P. australis
decreases with increasing latitude; and (3) invasive genotypes exhibit either no latitudinal gradient, or a nonparallel latitudinal gradient in herbivory and defenses compared to native genotypes. For the European genotypes, we tested two additional hypotheses: (4) defenses, nutritional condition, and herbivory would differ between the native (EU) and invasive ranges (NA) and (5) latitudinal gradients in defenses and herbivory would be similar between ranges. Within NA, chewing damage, internal stem-feeding incidence, and aphid abundance were 650%, 300%, and 70% lower, respectively, on invasive than native
P. australis
genotypes. Genotypes in NA also differed in nutritional condition (percent N, C:N ratio), but there was little support for invasive genotypes being better defended than native genotypes. For the European genotypes, herbivory was significantly lower in the invaded than native range, supporting the enemy-release hypothesis. Defense levels (leaf toughness and total phenolics) and tissue percent C and percent N were higher in the invaded than native range for European genotypes. Overall, latitudinal gradients in

nutritional condition, defenses, and herbivory were common. Interestingly, chewing damage and stem-feeder incidence decreased with latitude for native

suggest that the community may be more susceptible to invasion at lower than at higher latitudes. Our study points to the need for invasion biology to include a biogeographic perspective.
Plant-microbe interactions play crucial roles in species invasions but are rarely investigated at the intraspecific level. Here, we study these interactions in three lineages of a globally ...distributed plant, Phragmites australis. We use field surveys and a common garden experiment to analyze bacterial communities in the rhizosphere of P. australis stands from native, introduced, and Gulf lineages to determine lineage-specific controls on rhizosphere bacteria. We show that within-lineage bacterial communities are similar, but are distinct among lineages, which is consistent with our results in a complementary common garden experiment. Introduced P. australis rhizosphere bacterial communities have lower abundances of pathways involved in antimicrobial biosynthesis and degradation, suggesting a lower exposure to enemy attack than native and Gulf lineages. However, lineage and not rhizosphere bacterial communities dictate individual plant growth in the common garden experiment. We conclude that lineage is crucial for determination of both rhizosphere bacterial communities and plant fitness.Environmental factors often outweigh host heritable factors in structuring host-associated microbiomes. Plant–soil feedbacks (PSFs) influence plant competition via direct interactions with pathogens and mutualists or indirectly via apparent competition/mutualisms (i.e., spillover to co-occurring ...plants) and soil legacy effects. It is currently unknown how intraspecific variation in PSFs interacts with the environment (e.g., nutrient availability) to influence competition between native and invasive plants. We conducted a fully crossed multi-factor greenhouse experiment to determine the effects of Phragmites australis rhizosphere soil biota, interspecific competition, and nutrient availability on biomass of replicate populations from one native and two invasive lineages of common reed (P. australis) and a single lineage of native smooth cordgrass (Spartina alterniflora). Harmful soil biota consistently dominated PSFs involving all three P. australis lineages, reducing biomass by 10%. Indirect PSFs (i.e., soil biota spillover) from the two invasive P. australis lineages reduced S. alterniflora biomass by 7%, whereas PSFs from the native P. australis lineage increased S. alterniflora biomass by 6%. Interestingly, interspecific competition and PSFs interacted to weaken their respective impacts on S. alterniflora, whereas they exerted synergistic negative effects on P. australis. Phragmites australis soil biota decreased S. alterniflora biomass when grown alone (i.e., a soil legacy), but increased S. alterniflora biomass when grown with P. australis, suggesting that P. australis recruits harmful generalist soil biota or facilitates S. alterniflora via spillover (i.e., apparent mutualism). Soil biota also reduced interspecific competition impacts on S. alterniflora, although it remained competitively inferior to P. australis across all treatments. Competitive interactions and responses to nutrients did not differ among P. australis lineages, indicating that interspecific competition and nutrient deposition may not be key drivers of P. australis invasion in North America. Although soil biota, interspecific competition, and nutrient availability appear to have no direct impact on the success of invasive P. australis lineages in North America, intraspecific lineage variation in indirect spillover and soil legacies from P. australis occur and may have important implications for co-occurring native species and restoration of invaded habitats. Our study integrates multiple factors linked to plant invasions, highlighting that indirect interactions are likely commonplace in influencing plant community dynamics and invasion success and impacts.
