  • Metode za določanje homocisteina = Methods for the determination of homocysteine

    The increasing interest in measuring homocysteine has led to the development of new methods for determination. Determination of homocysteine is performed mostly on plasma. Concentration of ... homocysteine in collected blood samples increases at ambient temperature. The increase is inhibited at low temperatureand by inhibitors: citrate, EDTA and 3-deazaadenosine. The latter causes interference in immunoassays. Use of fluoride is not recommended because of its osmotic effects. Homocysteine is most frequently determined by HPLC method with fluorescence detection. The method requires derivatization inaddition to reduction of homocysteine. Laboratories without HPLC equipment commonly use immunoassays. The first automated immunoassay was based on detection of fluorescence polarization (FPIA). Some ELISA assays were also developed. Enzymatic methods with colorimetric detection could possibly be adapted to most of biochemical analyzers. However, these methods are still notdeveloped for routine use. Choice of appropriate method for the determination of homocysteine should be based on anticipated number of samplesand predicted application of the results.
