We present a strong form, meshless point collocation explicit solver for the numerical solution of the transient, incompressible, viscous Navier-Stokes (N-S) equations in two dimensions. We ...numerically solve the governing flow equations in their stream function-vorticity formulation. We use a uniform Cartesian embedded grid to represent the flow domain. We discretize the governing equations using the Meshless Point Collocation (MPC) method. We compute the spatial derivatives that appear in the governing flow equations, using a novel interpolation meshless scheme, the Discretization Corrected Particle Strength Exchange (DC PSE). We verify the accuracy of the numerical scheme for commonly used benchmark problems including lid-driven cavity flow, flow over a backward-facing step and unbounded flow past a cylinder. We have examined the applicability of the proposed scheme by considering flow cases with complex geometries, such as flow in a duct with cylindrical obstacles, flow in a bifurcated geometry, and flow past complex-shaped obstacles. Our method offers high accuracy and excellent computational efficiency as demonstrated by the verification examples, while maintaining a stable time step comparable to that used in unconditionally stable implicit methods. We estimate the stable time step using the Gershgorin circle theorem. The stable time step can be increased through the increase of the support domain of the weight function used in the DC PSE method.

The survival of bacteria in nature is greatly enhanced by their ability to grow within surface-associated communities called biofilms. Commonly, biofilms generate proliferations of bacterial cells, ...called microcolonies, which are highly recalcitrant, 3-dimensional foci of bacterial growth. Microcolony growth is initiated by only a subpopulation of bacteria within biofilms, but processes responsible for this differentiation remain poorly understood. Under conditions of crowding and intense competition between bacteria within biofilms, microevolutionary processes such as mutation selection may be important for growth; however their influence on microcolony-based biofilm growth and architecture have not previously been explored. To study mutation in-situ within biofilms, we transformed Pseudomonas aeruginosa cells with a green fluorescent protein gene containing a +1 frameshift mutation. Transformed P. aeruginosa cells were non-fluorescent until a mutation causing reversion to the wildtype sequence occurs. Fluorescence-inducing mutations were observed in microcolony structures, but not in other biofilm cells, or in planktonic cultures of P. aeruginosa cells. Thus microcolonies may represent important foci for mutation and evolution within biofilms. We calculated that microcolony-specific increases in mutation frequency were at least 100-fold compared with planktonically grown cultures. We also observed that mutator phenotypes can enhance microcolony-based growth of P. aeruginosa cells. For P. aeruginosa strains defective in DNA fidelity and error repair, we found that microcolony initiation and growth was enhanced with increased mutation frequency of the organism. We suggest that microcolony-based growth can involve mutation and subsequent selection of mutants better adapted to grow on surfaces within crowded-cell environments. This model for biofilm growth is analogous to mutation selection that occurs during neoplastic progression and tumor development, and may help to explain why structural and genetic heterogeneity are characteristic features of bacterial biofilm populations.
Time-domain science has undergone a revolution over the past decade, with tens of thousands of new supernovae (SNe) discovered each year. However, several observational domains, including SNe within ...days or hours of explosion and faint, red transients, are just beginning to be explored. Here we present the Young Supernova Experiment (YSE), a novel optical time-domain survey on the Pan-STARRS telescopes. Our survey is designed to obtain well-sampled griz light curves for thousands of transient events up to z 0.2. This large sample of transients with four-band light curves will lay the foundation for the Vera C. Rubin Observatory and the Nancy Grace Roman Space Telescope, providing a critical training set in similar filters and a well-calibrated low-redshift anchor of cosmologically useful SNe Ia to benefit dark energy science. As the name suggests, YSE complements and extends other ongoing time-domain surveys by discovering fast-rising SNe within a few hours to days of explosion. YSE is the only current four-band time-domain survey and is able to discover transients as faint as ∼21.5 mag in gri and ∼20.5 mag in z, depths that allow us to probe the earliest epochs of stellar explosions. YSE is currently observing approximately 750 deg2 of sky every 3 days, and we plan to increase the area to 1500 deg2 in the near future. When operating at full capacity, survey simulations show that YSE will find ∼5000 new SNe per year and at least two SNe within 3 days of explosion per month. To date, YSE has discovered or observed 8.3% of the transient candidates reported to the International Astronomical Union in 2020. We present an overview of YSE, including science goals, survey characteristics, and a summary of our transient discoveries to date.
We present a sample of 21 hydrogen-free superluminous supernovae (SLSNe-I) and one hydrogen-rich SLSN (SLSN-II) detected during the five-year Dark Energy Survey (DES). These SNe, located in the ...redshift range 0.220 < z < 1.998, represent the largest homogeneously selected sample of SLSN events at high redshift. We present the observed $g, r, i, z$ light curves for these SNe, which we interpolate using Gaussian processes. The resulting light curves are analysed to determine the luminosity function of SLSNe-I, and their evolutionary time-scales. The DES SLSN-I sample significantly broadens the distribution of SLSN-I light-curve properties when combined with existing samples from the literature. We fit a magnetar model to our SLSNe, and find that this model alone is unable to replicate the behaviour of many of the bolometric light curves. We search the DES SLSN-I light curves for the presence of initial peaks prior to the main light-curve peak. Using a shock breakout model, our Monte Carlo search finds that 3 of our 14 events with pre-max data display such initial peaks. However, 10 events show no evidence for such peaks, in some cases down to an absolute magnitude of <-16, suggesting that such features are not ubiquitous to all SLSN-I events. Finally, we also identify a red pre-peak feature within the light curve of one SLSN, which is comparable to that observed within SN2018bsz.
Homozygous familial hypercholesterolaemia (FH) causes severe premature coronary artery disease because of very high levels of low density lipoprotein (LDL)‐cholesterol. Standard lipid‐lowering drugs ...and LDL‐apheresis may not be sufficiently effective. Liver transplantation replaces defective LDL receptors and vastly improves the lipid profile, and we present the first report of an Australian adult to receive this treatment. Emerging drug treatments for FH may be alternatives to LDL‐apheresis and transplantation, but long‐term safety and efficacy data are lacking for all of these options.
In this work, we present the first Hubble diagram of superluminous supernovae (SLSNe) out to a redshift of two, together with constraints on the matter density, ΩM, and the dark energy ...equation-of-state parameter, w(≡p/ρ). We build a sample of 20 cosmologically useful SLSNe I based on light curve and spectroscopy quality cuts. We confirm the robustness of the peak–decline SLSN I standardization relation with a larger data set and improved fitting techniques than previous works. We then solve the SLSN model based on the above standardization via minimization of the χ2 computed from a covariance matrix that includes statistical and systematic uncertainties. For a spatially flat Λ cold dark matter (ΛCDM) cosmological model, we find $\Omega _{\rm M}=0.38^{+0.24}_{-0.19}$, with an rms of 0.27 mag for the residuals of the distance moduli. For a w0waCDM cosmological model, the addition of SLSNe I to a ‘baseline’ measurement consisting of Planck temperature together with Type Ia supernovae, results in a small improvement in the constraints of w0 and wa of 4 per cent. We present simulations of future surveys with 868 and 492 SLSNe I (depending on the configuration used) and show that such a sample can deliver cosmological constraints in a flat ΛCDM model with the same precision (considering only statistical uncertainties) as current surveys that use Type Ia supernovae, while providing a factor of 2–3 improvement in the precision of the constraints on the time variation of dark energy, w0 and wa. This paper represents the proof of concept for superluminous supernova cosmology, and demonstrates they can provide an independent test of cosmology in the high-redshift (z > 1) universe.

