    Cellular and molecular mechanisms of chronic obstructive pulmonary disease (COPD) are poorly understood. The active components of the complement system C3a and C5a might have a role in COPD by ... promoting inflammation and enhancing airway hyper responsiveness. C5a is involved in both, non-specific and specific immune responses. It is a potent chemotactic factor for many inflammatory cells, including lymphocytes T, which were proposed to be the keycells in COPD pathogenesis (autoimmune hypothesis of COPD), and was reported to stimulate the development of type 1 T cells. We sought to determine whether the levels of complement factors C3a, C4a, and C5a are elevated at the site of inflammation in COPD and asthmatic patients. We analysed the induced sputum of patients with COPD (n=7), asthma (n=10) and healthy non-smokers (n=12). The concentrations of anaphylatoxins in the induced sputum were measured by cytometric bead array. We found significantly increased CSa/CSa desArg concentrations in supernatants of the induced sputum of COPD (P = 0, 007) and asthmatic patients (P = 0,002) compared to the - control group. Furthermore, in COPD patients the CSa/CSa desArg concentrationswere significantly negatively correlated with lung diffusion coefficient (r = - 0,71, P = 0,035). There was no significant difference in C3a/C3a desArg or C4alC4a desArg measurements between the three groups of subjects. We failed to examine the correlation between C5a concentrations and percentage of type 1 T cells in induced sputum because of the small numbers ofdetected T cells in the samples. Second part of a study investigated the phenotype of lung tissue T cells in COPD patients and compared it to the phenotype of T cells in smokers that despite a similar smoking history, did not develop the disease.
