial epithelium. The anti-cytokeratin IHC evaluation showed marked diffuse intracytoplasmic staining in hyperplastic type II pneumocytes in the bronchiolar epithelium of the three evaluated sheep. The liver of the three sheep had mild multifocal centrilobular necrosis. It seems to be the second report of spontaneous poisoning by T. micrantha in sheep developing lung lesions described in Brazil and the first in the State of Santa Catarina.
RESUMO: A intoxicação por Trema micrantha, comumente causa em bovinos, ovinos e caprinos necrose hepatocelular e, edema e hemorragia cerebral em equinos. Essa planta em ovinos pode causar pneumopatia tóxica, existindo descrição apenas de um relato da forma natural e um da forma experimental no estado do Rio Grande do Sul. O objetivo deste trabalho é relatar, um surto, da forma respiratória de intoxicação natural por T. micranta em ovinos. Seis ovinos desenvolveram sinais clínicos respiratórios após o consumo da planta e destes quatro morreram e dois após o tratamento com dexametasona se recuperaram. Os ovinos apresentaram taquipneia, respiração ruidosa, edema de face, pálpebras, vulva e enfisema subcutâneo na face e pescoço. Na necropsia (Ovinos 2, 3 e 4) observou-se pulmões não colabados, pesados, vermelhos difusos com impressões costais evidentes e ao corte fluindo moderada quantidade de líquido serosanguinolento. O fígado apresentava padrão lobular evidente difuso moderado. Na histopatologia, em pulmões dos três ovinos necropsiados havia congestão e edema com formação de membranas hialinas no interior de alvéolos difusos acentuados, além de espessamento dos septos alveolares devido a hiperplasia de pneumócitos tipo II difuso discreta à moderada, havia ainda, infiltrado de macrófagos, linfócitos, plasmócitos e neutrófilos difuso discreto a moderado no lúmen de alvéolo, brônquios e bronquíolos. No Ovino 3, observou-se ainda pneumócitos tipo II com núcleos aumentados e hipercromáticos, por vezes binucleados com nucléolos evidentes, e em algumas regiões estes pneumócitos estavam descamados para a luz alveolar formando pequenos sincícios e no epitélio de brônquios havia hiperplasia multifocal discreta. A avaliação IHQ anti-citoqueratina demonstrou marcação intracitoplasmática difusa acentuada em pneumócitos tipo II hiperplásicos no epitélio bronquiolar dos três ovinos avaliados. No fígado dos três ovinos havia necrose centrolobular multifocal discreta. Este parece ser o segundo relato da intoxicação espontânea por T. micrantha em ovinos desenvolvendo lesões pulmonares, descrito no Brasil e o primeiro no estado de Santa Catarina.
Trema orientalis L. is a potential species to be developed as a biomass-based renewable energy source. The use of quality seedling that can be done through vegetative techniques is one of the issues ...that determine the success of plantation development. The purpose of this research is to get the best cutting material and an optimal growth regulator for multiplication of shoot cuttings. The shoot cuttings were obtained from 12-month-old seedlings. The design used was a complete randomized design (CRD) factorial pattern with two factors, namely cuttings positions (shoot, middle and bottom parts) and concentration of naphthalene acetic acid (NAA) growth regulator of 0, 50, 100, 150, 200, 250, and 300 ppm. The result showed that T. Orientalis propagation can be carried out well by using seedling shoot section soaked in NAA 300 ppm for 10 minutes. The treatment was able to have the percentage of rooting cutting of 71.67percent, number of roots of 6 pieces, root dry weight of 0.088 g, and shoot dry weight of 0.148 g.

L. (MFETO) was analysed through HRMS. In this in silico study, pharmacokinetic and physicochemical properties of the identified flavonoids from MFETO were screened out by ADMET analysis. Kaempferol and catechin followed Lipinski rules and showed no toxicity in Protox II. Targets of these compounds were taken from SwissTarget prediction and TCMSP whilst targets for pancreatic cancer were taken from GeneCards and DisGeNET databases. The protein–protein interaction (PPI) network of common genes was generated through STRING and then exported to the Cytoscape to get top 5 hub genes (AKT1, SRC, EGFR, TNF, and CASP3). The interaction between compounds and hub genes was analysed using molecular docking, and high binding affinity between them can be visualised by Biovia discovery studio visualizer. Our study shows that, five hub genes related to pancreatic cancer play an important role in tumour growth induction, invasion and migration. Kaempferol effectively check cell migration by inhibiting ERK1/2, EGFR-related SRC, and AKT pathways by scavenging ROS whilst catechin inhibited TNFα-induced activation and cell cycle arrest at G1 and G2/M phases by induction of apoptosis of malignant cells. Kaempferol and catechin containing MFETO can be used for formulation of potent drugs for pancreatic cancer treatment in future.
