Present day lead pollution is an environmental hazard of global proportions. A correct determination of natural lead levels is very important in order to evaluate anthropogenic lead contributions. In ...this paper, the anthropogenic signature of early metallurgy in Southern Iberia during the Holocene, more specifically during the Late Prehistory, was assessed by mean of a multiproxy approach: comparison of atmospheric lead pollution, fire regimes, deforestation, mass sediment transport, and archeological data. Although the onset of metallurgy in Southern Iberia is a matter of controversy, here we show the oldest lead pollution record from Western Europe in a continuous paleoenvironmental sequence, which suggests clear lead pollution caused by metallurgical activities since ~3900cal BP (Early Bronze Age). This lead pollution was especially important during Late Bronze and Early Iron ages. At the same time, since ~4000cal BP, an increase in fire activity is observed in this area, which is also coupled with deforestation and increased erosion rates. This study also shows that the lead pollution record locally reached near present-day values many times in the past, suggesting intensive use and manipulation of lead during those periods in this area.

Understanding the natural mechanisms that control fire occurrence in terrigenous ecosystems requires long and continuous records of past fires. Proxies, such as sedimentary charcoal and tree-ring fire scars, have temporal or spatial limitations and do not directly detect fire intensity. We show in this study that polycyclic aromatic hydrocarbons (PAHs) produced during wildfires record local fire events and fire intensity. We demonstrate that high performance liquid chromatography with fluorescence detector (HPLC-FLD) is superior to gas chromatography–mass spectrometry (GC–MS) for detecting the low concentrations of sedimentary PAHs derived from natural fires. The HPLC-FLD is at least twice as sensitive as the GC–MS in selective ion monitoring (SIM) mode for parent PAHs and five times as sensitive for retene. The annual samples extracted from varved sediments from Swamp Lake in Yosemite National Park, California are compared with the observational fire history record and show that PAH fluxes record fires within 0.5km of the lake. The low molecular weight (LMW) PAHs (e.g., fluoranthene, pyrene and benzaanthracene) are the best recorders of fire, whereas the high molecular weight (HMW) PAHs likely record fire intensity. PAHs appear to resolve some of the issues inherent to other fire proxies, such as secondary deposition of charcoal. This study advances our understanding of how PAHs can be used as markers for fire events and poses new questions regarding the distribution of these compounds in the environment.
The Kenai Peninsula of south‐central Alaska is a region of high topographic diversity with a complex glacial history. The sedimentary record of two small lakes Sunken Island (SIL; 76 m a.s.l.) in the ...Kenai Lowlands; Choquette (CL; 527 m a.s.l.) in the Caribou Hills upland exemplifies the postglacial development of the conifer–hardwood forest over an elevational range there. A herb–shrub tundra was established at both sites after deglaciation. By ~10.7 ka, poplar (Populus sp.) and alder (Alnus) dominated the lowland forest, while alder with minor poplar occurred at the upland site. Lake levels lower than today occurred during the early Holocene until ~8 ka. Subsequently at SIL, the near‐modern Kenai birch (Betula kenaica) – white spruce (Picea glauca) forest maintained prominence throughout the Holocene. However, at CL, alder dominated with dwarf birch and other subshrubs; small amounts of white spruce arrived ~5.2 ka. Black spruce (Picea mariana) grew around SIL by ~4 ka, but never gained prominence at CL. Fire, a prominent agent of disturbance in the Kenai Lowlands since ~8 ka, was essentially absent at the hardwood‐dominated upland site before ~6 ka, and rare thereafter. This suggests an important link between fire and spruce in Kenai forests.
The ectoparasitic mite Varroa destructor has emerged as the primary pest of domestic honey bees (Apis mellifera). Here we present an initial survey of the V. destructor genome carried out to advance ...our understanding of Varroa biology and to identify new avenues for mite control. This sequence survey provides immediate resources for molecular and population-genetic analyses of Varroa-Apis interactions and defines the challenges ahead for a comprehensive Varroa genome project.
The genome size was estimated by flow cytometry to be 565 Mbp, larger than most sequenced insects but modest relative to some other Acari. Genomic DNA pooled from ~1,000 mites was sequenced to 4.3× coverage with 454 pyrosequencing. The 2.4 Gbp of sequencing reads were assembled into 184,094 contigs with an N50 of 2,262 bp, totaling 294 Mbp of sequence after filtering. Genic sequences with homology to other eukaryotic genomes were identified on 13,031 of these contigs, totaling 31.3 Mbp. Alignment of protein sequence blocks conserved among V. destructor and four other arthropod genomes indicated a higher level of sequence divergence within this mite lineage relative to the tick Ixodes scapularis. A number of microbes potentially associated with V. destructor were identified in the sequence survey, including ~300 Kbp of sequence deriving from one or more bacterial species of the Actinomycetales. The presence of this bacterium was confirmed in individual mites by PCR assay, but varied significantly by age and sex of mites. Fragments of a novel virus related to the Baculoviridae were also identified in the survey. The rate of single nucleotide polymorphisms (SNPs) in the pooled mites was estimated to be 6.2 × 10-5 per bp, a low rate consistent with the historical demography and life history of the species.