The juxtaposition of plant-species invasions with latitudinal gradients in herbivore pressure is an important yet mostly unexplored issue in invasion biology. Latitudinal clines in defense and ...palatability to herbivores are expected to exist in native plant species but the evolution of these clines may lag behind for invasive plant species resulting in non-parallel latitudinal clines that may impact invasion success. Our study focused on a native and European invasive lineages of the common reed Phragmites australis in North America. Using native and invasive genotypes of P. australis collected across a 17° latitudinal range, we performed experiments in replicate northern and southern common gardens to investigate whether these two lineages exhibited different genetically based latitudinal clines in defenses, nutritional condition, and palatability to their herbivores, the aphid Hyalopterus pruni and the fall armyworm Spodoptera frugiperda. We also tested whether invasive genotypes are more phenotypically plastic than native genotypes and whether plasticity varies with latitude. Although invasive genotypes did not exhibit higher defense levels (leaf toughness, phenolics, percent carbon), they were considerably less palatable to their herbivores than native genotypes. Genetically based latitudinal clines were evident for both native and invasive P. australis and for all defenses, nutrients, and at least one palatability trait for each herbivore. In 36% of the cases where clines were evident, they were non-parallel between the two lineages. These data suggest that clines in the invasive genotypes of P. australis evolved within the past ~100 years. Moreover, our study showed that the occurrence and direction of latitudinal clines in plant traits were commonly dependent on where the study was conducted (north or south), indicating strong phenotypic plasticity in these genetic-based clines. Finally, traits for invasive genotypes of P. australis were 2.5 times more plastic than traits for native genotypes. Interestingly, plasticity for native but not invasive genotypes was strongly dependent on latitude of origin. Such spatial heterogeneity within and between the native and invasive lineages of P. australis with respect to their interactions with herbivores can generate substantial spatial variability in biotic resistance that can have important implications for the establishment and spread of invasive genotypes and species.
Psoriasis is a chronic autoimmune disease with complex genetic architecture. Previous genome-wide association studies (GWAS) and a recent meta-analysis using Immunochip data have uncovered 36 ...susceptibility loci. Here, we extend our previous meta-analysis of European ancestry by refined genotype calling and imputation and by the addition of 5,033 cases and 5,707 controls. The combined analysis, consisting of over 15,000 cases and 27,000 controls, identifies five new psoriasis susceptibility loci at genome-wide significance (P<5 × 10(-8)). The newly identified signals include two that reside in intergenic regions (1q31.1 and 5p13.1) and three residing near PLCL2 (3p24.3), NFKBIZ (3q12.3) and CAMK2G (10q22.2). We further demonstrate that NFKBIZ is a TRAF3IP2-dependent target of IL-17 signalling in human skin keratinocytes, thereby functionally linking two strong candidate genes. These results further integrate the genetics and immunology of psoriasis, suggesting new avenues for functional analysis and improved therapies.

Purpose The authors of this guideline reviewed the urologic trauma literature to guide clinicians in the appropriate methods of evaluation and management of genitourinary injuries. Materials and ...Methods A systematic review of the literature using the MEDLINE® and EMBASE databases (search dates 1/1/90-9/19/12) was conducted to identify peer-reviewed publications relevant to urotrauma. The review yielded an evidence base of 372 studies after application of inclusion/exclusion criteria. These publications were used to inform the statements presented in the guideline as Standards, Recommendations or Options. When sufficient evidence existed, the body of evidence for a particular treatment was assigned a strength rating of A (high), B (moderate) or C (low). In the absence of sufficient evidence, additional information is provided as Clinical Principles and Expert Opinions. Results Guideline statements were created to inform clinicians on the initial observation, evaluation and subsequent management of renal, ureteral, bladder, urethral and genital traumatic injuries. Conclusions Genitourinary organ salvage has become increasingly possible as a result of advances in imaging, minimally invasive techniques, and reconstructive surgery. As the field of genitourinary reconstruction continues to evolve, clinicians must strive to approach clinical problems in a creative, multidisciplinary, evidence-based manner to ensure optimal outcomes.
