West Greenland has had multiple episodes of human colonization and cultural transitions over the past 4,500 y. However, the explanations for these large-scale human migrations are varied, including ...climatic factors, resistance to adaptation, economic marginalization, mercantile exploration, and hostile neighborhood interactions. Evaluating the potential role of climate change is complicated by the lack of quantitative paleoclimate reconstructions near settlement areas and by the relative stability of Holocene temperature derived from ice cores atop the Greenland ice sheet. Here we present high-resolution records of temperature over the past 5,600 y based on alkenone unsaturation in sediments of two lakes in West Greenland. We find that major temperature changes in the past 4,500 y occurred abruptly (within decades), and were coeval in timing with the archaeological records of settlement and abandonment of the Saqqaq, Dorset, and Norse cultures, which suggests that abrupt temperature changes profoundly impacted human civilization in the region. Temperature variations in West Greenland display an antiphased relationship to temperature changes in Ireland over centennial to millennial timescales, resembling the interannual to multidecadal temperature seesaw associated with the North Atlantic Oscillation.
Metabolism has an essential role in biological systems. Identification and quantitation of the compounds in the metabolome is defined as metabolic profiling, and it is applied to define metabolic ...changes related to genetic differences, environmental influences and disease or drug perturbations. Chromatography-mass spectrometry (MS) platforms are frequently used to provide the sensitive and reproducible detection of hundreds to thousands of metabolites in a single biofluid or tissue sample. Here we describe the experimental workflow for long-term and large-scale metabolomic studies involving thousands of human samples with data acquired for multiple analytical batches over many months and years. Protocols for serum- and plasma-based metabolic profiling applying gas chromatography-MS (GC-MS) and ultraperformance liquid chromatography-MS (UPLC-MS) are described. These include biofluid collection, sample preparation, data acquisition, data pre-processing and quality assurance. Methods for quality control-based robust LOESS signal correction to provide signal correction and integration of data from multiple analytical batches are also described.
Summary Background The UK has had universal free health care and public health programmes for more than six decades. Several policy initiatives and structural reforms of the health system have been ...undertaken. Health expenditure has increased substantially since 1990, albeit from relatively low levels compared with other countries. We used data from the Global Burden of Diseases, Injuries, and Risk Factors Study 2010 (GBD 2010) to examine the patterns of health loss in the UK, the leading preventable risks that explain some of these patterns, and how UK outcomes compare with a set of comparable countries in the European Union and elsewhere in 1990 and 2010. Methods We used results of GBD 2010 for 1990 and 2010 for the UK and 18 other comparator nations (the original 15 members of the European Union, Australia, Canada, Norway, and the USA; henceforth EU15+). We present analyses of trends and relative performance for mortality, causes of death, years of life lost (YLLs), years lived with disability (YLDs), disability-adjusted life-years (DALYs), and healthy life expectancy (HALE). We present results for 259 diseases and injuries and for 67 risk factors or clusters of risk factors relevant to the UK. We assessed the UK's rank for age-standardised YLLs and DALYs for their leading causes compared with EU15+ in 1990 and 2010. We estimated 95% uncertainty intervals (UIs) for all measures. Findings For both mortality and disability, overall health has improved substantially in absolute terms in the UK from 1990 to 2010. Life expectancy in the UK increased by 4·2 years (95% UI 4·2–4·3) from 1990 to 2010. However, the UK performed significantly worse than the EU15+ for age-standardised death rates, age-standardised YLL rates, and life expectancy in 1990, and its relative position had worsened by 2010. Although in most age groups, there have been reductions in age-specific mortality, for men aged 30–34 years, mortality rates have hardly changed (reduction of 3·7%, 95% UI 2·7–4·9). In terms of premature mortality, worsening ranks are most notable for men and women aged 20–54 years. For all age groups, the contributions of Alzheimer's disease (increase of 137%, 16–277), cirrhosis (65%, −15 to 107), and drug use disorders (577%, 71–942) to premature mortality rose from 1990 to 2010. In 2010, compared with EU15+, the UK had significantly lower rates of age-standardised YLLs for road injury, diabetes, liver cancer, and chronic kidney disease, but significantly greater rates for ischaemic heart disease, chronic obstructive pulmonary disease, lower respiratory infections, breast cancer, other cardiovascular and circulatory disorders, oesophageal cancer, preterm birth complications, congenital anomalies, and aortic aneurysm. Because YLDs per person by age and sex have not changed substantially from 1990 to 2010 but age-specific mortality has been falling, the importance of chronic disability is rising. The major causes of YLDs in 2010 were mental and behavioural disorders (including substance abuse; 21·5% 95 UI 17·2–26·3 of YLDs), and musculoskeletal disorders (30·5% 25·5–35·7). The leading risk factor in the UK was tobacco (11·8% 10·5–13·3 of DALYs), followed by increased blood pressure (9·0 % 7·5–10·5), and high body-mass index (8·6% 7·4–9·8). Diet and physical inactivity accounted for 14·3% (95% UI 12·8–15·9) of UK DALYs in 2010. Interpretation The performance of the UK in terms of premature mortality is persistently and significantly below the mean of EU15+ and requires additional concerted action. Further progress in premature mortality from several major causes, such as cardiovascular diseases and cancers, will probably require improved public health, prevention, early intervention, and treatment activities. The growing burden of disability, particularly from mental disorders, substance use, musculoskeletal disorders, and falls deserves an integrated and strategic response. DNA can be preserved in marine and freshwater sediments both in bulk sediment and in intact, viable resting stages. Here, we assess the potential for combined use of ancient, environmental, DNA ...and timeseries of resurrected long-term dormant organisms, to reconstruct trophic interactions and evolutionary adaptation to changing environments. These new methods, coupled with independent evidence of biotic and abiotic forcing factors, can provide a holistic view of past ecosystems beyond that offered by standard palaeoecology, help us assess implications of ecological and molecular change for contemporary ecosystem functioning and services, and improve our ability to predict adaptation to environmental stress.