This survey has provided general tools for the research community and novel directions for investigating the biology and control of Varroa mites. Ongoing development of Varroa genomic resources will be a boon for comparative genomics of under-represented arthropods, and will further enhance the honey bee and its associated pathogens as a model system for studying host-pathogen interactions.
Ticks are currently among the most prevalent blood-feeding ectoparasites, but their feeding habits and hosts in deep time have long remained speculative. Here, we report direct and indirect evidence ...in 99 million-year-old Cretaceous amber showing that hard ticks and ticks of the extinct new family Deinocrotonidae fed on blood from feathered dinosaurs, non-avialan or avialan excluding crown-group birds. A †Cornupalpatum burmanicum hard tick is entangled in a pennaceous feather. Two deinocrotonids described as †Deinocroton draculi gen. et sp. nov. have specialised setae from dermestid beetle larvae (hastisetae) attached to their bodies, likely indicating cohabitation in a feathered dinosaur nest. A third conspecific specimen is blood-engorged, its anatomical features suggesting that deinocrotonids fed rapidly to engorgement and had multiple gonotrophic cycles. These findings provide insight into early tick evolution and ecology, and shed light on poorly known arthropod-vertebrate interactions and potential disease transmission during the Mesozoic.
Obliterative bronchiolitis (OB) is a major cause of mortality after lung transplantation. Depletion of airway stem cells (SCs) may lead to fibrosis in OB.
Two major SC compartments in airways are ...submucosal glands (SMGs) and surface airway p63 (also known as TP63 tumor protein 63)-positive/K5 (also known as KRT5 keratin 5)-positive basal cells (BCs). We hypothesized that depletion of these SC compartments occurs in OB.
Ferret orthotopic left lung transplants were used as an experimental model of OB, and findings were corroborated in human lung allografts. Morphometric analysis was performed in ferret and human lungs to evaluate the abundance of SMGs and changes in the expression of phenotypic BC markers in control, lymphocytic bronchiolitis, and OB airways. The abundance and proliferative capacity of proximal and distal airway SCs was assessed using a clonogenic colony-forming efficiency assay.
Ferret allografts revealed significant loss of SMGs with development of OB. A progressive decline in p63

and increase in K5
/K14
and K14
BC phenotypes correlated with the severity of allograft rejection in large and small ferret airways. The abundance and proliferative capacity of basal SCs in large allograft airways declined with severity of OB, and there was complete ablation of basal SCs in distal OB airways. Human allografts mirrored phenotypic BC changes observed in the ferret model.
SMGs and basal SC compartments are depleted in large and/or small airways of lung allografts, and basal SC proliferative capacity declines with progression of disease and phenotypic changes. Global airway SC depletion may be a mechanism for pulmonary allograft failure.
A high-resolution multi-proxy approach, integrating pollen, inorganic and organic geochemical and sedimentological analyses, has been carried out on the Holocene section of the Padul sedimentary ...record in the southern Iberian Peninsula reconstructing vegetation, environment and climate throughout the last ~ 11.6 cal kyr BP in the western Mediterranean. The study of the entire Holocene allows us to determine the significant climate shift that occurred during the middle-to-late Holocene transition. The highest occurrence of deciduous forest in the Padul area from ~ 9.5 to 7.6 cal kyr BP represents the Holocene humidity optimum probably due to enhanced winter precipitation during a phase of highest seasonal anomaly and maximum summer insolation. Locally, insolation maxima induced high evaporation, counterbalancing the effect of relatively high precipitation, and triggered very low water table in Padul and the deposition of peat sediments. A transitional environmental change towards more regional aridity occurred from ~ 7.6 to 4.7 cal kyr BP and then aridification enhanced in the late Holocene most likely related to decreasing summer insolation. This translated into higher water levels and a sedimentary change at ~ 4.7 cal kyr BP in the Padul wetland, probably related to reduced evaporation during summer in response to decreased in seasonality. Millennial-scale variability is superimposed on the Holocene long-term trends. The Mediterranean forest regional climate proxy studied here shows significant cold-arid events around ~ 9.6, 8.5, 7.5, 6.5 and 5.4 cal kyr BP with cyclical periodicities (~1100 and 2100 yr) during the early and middle Holocene. A change is observed in the periodicity of these cold-arid events towards ~1430 yr in the late Holocene, with forest declines around ~ 4.7–4, 2.7 and 1.3 cal kyr BP. The comparison between the Padul-15-05 data with published North Atlantic and Mediterranean paleoclimate records suggests common triggers for the observed climate variability, with the early and middle Holocene forest declines at least partially controlled by external forcing (i.e. solar activity) and the late Holocene variability associated with internal mechanisms (oceanic-atmospheric).
•We carried out a multi-proxy analysis for the last 11.6 cal kyr BP from a new sedimentary record from Padul (Sierra Nevada, Spain).•This record shows a long-term climate pattern mostly forced by insolation, showing a significant climate and environmental shift at 4.7 cal kyr BP.•Millennial-scale climate oscillations are also characterized in this study by the decrease in Mediterranean forest and local response in the lake level, showing possible atmospheric and climate links between the western Mediterranean and North Atlantic areas.