Psoriasis is a common and debilitating disease of the skin, nails, and joints, with an acknowledged but complex genetic basis. Early genome-wide linkage studies of psoriasis focused on segregation of ...microsatellite markers in families; however, the only locus consistently identified resided in the major histocompatibility complex. Subsequently, several groups mapped this locus to the vicinity of HLA-C, and two groups have reported HLA-Cw6 itself to be the major susceptibility allele. More recently, the development of millions of single-nucleotide polymorphisms, coupled with the development of high-throughput genotyping platforms and a comprehensive map of human haplotypes, has made possible a genome-wide association approach using cases and controls rather than families. Taking advantage of these developments, we participated in a collaborative genome-wide association study of psoriasis involving thousands of cases and controls. Initial analysis of these data revealed and/or confirmed association between psoriasis and seven genetic loci—HLA-C, IL12B, IL23R, IL23A, IL4/IL13, TNFAIP3, and TNIP1—and ongoing studies are revealing additional loci. Here, we review the epidemiology, immunopathology, and genetics of psoriasis, and present a disease model integrating its genetics and immunology.
We describe the application of non-negative matrix factorization to generate compact reconstructions of quasar spectra from the Sloan Digital Sky Survey (SDSS), with particular reference to broad ...absorption line quasars (BALQSOs). BAL properties are measured for Si ivλ1400, C ivλ1550, Al iiiλ1860 and Mg iiλ2800, resulting in a catalogue of 3547 BALQSOs. Two corrections, based on extensive testing of synthetic BALQSO spectra, are applied in order to estimate the intrinsic fraction of C iv BALQSOs. First, the probability of an observed BALQSO spectrum being identified as such by our algorithm is calculated as a function of redshift, signal-to-noise ratio and BAL properties. Secondly, the different completenesses of the SDSS target selection algorithm for BALQSOs and non-BAL quasars are quantified. Combining the detection probabilities with an intrinsic E(B−V) distribution capable of reproducing the observed increase in mean E(B−V) with increasing redshift, the intrinsic C iv BALQSO fraction is 41 ± 5 per cent. Our analysis of the selection effects allows us to measure the dependence of the intrinsic C iv BALQSO fraction on luminosity and redshift. We find a factor of 3.5 ± 0.4 decrease in the intrinsic fraction from the highest redshifts, z≃ 4.0, down to z≃ 2.0. The redshift dependence implies that an orientation effect alone is not sufficient to explain the presence of BAL troughs in some but not all quasar spectra. Our results are consistent with the intrinsic BALQSO fraction having no strong luminosity dependence, although with 3σ limits on the rate of change of the intrinsic fraction with luminosity of −6.9 and 7.0 per cent dex−1 we are unable to rule out such a dependence.

Chronic Refractory Cough (CRC) is a common condition that significantly impairs patients' quality of life. Unfortunately, in many situations patients continue to experience CRC in spite of following ...published guidelines for diagnosis and treatment.
99 patients were referred to National Jewish Health (NJH), a specialty respiratory center for evaluation of CRC (cough ≥ 8 weeks duration). Study duration occurred over 18 months. Intake evaluation for all patients included history, physical examination, spirometry and fiberoptic laryngoscopy. Testing to confirm causes of CRC were performed. Specific therapy for each potential cause was provided. A visual analog cough scale measured cough response.
Ten final diagnostic categories were found in the cohort of 99 patients with CRC: Obstructive sleep apnea (apnea/hypoxia index ≥ 5), rhinosinusitis, Tracheobronchomalacia (≥65% collapse of airway with dynamic expiratory imaging), esophageal dysmotility, gastroesophageal reflux, abnormal swallowing with laryngeal penetration, asthma, COPD, bronchiectasis and paradoxical vocal cord movement. In these patients there were 42 incorrect intake diagnoses and 101 new diagnoses established. Patients with CRC have had multiple diagnoses (3.8 ± 1.6) associated with chronic cough. With directed therapy 71/76 (93%) patients had resolution or improvement in cough symptoms.
Among patients referred to a specialty respiratory center with CRC multiple concomitant diagnoses for cough were common. Certain diagnoses such as OSA and TBM have not been reported in cough guidelines but in this study are commonly associated diagnoses. Targeted therapy for each recognized diagnosis improves patient response.
•Chronic refractory cough is associated with multiple diagnoses.•TBM and OSA are frequently found in patients with CRC.•All potential causes of CRC should be treated.