Methylation of DNA at CpG dinucleotides represents one of the most important epigenetic mechanisms involved in the control of gene expression in vertebrate cells. In this report, we conducted ...nucleosome reconstitution experiments in conjunction with high-throughput sequencing on 572 KB of human DNA and 668 KB of mouse DNA that was unmethylated or methylated in order to investigate the effects of this epigenetic modification on the positioning and stability of nucleosomes. The results demonstrated that a subset of nucleosomes positioned by nucleotide sequence was sensitive to methylation where the modification increased the affinity of these sequences for the histone octamer. The features that distinguished these nucleosomes from the bulk of the methylation-insensitive nucleosomes were an increase in the frequency of CpG dinucleotides and a unique rotational orientation of CpGs such that their minor grooves tended to face toward the histones in the nucleosome rather than away. These methylation-sensitive nucleosomes were preferentially associated with exons as compared to introns while unmethylated CpG islands near transcription start sites became enriched in nucleosomes upon methylation. The results of this study suggest that the effects of DNA methylation on nucleosome stability in vitro can recapitulate what has been observed in the cell and provide a direct link between DNA methylation and the structure and function of chromatin.
Northern forests are important ecosystems for carbon (C) cycling and lakes within them process and bury large amounts of organic-C. Current burial estimates are poorly constrained and may discount ...other shifts in organic-C burial driven by global change. Here we analyse a suite of northern lakes to determine trends in organic-C burial throughout the Anthropocene. We found burial rates increased significantly over the last century and are up to five times greater than previous estimates. Despite a correlation with temperature, warming alone did not explain the increase in burial, suggesting the importance of other drivers including atmospherically deposited reactive nitrogen. Upscaling mean lake burial rates for each time period to global northern forests yields up to 4.5 Pg C accumulated in the last 100 years--20% of the total burial over the Holocene. Our results indicate that lakes will become increasingly important for C burial under future global change scenarios.
DNA methylation is an epigenetic modification that is enriched in heterochromatin but depleted at active promoters and enhancers. However, the debate on whether or not DNA methylation is a reliable ...indicator of high nucleosome occupancy has not been settled. For example, the methylation levels of DNA flanking CTCF sites are higher in linker DNA than in nucleosomal DNA, while other studies have shown that the nucleosome core is the preferred site of methylation. In this study, we make progress toward understanding these conflicting phenomena by implementing a bioinformatics approach that combines MNase-seq and NOMe-seq data and by comprehensively profiling DNA methylation and nucleosome occupancy throughout the human genome.
The results demonstrated that increasing methylated CpG density is correlated with nucleosome occupancy in the total genome and within nearly all subgenomic regions. Features with elevated methylated CpG density such as exons, SINE-Alu sequences, H3K36-trimethylated peaks, and methylated CpG islands are among the highest nucleosome occupied elements in the genome, while some of the lowest occupancies are displayed by unmethylated CpG islands and unmethylated transcription factor binding sites. Additionally, outside of CpG islands, the density of CpGs within nucleosomes was shown to be important for the nucleosomal location of DNA methylation with low CpG frequencies favoring linker methylation and high CpG frequencies favoring core particle methylation. Prominent exceptions to the correlations between methylated CpG density and nucleosome occupancy include CpG islands marked by H3K27me3 and CpG-poor heterochromatin marked by H3K9me3, and these modifications, along with DNA methylation, distinguish the major silencing mechanisms of the human epigenome.
Thus, the relationship between DNA methylation and nucleosome occupancy is influenced by the density of methylated CpG dinucleotides and by other epigenomic components in chromatin.