≈ 0.5. We present relative SN rates from YSE’s magnitude- and volume-limited surveys, which are consistent with previously published values within estimated uncertainties for untargeted surveys. We combine YSE and ZTF data, and create multisurvey SN simulations to train the ParSNIP and SuperRAENN photometric classification algorithms; when validating our ParSNIP classifier on 472 spectroscopically classified YSE DR1 SNe, we achieve 82% accuracy across three SN classes (SNe Ia, II, Ib/Ic) and 90% accuracy across two SN classes (SNe Ia, core-collapse SNe). Our classifier performs particularly well on SNe Ia, with high (>90%) individual completeness and purity, which will help build an anchor photometric SNe Ia sample for cosmology. We then use our photometric classifier to characterize our photometric sample of 1483 SNe, labeling 1048 (∼71%) SNe Ia, 339 (∼23%) SNe II, and 96 (∼6%) SNe Ib/Ic. YSE DR1 provides a training ground for building discovery, anomaly detection, and classification algorithms, performing cosmological analyses, understanding the nature of red and rare transients, exploring tidal disruption events and nuclear variability, and preparing for the forthcoming Vera C. Rubin Observatory Legacy Survey of Space and Time.

. First, our study reveals a unified molecular genetic landscape of cortical cell types that integrates their transcriptome, open chromatin and DNA methylation maps. Second, cross-species analysis achieves a consensus taxonomy of transcriptomic types and their hierarchical organization that is conserved from mouse to marmoset and human. Third, in situ single-cell transcriptomics provides a spatially resolved cell-type atlas of the motor cortex. Fourth, cross-modal analysis provides compelling evidence for the transcriptomic, epigenomic and gene regulatory basis of neuronal phenotypes such as their physiological and anatomical properties, demonstrating the biological validity and genomic underpinning of neuron types. We further present an extensive genetic toolset for targeting glutamatergic neuron types towards linking their molecular and developmental identity to their circuit function. Together, our results establish a unifying and mechanistic framework of neuronal cell-type organization that integrates multi-layered molecular genetic and spatial information with multi-faceted phenotypic properties.