Benthic diatoms are commonly used for palaeoenvironmental reconstruction in Arctic regions, but interpretation of their ecology remains challenging. We studied epilithic diatom assemblages from the ...shallow margins of 19 lakes from three areas (coast-inland-ice sheet margin) along a climate gradient in Kangerlussuaq, West Greenland during two periods; shortly after ice-off (spring) and in the middle of the growth season (summer). We aimed to understand the distribution of Arctic epilithic diatoms in relation to water chemistry gradients during the two seasons, to investigate their incorporation into lake sediments and to assess their applicability as palaeoenvironmental indicators. Diatoms were correlated with nutrients in the spring and alkalinity/major ions in the summer, when nutrients were depleted; approximately half of the variance explained was independent of spatial factors. When categorised by functional attributes, diatom seasonal succession differed among regions with the most obvious changes in inland lakes where summer temperatures are warmer, organic nutrient processing is prevalent and silicate is limiting. These conditions led to small, motile and adnate diatoms being abundant in inland lakes during the summer (
Nitzschia
spp.,
Encyonopsis microcephala
), as these functional attributes are suited to living within complex mats of non-siliceous microbial biofilms. Seasonal succession in silica-rich lakes at the coast was less pronounced and assemblages included
Tabellaria flocculosa
(indicating more acidic conditions) and
Hannaea arcus
(indicating input from inflowing rivers). The nitrogen-fixing diatom
Epithemia sorex
increased from the coast to the ice sheet, negatively correlating with a gradient of reactive nitrogen. The presence of this diatom in Holocene sediment records alongside cyanobacterial carotenoids during arid periods of low nitrogen delivery, suggests that it is a useful indicator of nitrogen limitation.
Nitzschia
species appear to be associated with high concentrations of organic carbon and heterotrophy, but their poor representation in West Greenland lake sediments due to taphonomic processes limits their palaeoenvironmental application in this region. Proportions of epilithic taxa in lake sediment records of coastal lakes increased during some wetter periods of the Holocene, suggesting that snowpack-derived nutrient delivery may offer diatom taxa living at lake margins a competitive advantage over planktonic diatoms during the “moating” ice melt period. Thus, further research investigating linkages between epilithic diatoms, snowpack and nutrient delivery in seasonally frozen lakes is recommended as these taxa live on the ‘front-line’ during the spring and may be especially sensitive to changes in snowmelt conditions.
Diurnal and seasonal cycles of aeolian activity are well‐constrained for low latitude dust source regions and provide valuable insights into relationships between dust emissions and environmental ...drivers. Such cycles have received little systematic attention in high latitude dust source areas (≥50°N and ≥40°S), and understanding them will aid the modelling of atmospheric dust over different timescales. This paper examines the timing and drivers of atmospheric dust concentration close to source at ice‐free locations c. 6 km and c. 37 km from the Greenland Ice Sheet margin. Dust concentration, and associated environmental drivers including wind speed and velocity, air temperature and humidity, was measured from April–October 2018 and April–August 2019. Measured dust concentrations were a similar order of magnitude to source‐proximal values measured globally and varied from 0 to ≥1000 μg m−3. Diurnal cycles in the environmental drivers of dust emission were similar to those observed in low latitudes, but unlike in the low latitudes, there was no clear diurnal pattern of dust concentration. Only 3 out of 17 recorded dust events showed a significant positive relationship between wind speed and dust concentration, and factors such as wind direction, humidity and sediment availability are the over‐riding controls on dust activity at the event and seasonal scales. Whereas 7 dust events are attributed to down‐valley katabatic winds, 10 events were associated with a westerly sea breeze blowing up‐valley towards the ice sheet and high atmospheric moisture content. If deposited on the ice, this dust will alter ice albedo both directly and indirectly, (e.g. through the promotion of algal blooms) affecting cryospheric melt rates. Our results demonstrate an intricate relationship between first‐order controls and dust concentration that raises challenges for modelling the uplift and subsequent dust transport from high latitude sources, especially the appropriateness of assumptions based on emissions behaviour at low latitudes.

Lakes may store globally significant quantities of organic carbon (OC) in their sediments, but the extent to which burial rates vary across space and time is not well described. Using 210Pb‐dated ...sediment cores, we explored patterns of OC burial in 116 lakes spanning several ecoregions and land use regimes in Minnesota, USA during the past 150–200 years. Rates for individual lakes (across all time periods) range from 3 to 204 g C m−2 yr−1 (median 33 g C m−2 yr−1) and show strong geographic separation in accordance with the degree of catchment disturbance and nutrient enrichment. Climate and basin morphometry exercise subordinate control over OC burial patterns, and diagenetic gradients introduce little bias to estimated temporal trends. Median burial rates in agricultural lakes exceed urban lakes and have increased fourfold since Euro‐American settlement. The greatest increase in OC burial occurred prior to the widespread adoption of industrial fertilizers, during an era of land clearance and farmland expansion. Northern boreal lakes, impacted by historical logging and limited cottage development yet comparatively undisturbed by human activity, bury OC at rates 3X lower than agricultural lakes and exhibit much smaller increases in OC burial. Scaling up modern OC burial estimates to the entire state, we find that Minnesota lakes annually store 0.40 Tg C in their sediments, equal to 1.5% of annual statewide CO2 emissions from fossil fuel combustion. During the period of Euro‐American settlement (circa 1860–2000), cumulative OC burial amounted to 36 Tg C, 40% of which can be attributed to anthropogenic enhancement.
